Toxoplasma gondii, a globally prevalent apicomplexan parasite, causes severe morbidity and mortality in immunocompromised individuals and livestock. Current therapies exhibit limited efficacy against chronic bradyzoite stages and face drug resistance. Here, we screened imidocarb and its derivatives for anti-Toxoplasma activity. Among 20 synthesized compounds, squaramide derivative 18 (SA-18) emerged as a promising candidate with nanomolar EC50 against tachyzoites, potent inhibition of bradyzoite differentiation, and > 100-fold selectivity index. Transcriptomic analysis revealed transcriptional

Babesiosis is a zoonosis caused by protozoan parasites of the genus Babesia. It has a worldwide distribution and affects many kinds of mammals, principally domestic animals and humans. Because there are no safe and effective vaccines available, the treatment and control for babesiosis continues to involve the use of chemotherapeutics. For years, only a few drugs have been used for clinical treatment, namely atovaquone plus azithromycin or clindamycin plus quinine for human, and imidocarb dipropionate and diminazene aceturate for domestic animals. Although screening and developing alternative d

This study investigated the practicality and potential of the gerbil, Meriones unguiculatus, as a source of live Babesia divergens vaccine and also as a model for the use of the vaccine in cattle. A series of experiments with gerbils concerning vaccine infectivity, immunogenicity and safety were carried out. It was concluded that the use of RPMI medium/40% foetal calf serum as a diluent improved vaccine infectivity, but that the parasitaemia of the blood obtained from donor gerbils had little or no effect. The immunostimulants levamisole and killed Corynebacterium parvum improved vaccine immun

A complete necropsy performed on 2 sable antelope (Hippotragus niger), revealed lesions concomitant with a massive haemolytic crisis. These included widespread oedema and anaemia of the carcass, severe oedema of the lungs, petechiae and echymoses of the epicardium, a moderate splenomegaly and a severe haemoglobinuria. The histopathological lesions included a moderate alveolar oedema, the presence of haemosiderin in the spleen and lymph nodes, and mild degenerative changes of the renal tubular epithelium. Peripheral blood and brain smears contained numerous parasitised red blood cells. The para

The hydroxynaphthoquinone, atovaquone (Wellvone, Glaxo-Wellcome Ltd.) was found to have significant activity against Babesia microti, the main cause of human babesiosis in the U.S.A. This activity compares well with that of the most effective babesicide currently available for use in animals, imidocarb dipropionate, that unlike atovaquone is not licensed for use in humans. Treatment with well tolerated doses of atovaquone results in a rapid reduction in parasitemias and an early disappearance of parasites from blood smears. However, in common with all the other babesicides tested, atovaquone d

An Irish setter from the Oslo area was presented to the clinic with signs of babesiosis, a few days after a tick bite. Blood analysis confirmed babesiosis. Microscopic examination of thin blood film revealed large, basophilic, bodies inside erythrocytes, indicative of a large Babesia sp. Molecular analysis using PCR, indicated the presence of a Babesia spp. in the blood. Sequencing and phylogenetic analysis of the PCR fragment revealed a sequence which was 100% identical to Babesia canis canis 18S. As this dog had never been abroad, it can be concluded that this is the first report of an autoc

A six-year old pit bull terrier, female, was presented to a veterinary clinic in Mocsow, Russia. The leading clinical signs were a decrease in the physical activity of the dog with weight loss despite normal appetite. The dog had never traveled to other regions, but there was a history of a fight with a dog from Serbia about two years before. A severe anemia and azotemia were revealed during the examination. The merozoites of small Babesia were detected in a blood smear, which were identified as Babesia vulpes by PCR. The treatment with imidocarb dipropionate and atovaquone in combination with

Piroplasmosis, a disease endemic to most tropical and subtropical areas, appears to be spreading to more temperate zones. This article gives a review of equine piroplasmosis and describes an acute case of infection with Babesia caballi in a Dutch Standard bred foal after a short stay at a stud in Normandy (France). A 3-month-old stallion foal was presented with lethargy, fever of 41 degrees C, and pale mucosal membranes. Haematology revealed a low packed cell volume (14 l/l) leucytosis (25 G/l) and a high blood urea nitrogen concentration (20.1mmol/l). Infection with B. caballi was diagnosed o

BACKGROUND: Equine piroplasmosis (EP) is a tick-borne disease of equids caused by the intraerythrocytic apicomplexan parasites Theileria equi, Babesia caballi and the recently identified Theileria haneyi. Acute cases can be severe, with anemia, jaundice, abortion or sudden death. Survivors remain lifelong carriers, serving as reservoirs for tick-borne and iatrogenic transmission. No vaccines are currently available, and control strategies rely heavily on accurate diagnostics and chemotherapeutic intervention. Imidocarb dipropionate (ID) is the current standard of care for both acute treatment

A 12-month-old male neutered mixed breed dog was presented with a history of diarrhea, lethargy, emaciation, polydypsia, and sniffling. Physical examination findings included pale mucous membranes, increased heart and respiratory rates, and normal rectal temperature (38°C). Hematologic abnormalities included anemia and thrombocytopenia. Biochemical abnormalities included hypoalbuminemia, hyperbilirubinemia, and elevated ALP and ALT activities. A SNAP 4Dx test result was positive for Ehrlichia canis. Babesia canis vogeli organisms were found in the peripheral blood films, while morulae of E can

Babesiosis/theileriosis is a tick-borne disease caused by protozoan parasites of the genus Babesia and Theileria. Tick species of genera, such as Ixodes, Rhipicephalus, Haemaphysalis and Hyalomma, transmit these parasites. These diseases threaten livestock productivity and health, particularly in tropical and subtropical areas. These diseases have a significant economic impact due to reduced milk and meat production, reproductive losses, higher veterinary expenses, and, in extreme cases, animal mortality. Even after decades of research, piroplasmosis is still difficult to control and manage. T

A Doberman Pinscher contracted babesiosis after receiving a fresh blood transfusion from a Greyhound blood donor. Hypotensive shock syndrome was suspected on the basis of arterial hypotension, weakness, and pyrexia in the absence of detectable hemolysis and within hours of detection of low numbers of circulating Babesia canis organisms. Treatment with imidocarb dipropionate appears to have been effective in eliminating circulating B canis organisms and clinical disease. The blood donor, recently acquired from a race track, was healthy and lacked any abnormalities on initial laboratory evaluati

Hepatozoon felis is the most prevalent Hepatozoon species infecting domestic cats worldwide; however, clinical disease is considered uncommon. This study describes the first clinically confirmed case of H. felis infection in a domestic cat from Panama. A 3-year-old male cat presented with painful periostitis of long bones, poor body condition and leukocytosis with marked neutrophilia. Hepatozoon gamonts were detected in neutrophils, and infection was confirmed by PCR and sequencing of the 18S rRNA gene. Phylogenetic analysis revealed a highly divergent H. felis lineage clustering separately fr

Equine piroplasmosisis a tick-borne haemoprotozoan disease of equine population (horses, mules, donkeys and zebras) caused either by T. equi or B. caballi. Clinically it is characterized by pyrexia, hemolytic anemia and jaundice. Intravascular hemolysis and development of oxidative stress are key pathological alterations. Disease is endemic in almost all part of world (tropical, subtropical, temperate regions) except some courtiers which are EP free like US, UK, Northern Europe, Iceland and New Zealand. These countries follow strict quarantine measures and T. equi infection leads to developmen

BACKGROUND: An innovative approach has been introduced for identifying and developing novel potent and safe anti-Babesia and anti-Theileria agents for the control of animal piroplasmosis. In the present study, we evaluated the inhibitory effects of Malaria Box (MBox) compounds (n = 8) against the growth of Babesia microti in mice and conducted bioinformatics analysis between the selected hits and the currently used antibabesial drugs, with far-reaching implications for potent combinations.
METHODS: A fluorescence assay was used to evaluate the in vivo inhibitory effects of the selected compou
