The impact of peroxisome proliferator-activated receptor-γ activating angiotensin receptor blocker on outcomes of patients receiving immunotherapy.

BACKGROUND: Certain angiotensin receptor blockers (ARBs) have peroxisome proliferator-activated receptor-γ (PPAR-γ) activation property, which has been associated with improved programmed cell death ligand 1 blockade and cytotoxic T lymphocyte-mediated antitumor activity. METHODS: We conducted a retrospective cohort study to investigate the impact of PPAR-γ-activating ARBs on patient survival in patients treated with immune checkpoint inhibitors (ICIs) across all types of cancers. RESULTS: A total of 167 patients receiving both angiotensin receptor blockers (ARBs) and immune checkpoint inhib

Understanding the Mode of Action of a Micro-Immunotherapy Formulation: Pre-Clinical Evidence from the Study of 2LEBV® Active Ingredients.

BACKGROUND: Epstein-Barr virus (EBV) is often kept silent and asymptomatic; however, its reactivation induces a chronic and/or recurrent infection that is associated with numerous diseases, including cancer and inflammation-related disorders. As no specific treatment is currently available, the immune factors-based micro-immunotherapy (MI) medicine 2LEBV® could be considered a valuable therapeutic option to sustain the immune system in EBV reactivation. METHODS: The present work aimed to investigate, for the first time, the effect of 2LEBV® in several in vitro models of uninfected immune-rela

Opportunities and challenges of active immunotherapy in dogs with B-cell lymphoma: a 5-year experience in two veterinary oncology centers.

BACKGROUND: Pet dogs spontaneously develop lymphoma. An anthracycline-based multidrug chemotherapy regimen represents the treatment cornerstone; however, cure is rarely achieved. We have been treating dogs with B-cell lymphoma with an autologous vaccine (APAVAC®) and CHOP-based chemotherapy since 2011. METHODS: To better characterize the safety and efficacy of APAVAC®, and to find the best candidates for immunotherapy, we designed a retrospective study on all dogs treated with chemo-immunotherapy to date and compared them with those dogs treated with chemotherapy only. All dogs were completel

Molecular Engineering Boosts Photon-Activated Immunotherapy for Prostate Cancer Through Concurrent Pyroptosis and cGAS-STING Pathway Activation.

Prostate cancer therapy is often limited by metastasis, drug resistance, and systemic toxicity. Photodynamic immunotherapy (PDIT) offers a promising alternative, yet its efficacy depends on photosensitizers that can simultaneously generate reactive oxygen species (ROS) and activate antitumor immunity. Herein, three thiophene (T) dyes based on triphenylamine (D) and N-ethyl-benzoselenazolium iodide (Se) (DTSe)-based photosensitizers were molecularly engineered by modulating the π-conjugated structure. Incorporation of a carbazole unit affords DZTSe with suppressed fluorescence, enlarged Huang-R

pH-Responsive liposomes co-delivering a membrane-active peptide and the PI3Kγ inhibitor to reconstruct the tumor microenvironment for enhanced antitumor immunotherapy.

Immunotherapy has shown enormous promise for cancer treatment, yet its efficacy is often hindered by the highly immunosuppressive tumor microenvironment (TME). Here, we developed a pH-responsive and cRGD-modified multifunctional liposomal system (FP2@PSLR) to co-deliver an immunogenic cytotoxic peptide (FR) and the PI3Kγ inhibitor IPI549 for coordinated TME remodeling. FR induces immunogenic cell death while exerting direct cytolytic activity, whereas IPI549 promotes macrophage repolarization toward the pro-inflammatory M1-like phenotype, enabling synergistic relief of immune suppression. Multicentric prognostic observational study on biomarker profile in immunotherapy-naïve patients with highly active generalized myasthenia gravis (PROGNO-MG): a study protocol.

BACKGROUND: Myasthenia gravis has a variable disease course. Early identification of patients with a highly active disease is crucial to prevent myasthenic exacerbations and crisis, necessitating more aggressive immunotherapy. However, no biomarkers currently exist to identify patients at risk for highly active disease course. OBJECTIVE: The PROGNO-MG study aims to identify blood-based biomarker signatures associated with a highly active disease course in immunotherapy-naïve patients with acetylcholine-receptor-antibody positive (AChR+) generalized Myasthenia gravis (gMG). 5,6-Dimethylxanthenone-4-acetic acid treatment of a non-immunogenic tumour does not synergize with active or passive CD8+ T-cell immunotherapy.

The chemotherapeutic drug 5,6-dimethylxanthenone-4-acetic acid (DMXAA) inhibits intratumoural blood flow, causing hypoxia, haemorrhagic necrosis, vascular collapse and tumour cell death. Production of TNF-alpha and IFN is also induced, causing local inflammation and activation of immune cells including CD8+ T cells. We used the tumour cell line LL-LCMV, which expresses the gp33 epitope of lymphocytic choriomeningitis virus in a non-immunogenic form, to investigate whether tumour cell death caused by treatment with DMXAA may improve the success of tumour immunotherapy mediated by CD8+ T cells.

Radiotherapy and tertiary lymphoid structures: balancing immune activation and immune damage in cancer immunotherapy.

Cancer immunotherapy, exemplified by immune checkpoint blockade (ICB), remains strongly influenced by the pre-existing immune organization of the tumor microenvironment. Tertiary lymphoid structures (TLSs) are ectopic lymphoid aggregates whose density, maturation state, and spatial localization correlate with clinical prognosis and ICB response across several malignancies. Radiotherapy (RT) can reshape this immune context in opposing directions. By inducing immunogenic cell death, antigen release, cGAS-STING/type I interferon signaling, vascular remodeling, and lymphocyte recruitment, RT can c

Hollow RuO2 nanozymes sensitized by carbon dot sonosensitizers for sonodynamic/chemodynamic-activated immunotherapy.

Background: Regulating the morphology structure of sonosensitizers and nanozymes is crucial to improve sonodynamic and enzyme-mimic activities. Methods: We report for the first time the utilization of Cu2O nanospheres as the sacrificial templates for the synthesis of hollow RuO2 nanospheres (H-RuO2) for high-efficiency sonodynamic and chemodynamic therapy (SDT/CDT). We then utilized NIR phosphorescence carbon dots (CDs) as the auxiliary sonosensitizers to sensitize H-RuO2 for the construction of CD@H-RuO2 heterojunctions. Results: Compared with solid nanoparticles, nanosheets, and other struct

ADAR1 Deficiency in Pancreatic Cancer Cells Potentiates cDC1 Activation and Promotes Sensitization to Immunotherapy.

Pancreatic cancer responds poorly to immunotherapy, primarily due to poor activation of CD8+ T cells and their limited infiltration into the tumor tissue. Strategies to enhance CD8+ T-cell priming and function are therefore critical for improving immunotherapeutic efficacy in pancreatic cancer. The RNA-editing enzyme adenosine deaminase acting on RNA 1 (ADAR1) plays a pivotal role in maintaining immune homeostasis. In this study, we found that ADAR1 expression positively correlates with pancreatic cancer progression. Depletion of ADAR1 in tumor cells resulted in excessive production of interfe

D-peptide-nanodrug conjugates synergize VISTA blockade and innate activation for cancer immunotherapy.

Effective cancer immunotherapy is often hindered by V-domain Ig suppressor of T cell activation (VISTA) signaling and myeloid-derived suppressor cells (MDSCs) within tumor microenvironment (TME). Here, we report D-peptide-nanodrug conjugates that synergize VISTA blockade and innate activation to overcome these constraints. We developed Reorpn7, a proteolytically stable D-peptide VISTA inhibitor with high binding affinity and in vitro serum stability. This inhibitor was co-loaded with a toll-like receptor 7/8 (TLR7/8) agonist (R848) into an matrix metalloproteinase-2 (MMP-2) responsive poly lac

cGAS-STING pathway activation drives the cold-to-hot tumor transition and sensitizes immunotherapy.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a central sensor of innate immunity that plays critical roles in recognizing cytosolic DNA and initiating antitumor immune responses. However, this pathway exhibits extensive spatiotemporal duality and context dependency in tumor regulation. In recent years, modulating this pathway to convert immunologically "cold" tumors into immunologically "hot," inflamed tumors has emerged as a cutting-edge strategy to reverse resistance to immune checkpoint inhibitors (ICIs). This review outlines the molecular mechanisms un

Sonogenetics Nanomedicine Activates the cGAS-STING Pathway for Immunotherapy of Triple-Negative Breast Cancer.

Due to pronounced tumor heterogeneity and the lack of effective therapeutic options, triple-negative breast cancer (TNBC) remains a formidable clinical challenge for the development of effective medicines. Here, we report a HER3-targeted sonogenetic mechano-immunomodulatory nanoplatform (HSMIN-LNP), achieving integrated synergistic therapy for TNBC. HSMIN-LNP employs a de novo AI-designed HER3-targeting miniprotein (HTIM) and co-delivers the TRPV4 (mechanosensory) plasmid together with an NFAT-promoted IL-15 gene. The results demonstrated that HTIM directly bound and suppressed HER3 signaling,

Micron-scale lactoferrin proteinosomes potentiate immune activation and chemo-immunotherapy in breast cancer.

Efficient uptake by antigen-presenting cells (APCs) is critical for the immunostimulatory performance of protein-based adjuvants, yet soluble proteins often suffer from poor cellular internalization and transient immune activation. Here, we report a hierarchical micron-scale assembly strategy to construct lactoferrin proteinosomes (LPs) via interfacial self-assembly. The resulting spherical LPs, with an average diameter of ∼2 μm, exhibit robust structural stability while retaining the native secondary structure of lactoferrin. Compared with monomeric lactoferrin, LPs markedly enhance internali

Hybrid membrane nanosystems for cancer immunotherapy: Biomimetic design, immune activation, and translational potential.

Conventional nanocarriers used in cancer immunotherapy face intrinsic limitations, such as suboptimal targeting efficiency, rapid reticuloendothelial clearance, and insufficient functionality, which have severely hindered the clinical translation of immunotherapeutic agents. Hybrid membrane-based biomimetic nanosystems have emerged as a promising solution to overcome these constraints by integrating the biological properties of heterogeneous cell membrane components to achieve synergistic functions encompassing "long circulation-active targeting-immune regulation". This represents an innovativ
