Psoriasis is largely mediated by interleukin (IL)-23/T helper (Th) 17 axis, and IL-21 is a pleiotropic cytokine expressed by Th17 cells. Despite previously reported possible pathogenic roles of IL-21 in human psoriasis, we found that IL-21 receptor (IL-21R) signalling was not crucial for imiquimod-induced psoriatic inflammation, using IL-21R-/- mice. The severity of imiquimod-induced psoriatic manifestation and pro-inflammatory Th17 cytokine levels, IL-17A-producing γδ T cells and CD4+ T cells, and in vitro IL-17A production by γδ T cells after IL-23 stimulation was comparable between wild-typ

OBJECTIVES: The central nervous system disorder in systemic lupus erythematosus (SLE), called neuropsychiatric lupus (NPSLE), is one of the most severe phenotypes with various clinical symptoms, including mood disorder, psychosis and delirium as diffuse neuropsychological manifestations (dNPSLE). Although stress is one of the aggravating factors for neuropsychiatric symptoms, its role in the pathogenesis of dNPSLE remains to be elucidated. We aimed to investigate stress effects on the neuropsychiatric pathophysiology in SLE using lupus-prone mice and patients' data.

INTRODUCTION: The therapeutic landscape for psoriasis is ever-changing. Risankizumab is the newest approved biologic and one of three currently licensed that targets the p19 subunit of interleukin-23 (IL-23). It is increasingly clear that different biologics vary in their efficacy, effectiveness, and safety profiles, highlighting that there is a need to understand for which patients and in which circumstances to use each drug.
AREAS COVERED: This article summarizes original clinical trial data, and reviews in more detail recent post-marketing studies and meta-analyses that differentiate risan

Listeria monocytogenes (LM) is a Gram-positive, intracellular bacterium that can induce spontaneous abortion, septicemia, and meningitis. Although it is known that neutrophils are required for elimination of the bacteria and for survival of the host, the mechanisms governing the recruitment of neutrophils to LM-infected tissues are not fully understood. We demonstrate here that IL-23 and the IL-17 receptor A (IL-17RA), which mediates both IL-17A and IL-17F signaling, are necessary for resistance against systemic LM infection. LM-infected IL-23p19 knockout (KO) mice have decreased production of

In mice and women, terminal differentiation of uterine natural killer (uNK) cells commences during endometrial decidualization. Both proliferation and interferon (IFN)-gamma are induced. Uterine NK cell precursors appear to home from secondary lymphoid organs to decidualizing uteri and localize mesometrially to the central decidua basalis, the site of maternal arterial modification at Gestation Days (gd) 9.5-10. In mice, genetic absence of uNK cells results in absence of pregnancy-induced spiral artery modification. Administration of IFN-gamma to uNK-negative pregnant females induces arterial

Psoriasis is an autoimmune chronic inflammatory skin condition with a strong genetic predisposition. Pathogenesis of psoriasis is complex and multifactorial; it is known that genetic, immunological, and environmental factors play significant roles in its development. Treatment options vary and include topical therapy (e.g., corticosteroids, vitamin D analogs, and calcineurin inhibitors), phototherapy (e.g., narrowband ultraviolet radiation (NB-UVB)), and systemic therapy (e.g., methotrexate and retinoids). Several new treatments have emerged in recent years, including biological treatments. Bi

Interleukin (IL) 12 and IL23 are two related heterodimeric cytokines produced by antigen-presenting cells. The balance between these two cytokines plays a crucial role in the control of Th1/Th17 responses and autoimmune inflammation. Most studies focused on their transcriptional regulation. Herein, we explored the role of the adenine and uridine-rich element (ARE)-binding protein tristetraprolin (TTP) in influencing mRNA stability of IL12p35, IL12/23p40, and IL23p19 subunits. LPS-stimulated bone marrow-derived dendritic cells (BMDCs) from TTP(-/-) mice produced normal levels of IL12/23p40. Pro

The live-attenuated oral vaccine intrOv is an IFNγ-susceptible mutant of C. muridarum that is rapidly cleared from the large intestine by IFNγ-producing group 3 innate lymphoid cells (IFNγ+ILC3s), while the wild-type C. muridarum maintains long-lasting colonization. Although we have previously shown that IL-23 receptor signaling is essential for IFNγ+ILC3s to clear intrOv, the cellular source of IL-23 remains unknown. In the current study, we found that intrOv induces IL-23 in the large intestine, which correlates with intrOv inhibition. Mice deficient in IL-23p19 fail to inhibit intrOv, demon

Unresolved tissue damage is a common feature of Inflammatory Bowel Disease (IBD) that facilitates disease progression. Here, we showed that high animal fat diets (HFD), an environmental risk factor associated with IBD pathogenesis, suppress intestinal macrophage production of critical tissue repair responses after damage. This includes reduced IL-23 production, which drives downstream production of IL-22, which is needed for barrier repair. Indicating that dietary lipids interfere with responses to microbial molecules needed to induce barrier protective functions, we found oleic acid could dir

BACKGROUND: Interleukin-23 (IL-23) and IL-17 may be involved in the pathogenesis of allergic rhinitis (AR). However, a differentiation of the role of IL-23 and IL-17 has not been performed yet.
OBJECTIVE: The aim of this study was therefore to investigate the immunomodulatory effects of IL-23 and IL-17, using a mouse model of AR.
METHODS: Anti-IL-23p19 and anti-IL-17 Abs were administrated intranasally during rechallenge in ovalbumin (OVA)-induced AR in BALB/c mice. Immunomodulatory effects were evaluated by measuring nasal rubbing and sneezing occurrences, serum OVA-specific antibodies, Th2

Interleukin (IL)-23, a cytokine produced by antigen presenting cells, targets both T cells and non-T cell types with the downstream effect of enhancing inflammatory pathways. Genome-wide association studies and data from human and mouse models of intestinal inflammation support the pathogenic role of IL-23 in Crohn's disease (CD), an immune-mediated disorder that can involve any part of the gastrointestinal tract. Areas covered: This review summarizes the available data on the role of IL-23 in CD and discusses the therapeutic relevance of blocking the function of IL-23 in this disorder. Expert

Burkholderia pseudomallei, a gram-negative intracellular bacterium, is a causative agent of melioidosis. The bacterium has been shown to induce the innate immune response, particularly pro-inflammatory cytokine production in several of both mouse and human cell types. In the present study, we investigate host immune response in B. pseudomallei-infected primary human monocytes. We discover that wild-type B. pseudomallei is able to survive and multiply inside the primary human monocytes. In contrast, B. pseudomallei LPS mutant, a less virulent strain, is susceptible to host killing during bacter

The inflammatory disorders collectively termed the seronegative spondyloarthropathies (SpA) include ankylosing spondylitis (AS), psoriatic arthritis (PsA), reactive arthritis, the arthritis associated with inflammatory bowel disease including Crohn's disease and ulcerative colitis, the arthritis related to anterior uveitis, and finally, somewhat controversially Behcet's disease. All of these diseases are associated with SNPs in the IL-23R or the interleukin-23 (IL-23) cytokine itself and related downstream signaling JAK pathway genes and the interleukin-17 (IL-17) pathway. Interleukin 23 (IL-23) is a key mediator of immune-mediated intestinal inflammation and plays an important role in the pathogenesis of inflammatory bowel disease. Inhibition of IL-23 signaling has therefore been validated as a therapeutic target in IBD. Here, we tested small binding proteins of non-immunoglobulin origin targeting either the IL-23 cytokine p19 subunit or the IL-23 receptor subunit IL-23R, designated ILP and REX binders, respectively. ILP317 and REX115 variants of formerly identified protein collections were tested in murine experimental colitis. Lactococcus lactis bacteria engi
