Role for a somatically diversified lectin in resistance of an invertebrate to parasite infection.

Invertebrates lack adaptive immune systems homologous to those of vertebrates, yet it is becoming increasingly clear that they can produce diversified antigen recognition molecules. We have previously noted that the snail Biomphalaria glabrata produces a secreted lectin, fibrinogen-related protein 3 (FREP3), unusual among invertebrate defense molecules because it is somatically diversified by gene conversion and point mutation. Here we implicate FREP3 in playing a central role in resistance to a major group of snail pathogens, digenetic trematodes. FREP3 is up-regulated in three models of resi

Characterization of the allergen filarial tropomyosin with an invertebrate specific monoclonal antibody.

Tropomyosins of invertebrates are pan-allergens responsible for wide spread allergic reactions against seafood and arthropods. As invertebrate tropomyosins are highly conserved, helminth tropomyosins are likely to show properties similar to these medically important allergens. Studies with a monoclonal antibody, NR1, raised against tropomyosin of the rodent filarial nematode Acanthocheilonema viteae revealed a B cell epitope common to helminths and marine mollusks, which does not occur in vertebrate tropomyosin. This antibody detected tropomyosin of A. viteae, other filariids, nematodes, trema

Canine piroplasmosis in Italy: epidemiological aspects in vertebrate and invertebrate hosts.

In Italy, canine piroplasmosis is believed to be widespread, but few data are available on its presence in most areas. In 2005 and 2006, vertebrate and invertebrate hosts were investigated in Central and Northern Regions of the Country. Microscopy on blood smears, molecular tools and serological tests were applied to 420 blood samples collected from dogs, in order to evaluate the presence of these protozoa and to identify possible risk factors. Moreover, ticks were analyzed by molecular techniques. Microscopy identified as positive 2.8% of the animals, all from Central Italy, and PCR detected

Antifungal drug discovery through the study of invertebrate model hosts.

There is an urgent need for new antifungal agents that are both effective and non-toxic in the therapy of systemic mycoses. The model nematode Caenorhabditis elegans has been used both to elucidate evolutionarily conserved components of host-pathogen interactions and to screen large chemical libraries for novel antimicrobial compounds. Here we review the use of C. elegans models in drug discovery and discuss caffeic acid phenethyl ester, a novel antifungal agent identified using an in vivo screening system. C. elegans bioassays allow high-throughput screens of chemical libraries in vivo. This

Genomic resources for invertebrate vectors of human pathogens, and the role of VectorBase.

High-throughput genome sequencing techniques have now reached vector biology with an emphasis on those species that are vectors of human pathogens. The first mosquito to be sequenced was Anopheles gambiae, the vector for Plasmodium parasites that cause malaria. Further mosquitoes have followed: Aedes aegypti (yellow fever and dengue fever vector) and Culex pipiens (lymphatic filariasis and West Nile fever). Species that are currently in sequencing include the body louse Pediculus humanus (Typhus vector), the triatomine Rhodnius prolixus (Chagas disease vector) and the tick Ixodes scapularis (L

[Some peculiarities of the relationships between parasitic copepods and their invertebrate hosts].

According to the rule of academican E. N. Pavlovskiy, any organism of host is an environment of inhabit for a parasite (Pavlovskiy, 1934). It was analysed, which "ecological niche" or microbiotop (= microhabitat) is occupied by this or that species of symbiotic (parasitic) copepods in organisms of different groups invertebrate-hosts. The assumption lying in a basis of the given analysis means that each group of hosts may give to cohabitants only certain variants of microbiotopes independently on the general morphological structure and life mode of hosts. Five types of microbiotops offered by v

Invertebrate immune recognition, natural immunity and the evolution of positive selection.

Invertebrate immunity exhibits immediate recognition without prior contact. Here, Tom Humphreys and Ellis Reinherz propose that receptors capable of actively recognizing self histocompatibility antigens could be generated by a process similar to positive selection, and that these could guide 'natural immunity' to foreign cells. These ideas provide a conceptual basis for invertebrate immunity and for understanding the evolutionary precedents for T-cell-mediated antigen recognition.

Filling the gaps - New molecular and morphological data of gregarine apicomplexans colonising freshwater invertebrates.

Gregarines (Apicomplexa: Gregarinasina) are widespread protist symbionts of invertebrates, occupying roles across the symbiotic spectrum from mutualism to parasitism. Despite their ecological importance, they remain far less studied than other apicomplexans, leaving many aspects of their diversity, host specificity, and evolutionary history unresolved. This is particularly true for freshwater taxa for which only a handful of small subunit (SSU) rDNA sequences from species colonising freshwater hosts are available in public databases. In this study, we screened ten freshwater invertebrates (Art

Phenytoin as an emerging pollutant on non-target aquatic invertebrates: effects on Lamellidens marginalis.

Pharmaceutical drugs have detrimental effects on aquatic organisms and change the ecological implications of the aquatic ecosystem. The present study was designed to examine the acute exposure (seven days) effect of phenytoin on the physiological parameters (condition index, filtration rate (FR), and clearance rate (CR)), metabolic capacity (electron transport system activity), energy reserves content (glycogen and protein concentrations), and oxidative biomarkers (antioxidant and biotransformation enzymes activities, lipid peroxidation levels) in the freshwater mussels Lamellidens marginalis.

Effect of copper mill waste material on benthic invertebrates and zooplankton diversity and abundance.

Copper (Cu) stamp mill mining in North America from the early 1900s produced a pulverized ore by-product now known as stamp sands (SS). In a mining operation near the city of Gay (Michigan, USA), SS were originally deposited near a Lake Superior beach, but erosion and wave action have moved many SS into beaches and reefs that are critical spawning and nursery areas for native fish (e.g., Lake Whitefish). Larval and juvenile native fish consume zooplankton and benthic invertebrates during their development, and many of these invertebrate taxa may be sensitive to metal contamination from the SS.

Fluctuating seawater pCO2/pH induces opposing interactions with copper toxicity for two intertidal invertebrates.

Global ocean pCO2 is increasing as a result of anthropogenic CO2 emissions, driving a decline in seawater pH. However, coastal waters already undergo fluctuations in pCO2/pH conditions over far shorter timescales, with values regularly exceeding those predicted for the open ocean by the year 2100. The speciation of copper, and therefore its potential toxicity, is affected by changing seawater pH, yet little is known concerning how present-day natural fluctuations in seawater pH affect copper toxicity to marine biota. Here, we test the hypothesis that a fluctuating seawater pCO2/pH regime will

Screening for Small Molecule Inhibitors of BMP-Induced Osteoblastic Differentiation from Indonesian Marine Invertebrates.

Fibrodysplasia ossificans progressiva (FOP) is a rare congenital disorder with heterotopic ossification (HO) in soft tissues. The abnormal activation of bone morphogenetic protein (BMP) signaling by a mutant activin receptor-like kinase-2 (ALK2) leads to the development of HO in FOP patients, and, thus, BMP signaling inhibitors are promising therapeutic applications for FOP. In the present study, we screened extracts of 188 Indonesian marine invertebrates for small molecular inhibitors of BMP-induced alkaline phosphatase (ALP) activity, a marker of osteoblastic differentiation in a C2C12 cell

Secondary metabolites from Vietnamese marine invertebrates with activity against Trypanosoma brucei and T. cruzi.

Marine-derived natural products from invertebrates comprise an extremely diverse and promising source of the compounds from a wide variety of structural classes. This study describes the discovery of five marine natural products with activity against Trypanosoma species by natural product library screening using whole cell in vitro assays. We investigated the anti-trypanosomal activity of the extracts from the soft corals and echinoderms living in Vietnamese seas. Of the samples screened, the methanolic extracts of several marine organisms exhibited potent activities against cultures of Trypan

Methylmercury in water, sediment, and invertebrates in created wetlands of Rouge Park, Toronto, Canada.

Thousands of hectares of wetlands are created annually because wetlands provide beneficial ecosystem services. Wetlands are also key sites for production of the bioaccumulative neurotoxin methylmercury (MeHg), but little is known about MeHg production in created systems. Here, we studied methylmercury in sediment, water, and invertebrates in created wetlands of various ages. Sediment MeHg reached 8 ng g(-1) in the newest wetland, which was significantly greater than in natural, control wetlands. This trend was mirrored in several invertebrate taxa, whose concentrations reached as high as 1.6 μ

Influence of metal exposure history on trace metal uptake and accumulation by marine invertebrates.

This paper reviews recent studies on the influence of trace metal exposure history on subsequent metal uptake in marine invertebrates, particularly bivalves and barnacles. Trace metal exposure may induce specific metal-binding ligands. Metallothionein-like proteins appear to play an important role in mediating metal uptake and hence accumulation, especially for Cd. Other ligands such as sulfide are important for Ag biokinetic changes in bivalves. The incorporation of trace metals into metal-rich granules appears to have little if any effect in modifying trace metal uptake. Furthermore, metals

Development of biotic ligand models for chronic manganese toxicity to fish, invertebrates, and algae.

Ecotoxicity tests were performed with fish, invertebrates, and algae to investigate the effect of water quality parameters on Mn toxicity. Models were developed to describe the effects of Mn as a function of water quality. Calcium (Ca) has a protective effect on Mn toxicity for both fish and invertebrates, and magnesium (Mg) also provides a protective effect for invertebrates. Protons have a protective effect on Mn toxicity to algae. The models derived are consistent with models of the toxicity of other metals to aquatic organisms in that divalent cations can act as competitors to Mn toxicity

A relationship between avian carcasses and living invertebrates in the epizootiology of avian botulism.

A survey of the sources of Clostridium botulinum type C toxin possibly utilized as food by aquatic birds in an epizootic area of avian botulism in northern Utah showed that living aquatic and terrestrial invertebrates normally found in close association with dead, decomposing birds commonly carried the toxin. Of 461 samples associated with 21 species of avian carcasses, 198 were toxin-positive. Invertebrate species not normally scavengers of vertebrate tissues were less commonly and less highly toxic, particularly when captured 30 cm or more from a carcass; six of 237 samples of such aquatic i

Traits retained in arthropod corpses influence the scavenging behavior of invertebrate scavengers.

Unveiling the enigmatic processes underlying the rapid disappearance of arthropod corpses in nature is not only fascinating but also essential for understanding their ecological fate. Functional traits retained in freshly dead arthropods may influence their consumption by invertebrate scavengers. To validate this idea, we conducted a field trapping survey using four types of baits (arthropod corpses, yak carrion, yak dung, and horse dung) to investigate the attraction of arthropod corpses to necrophagous beetles in a Tibetan alpine meadow, complemented by a laboratory experiment to further exa

Assessment of leaf-litter invertebrate biodiversity using high throughput sequencing.

Leaf litter ecosystems and their fauna are largely understudied, despite their critical ecological roles. Here, we investigate challenges associated with estimating biodiversity in terrestrial leaf litter. Current methodologies for biodiversity assessment are fraught with limitations; amongst the most significant is a decline in taxonomic expertise, complicating the process of species identification and the significant costs associated with species-level morphological identifications. DNA barcoding employs molecular markers to identify animal species, and the mitochondrial gene cytochrome c ox
