Polypyrrole-supported membrane proteins for bio-inspired ion channels.

Biomedical platforms constructed by immobilizing membrane proteins in matrixes made of synthetic organic polymers is a challenge because the structure and function of these proteins are affected by environmental conditions. In this work, an operative composite that regulates the diffusion of alkali ions has been prepared by functionalizing a supporting matrix made of poly(N-methylpyrrole) (PNMPy) with a β-barrel membrane protein (Omp2a) that forms channels and pores. The protein has been unequivocally identified in the composite, and its structure has been shown to remain unaltered. The PNMPy-

Ion channels and osteoarthritic pain: potential for novel analgesics.

Osteoarthritis (OA) is a debilitating chronic condition widely prevalent in ageing populations. Because the pathology of the disease includes cartilage erosion and joint remodelling, OA patients experience a great deal of pain. Despite numerous studies, details of OA are frequently inseparable from other types of chronic pain, and its causes are unknown. In most circumstances in OA, the cartilage lacks afferent innervation, although other joint tissues contain nociceptive neurones. In addition to physical joint damage, there is a strong element of joint inflammation. Genetic studies have ident

Mouse middle ear ion homeostasis channels and intercellular junctions.

HYPOTHESIS: The middle ear contains homeostatic mechanisms that control the movement of ions and fluids similar to those present in the inner ear, and are altered during inflammation. BACKGROUND: The normal middle ear cavity is fluid-free and air-filled to allow for effective sound transmission. Within the inner ear, the regulation of fluid and ion movement is essential for normal auditory and vestibular function. The same ion and fluid channels active in the inner ear may have similar roles with fluid regulation in the middle ear. METHODS: Middle and inner ears from BALB/c mice were process

Upregulation of colonic ion channels in APC ( Min/+ ) mice.

The adenomatous polyposis coli (APC) tumor suppressor gene is mutated in almost all human colonic cancers. Disturbances in Na(+) absorption have been observed in colonic cancer, and ion channels such as ether a go-go (Eag) or Ca(2+)-sensitive BK channels have been recognized for their oncogenic potential. APC ( Min/+ ) mice have reduced APC expression and develop multiple intestinal neoplasias (Min). Ion channels in the colonic epithelium were examined using electrophysiology and molecular techniques. APC ( Min/+ ) mice developed intestinal neoplasia and experienced a significant weight loss.

Human muscle voltage-gated ion channels and hereditary disease.

Insights in the field of ion channels were made possible by the Nobel-prize-winning patch-clamp technique that enables characterization of channel function, and have greatly been inspired by associated diseases pointing to regions of functional significance. These so-called ion channelopathies have common clinical features, recurrent patterns of mutations, and almost predictable mechanisms of pathogenesis. In skeletal muscle, disorders are associated with mutations in Na+, K+, Ca2+, and Cl- channels that lead to hypoexcitability (causing periodic paralysis) and to hyperexcitabilty (causing myo

Electroactive biomaterials modulating ion channels in nervous system regeneration.

Traumatic injury to the central and peripheral nervous systems disrupts the electrophysiological microenvironment through persistent ionic dysregulation and maladaptive ion channel remodeling, creating conditions that are fundamentally hostile to endogenous repair. Conventional biomaterials, while offering structural support, remain electrically inert and unable to reconstitute the bioelectric signaling context essential for functional neural regeneration. Here, we review the emerging class of electroactive biomaterials, encompassing conductive and piezoelectric platforms that are engineered t

Chemically induced cardiotoxicity: Role of voltage dependent ion channels.

Chemicals pervade human environments, with> 350,000 synthetic compounds driving unintentional exposures and pollution-linked deaths exceeding those from infectious diseases or violence. This review discusses the mechanisms of chemically induced cardiotoxicity (including pesticides, pollutants, heavy metals, and chemotherapeutics), emphasizing modulation of voltage-gated ion channels in cardiomyocytes, which disrupts action potential (AP) dynamics, excitation-contraction coupling (ECC), and predisposes to arrhythmogenesis. Key currents, sodium (INa), calcium (ICaL, ICaT), and potassium (Ito, IK

Migraine is a dysfunction of neuronal potassium ion channels.

Migraine is a primary headache disorder characterized by unilateral pain usually with aura, that affects approximately one in six individuals in India. The underlying biomechanical processes of migraine are still poorly understood, and new research is constantly being published. One of the major factors in migraine pathogenesis is the dysfunction of ion channels in the trigeminal nuclei and sensory cortices. Potassium channels are modulators and regulators of neuronal signaling and conductance, playing an important role in maintenance of the membrane potential and neuronal conduction. Therefor

Delta-type glutamate receptors are ligand-gated ion channels.

Delta-type ionotropic glutamate receptors (iGluRs, also known as GluDs) are members of the iGluR ligand-gated ion channel family, yet their function remains unknown1. Although GluDs are widely expressed in the brain, have key roles in synaptic organization, and harbour disease-linked mutations, whether they retain iGluR-like channel function is debated as currents have not been directly observed2,3. Here we define GluDs as ligand-gated ion channels that are tightly regulated in cellular contexts by purifying human GluD2 (hGluD2) and directly characterizing its structure and function using cryo

Ion channels in respiratory rhythm generation and sensorimotor integration.

Breathing movements depend on rhythmic neural activity in brainstem nuclei whose constituent neurons are well characterized. Knowing the sites and cells underlying the behavior enables us to identify the roles of individual ion channels. They accomplish three tasks: regulate excitability via the balance of intrinsic currents that govern baseline membrane potential and tonic firing; generate bursts to drive the motor output pattern; and transduce blood-gas levels, lung volume, and air qualities. Here, we explain how sodium and mixed cation channels (sodium leak channel non-selective [NALCN], Na

Statins activate temperature-gated transient receptor potential ion channels.

Statins are HMG-CoA reductase inhibitors administered to decrease levels of LDL cholesterol and to lower the risk of cardiovascular disease. Although statins are relatively well tolerated, adverse effects such as myalgia and painful peripheral neuropathy have been reported. While the underlying cause has not been fully elucidated, accumulating evidence shows that statins have numerous pleiotropic effects including anti-inflammatory and analgesic actions in several animal pain models. Here we report that some of the most extensively used statins activate members of the temperature-gated transie

The role of potassium ion channels in chronic sinusitis.

Chronic sinusitis is a common inflammatory disease of the nasal and sinus mucosa, leading to symptoms such as nasal congestion, runny nose, decreased sense of smell, and headache. It often recurs and seriously affects the quality of life of patients. However, its pathological and physiological mechanisms are not fully understood. In recent years, the role of potassium ion channels in the regulation of mucosal barrier function and inflammatory cell function has received increasing attention. In chronic sinusitis, there are often changes in the expression and function of potassium channels, lead

The Variety of Mechanosensitive Ion Channels in Retinal Neurons.

Alterations in intraocular and external pressure critically involve the pathogenesis of glaucoma, traumatic retinal injury (TRI), and other retinal disorders, and retinal neurons have been reported to express multiple mechanical-sensitive channels (MSCs) in recent decades. However, the role of MSCs in visual functions and pressure-related retinal conditions has been unclear. This review will focus on the variety and functional significance of the MSCs permeable to K+, Na+, and Ca2+, primarily including the big potassium channel (BK); the two-pore domain potassium channels TRAAK and TREK; Piezo

Bacterial spore germination receptors are nutrient-gated ion channels.

Bacterial spores resist antibiotics and sterilization and can remain metabolically inactive for decades, but they can rapidly germinate and resume growth in response to nutrients. Broadly conserved receptors embedded in the spore membrane detect nutrients, but how spores transduce these signals remains unclear. Here, we found that these receptors form oligomeric membrane channels. Mutations predicted to widen the channel initiated germination in the absence of nutrients, whereas those that narrow it prevented ion release and germination in response to nutrients. Expressing receptors with widen

New Insights into Ion Channels: Predicting hERG-Drug Interactions.

Drug-induced long QT syndrome can be a very dangerous side effect of existing and developmental drugs. In this work, a model proposed two decades ago addressing the ion specificity of potassium channels is extended to the human ether-à-gogo gene (hERG). hERG encodes the protein that assembles into the potassium channel responsible for the delayed rectifier current in ventricular cardiac myocytes that is often targeted by drugs associated with QT prolongation. The predictive value of this model can guide a rational drug design decision early in the drug development process and enhance NCE (New

Ligand-gated ion channels as targets of neuroactive insecticides.

The Cys-loop superfamily of ligand-gated ion channels (Cys-loop receptors) is one of the most ubiquitous ion channel families in vertebrates and invertebrates. Despite their ubiquity, they are targeted by several classes of pesticides, including neonicotinoids, phenylpyrazols, and macrolides such as ivermectins. The current commercialized compounds have high target site selectivity, which contributes to the safety of insecticide use. Structural analyses have accelerated progress in this field; notably, the X-ray crystal structures of acetylcholine binding protein and glutamate-gated Cl channel

Glycogen Synthase Kinase 3: Ion Channels, Plasticity, and Diseases.

Glycogen synthase kinase 3β (GSK3) is a multifaceted serine/threonine (S/T) kinase expressed in all eukaryotic cells. GSK3β is highly enriched in neurons in the central nervous system where it acts as a central hub for intracellular signaling downstream of receptors critical for neuronal function. Unlike other kinases, GSK3β is constitutively active, and its modulation mainly involves inhibition via upstream regulatory pathways rather than increased activation. Through an intricate converging signaling system, a fine-tuned balance of active and inactive GSK3β acts as a central point for the ph

Transient Receptor Potential (TRP) Ion Channels in Orofacial Pain.

Orofacial pain, including temporomandibular joint disorders pain, trigeminal neuralgia, dental pain, and debilitating headaches, affects millions of Americans each year with significant population health impact. Despite the existence of a large body of information on the subject, the molecular underpinnings of orofacial pain remain elusive. Two decades of research has identified that transient receptor potential (TRP) ion channels play a crucial role in pathological pain. A number of TRP ion channels are clearly expressed in the trigeminal sensory system and have critical functions in the tran
