PRM-LIVE with Trapped Ion Mobility Spectrometry and Its Application in Selectivity Profiling of Kinase Inhibitors.

Parallel reaction monitoring (PRM) has emerged as a popular approach for targeted protein quantification. With high ion utilization efficiency and first-in-class acquisition speed, the timsTOF Pro provides a powerful platform for PRM analysis. However, sporadic chromatographic drift in peptide retention time represents a fundamental limitation for the reproducible multiplexing of targets across PRM acquisitions. Here, we present PRM-LIVE, an extensible, Python-based acquisition engine for the timsTOF Pro, which dynamically adjusts detection windows for reproducible target scheduling. In this i

Stepwise optimization of a Flexible Microtube Plasma (FµTP) as an ionization source for Ion Mobility Spectrometry.

The ionization source is the central system of analytical devices such as mass spectrometers or ion mobility spectrometers. In this study, a recently developed flexible microtube plasma (FμTP) is applied as an ionization source for a custom-made drift tube ion mobility spectrometer (IMS) for the first time. The FµTP is based on a highly miniaturized, robust and a small-footprint dielectric barrier discharge design with an outstanding ionization efficiency. In this study, the experimental setup of the FµTP was further improved upon to achieve optimal coupling conditions in terms of the ion mobi

Detection of malignant pleural mesothelioma in exhaled breath by multicapillary column/ion mobility spectrometry (MCC/IMS).

Malignant pleural mesothelioma (MPM) is predominantly caused by previous asbestos exposure. Diagnosis often happens in advanced stages restricting any therapeutic perspectives. Early stage detection via breath analysis was explored using multicapillary column/ion mobility spectrometry (MCC/IMS) to detect volatile organic compounds (VOCs) in the exhaled breath of MPM patients in comparison to former occupational asbestos-exposed and non-exposed controls. Breath and background samples of 23 MPM patients, 22 asymptomatic former asbestos (AEx) workers and 21 healthy non-asbestos exposed persons we

The potential of ion mobility spectrometry (IMS) for detection of 2,4,6-trichloroanisole (2,4,6-TCA) in wine.

The off-flavor of "tainted wine" is attributed mainly to the presence of 2,4,6-trichloroanisole (2,4,6-TCA) in the wine. In the present study the atmospheric pressure gas-phase ion chemistry, pertaining to ion mobility spectrometry, of 2,4,6-trichloroanisole was investigated. In positive ion mode the dominant species is a monomer ion with a lower intensity dimer species with reduced mobility values (K(0)) of 1.58 and 1.20 cm(2)V(-1) s(-1), respectively. In negative mode the ion with K(0) =1.64 cm(2)V(-1)s(-1) is ascribed to a trichlorophenoxide species while the ions with K(0) =1.48 and 1.13 c

Detection of chlorinated and brominated byproducts of drinking water disinfection using electrospray ionization-high-field asymmetric waveform ion mobility spectrometry-mass spectrometry.

The lower limit of detection for low molecular weight polar and ionic analytes using electrospray ionization-mass spectrometry (ESI-MS) is often severely compromised by an intense background that obscures ions of trace components in solution. Recently, a new technique, referred to as high-field asymmetric waveform ion mobility spectrometry (FAIMS), has been shown to separate gas-phase ions at atmospheric pressure and room temperature. A FAIMS instrument is an ion filter that may be tuned, by control of electrical voltages, to continuously transmit selected ions from a complex mixture. This cap

Utilizing ion mobility spectrometry and mass spectrometry for the analysis of polycyclic aromatic hydrocarbons, polychlorinated biphenyls, polybrominated diphenyl ethers and their metabolites.

Polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are persistent environmental pollutants originating from incomplete combustion of organic materials and synthetic sources. PAHs, PCBs, and PBDEs have all been shown to have a significant effect on human health with correlations to cancer and other diseases. Therefore, measuring the presence of these xenobiotics in the environment and human body is imperative for assessing their health risks. To date, their analyses require both gas chromatography and liquid chromatography separa

Rapid Analysis of Anabolic Steroid Metabolites in Urine by Combining Field Asymmetric Waveform Ion Mobility Spectrometry with Liquid Chromatography and Mass Spectrometry.

The combination of field asymmetric waveform ion mobility spectrometry with liquid chromatography-mass spectrometry (LC-FAIMS-MS) has been developed for the analysis of glucuronide and sulfate metabolites of seven anabolic-androgenic steroids in urine. Separation by FAIMS-MS was investigated in positive ion mode for selected cationic adducts (H+, NH4+, Na+, K+, and Cs+). LC-FAIMS-MS analysis of the doubly sodiated adducts ([M + 2Na - H]+) of isobaric and coeluting steroid metabolites allowed their rapid (8 min) qualitative and quantitative determination in spiked urine using hydrophilic intera

High-Throughput Screening of Isomeric Reaction Products by Droplet Microfluidics Coupled to Cyclic Ion Mobility-Mass Spectrometry.

Engineered biocatalysts enable highly selective chemical transformations with low environmental impact. Development of biocatalysts by directed evolution requires screening many enzyme variants for improved catalytic properties. The low throughput of commonly used label-free screening methods, e.g., liquid chromatography-mass spectrometry (LC-MS), becomes the rate-limiting step in biocatalyst development, limiting the coverage of protein sequence space explored. Direct MS methods have been applied to biocatalyst screening; however, these methods cannot be used to evaluate isomer selectivity. I

Dielectric barrier discharge ionization (DBDI) enables rapid analysis of new psychoactive substances with ion mobility-mass spectrometry.

New psychoactive substances (NPS) present a major public health crisis across the world due to their variable potency, constant evolution within the recreational drug community, and ability to skirt legal/policy regulations. Because of the frequency with which new substances are introduced, it is increasingly difficult for clinical, toxicological, and forensic laboratories to keep up with the newest drug threats, presenting a critical need for development of rapid analytical methods capable of confident structural characterization. In the present work, we demonstrate successful coupling of a c

Ion Mobility-Constrained Nontargeted Screening of PFAS in Complex Environmental Matrices Using 18-T FTICR Mass Spectrometry.

Reliable screening of per- and polyfluoroalkyl substances (PFASs) in complex environmental matrices remains challenging due to severe spectral congestion and matrix-derived interferences under nontargeted conditions. Accurate-mass criteria from high-resolution mass spectrometry (HRMS) alone are often insufficient to suppress putative PFAS candidates arising from natural organic matter (NOM), leading to elevated false-positive rates. Here, we develop a mobility-resolved PFAS screening strategy by integrating direct-infusion gated trapped ion mobility spectrometry with Fourier transform ion cycl

Revelation of the discrepancy of volatile compounds in fig (Ficus carica) via gas chromatography ion-mobility spectrometry.

The volatile compounds of fig (Ficus carica) are influenced by various factors. To explore the composition and difference of volatile compounds among figs, gas chromatography ion mobility spectrometry (GC-IMS) was used to study the volatiles of figs from various regions, diverse cultivars, and after treatment with different drying methods. Aldehydes were the main volatile compounds in Bojihong from Shandong, while esters, ketones, and alcohols were the main volatile compounds in Bojihong from Sichuan and Guangdong. The volatiles of Branswick and Banane were similar, but differed significantly

Diagnosis of COVID-19 by analysis of breath with gas chromatography-ion mobility spectrometry - a feasibility study.

BACKGROUND: There is an urgent need to rapidly distinguish COVID-19 from other respiratory conditions, including influenza, at first-presentation. Point-of-care tests not requiring laboratory- support will speed diagnosis and protect health-care staff. We studied the feasibility of using breath-analysis to distinguish these conditions with near-patient gas chromatography-ion mobility spectrometry (GC-IMS). METHODS: Independent observational prevalence studies at Edinburgh, UK, and Dortmund, Germany, recruited adult patients with possible COVID-19 at hospital presentation. Participants gave a

Utilizing Untargeted Ion Mobility-Mass Spectrometry To Profile Changes in the Gut Metabolome Following Biliary Diversion Surgery.

Obesity and obesity-related disorders are a global epidemic affecting over 10% of the world's population. Treatment of these diseases has become increasingly challenging and expensive. The most effective and durable treatment for Class III obesity (body mass index ≥35 kg/m2) is bariatric surgery, namely, Roux-en-Y gastric bypass (RYGB) and vertical sleeve gastrectomy. These procedures are associated with increased circulating bile acids, molecules that not only facilitate intestinal fat absorption but are also potent hormones regulating numerous metabolic pathways. We recently reported on a no

TIMSImaging: an open and interoperable workflow for trapped ion mobility mass spectrometry imaging data processing and visualization.

SUMMARY: Mass spectrometry imaging (MSI) enables high-throughput spatial mapping of molecules, but the lack of chromatographic separation limits its utility for complex biological samples. Ion mobility (IM) provides key orthogonal separation. However, open-source tools dedicated for IM-MSI data analysis remain scarce and the integration of ion mobility information into downstream analysis is underexplored. Here, we present TIMSImaging, an open-source workflow for processing and visualization of MALDI-TIMS-MS data from Bruker timsTOF instruments. TIMSImaging incorporates a graph-based two-dimen

Machine Learning-Assisted Headspace Gas Chromatography Ion Mobility Spectrometry for Characterization and Geographical Authentication of Perillae Folium.

Perillae Folium, derived from the leaves of Perilla frutescens (L.) Britt., is recognized as both a medicinal substance and an edible ingredient in China, with its market demand steadily rising. The nutritional value of Perillae Folium varies markedly across different geographical origins, posing a constraint on its further industrial development. To date, there remains a paucity of robust and reliable approaches for tracing the geographical origin of Perillae Folium samples. This study employed headspace gas chromatography ion mobility spectrometry to profile VOCs in Perillae Folium, revealin

Characterization of Alkane Oxidation Products in a Corona-Discharge Reactor Using Ammonia-Doped Ion Mobility-Mass Spectrometry.

This study examines the oxidation of C5-C8 alkanes in an external corona-discharge reactor. Oxidation products were characterized using ion mobility spectrometry (IMS) and tandem IMS-mass spectrometry (IMS-MS) with an internal corona-discharge APCI source. This configuration enables structural isomer resolution and provides deeper insight into the molecular composition of the oxidation products. IMS-MS analysis showed that alkane (M) oxidation produces mainly ketones ([M+O-2H]) and diketones ([M+2O-4H]), with 3-ketone isomers dominating. Under standard APCI conditions, H+(H2O)n reactant ions p

Tiny Droplets, Large Insights: Studying Peptide Aggregation with Droplet-Based Microfluidics Hyphenated with Ion Mobility Mass Spectrometry.

Peptide and protein aggregation are hallmarks of numerous neurodegenerative diseases. However, the structural characterization of oligomeric intermediates remains challenging due to their low abundance, heterogenic nature, and complexity of coinciding and transient reaction networks. Confining reactions to microenvironments, such as picoliter droplets in droplet-based microfluidics (DBMFs), simplifies these networks, thereby enabling high-throughput analysis and controlled reaction kinetics. This study explores the integration of DBMF with ion mobility mass spectrometry (IM-MS) to probe the ag

Determination of trace methanol in wine by improved distillation-dopant-assisted high-field asymmetric ion mobility spectrometry.

Methanol is a harmful component in wine, and excessive intake is a serious health hazard. In this study, an improved distillation enrichment coupled with dopant-assisted high-field asymmetric waveform ion mobility spectrometry (FAIMS) detection system was constructed for the sensitive determination of methanol in wine. The methanol in wine was effectively separated and enriched by improved distillation pretreatment to reduce matrix interference; 2-butanone was introduced as a dopant to generate ions and free electrons under UV lamp ionization. A new reaction channel was provided for the effici

Investigation of corona discharge ionization of barbituric acid using ion mobility spectrometry along with quantum chemical calculations.

This study aimed at examining atmospheric-pressure chemical ionization of barbituric acid through the corona discharge ion mobility spectrometry (CD-IMS) and the quantum chemical calculations. The results indicated two product ion peaks in the IMS spectrum of barbituric acid. The thermal decomposition of the barbituric acid sample was investigated by scanning the temperature of the injection port and analyzing the temporal evolution of the IMS peaks over elapsed time. It was found that the barbituric acid sample was not thermally decomposed in the injection port of the instrument. Experimental
