[Free gingival graft for augmenting peri-implant keratinized mucosa].

Insufficient keratinized mucosa is a common clinical problem. Although there is no consistent data on the long-term success of implant restorations relative to the presence or absence of peri-implant keratinized mucosa, it is widely accepted that the presence of keratinized mucosa width<2 mm is associated with increased biofilm accumulation, soft-tissue inflammation, and mucosal recession. Free gingival graft (FGG) is the standard surgical intervention for augmenting the width of keratinized gingiva. However, the clinical application of FGG is technique-sensitive. Therefore, this article descr

Trichilemmal cysts. Ultrastructural similarities to the trichilemmal sac.

Recently experienced cases of trichilemmal cysts were studied by light and electron microscopy. Although ultrastructural findings in the present case were basically consistent with those reported in the literature, they were in contrast to them in some important respects: (1) a few, oval and small-sized keratohyalin and a large number of membrane-coating granules were observed in the cytoplasm of keratinizing cells beneath the keratinized cells and (2) the junctional structure between the keratinized cells and the keratinizing cells beneath them showed interdigitation. From these findings, it

Electrospun PVA-PCL/HA nanofiber scaffolds containing mupirocin for surgical wound healing in Sprague Dawley rats.

Surgical site infections (SSIs) and wound dehiscence are significant postoperative complications that prolong hospital stays and increase treatment costs thus highlighting the need for effective wound dressing options to support healing and minimize scar formation. This study seeks to produce biodegradable electrospun scaffolds for initial burst release and subsequent prolonged release of bioactive substances to enhance wound closure after surgical procedures. Five nanofibrous scaffolds were fabricated using keratin, PCL-PVA blend with CoQ10, mupirocin, and hyaluronic acid at a voltage of 25 -

Keratin K6c mutations cause focal palmoplantar keratoderma.

The palmoplantar keratodermas (PPKs) are a large group of clinically and genetically heterogeneous genodermatoses. The gene defects underlying many PPKs still need to be resolved to facilitate definitive molecular diagnosis and genetic counseling. Dominant-negative mutations in any of the four identified keratin genes, KRT6A, KRT6B, KRT16, or KRT17, cause pachyonychia congenita (PC), characterized by hypertrophic nail dystrophy and other ectodermal features. In PC, focal PPK (FPPK) is the most painful and debilitating phenotypic feature. Some families presenting with FPPK alone, or with minima

Impact of connective tissue graft thickness on surgical outcomes: A pilot randomized clinical trial.

BACKGROUND: The aim of this study was to compare thick versus thin connective tissue grafts (CTG) for the treatment of gingival recession, over a 3-month period. METHODS: Forty-two CTG procedures were performed on single tooth Miller Class I or II recession defects at either premolar or anterior sites in 30 individuals. Procedures were randomized (1:1 ratio) to CTG thickness of 1 or 2 mm (parallel group design). Primary outcomes were the change in the width of the zone of keratinized tissue and the amount of root coverage achieved 3 months postoperatively at the recipient site. Secondary outc

A spectrum of mutations in keratins K6a, K16 and K17 causing pachyonychia congenita.

BACKGROUND: Pachyonychia congenita (PC) is a rare autosomal dominant keratin disorder, subdivided into two major variants, PC-1 and PC-2. Predominant characteristics include hypertrophic nail dystrophy, focal palmoplantar keratoderma and oral leukokeratosis. Multiple steatocystomas that develop during puberty are a useful feature distinguishing PC-2 from PC-1. At the molecular level it has been shown that mutations in keratin K6a or K16 cause PC-1 whereas those in K6b or K17 lead to PC-2. OBJECTIVE: To identify mutations in 22 families presenting with clinical symptoms of either PC-1/focal no

Keratin mutation predisposes to mouse liver fibrosis and unmasks differential effects of the carbon tetrachloride and thioacetamide models.

BACKGROUND & AIMS: Keratins 8 and 18 (K8/K18) are important hepatoprotective proteins. Animals expressing K8/K18 mutants show a marked susceptibility to acute/subacute liver injury. K8/K18 variants predispose to human end-stage liver disease and associate with fibrosis progression during chronic hepatitis C infection. We sought direct evidence for a keratin mutation-related predisposition to liver fibrosis using transgenic mouse models because the relationship between keratin mutations and cirrhosis is based primarily on human association studies. METHODS: Mouse hepatofibrosis was induced by

Giant Cell Tumor of Soft Tissue Involving the Common Hepatic Duct: A Case Report and Review of the Literature.

Giant cell tumor of soft tissue is a low-malignant behavior neoplasm, with histopathological features and immunophenotype similar to its bone counterpart. Primary giant cell tumor of soft tissue in the biliary tract is an exceedingly rare entity. Here, we describe the first patient of giant cell tumor of soft tissue involving the common hepatic duct in a 66-year-old man who developed a biliary obstruction. Imaging revealed an ill-defined 34 × 21 mm intraluminal mass at the liver hilum, initially suggestive of a Klatskin tumor. A right hepatectomy with excision of the common hepatic duct and ga

Uncovering of transplanted connective tissue graft: Clinical and histological evaluation.

BACKGROUND: Periodontal plastic surgery aims to restore recessions and dehiscence around teeth and implants. Several techniques, such as subepithelial connective tissue graft (CTG), were proposed with the main outcome of improving volume and root coverage. Nevertheless, this surgery might not improve the keratinized tissue width. Thus, the primary aim of this case report was to describe the possible increase in keratinized tissue after a subepithelial CTG and simultaneously use the previously harvested graft as a source for covering an adjacent tooth. METHODS: A 38-year-old patient presented

Biochemical examination of fetal skin biopsy specimens obtained by fetoscopy: use of the method for analysis of keratins and filaggrin.

This paper describes a method for biochemical analysis of proteins from fetal skin biopsy samples. The method has wide potential application for diagnosis of disorders with a known protein abnormality detectable by protein staining or a specific antibody. Analysis requires a single 1 mm biopsy, is rapid (2 days) and extremely sensitive. In the present study, fetal skin biopsies from normal fetuses and a fetus at risk for lamellar ichthyosis were obtained. The epidermis or hairs with attached follicular cells were dissected from the remaining skin. Proteins were extracted and separated by SDS-p

Treatment of gingival recession in 2 surgical stages: free gingival graft plus coronally positioned flap.

Gingival recession is a common clinical condition that creates unfavorable esthetics and dentinal hypersensitivity. Treatment of gingival recession aims to create or increase keratinized tissue and provide root coverage. The aim of this article is to present 2 clinical cases of severe Miller Class II gingival recession with absence of keratinized tissue. Each patient was treated with a free gingival graft associated with a coronally positioned flap. Follow-up indicated that the use of the free gingival graft was effective in increasing keratinized tissue and in covering the recession in a sati

A mechanically tunable photo-cured hydrogel system based on methacrylated collagen-keratin inter-penetrating networks for rapid and enhanced wound healing.

Conventional hydrogel wound dressings have non-tunable mechanical properties and serve only as passive barriers, resulting in slow wound healing. Advanced hydrogel dressings that can stimulate wound contraction and tissue deformation can potentially accelerate the healing of chronic wounds characterized by long healing times and high infection risk. In this study, we developed injectable photo-cured methacrylated collagen-keratin interpenetrating network (ColMA-K@IPN) hydrogels for rapid enhanced wound healing. Fourier transform infrared spectroscopy (FTIR) and 1H nuclear magnetic resonance (N

Molecular and immunohistochemical profile of a basaloid (cloacogenic) carcinoma of the sigmoid colon: possible predictive value for clinical outcomes.

A 61-year-old woman was hospitalized with a 5-week history of abdominal discomfort, change in bowel habits, and weight loss. Colonoscopy showed a protruded tumor of the sigmoid colon first diagnosed as undifferentiated carcinoma. Surgical resection of the sigmoid colon was performed. Histological examination of the surgical specimen showed a proliferation of basaloid cells arranged in tumor clusters with central comedonecrosis and peripheral palisading of the nuclei. The tumor invaded the subserosa and presented liver metastasis without lymph node metastases. The tumor cells were marked by ker

Role of miR-182 in response to oxidative stress in the cell fate of human fallopian tube epithelial cells.

BACKGROUND: Epidermolysis bullosa simplex (EBS) is an inherited skin fragility disorder caused by mutations in keratin intermediate filament proteins. While discoveries of these mutations have increased understanding of the role of keratins and other intermediate filaments in epithelial tissues, progress towards the development of therapy for these disorders is much slower. OBJECTIVES: Cell culture model systems that display these structural defects are needed for analysis of the cellular consequences of the mutations and to enable possible therapeutic strategies to be developed. Our aim was

Keratin 8 modulates β-cell stress responses and normoglycaemia.

Keratin intermediate filament (IF) proteins are epithelial cell cytoskeletal components that provide structural stability and protection from cell stress, among other cellular and tissue-specific functions. Numerous human diseases are associated with IF gene mutations, but the function of keratins in the endocrine pancreas and their potential significance for glycaemic control are unknown. The impact of keratins on β-cell organisation and systemic glucose control was assessed using keratin 8 (K8) wild-type (K8(+/+)) and K8 knockout (K8(-/-)) mice. Islet β-cell keratins were characterised under
