Evaluation of histophysiological alterations associated with ketoprofen administration in albino NMRI mice.

The aim of this study was to investigate the changes caused by the administration of ketoprofen to albino NMRI mice on some hematological, biochemical, and structural parameters. For this purpose, the mice were divided into two lots: a control batch and an experimental batch to which ketoprofen was administered subcutaneously at a dose of 10 mg/kg body weight per day for 7 days. A decrease in erythrocyte number and hemoglobin was observed altogether with the increase in white blood cells. Blood biochemistry indicates increased blood glucose, cholesterol, and triglyceride levels. Enzyme values

Ketoprofen affects the mammary immune response in dairy cows in vivo and in vitro.

Nonsteroidal anti-inflammatory drugs are commonly administered parenterally in addition to antimicrobial mastitis therapy to increase the well-being of the diseased animal. As mastitis is usually a localized infection of mammary tissue, we tested the hypothesis that a local administration of nonsteroidal anti-inflammatory drugs through the teat canal could have anti-inflammatory effects on the affected area. We investigated the effects of intramammarily administered ketoprofen (KET) during an LPS-induced immune response on somatic cell count (SCC) and blood-milk barrier integrity. In addition,

Prophylactic effect of plaster and cataplasm contained ketoprofen in rats with adjuvant arthritis.

This study demonstrates that prophylactic effect of plaster and cataplasm contained ketoprofen in adjuvant arthritis therapy by X-ray. Adjuvant arthritis was induced by a single injection of Freund's complete adjuvant. Mature female Sprague-Dawley rats were designated to 3 groups such as nontreated control, plaster-treated (PT) and cataplasm-treated (CT), each of which was composed of ten animals. The PT and the CT groups showed reduced primary paw swelling, but secondary paw swelling was not affected. Bony changes were observed in all regions of the femur and tibia of the nonadjuvant-injected

Total and free ketoprofen in serum and synovial fluid after intramuscular injection.

Free and total ketoprofen levels in serum and synovial fluid were determined in 37 patients after a single intramuscular injection of ketoprofen, 100 mg. Free drug was separated by equilibrium dialysis. Ketoprofen was assayed by HPLC. Ketoprofen penetrated into the joints rapidly and significant concentrations were found at 15 minutes. The equilibrium time was about 3 1/2 hours. The AUC for total ketoprofen was greater in serum than in synovial fluid. On the other hand, the free fraction AUC in the serum and synovial fluid were quite similar. The mean residence time in the joint was about thre

The Hidden Threat of Pharmaceuticals: Ketoprofen Degradation and Toxicity to Non-Target Organisms.

Ketoprofen is a widely prescribed non-steroidal anti-inflammatory drug whose extensive global use, combined with limited biodegradability, has led to its increasing detection as a micropollutant in aquatic and terrestrial environments. Incomplete removal during wastewater treatment results in its continuous release into surface waters and soils, creating conditions for chronic, low-dose exposure of non-target organisms. This review synthesizes current knowledge on the physicochemical characteristics of ketoprofen, its mechanism of action, environmental occurrence, degradation pathways, and eco

An Unusual Localized Reaction of Conjunctivitis with Systemic Use of Ketoprofen: A Case Report.

INTRODUCTION: Although the eye is rarely the sole target of an immediate allergic-type response, ocular signs and symptoms often represent the most prominent features of systemic allergic reactions. Nonsteroidal anti-inflammatory drugs (NSAIDs), due to their cyclooxygenase inhibitory activity, can trigger nonallergic hypersensitivity responses. Ketoprofen, in particular, is one of the most potent cyclooxygenase inhibitors at therapeutic plasma concentrations and is recognized as a frequent photoallergic agent. However, localized hypersensitivity reactions following systemic administration are

Use of Meloxicam or Ketoprofen for Piglet Pain Control Following Surgical Castration.

Surgical castration of piglets is performed routinely on commercial pig farms, to prevent boar taint and minimize aggression. While this procedure is known to be painful, piglets are generally not provided any analgesic for pain relief, leading to welfare concerns. The objectives of this study were to assess the efficacy of two non-steroidal anti-inflammatory drugs (NSAIDs), meloxicam (MEL) (0.4 mg/kg or 1.0 mg/kg) and ketoprofen (KET) (6.0 mg/kg) in reducing behavioral indicators of pain in castrated piglets. This study also examined the utility of the Piglet Grimace Scale (PGS) as a pain ass

Single dose oral ketoprofen or dexketoprofen for acute postoperative pain in adults.

BACKGROUND: This review is an update of "Single dose oral ketoprofen and dexketoprofen for acute postoperative pain in adults" last updated in Issue 4, 2009. Ketoprofen is a non-selective nonsteroidal anti-inflammatory drug (NSAID) used to treat acute and chronic painful conditions. Dexketoprofen is the (S)-enantiomer, which is believed to confer analgesia. Theoretically dexketoprofen is expected to provide equivalent analgesia to ketoprofen at half the dose, with a consequent reduction in gastrointestinal adverse events. This review is one of a series on oral analgesics for acute postoperativ

Modulation by gamithromycin and ketoprofen of in vitro and in vivo porcine lipopolysaccharide-induced inflammation.

The immunomodulatory properties of gamithromycin (GAM), ketoprofen (KETO) and their combination (GAM-KETO) were investigated after both in vitro and in vivo lipopolysaccharide (LPS)-induced inflammation. The influence of these drugs was measured on the production of prostaglandin E2 (PGE2) and the pro-inflammatory cytokines tumour necrosis factor (TNF)-α, interleukin (IL)-6 and IL-1β in both LPS-stimulated porcine peripheral blood mononuclear cells (PBMCs) and LPS-challenged pigs. Additionally, effects on the production of acute phase proteins (APPs), including pig major acute phase protein (p

Ecotoxicity of ketoprofen, diclofenac, atenolol and their photolysis byproducts in zebrafish (Danio rerio).

The occurrence of pharmaceutical compounds in wastewater treatment plants and surface waters has been detected worldwide, constituting a potential risk for aquatic ecosystems. Adult zebrafish, of both sexes, were exposed to three common pharmaceutical compounds (atenolol, ketoprofen and diclofenac) and their UV photolysis by-products over seven days. The results show that diclofenac was removed to concentrations<LOD after 5 min of UV irradiation. The oxidative stress response of zebrafish to pharmaceuticals and their photolysis by-products was evaluated through oxidative stress enzymes (glutat

Titanium dioxide inclusion in backing reduce the photoallergenicity of ketoprofen transdermal patch.

Ketoprofen (KP) is a widely used transdermal non-steroidal anti-inflammatory drug. However, increasing number of adverse effect case reports suggests that KP transdermal formulation can cause photoallergic reaction. The photoallergic potential of KP is attributable to the instability of KP under UV/visible light and subsequent formation of reactive degradation products. In this study, we investigated whether the inclusion of titanium dioxide (TiO(2)), a well-known mineral sunscreen agent, in the KP transdermal patch can prevent the photodegradation of KP and ultimately, can reduce photoallergi

In vivo pharmacokinetics of ketoprofen after patch application in the Mexican hairless pig.

To evaluate the pharmacokinetics of topical drugs, in vitro permeation studies are performed using sacrificed pig skin or human tissues resected at surgery; however, these methods have their limitations in in vivo pharmacokinetics. This study examined the usefulness of Mexican hairless pigs for in vivo pharmacokinetic study, especially the drug concentration in the tissues. A ketoprofen patch was applied on the back of Mexican hairless pigs for 24 h, followed by sequential collection of blood specimens from 0 to 36 h (n=3). Also, the skin, subcutaneous fat, fascia and muscle from the center of
