General and histological indicators of health in wild fishes from a biological mercury hotspot in northeastern North America.

Kejimkujik National Park and National Historic Site, Nova Scotia, Canada, is considered a biological mercury (Hg) hotspot because the tissues of yellow perch (Perca flavescens) and common loons (Gavia immer) inhabiting the lakes frequently exceed so-called safe levels of Hg. In the present study, the relationships between Hg and overall health of males and females of 3 forage fish species (brown bullhead Ameirus nebulosus, banded killifish Fundulus diaphanus, and golden shiner Notemigonus crysoleucas; n = 6-18/sex/lake) in 6 lakes at the park were assessed using condition factor, liversomatic

An integrative taxonomic characterization of extremophile fish Aphanius almiriensis (Teleostei: Cyprinodontiformes) in a hypersaline geothermal system (NW Anatolia).

The small killifish Aphanius almiriensis (Kottelat, Barbieri & Stoumboudi, 2007) inhabits from brackish springs to estuaries and coastal lagoons in the eastern Mediterranean basin. This study focused on the detailed characterization of Aphanius almiriensis from a hypersaline geothermal spring (Tuzla, Çanakkale, NW Anatolia), where extreme salinity (61.14 ppt in winter, January 2023) and temperature (42.7°C in summer, July 2023) conditions were observed. The obtained specimens of killifish in the sampling site, with a slightly convex dorsal head profile and a more angular ventral head profile,

Adverse effects of BDE-47 on in vivo developmental parameters, thyroid hormones, and expression of hypothalamus-pituitary-thyroid (HPT) axis genes in larvae of the self-fertilizing fish Kryptolebias marmoratus.

2,2',4,4'-tetrabromodiphenylether (BDE-47) is known to have the potential to disrupt the thyroid endocrine system in fishes due to its structural similarity to the thyroid hormones triiodothyronine (T3) and thyroxine (T4). However, the effects of BDE-47 on thyroid function in fishes remain unclear. In this study, abnormal development (e.g. deformity, hemorrhaging) and an imbalance in thyroid hormone (TH) homeostasis was shown in the early developmental stages of the mangrove killifish Kryptolebias marmoratus in response to BDE-47 exposure. To examine the thyroid endocrinal effect of BDE-47 exp

Regional Distribution of a Brain-Encysting Parasite Provides Insight on Parasite-Induced Host Behavioral Manipulation.

Some parasite species alter the behavior of intermediate hosts to promote transmission to the next host in the parasite's life cycle. This is the case for Euhaplorchis californiensis, a brain-encysting trematode parasite that causes behavioral changes in the California killifish (Fundulus parvipinnis). These manipulations increase predation by the parasite's final host, piscivorous marsh birds. The mechanisms by which E. californiensis achieves this manipulation remain poorly understood. As E. californiensis cysts reside on the surface of the killifish's brain, discerning regional differences

Age-related biochemical profiling reveals multi-system physiological decline in the annual fish Nothobranchius guentheri.

Annual killifishes of the genus Nothobranchius compress a vertebrate life cycle into a few months, which is what makes them useful in aging research, yet no broad biochemical description of ageing exists for N. guentheri. In a cross-sectional design, we measured 27 clinical chemistry analytes in whole-body homogenates of male and female fish sampled at 3, 6 and 9 months of age (20 analytical samples). Age trends were assessed by Spearman rank correlation, and age, sex and their interaction by two-way ANOVA. Twenty-four of the 27 analytes fell with age. Calcium declined most steeply (r = - 0.57

Mitochondrial physiology and responses to elevated hydrogen sulphide in two isogenic lineages of an amphibious mangrove fish.

Hydrogen sulphide (H2S) is toxic and can act as a selective pressure on aquatic organisms, facilitating a wide range of adaptations for life in sulphidic environments. Mangrove rivulus (Kryptolebias marmoratus) inhabit mangrove swamps and have developed high tolerance to environmental H2S. They are hermaphroditic and can self-fertilize, producing distinct isogenic lineages with different sensitivity to H2S. Here, we tested the hypothesis that observed differences in responses to H2S are the result of differences in mitochondrial functions. For this purpose, we performed two experimental series

A cross-species drug-discovery platform to accelerate the identification of lifespan-extending interventions.

We develop an integrated, high-throughput platform to evaluate pharmacological lifespan-extending interventions across five model organisms spanning unicellular to mammalian biology including yeast, nematodes, fruit flies, killifish, and mice. By combining automated imaging, miniaturized assays, and deep-learning-based death detection, we evaluate over 400 compounds across thousands of conditions. In S. cerevisiae, a miniaturized PI/flow cytometry CLS assay enables scalable screening and identifies key assay confounders, while in C. elegans and D. melanogaster, compact imaging platforms couple

Histopathology of laboratory-reared Nothobranchius fishes: Mycobacterial infections versus neoplastic lesions.

Short-lived killifishes of the genus Nothobranchius Peters, 1868 (Cyprinodontiformes) are considered promising model organisms for biomedical research on ageing and tumorigenesis. We conducted histopathological analysis of 411 adult individuals from three Nothobranchius species to study details on spontaneous age-related neoplastic lesions. Light microscopy based on H&E and toluidine blue-stained sections revealed (a) non-proliferative liver changes with pronounced vacuolation of hepatocytes; (b) proliferation of kidney haemopoietic tissue contributing to excretory system damage; (c) prolifera

Differences of oxidative damage in the brain of short- and long-lived Nothobranchius furzeri strains.

BACKGROUND: The turquoise killifish (Nothobranchius furzeri), while being reported as the vertebrate with the shortest lifespan in captivity, includes many strains that diverge in their lifespans. Underlying mechanisms for these differences are believed to occur in part at the level of mitochondria, which contribute to aging through their role in energy metabolism and oxidative stress. METHODS: We used the short-lived GRZ strain and the longer-lived MZCS-222 strain to determine whether lifespan divergence is associated with distinct mitochondrial maintenance, oxidative damage, and mitigating

A scalable and tunable platform for functional interrogation of peptide hormones in fish.

Pituitary hormones play a central role in shaping vertebrate life history events, including growth, reproduction, metabolism, and aging. The regulation of these traits often requires precise control of hormone levels across diverse timescales. However, fine tuning circulating hormones in-vivo has traditionally been experimentally challenging. Here, using the naturally short-lived turquoise killifish (N. furzeri), we describe a high-throughput platform that combines loss- and gain-of-function of peptide hormones. Mutation of three primary pituitary hormones, growth hormone (gh1), follicle stimu

Low-conductance anion channel activated by cAMP in teleost Cl- -secreting cells.

We studied characteristics and modulation of ion channels in primary cultures of opercular epithelium from the euryhaline marine killifish Fundulus heteroclitus. Primary cultures, 17-28 h old, retain mitochondria-rich Cl- cells identifiable by fluorescence microscopy. Cell-attached patches revealed frequent low-conductance 8.1 +/- 0.35 pS channels that usually became inactive on excision; high-conductance anion channels were not apparent. Ion substitution experiments demonstrated selectivity for Cl- over gluconate of 1:0.07. With addition of 1-isobutyl-3-methylxanthine (0.1 mM) and dibutyrylad

Ultrastructural alterations of hepatocytes in short-lived model fish, Nothobranchius furzeri Jubb, 1971.

The hepatocytes from 40 Nothobranchius furzeri, a promising model in biogerontology, were subjected to ultrastructural examination to provide a visual guide to the repertoire of hepatocyte alterations recognized in this species. The observed hepatocyte alterations were mostly regressive in nature varying in the level of reversibility. In addition to the frequent occurrence of cytoplasmic deposits of lipids and glycogen, this study highlights the first findings of giant mitochondria with paracrystalline structures in hepatocytes and monocytic cells of the spleen in N. furzeri, which are current

Temperature Effects on Swimming Performance, Oxygen Consumption, and Skeletal Muscle Metabolic Phenotype in Amargosa River Pupfish, Cyprinodon nevadensis amargosae.

AbstractClimate change is raising temperatures in aquatic habitats and shifting the frequency and severity of precipitation events. In fishes, metabolic demands increase under warmer conditions, which may limit the ability of fishes to survive the physical and metabolic demands of high-water flow events if extreme rainfall occurs during periods of high temperature. Here, we investigated how temperature affected the swimming performance, aerobic metabolism, and skeletal muscle phenotype of Amargosa River pupfish (Cyprinodon nevadensis amargosae) to explore how warmer temperatures may reduce the

Fish Models of Ageing Retinal Disease.

Age-related macular degeneration (AMD) is a common cause of vision loss with few effective treatments available to prevent retinal degeneration long-term. Challenges associated with treatment development are due in part to a lack of understanding of the mechanisms underlying disease onset. However, diseases of ageing are challenging to model. Here, features of diurnal fish models that are beneficial for studying ageing retinal diseases are highlighted, and the unique characteristics of ageing zebrafish, turquoise killifish, and medaka retinas are summarised.

Mercury isotope study of sources and exposure pathways of methylmercury in estuarine food webs in the Northeastern U.S.

We measured mercury (Hg) isotope ratios in sediments and various estuarine organisms (green crab, blue mussel, killifish, eider) to investigate methylmercury (MMHg) sources and exposure pathways in five Northeast coast (U.S.) estuaries. The mass independent Hg isotopic compositions (MIF; Δ(199)Hg) of the sediments were linearly correlated with the sediment 1/Hg concentrations (Δ(199)Hg: r(2) = 0.77, p < 0.05), but the mass dependent isotope compositions (MDF; δ(202)Hg) were not (r(2) = 0.26, p = 0.16), reflecting inputs of anthropogenic Hg sources with varying δ(202)Hg. The estuarine organisms

Nothobranchius as a model for anorexia of aging research: an evolutionary, anatomical, histological, immunohistochemical, and molecular study.

BACKGROUND: Anorexia of aging, defined as a decrease in appetite and a preponderant loss of body weight occurring in late life, is one of the most common diseases affecting older people. The peptide hormone cholecystokinin (Cck) is known to play a key role in regulating food intake and satiety in higher vertebrates. In humans as well as in rats, an increased concentration of Cck was described as the basis of appetite loss in elderly. However, the role of increased plasma Cck concentrations in mediating the age-related decrease in appetite remains to be established. Although in vitro studies ar

Effects of methylmercury on the early life stages of an estuarine forage fish using two different dietary sources.

Marine fish accumulate methylmercury (MeHg) to elevated concentrations, often higher than in freshwater systems. As a neurotoxic compound, high MeHg tissue concentrations could affect fish behavior which in turn could affect their populations. We examined the sublethal effects of MeHg on larvae of the Sheepshead minnow (Cyprinodon variegatus), an estuarine fish, using artificial or natural diets with varying MeHg concentrations (0-4.8 ppm). Larvae were fed control and MeHg-contaminated diets at low or normal (10% of their body mass) daily food rations from 7 to 29 days when they reached juveni

An mTOR-Tfeb-Fabp7a Axis Ameliorates bag3 Cardiomyopathy via Decelerating Cardiac Aging.

While BAG3 has been identified as a causative gene for dilated cardiomyopathy, the major pathological events in BAG3-related cardiomyopathy that could be targeted for therapeutic benefit remain to be discovered. Here, we aim to uncover novel pathological events through genetic studies in a zebrafish bag3 cardiomyopathy model. Given the known cardioprotective effects of mtor inhibition and the fact that transcription factor EB (tfeb) encodes a direct downstream phosphorylation target of mTOR signaling, we generated a cardiomyocyte-specific transgenic line overexpressing tfeb (Tg[cmlc2:tfeb]). The Utility of Small Fishes for the Genetic Study of Human Age-Related Disorders.

Animal models have been used to model human diseases, and among them, small fishes have been highlighted for their usefulness in various ways, such as the low cost of maintenance, ease of genetic modification, small size for easy handling, and strength in imaging studies due to their relative transparency. Recently, the use of turquoise killifish, Nothobranchius furzeri, which is known to exhibit various aging phenotypes in a short period, has attracted attention in research on aging and age-related diseases. However, when using animal models, it is important to keep their genetic background a

Lifelong behavioral screen reveals an architecture of vertebrate aging.

Mapping behavior of individual vertebrate animals across lifespan could provide an unprecedented view into the lifelong process of aging. We created a platform for high-resolution continuous behavioral tracking of the African killifish across natural lifespan from adolescence to death. We found that animals follow distinct individual aging trajectories. The behaviors of long-lived animals differed markedly from those of short-lived animals, even relatively early in life, and were linked to organ-specific transcriptomic shifts. Machine-learning models accurately inferred age and even forecasted
