Paracellular entry of interleukin-10 producing Lactococcus lactis in inflamed intestinal mucosa in mice.

BACKGROUND: Genetically modified Lactococcus lactis secreting interleukin-10 (IL-10) has been demonstrated to provide localized delivery of a therapeutic agent through active in situ synthesis in murine colitis. At present, many aspects of the exact mechanism by which the beneficial effect of the IL-10-producing L. lactis on the mucosa is mediated remain to be clarified. METHODS: Our aim was to determine the interaction of L. lactis with the intestinal mucosa. Therefore, we administered IL-10-producing L. lactis to healthy mice and in 2 mouse models of chronic colitis. Paraffin sections of il

Mucosal delivery of a pneumococcal vaccine using Lactococcus lactis affords protection against respiratory infection.

BACKGROUND: Economical and effective vaccines against Streptococcus pneumoniae (pneumococcus) are needed for implementation in poorer countries where the disease burden is highest. Here, we evaluated Lactococcus lactis intracellularly producing the pneumococcal surface protein A (PspA) as a mucosal vaccine in conferring protection against pneumococcal disease. METHODS: Mice were intranasally (inl) immunized with the lactococcal vaccine. Control groups were also immunized with similar amounts of recombinant PspA administered inl or subcutaneously with alum. PspA-specific antibodies in serum sa

Impact of a GABA-Producing Lactococcus lactis on Microbiota and Mycobiota During CNS Inflammatory Demyelination.

Gut microbes are key regulators of immune homeostasis. Their composition fluctuates over time and between individuals and is also influenced by disease. We and others have reported changes in gut bacterial composition following induction of experimental autoimmune encephalomyelitis (EAE), a well-established model for multiple sclerosis (MS). Specifically, we observed reductions in the abundance of bacteria capable of producing gamma-aminobutyric acid (GABA). Because GABA regulates immune cell function, we genetically engineered a Lactococcus lactis strain to overproduce GABA (P8s-GAD L. lactis

Recombination of miR-146b by Lactococcus lactis for Remolding Macrophages and the Microbiome in the Treatment of Murine Colitis.

MicroRNA 146b (miR-146b) mitigates the progression of inflammatory bowel disease (IBD) by reprogramming macrophage polarization through the induction of interleukin-10 (IL-10), thereby eliciting an anti-inflammatory response, but its application is currently hindered by the lack of safe delivery systems that allow sustained miR-146b expression. Inspired by the probiotic Lactococcus lactis, which can produce extracellular vesicles (EVs) that carry a variety of biomolecules, we successfully constructed a genetically modified L. lactis strain that expresses miR-146b (LL-miR-146b) in a nisin-depen

Lactococcus lactis endocarditis and liver abscess in an immunocompetent patient: a case report and review of the literature.

BACKGROUND: Over the last two decades, several cases of infections caused by Lactococcus lactis have been reported. This Gram-positive coccus is considered non-pathogenic for humans. However, in some rare cases, it can cause serious infections such as endocarditis, peritonitis, and intra-abdominal infections. CASE PRESENTATION: A 56-year-old Moroccan patient was admitted to the hospital because of diffuse abdominal pain and fever. The patient had no past medical history. Five days before his admission, he developed abdominal pain in the right lower quadrant along with chills and feverish sens

Unraveling the role of surface mucus-binding protein and pili in muco-adhesion of Lactococcus lactis.

Adhesion of bacteria to mucus may favor their persistence within the gut and their beneficial effects to the host. Interactions between pig gastric mucin (PGM) and a natural isolate of Lactococcus lactis (TIL448) were measured at the single-cell scale and under static conditions, using atomic force microscopy (AFM). In parallel, these interactions were monitored at the bacterial population level and under shear flow. AFM experiments with a L. lactis cell-probe and a PGM-coated surface revealed a high proportion of specific adhesive events (60%) and a low level of non-adhesive ones (2%). The st

Cryo-EM structure of bacteriophage Nocturne116: insights into the architecture of c2-like Lactococcus viruses.

UNLABELLED: Nocturne116 is a small bacterial virus with a prolate head and a flexible, non-contractile tail that infects an insect-associated strain of Lactococcus lactis. Nocturne116 has little sequence similarity to other bacteriophages and represents a diverged virus lineage that includes the Lactococcus phage Q54 and phages of the Ceduovirus genus. To better understand the molecular architecture of this group of viruses, we determined the three-dimensional structure of the Nocturne116 virion. The head of the Nocturne116 phage is composed of two types of capsid proteins: the major capsid pr

Is Lactococcus lactis a Suitable Candidate for Use as a Vaccine Delivery System Against Helicobacter pylori?

Helicobacter pylori was described in 1979. This bacterium, which thrives in the harsh conditions of the stomach, is typically acquired during childhood and can remain colonized for life. Approximately, 90% of the global population is affected, and H. pylori is linked to various conditions, including gastritis, peptic ulcers, lymphoproliferative gastric lymphoma, and even gastric cancer. Currently, antibiotics are the primary treatment method, but the associated challenges of antibiotic use have led to the consideration of oral vaccination as a viable preventive measure against this infection.

Formation of intracellular vesicles within the Gram+ Lactococcus lactis induced by the overexpression of Caveolin-1β.

BACKGROUND: Caveolae are invaginated plasma membrane domains of 50-100 nm in diameter involved in many important physiological functions in eukaryotic cells. They are composed of different proteins, including the membrane-embedded caveolins and the peripheric cavins. Caveolin-1 has already been expressed in various expression systems (E. coli, insect cells, Toxoplasma gondii, cell-free system), generating intracellular caveolin-enriched vesicles in E. coli, insect cells and T. gondii. These systems helped to understand the protein insertion within the membrane and its oligomerization. There is

Enhancing bile tolerance improves survival and persistence of Bifidobacterium and Lactococcus in the murine gastrointestinal tract.

BACKGROUND: The majority of commensal gastrointestinal bacteria used as probiotics are highly adapted to the specialised environment of the large bowel. However, unlike pathogenic bacteria; they are often inadequately equipped to endure the physicochemical stresses of gastrointestinal (GI) delivery in the host. Herein we outline a patho-biotechnology strategy to improve gastric delivery and host adaptation of a probiotic strain Bifidobacterium breve UCC2003 and the generally regarded as safe (GRAS) organism Lactococcus lactis NZ9000. RESULTS: In vitro bile tolerance of both strains was signif

Previous administration of recombinant or wild type Lactococcus lactis NCDO2118 prevented severe mucositis in mice.

Mucositis is an inflammatory ulcerative condition that affects the mucous membranes of the entire gastrointestinal tract caused by anti-cancer chemotherapy. In the search for strategies to prevent or control this condition, many probiotic bacteria have been described as promising therapeutic tools. However, giving live bacteria during gut inflammatory conditions is a clinical concern and preventive treatments are the most suitable design namely when the disease onset is known as in the case of chemotherapy-induced mucositis. In the present study, we tested the effects of Lactococcus lactis NCDO

Lactococcus garvieae prosthetic aortic and mitral valve endocarditis with multiple embolic complications: a case report.

Lactococcus garvieae is a facultatively anaerobic Gram-positive coccus which causes lactococcosis, a septicaemic illness in fish of major aquacultural significance. This pathogen has emerged in recent years as a cause of zoonotic infections including infective endocarditis, primary bacteraemia, peritonitis and orthopaedic infections, with putative risk factors such as raw seafood ingestion and underlying gastrointestinal disease. We report a case of L. garvieae bioprosthetic aortic and mitral valve endocarditis in a 75-year-old man, complicated by multiple cerebral septic emboli, lumbar discit

Butyrate-producing Lactococcus lactis-fermented goat cheese enhances bone health and reduces osteoporotic bone loss.

Osteoporosis has emerged as a major public health concern, exacerbated by a global aging population. Although pharmaceutical agents, dairy products, and probiotics have shown potential in managing osteoporosis, safer and more effective therapeutic options remain limited. In this study, Lactococcus lactis 3-1 was identified as a bone health-promoting strain, exhibiting high butyrate production, upregulation of osteogenic genes (RUNX2, BGLAP, and ALP), and enhancement of pre-osteoblast MC3T3-E1 differentiation and calcium nodule formation. Moreover, L. lactis 3-1 demonstrated cheese-making poten

Lactococcus lactis blood products contamination resulting in fatal human case: insights from a forensic case.

Lactococcus species are micro-aerophilic Gram positive bacteria characterized by low virulence features and other biotechnological properties of industrial interest. They are thus widely employed in food fermentation processes. Despite its low pathogenic potential and food grade safety, L. lactis may, however, rarely cause infections, especially among immunocompromised hosts. Moreover, the growing complexity of patients implies increased detections of such infections. This said, there is a paucity of data concerning L. lactis infections from infusion of blood transfusion products. To our knowl
