High-level resistance to oxidative stress in Lactococcus lactis conferred by Bacillus subtilis catalase KatE.

Lactococcus lactis, a lactic acid bacterium widely used for food fermentations, is often exposed to damaging stress conditions. In particular, oxidative stress leads to DNA, protein and membrane damages that can be lethal. As L. lactis has no catalase, the impact of production of the Bacillus subtilis haem catalase KatE on its oxidative stress resistance was tested. This cytoplasmic catalase was engineered for extracellular expression in L. lactis with an optimization strategy based on fusion to the nisin-inducible promoter and a lactococcal signal peptide (SP(Usp45)). The production of KatE b

Production of Brucella melitensis Omp16 protein fused to the human interleukin 2 in Lactococcus lactis MG1363 toward developing a Lactococcus-based vaccine against brucellosis.

The use of the food-grade bacterium Lactococcus lactis as a new cell factory is a promising alternative expression system for producing a desired protein. The Omp16-IL2 fusion protein antigen was cloned, expressed, and purified in this study. The Omp16-IL2 fusion gene was designed and cloned in pGH plasmid with appropriate restriction sites and subcloned in pAMJ2008 expression vector digested with the same enzymes. The purified recombinant constructed pAMJ-rOmp-IL2 was introduced into L. lactis subsp. cremoris MG1363 by electrotransformation. Finally, the expression and purification of Omp16-I

A novel "laccase-degradation and GABA-compensation" strategy mediated by Lactococcus lactis subsp. lactis GLM-1 for the safe and targeted detoxification of ginkgo seeds.

Ginkgo biloba L. seeds are valued for their bioactive compounds but contain the neurotoxin 4'-O-methylpyridoxine (MPN), limiting their safe use. This study developed a novel "laccase-degradation and GABA-compensation" strategy using a food-grade bacterium, Lactococcus lactis subsp. lactis GLM-1, to achieve detoxification with bioactive preservation. The study initially confirmed that while free laccase effectively degrades MPN, it nonspecifically oxidizes total flavonoids and terpene lactones. Furthermore, using a model Caenorhabditis elegans, it was elucidated that MPN impedes GABA production

A comparative analysis of signal peptide signatures in the bacterial cell factories Bacillus subtilis and Lactococcus lactis.

Secreted recombinant proteins are of great significance for industry, healthcare and a sustainable bio-based economy. Due to their relatively simple cell envelope structure and effective protein secretion machinery, monoderm Gram-positive bacteria, such as Bacillus subtilis and Lactococcus lactis, are attractive cell factories for the secretory production of recombinant proteins of interest. Equally important for recombinant protein production as the secretion machinery are effective secretion signals, which direct proteins from the cytoplasm across the cytoplasmic membrane and cell wall into

The chemotaxis disruption of Aeromonas veronii by Lactococcus lactis and its probiotic effect on loach (Misgurnus anguillicaudatus).

Aeromonas veronii is a pathogen that is widely found in aquaculture environments. It has been observed to colonize the gastrointestinal tract of fish and other aquatic animals, with the potential to cause severe infections. Previous studies have demonstrated that probiotics, such as Lactococcus lactis 01, Lactococcus lactis 02, Enterococcus pseudoavium 03, and Bacillus sp. 04, can reduce the colonization capability of A. veronii. In this study, we sought to ascertain whether these four probiotics could affect chemotaxis by interfering with N-acyl-homoserine lactones (AHLs), thereby regulating

The irpT gene influences the multiple stresses tolerance by affecting cytochrome c in Lactococcus lactis N8.

Lactococcus lactis, an internationally recognized safe species of Gram-positive bacteria, produces a variety of antimicrobial peptides. In this study, we examined the irpT gene of L. lactis N8 and its influence on the bacterial stress response. We used the Cre-loxP system to knock out the irpT gene from the L. lactis N8Δply10300 genome. The mutant strains L. lactis N8Δply10300 and L. lactis N8Δply10300ΔirpT showed higher nisin tolerance and nisin production than the wild-type strain L. lactis N8, and also obtained tolerance for multiple stressors. The biological characteristics of the irpT gen

Intra-bone marrow injection with engineered Lactococcus lactis for the treatment of metastatic tumors: Primary report.

Bone marrow has the capacity to produce different types of immune cells, such as natural killer cells, macrophages, dendritic cells (DCs) and T cells. Improving the activation of immune cells in the bone marrow can enhance the therapy of bone metastases. Previously, we designed an engineered probiotic Lactococcus lactis, capable of expressing a fusion protein of Fms-like tyrosine kinase 3 ligand and co-stimulator OX40 ligand (FOLactis), and proved that it can induce the activation and differentiation of several immune cells. In this research, we successfully establish mouse models of bone meta

Modulating effects of the probiotic Lactococcus lactis on the hepatic fibrotic process induced by CCl4 in Wistar rats.

Hepatic cirrhosis is a chronic disease that affects one fifth of the World's population and is the third leading cause of death in Mexico. Attempts have been made to develop treatments for this hepatic cirrhosis, which include manipulating the intestinal microbiota and thus decreasing the early inflammatory response. The microbiota is reportedly altered in patients with cirrhosis. Due to its immunomodulatory properties and its ability to survive in the gastrointestinal tract, Lactococcus lactis (L. lactis) has been used as a therapeutic measure in inflammatory disorders of the colon. The objec

Complementation of the Lactococcus lactis secretion machinery with Bacillus subtilis SecDF improves secretion of staphylococcal nuclease.

Unlike Bacillus subtilis and Escherichia coli, the gram-positive lactic acid bacterium Lactococcus lactis does not possess the SecDF protein, a component of the secretion (Sec) machinery involved in late secretion stages and required for the high-capacity protein secretion in B. subtilis. In this study, we complemented the L. lactis Sec machinery with SecDF from B. subtilis and evaluated the effect on the secretion of two forms of staphylococcal nuclease, NucB and NucT, which are efficiently and poorly secreted, respectively. The B. subtilis SecDF-encoding gene was tested in L. lactis at diffe

Ingestion of milk fermented by genetically modified Lactococcus lactis improves the riboflavin status of deficient rats.

Riboflavin deficiency is common in many parts of the world, particularly in developing countries. The use of riboflavin-producing strains in the production of dairy products such as fermented milks, yogurts, and cheeses is feasible and economically attractive because it would decrease the costs involved during conventional vitamin fortification and satisfy consumer demands for healthier foods. The present study was conducted to assess in a rat bioassay the response of administration of milk fermented by modified Lactococcus lactis on the riboflavin status of deficient rats. Rats were fed a rib

Intravenous iRGD-Guided, RBC-Membrane Camouflaged Lactococcus Lactis Remodels Cold NSCLC and Enhances PD-1 Blockade.

Resistance to programmed-death-1/programmed-death-ligand-1 (PD-1/PD-L1) blockade in non-small-cell lung cancer (NSCLC) arises mainly from weak tumor immunogenicity and limited effector T-cell infiltration. Here, this work presents an intravenously deliverable "living medicine" that addresses these barriers through biomimetic cloaking, tumor-penetrating guidance, and synthetic-biology-driven cytokine release. Lactococcus lactis is engineered to co-secrete Flt3L and OX40L (FOLactis) and then camouflage with red-blood-cell membranes, producing long-circulating mRBC@FOLactis. Conjugation of the iR

Valorisation of acid whey permeate for high-purity nisin Z production using artisanal Lactococcus lactis isolates.

The growing demand for sustainable bioprocessing has positioned whey, a side stream of the dairy industry, as a valuable substrate for producing high-value compounds. This study investigated the production and purification of nisin, a potent lantibiotic, using permeate (10 kDa cut-off) of ultrafiltered acid whey (PUFAW) devoid of lactoferrin as growth medium. Three strains of Lactococcus lactis subsp. lactis, including two artisanal isolates (IM 143 and IM 145) and a reference strain (IM 407), were evaluated for nisin production in PUFAW supplemented with yeast extract (YE) and mineral salts.

Efficacy of a Lactococcus lactis ΔpyrG vaccine delivery platform expressing chromosomally integrated hly from Listeria monocytogenes.

Listeria monocytogenes is a significant food-borne pathogen and the causative agent of listeriosis, a disease which manifests as meningitis in immunocompromised adults or infection of the fetus and miscarriage in pregnant women. We have previously used Lactococcus lactis, a GRAS (Generally Regarded As Safe) organism, as a vaccine vector against listeriosis by engineering plasmid-mediated expression of the immunodominant antigen from L. monocytogenes, listeriolysin O (LLO). However, the environmental release of an engineered vaccine vector carrying a replicating plasmid during clinical usage ma

A Lactococcus lactis Starter Culture Reduces the Bloater Defect in Cucumber Fermentations Brined With Low Salt.

The cucumber intrinsic Enterobacter and Leuconostoc generate sufficient carbon dioxide (CO2) in early fermentation, particularly those brined without sodium chloride, to induce bloater defects prior to the onset of Lactiplantibacillus pentosus. Because of the Lactococcus lactis short doubling time, it is proposed as a starter culture for cucumber fermentation brined with 342 mM (2%) sodium chloride to prevent bloater defects. A screening of putative decarboxylases (27) in the cucumber fermentation bacteriome identified substantially more of such enzymes in Enterobacteriaceae (24 or 89%) relati

Development of interleukin-27 recombinant Lactococcus lactis and its efficacy in treating psoriasis and colitis in mice.

Psoriasis and inflammatory bowel disease (IBD) are chronic immune-mediated diseases that adversely affect patients' quality of life. Interleukin (IL)-27 plays an important role in a variety of infectious diseases, autoimmune disorders, and cancers. However, its therapeutic effects in psoriasis and colitis remain underexplored. In this study, we evaluated the therapeutic potential of recombinant Lactococcus lactis (L. lactis) expressing IL-27 (pIL-27) in imiquimod-induced psoriasis and dextran sodium sulfate-induced colitis mouse models. Suppressive Effect of Lactococcus lactis subsp. cremoris YRC3780 on a Murine Model of Japanese Cedar Pollinosis.

Accumulating evidence suggests that Lactococcus lactis subsp. cremoris YRC3780 isolated from kefir has the potential to alleviate allergic responses. Herein, we investigated the effect of YRC3780 on a murine model of Japanese cedar pollinosis (JCP). BALB/c mice immunized with cedar pollen extract (CPE) exhibited an increase in serum immunoglobulin E and developed nasal inflammatory responses including sneezing, nasal hyperresponsiveness, and nasal eosinophil accumulation upon intranasal allergen challenge. These responses were suppressed by the oral administration of YRC3780, although the effe

Immunopathological evaluation of recombinant mycobacterial antigen Hsp65 expressed in Lactococcus lactis as a novel vaccine candidate.

Bovine tuberculosis (TBB) is a zoonotic disease distributed worldwide and is of great importance for public health and the livestock industry. Several experimental vaccines against this disease have been evaluated in recent years, yielding varying results. An example is the Bacillus Calmette-Guérin (BCG) vaccine, which has been used extensively in humans and tested in cattle showing mixed results related to protection (0-80%) against Mycobacterium bovis. In this study, we used the food-grade bacterium Lactococcus lactis as an expression system for production of mycobacterial protein Hsp65. For
