Lamotrigine is a clinically used drug, which inhibits Na(+) channel activity that in turn reduces glutamate release. Its downstream activity on other neurotransmitter systems such as serotonergic and dopaminergic has also been reported. Since both glutamate and monoamines might be involved in alcohol craving and relapse, our aim was to examine the effects of lamotrigine on alcohol seeking and relapse-like drinking behaviour. Thus, lamotrigine (5 and 15mg/kg) was tested in two behavioural models for alcohol seeking and relapse - the model of cue-induced reinstatement of alcohol-seeking behaviou

Rett syndrome (RS) is a particular neurodevelopmental disorder in which symptoms develop in early infancy and manifests particular symptoms at certain ages. Two girls aged 4.5 and 2.5-years-old were admitted with complaints of seizures, regression of language, psychomotor development, and autistic behavior. They had fulfilled the criteria of classic RS. Lamotrigine was used in the treatment of the convulsions. After lamotrigine therapy, the convulsions were successfully controlled and the stereotyped hand movements and autistic behaviors markedly decreased. We would like to stress that, aside

PURPOSE: Lamotrigine (LTG), a newer antiepileptic drug (AED), has activity against both partial-onset and generalized seizures. Its reported benefits for behavior, and its effectiveness in Lennox-Gastaut syndrome and other forms of refractory epilepsy, make it a logical choice for treatment of epilepsy in tuberous sclerosis complex (TSC). We present our experience with LTG therapy of epilepsy in 57 patients with TSC.
METHODS: Patients fulfilled the diagnostic criteria for clinically definite TSC. LTG was initiated and increased until improvement in seizure frequency was noted, intolerable sid

PURPOSE: To examine the safety of lamotrigine (LTG) used in general practice to treat epilepsy.
METHODS: Information was collected on 11,316 patients who were included in a noninterventional observational cohort study conducted by means of Prescription-Event Monitoring (PEM). A follow-up study provided information on the first 3,994 patients who had taken LTG for > or = 6 months. Incidence density (ID) measurements were used to rank the frequency of the reported events.
RESULTS: Rash was the most frequently reported nonepileptiform event (ID, 19.7/1,000 patient-months) in the first month of

Lamotrigine (LTG) was used in 12 patients with juvenile myoclonic epilepsy (JME) who either had sodium valproate (VPA) side effects or did not wish to take VPA. Five patients are now fully controlled on LTG monotherapy. Side-effects were corrected by VPA withdrawal or reduction except for severe weight gain. In three patients it was not possible to withdraw VPA due to the re-emergence of myoclonus, suggesting LTG-VPA synergism. Two patients had successful pregnancies whilst taking LTG. Lamotrigine is a useful alternative in the management of JME.

Product license applications for lamotrigine have been filed or approved in more than 50 countries throughout the world. In preclinical models, this drug demonstrates activity against both maximal electroshock and pentylenetetrazol seizures. The primary action of lamotrigine is, most likely, blockade of sodium channels, with resultant inhibition of glutamate and aspartate release. As would be expected from this spectrum of activity, the drug has shown clinical efficacy in both partial and generalized seizures and in a variety of epilepsy syndromes. Particularly promising results have been demo

Several drugs can unmask clinically silent systemic lupus erythematosus (SLE), induce flares in patients with already known SLE diagnosis or lead to the development of lupus-like syndromes. We describe a rare case of juvenile SLE triggered by lamotrigine. A 17-year-old female, who was recently medicated with lamotrigine, presented with a 3 week history of daily fever, fatigue, odynophagia, arthralgia, myalgia, diffuse erythematous maculopapular rash, vasculitic rash of the fingers, and lips and vulvar edema. Leukopenia, lymphopenia, elevated C-reactive protein, low serum C3 and C4 levels, anti

BACKGROUND: Lamotrigine (LMT) is a phenyltriazine derivative that was originally described as an antiepileptic drug.
OBJECTIVE: This literature review aims to evaluate the clinical epidemiological profile, pathological mechanisms, and management of lamotrigine-associated movement disorders.
METHODS: Relevant reports in six databases were identified and assessed by two reviewers without language restriction. Reports that the individuals only developed tremor or ataxia after LMT use were not included.
RESULTS: In total 48 reports of 108 cases from 19 countries were assessed. The movement diso

BACKGROUND: Although lamotrigine may be useful for treating patients with treatment-resistant bipolar disorder, some lamotrigine-associated adverse effects, including mild to moderate skin rash, may prevent the continuation of treatment.
METHODS: We investigated lamotrigine rechallenge for the treatment of bipolar disorder. The present study was based on retrospective chart review of outpatients with bipolar disorder (DSM-5 criteria) who visited the hospital's psychiatric department between July 2011 and August 2017. The review revealed 12 patients with bipolar disorder who underwent lamotrig

Aseptic meningitis could be a rare side effect of lamotrigine. Aseptic meningitis is a clinical condition where symptoms, signs and laboratory findings are consistent with meningeal inflammation but routine bacterial cultures are negative. Clinical presentation of aseptic meningitis is similar to bacterial meningitis with prominent symptoms of fever, headache and neck stiffness. Drug-induced aseptic meningitis is an uncommon adverse effect of several drugs. The most common ones are nonsteroidal anti-inflammatory drugs, intravenous immunoglobulins, intrathecal agents and vaccines. Disease manif

OBJECTIVE: To describe the laboratory and physical manifestations of lamotrigine toxicity presenting as anticonvulsant hypersensitivity syndrome.
CASE SUMMARY: A 49-year-old white man presented to our institution with a two-day history of low-grade fever, erythema, and edema involving the periorbital area. Five days earlier, he had been placed on lamotrigine treatment for bipolar disorder. He inadvertently received four daily doses of lamotrigine 2700 mg each. Physical examination was significant for periorbital edema and discrete and confluent blanching red macules and papules involving the

PURPOSE: To evaluate the effects of lamotrigine (LTG) therapy on epileptic seizures and general well-being in patients with juvenile neuronal ceroid lipofuscinosis (JNCL).
METHODS: LTG was initiated in 28 patients with JNCL. The mean age of the patients at the initiation of LTG was 13.7 years (range, 6.7-28.2 years). LTG was started at a dosage of 0.1-0.5 mg/kg/day and increased every 2 weeks until a maintenance dose of 1.25-15 mg/kg/day was reached. On the basis of the indication for LTG therapy, the patients could be divided into four groups. In the first group, LTG was initiated on an add-

Lamotrigine toxicity may be associated with a variety of nonspecific clinical findings, and rarely with psychosis. Many previous studies have reported that the use of lamotrigine with other agents, such as valproic acid, or an overdose of lamotrigine precipitated symptoms of lamotrigine toxicity. This case involved a 31-year-old woman who experienced dizziness, emesis, and psychosis after her lamotrigine prescription had recently been increased. These symptoms resolved when lamotrigine was held after the diagnosis of lamotrigine toxicity had been made. To our knowledge, this is a rare document

OBJECTIVE: Clinical management of bipolar disorder (BPD) patients during pregnancy is a major challenge. The high risk of bipolar depression during pregnancy encourages consideration of lamotrigine (LTG). We therefore compared recurrence risks among pregnant women with BPD treated with LTG to those discontinuing mood stabilizer therapies.
METHODS: We compared risks and weeks to new DSM-IV illness-episodes among 26 initially clinically stable pregnant women diagnosed with DSM-IV BPD who continued LTG treatment to those discontinuing all mood stabilizer treatment during pregnancy.
RESULTS: The

Erythema multiforme is an acute, hypersensitivity reaction of the skin often secondary to medications. Lamotrigine is a relatively new anticonvulsant medication approved for seizure and psychiatric disorders. Although the overall incidence of cutaneous reactions to lamotrigine is high, the incidence of serious eruptions such as erythema multiforme, Stevens-Johnson syndrome, and toxic epidermal necrolysis is low. Vigilant surveillance for any cutaneous eruption in patients on lamotrigine is important, particularly in the first 8 weeks, as prompt discontinuation of the medication can prevent pro

A case of toxic epidermal necrolysis following treatment with lamotrigine is presented. A 20 year old male suffering from epilepsy was treated with lamotrigine in addition to valproic acid. After three weeks he developed cutaneous manifestations of Steven-Johnson's syndrome followed by toxic epidermal necrolysis, mucosal lesions and liver symptoms. He was treated with systemic corticosteroids, antibiotics and intravenous fluid, and recovered after a few weeks.
