Cutting wild grapevines as a cultural control strategy for grape berry moth (Lepidoptera: Tortricidae).

A 3-yr field study was conducted at commercial grape farms to evaluate cutting wild grapevines as a cultural control strategy for grape berry moth, Paralobesia viteana (Clemens). At each farm, wild grapevines were cut in the woods adjacent to one vineyard for control of P. viteana, whereas the comparison vineyard received no such cutting. Both vineyards received a standard broad-spectrum insecticide program for control of P. viteana and other vineyard insect pests. Monitoring with pheromone traps showed no differences between treatments in the total number of male moths trapped in both woods a

Cephalic sensory pathways in the central nervous system of larval Manduca sexta (Lepidoptera : Sphingidae).

Central projections of neurons innervating sensory structures on the head of larval Manduca sexta were traced by using methods of anterograde cobalt-diffusion. Regions of the deutocerebrum and tritocerebrum in the brain receive input from the antenna, labrum, maxilla, labial palps, hypopharynx and other unidentified regions of the buccal cavity. Antennal, maxillary and labial inputs project to the larval antennal centre (LAC) of the deutocerebrum. Stemmatal neurons and a few antennal neurons project into the protocerebrum. The suboesophageal ganglion (SEG) receives input from mechanosensory ne

Gene Expression of GqBursicon in Gynaephora qinghaiensis (Lepidoptera: Lymantriidae) and Its Impact on Wing Expansion.

The tanning hormone (Bursicon) is a heterodimer secreted by the insect nervous system, playing a crucial role in the process of cuticle darkening and hardening and wing expansion in insects. In this study, we knocked out the GqBurs gene using RNAi, thereby impairing the flight ability of adult males by affecting wing development, laying the groundwork for further control of the damage they cause to alpine meadows. The GqBurs gene was successfully characterized and expressed at all developmental stages of Gynaephora qinghaiensis, with higher expression in 5th instar larvae and wing primordia. I

Selection and Validation of Stable Reference Genes for RT-qPCR in Scotogramma trifolii (Lepidoptera: Noctuidae).

The clover cutworm, Scotogramma trifolii Rottemberg (Lepidoptera: Noctuidae), is a globally distributed polyphagous pest causing significant economic losses to agricultural crops. RT-qPCR is a gold-standard technique for gene expression analysis, yet its accuracy depends critically on stable reference genes for data normalization. To address the lack of validated reference genes in S. trifolii, we evaluated six candidate genes (β-actin, RPL9, GAPDH, RPL10, EF1-α, and TUB) across four developmental stages (egg, larva, pupa, and adult) and six adult tissues (head, thorax, abdomen, wings, legs, a

Field Evaluation of a Binary Sex Pheromone for Sweetpotato Vine Borer (Lepidoptera: Crambidae) in Hawaii.

The sweetpotato vine borer, Omphisa anastomosalis (Guenée), is a primarily Asian pest of sweetpotato, Ipomoea batatas L. Damage by O. anastomosalis infestation can cause root yield losses of 30%-50%. A binary sex pheromone for O. anastomosalis, consisting of Type I [(10E,14E)-10,14-hexadecadienal (E10,E14-16:Ald)] and Type II [(3Z,6Z,9Z)-3,6,9-tricosatriene (Z3,Z6,Z9-23:H)] components, was identified in Vietnam from extracts of female pheromone glands. A structurally similar Type II compound [(3Z,6Z,9Z)-3,6,9-docosatriene (Z3,Z6,Z9-22:H)], not recovered from female pheromone glands, was also f

Baseline Susceptibility of Helicoverpa armigera (Lepidoptera: Noctuidae) to Indoxacarb, Emamectin Benzoate, and Chlorantraniliprole in Australia.

Baseline susceptibility of Helicoverpa armigera (Hübner) to emamectin benzoate, chlorantraniliprole, and indoxacarb was determined in feeding assays on insecticide-incorporated artificial diet in the laboratory. The intraspecific variation of H. armigera was established from field populations collected between September 2012 and March 2013, primarily from commercial farms across eastern Australia. Emamectin benzoate had the highest toxicity with a median lethal concentration (LC50) of 0.01 µg/ml diet (n=20 strains). The LC50 for chlorantraniliprole was 0.03 µg/ml diet (n=21 strains), while ind

Eyes of male and female Orgyia antiqua (Lepidoptera; Lymantriidae) react differently to an exposure with UV-A.

The structural organization of the eyes belonging to 12 winged male and 12 wingless female Orgyia antiqua moths, exposed for 1 h to UV-radiation (lambda(max)=351 nm) of 1.4 kW/m2, was compared with that of 12 male and 12 female non-irradiated control specimens. Following the UV-exposure, the screening pigments were found in a position indicative of extreme light-adaptation. Extensive formations of vesicles along the perimeter of the cones as well as disintegrating ER in the cone cytoplasm were noticeable, especially in the eye of the female. On the retinal side of the clearzone, the microvilli

Sentinel monitoring for resistance to Bt toxins in European corn borer (Lepidoptera: Crambidae) in Canada.

Transgenic corn Zea mays (L.) producing Bacillus thuringiensis insecticidal toxins (Bt corn) has successfully controlled the European corn borer Ostrinia nubilalis (Lepidoptera: Crambidae), one of the most destructive corn pests in North America, for over 25 years. However, field-evolved resistance to Cry1Fa toxin was first documented in Nova Scotia, Canada, in 2018, with subsequent detections in Quebec and Manitoba. Before these discoveries, no resistance had been reported in O. nubilalis populations to Bt toxins, despite ongoing resistance monitoring mandated by regulatory agencies. Sentinel

Composition and Diversity Characteristics of Gut Microbiota during the Development of Telchinia issoria (Lepidoptera: Nymphalidae).

Ramie (Boehmeria nivea) was a traditional economic crop of high commercial value, whose cultivation was threatened by the leaf-feeding pest Telchinia issoria. This study investigated how the gut microbiota of T. issoria shifted across its larval, pupal, and adult stages using 16S rRNA amplicon sequencing. We found that Pseudomonadota and Bacillota dominated across all stages, with stage-specific enrichments of key genera: Burkholderia-Caballeronia-Paraburkholderia in early larvae, Acinetobacter and Culicoidibacter in mid-instars, Serratia in late larvae, Enterococcus in pupae, and Pseudomonas

Influence of metal ions on the economic parameters of mulberry silkworm Bombyx mori (Lepidoptera: Bombycidae).

Micronutrients contribute significantly to homeostasis at the cellular, physiological, and biological interface of silkworm growth and development. The silkworm cocoon traits are highly physiologically dependent, controlled by the optimum availability of micronutrient concentrations. Keeping this in view, two micronutrients, i.e., copper and iron, were investigated to understand their role in economic traits in popular bivoltine silkworm double-hybrid FC1 × FC2. The mulberry leaves were bio-fortified with copper and iron in the form of copper chloride and ferric chloride with concentrations of

Effects of Dietary Iron Quantities and Sources on Lymantria dispar (Lepidoptera: Erebidae) Survival and Development.

Lymantria dispar L. is a serious invasive defoliator of broadleaf trees in both North America and Eurasia. When rearing L. dispar for research, the artificial diet must have a source of iron that is bioavailable and in the quantity needed to ensure normal development and survival. The standard iron supplement in L. dispar diet, amorphous ferric phosphate, is no longer commercially available, which motivated studies on alternative iron sources and concentrations. The responses of three L. dispar populations (including an L. dispar asiatica Vnukovskij population) to seven different iron doses we

Sublethal and transgenerational effects of spinetoram on the biological traits of Plutella xylostella (L.) (Lepidoptera: Plutellidae).

Spinetoram, a spinosyn insecticide is used to manage lepidopteran pests, including diamondback moth, Plutella xylostella. In addition to determining the lethal effects, identifying low and/or sublethal effects of an insecticide is crucial to understanding the total impact of an insecticide. We assessed the low lethal and sublethal effects of spinetoram on two successive generations of P. xylostella. The initial bioassay results showed that spinetoram exhibited high toxicity against P. xylostella with an LC50 of 0.114 mg L-1 after 48 h exposure. The low lethal (LC25) and sublethal (LC10) concen

Efficacy of a cry1Ab Gene for Control of Maruca vitrata (Lepidoptera: Crambidae) in Cowpea (Fabales: Fabaceae).

Cowpea [Vigna unguiculata (L) Walp.] is an important staple legume in the diet of many households in sub-Saharan Africa. Its production, however, is negatively impacted by many insect pests including bean pod borer, Maruca vitrata F., which can cause 20-80% yield loss. Several genetically engineered cowpea events that contain a cry1Ab gene from Bacillus thuringiensis (Bt) for resistance against M. vitrata were evaluated in Nigeria, Burkina Faso, and Ghana (West Africa), where cowpea is commonly grown. As part of the regulatory safety package, these efficacy data were developed and evaluated by

Identification of the sex pheromone of the tree infesting Cossid Moth Coryphodema tristis (Lepidoptera: Cossidae).

The cossid moth (Coryphodema tristis) has a broad range of native tree hosts in South Africa. The moth recently moved into non-native Eucalyptus plantations in South Africa, on which it now causes significant damage. Here we investigate the chemicals involved in pheromone communication between the sexes of this moth in order to better understand its ecology, and with a view to potentially develop management tools for it. In particular, we characterize female gland extracts and headspace samples through coupled gas chromatography electro-antennographic detection (GC-EAD) and two dimensional gas

Flavin-dependent monooxygenases as a detoxification mechanism in insects: new insights from the arctiids (lepidoptera).

Insects experience a wide array of chemical pressures from plant allelochemicals and pesticides and have developed several effective counterstrategies to cope with such toxins. Among these, cytochrome P450 monooxygenases are crucial in plant-insect interactions. Flavin-dependent monooxygenases (FMOs) seem not to play a central role in xenobiotic detoxification in insects, in contrast to mammals. However, the previously identified senecionine N-oxygenase of the arctiid moth Tyria jacobaeae (Lepidoptera) indicates that FMOs have been recruited during the adaptation of this insect to plants that

A megalin-like receptor is involved in protein endocytosis in the midgut of an insect (Bombyx mori, Lepidoptera).

The effect of endotoxemia (lipopolysaccharide, 2.5 mg/kg ip) was investigated in aquaporin (AQP) 1 knockout (KO) compared with wild-type (WT) mice. At baseline, KO mice exhibited higher water intake (WI) and urine output (UO). After endotoxemia, WI and UO remained higher in the KO than WT mice, and urine osmolality was lower. The higher serum osmolality in AQP1-KO mice during endotoxemia was associated with higher AQP2 (133 +/- 8 vs. 100 +/- 3%, P < 0.01), AQP3 (140 +/- 8 vs. 100 +/- 4%, P < 0.001) and Na(+)-K(+)-2Cl(-) cotransporter type 2 (NKCC2; 152 +/- 14 vs. 100 +/- 15%, P < 0.05) express

Use of nucleotide sequence data to identify a microsporidian pathogen of Pieris rapae (Lepidoptera, Pieridae).

Nucleotide sequence was determined for a portion of genomic DNA which spans the V4 variable region of the small subunit ribosomal RNA gene of an unidentified microsporidium from the cabbage white butterfly, Pieris rapae (174 base pairs). Comparison with equivalent sequence data obtained here for two other microsporidian species, Nosema bombycis (240 base pairs) and Nosema bombi (200 base pairs), and from the GenBank database for 11 other microsporidian species suggests that the unidentified species from P. rapae is most closely related to some Vairimorpha species. High-temperature acclimation during development modulates efficacy of emamectin benzoate against Mythimna separata (Lepidoptera: Noctuidae).

Global temperature increases require the reevaluation of pesticide efficacy under thermal stress conditions; however, most studies have focused on acute high-temperature exposure, neglecting the effects of natural diurnal temperature fluctuations and cumulative acclimation. This study investigated the effect of thermal acclimation during developmental on the sensitivity of Mythimna separata (Walker) (Lepidoptera: Noctuidae) to emamectin benzoate (EB). Larvae were reared under cyclic thermal regimes (25→34/38→25 °C), experiencing daily high-temperature exposures of 2, 4, or 6 hours from the egg
