Host-Directed Discovery of Dynamic-Covalent Guests From Dynamic Combinatorial Libraries.

Dynamic covalent chemistry and host-guest recognition are complementary tools for constructing adaptive molecular systems and functional materials, yet their integration has been hindered by the lack of general strategies for efficiently discovering host-compatible dynamic-covalent guests. Here we report a host-directed screening strategy that directly identifies optimal imine guests from dynamic combinatorial libraries. Upon addition of the macrocyclic host, selective host-guest binding drives thermodynamically controlled library redistribution, leading to significant constitutional amplifica

Genome-Wide Libraries for Protozoan Pathogen Drug Target Screening Using Yeast Surface Display.

The lack of genetic tools to manipulate protozoan pathogens has limited the use of genome-wide approaches to identify drug or vaccine targets and understand these organisms' biology. We have developed an efficient method to construct genome-wide libraries for yeast surface display (YSD) and developed a YSD fitness screen (YSD-FS) to identify drug targets. We show the efficacy of our method by generating genome-wide libraries for Trypanosoma brucei, Trypanosoma cruzi, and Giardia lamblia parasites. Each library has a diversity of ∼105 to 106 clones, representing ∼6- to 30-fold of the parasite's

Genome-wide sequence transposon insertion sites and analyze the essential genes of Brucella melitensis.

A transposon mutant library of B. melitensis NI including 32,640 transposon mutants was established. By sequencing the transposon insertion sites, 10,832 mutants were successfully defined for their insertion sites. Analysis of the mutants with defined transposon insertion sites (DTIS) indicated that the insertions were well spread through the two genomes. In addition, 948 genes with no detectable transposon insertions were taken as the candidate for identification of essential genes. In comparison with the Bacterial Database of Essential Genes and by using comparative genomics analysis, 183 po

Folate and global health review series, part 4: syntheses on folate and autoimmune diseases and skeletal outcomes.

BACKGROUND: Autoimmune diseases and bone density loss (osteopenia and osteoporosis) are chronic conditions of complex aetiology that affect diverse populations. Folate may be associated with a higher risk of these disorders due to its essential role in one-carbon metabolism required for nucleotide synthesis, homocysteine metabolism, and methylation processes. However, the evidence on this association is inconclusive. METHODS: We searched MEDLINE, Embase, CINAHL, the Cochrane Library, and the Database of Abstracts of Reviews of Effects from inception to February 2024 for systematic reviews and

Improving the management of Inherited Retinal Dystrophies by targeted sequencing of a population-specific gene panel.

Next-generation sequencing (NGS) has overcome important limitations to the molecular diagnosis of Inherited Retinal Dystrophies (IRD) such as the high clinical and genetic heterogeneity and the overlapping phenotypes. The purpose of this study was the identification of the genetic defect in 32 Spanish families with different forms of IRD. With that aim, we implemented a custom NGS panel comprising 64 IRD-associated genes in our population, and three disease-associated intronic regions. A total of 37 pathogenic mutations (14 novels) were found in 73% of IRD patients ranging from 50% for autosom

Multifactorial determinants of visual outcome in very and extremely preterm infants: A systematic literature review of risk factors for visual and oculomotor dysfunctions.

Preterm birth is often associated with neurological, neurodevelopmental and visual impairment arising from the complex interplay of ocular, visual pathway, and cerebral vulnerability in the immature infant. Visual dysfunction in premature infants may reflect ocular, anterior visual pathways, retrogeniculate pathways and visual associative areas abnormalities, as seen in Cerebral Visual Impairment. This systematic review synthesized evidence on the association between pre-, peri-, i and postnatal factors on basic visual (visual acuity, contrast sensitivity, visual field) and oculomotor (fixatio
