Robust DNA Cross-Linked Polymeric Lighting-Up Nanogel Facilitates Sensitive Live Cell MicroRNA Imaging.

Accurate and specific imaging of low-abundance intracellular microRNAs (miRNAs) is crucial for monitoring cellular processes and disease diagnosis. Despite the fact that nucleic acid amplification technologies have shown great advantages for the detection of trace targets, their live cell imaging applications remain a major challenge because of the insufficient stability and slow reaction kinetics of the probes in cellular delivery and imaging. Herein, we demonstrate the synthesis of DNA-cross-linked polymeric lighting-up nanogels (DPLNs) through the DNA hairpin-based hybridization chain react

Lighting Up Mutation: a New Unbiased System for the Measurement of Microbial Mutation Rates.

Although mutation drives evolution over long and short terms, measuring and comparing mutation rates accurately have been particularly difficult. This is especially true when mutations lead to an alteration in fitness. E. Shor, J. Schuyler, and D. S. Perlin (https://doi.org/10.1128/mBio.00120-19) present a new method to compare mutation rates across fungal strains and under different growth conditions: they employ the green fluorescent protein (GFP) as the reporter and count mutations using fluorescence-activated cell sorting (FACS). The estimates of mutation rates using the GFP-FACS approach

The Role of Exercise-Induced Arousal and Exposure to Blue-Enriched Lighting on Vigilance.

It is currently assumed that exposure to an artificial blue-enriched light enhances human alertness and task performance, but recent research has suggested that behavioral effects are influenced by the basal state of arousal. Here, we tested whether the effect of blue-enriched lighting on vigilance performance depends on participants' arousal level. Twenty-four participants completed four sessions (blue-enriched vs. dim light × low vs. high arousal) at 10 pm on four consecutive days, following a repeated-measures design. Participants' arousal was manipulated parametrically through the executio

Presence of the pineal hormone melatonin in rat cochlea: its variations with lighting conditions.

Within the normal inner ear, there are elements that belong to the immune system. Different inner ear disorders can be explained by autoimmune mechanisms, affecting both humoral and cellular immunity. Melatonin, the principal hormone of the pineal gland, modulates the immune system and extensively participates in the autoimmune processes related to type II collagen. Therefore, we have studied the presence of melatonin in rat cochlea, proving that its concentrations change depending on lighting conditions. Rats show high levels when confined to darkness and low levels when subject to continuous

Blue light, green light, white light, more light: treatment of neonatal jaundice.

This article reviews the current understanding of how phototherapy, the most widely used treatment of neonatal jaundice, uses light energy to pump bilirubin out of a jaundiced infant. It provides a framework for understanding the often conflicting results of clinical comparisons of various phototherapeutic regimens, and stresses the importance of light intensity in the overall effectiveness of phototherapy.

Robotic Arm Trajectory Planning for Tunnel Lighting Cleaning Based on the CAW-PSO Algorithm.

Tunnel lighting cleaning is of significant practical importance for improving driving safety. To address the low operational efficiency of tunnel lighting cleaning tasks, a trajectory planning method based on the chaotic adaptive whale-particle swarm optimization (CAW-PSO) algorithm is proposed. Taking the SIASUN GCR16-2000 robotic arm as the research object, the trajectory is constructed using a 3-5-3 polynomial interpolation, with the objective of achieving time-optimal trajectory planning. In the CAW-PSO algorithm, a tent chaotic map is introduced to improve the quality of the population; a

Lighting-driven behavioral responses in zebrafish for water toxicity monitoring and ecological risk assessment.

Zebrafish exhibit innate phototaxis, making light stimulation a promising tool for water toxicity monitoring. This study systematically investigated the effects of light color (red, yellow, green, blue), intensity (0-2400 lx), and stimulation duration (1-3 min) on zebrafish phototactic and locomotor behaviors, and established an optimized photobehavioral assay. The results showed that zebrafish exhibited strong positive phototaxis toward red light (612 nm, peak 78.74%) and clear negative phototaxis toward blue light (450 nm, peak -36.23%), with peak behavioral responses at 700 lx and 300 lx, r

Beyond visual comfort: comparing pupillometry and subjective measures of cognitive strain in workplace lighting.

INTRODUCTION: This study evaluated the discriminative sensitivity of pupillometry, relative to subjective self-report measures, as an objective indicator of cognitive load during screen-based tasks performed under two workplace illuminance levels (300 and 500 lux). Inadequate workplace illuminance is associated with visual fatigue, stress, and impaired attention, yet its effect on cognitive strain during screen-based tasks remains underexplored through objective physiological indicators, particularly given the confounding influence of the pupillary light reflex (PLR) on pupillometric measures.

Design of a smart contact lens for controlled retinal lighting in red light therapy.

Current red-light therapy devices lack control over light distribution on the retina, limiting their evaluation and use while presenting potential risks. We designed a scleral lens embedding 625 nm LEDs strategically placed to provide wide-angle retinal illumination while leaving the pupil free. Two configurations were tested: one with a metalized inner part to reflect light from the LEDs towards the retina and another with guiding micro-optics for better efficiency. Experimental irradiance maps obtained with both configurations demonstrate retinal illumination over 130°. We also discuss how t

An automated high-throughput lighting system for screening photosynthetic microorganisms in plate-based formats.

The capacity of photosynthetic microorganisms to fix carbon dioxide into biomass positions them as promising cell factories for sustainable biomanufacturing. However, limitations in screening throughput hinder the identification of enzymes, strains, and growth conditions needed to realize this potential. Here we present a microplate-based high-throughput cultivation system that can be integrated into existing automation infrastructure and supports growth of both prokaryotic and eukaryotic photosynthetic microorganisms. We validate this system by optimizing BG-11 medium compositions for Synecho

Light discomfort thresholds under different lighting conditions in healthy subjects and dry eye patients.

Light-related visual disturbances are common symptoms among patients with dry eye disease (DED) but difficult to be evaluated in the clinical practice. This cross-sectional, multicenter, controlled study aimed at comparing light discomfort thresholds in healthy individuals and patients with DED, and to further correlate thresholds with ocular surface parameters. The Lumiz 100 device (Essilor International, Paris, France) was employed to measure light discomfort thresholds in 25 patients with DED and 25 sex- and age-matched healthy subjects under different lighting conditions: continuous warm,

Artificial nighttime lighting impacts Plasmodium falciparum mature stage V gametocytes infectivity in Anopheles stephensi.

BACKGROUND: Malaria is one of the most important vector-borne diseases of humans with an estimated 241 million cases worldwide in 2020. As an urban and periurban mosquito species, Anopheles stephensi is exposed to artificial human stimuli like light that can alter many aspects of mosquito behaviour, physiology and metabolism. Therefore, fluctuations in the light environment may influence the host, parasite and/or mosquito biology and hence modulate risk for disease transmission. In this study, the effect of artifitial light at night on mosquito infectivity by Plasmodium falciparum during the f

Lighting the way: an economical alternative to feeder cell irradiation for T-cell expansion.

A robust T-cell expansion process involves co-culturing T-cells with non-proliferating feeder cells combined with anti-CD3 antibody and IL-2. Although ionizing irradiation effectively inhibits feeder cell proliferation, the high operating costs limit cell therapy research to well-funded institutions. UVC, known for causing DNA damage-induced cell death and commonly used for environmental sterilization, presents a cost-effective alternative to ionizing irradiation for generating non-proliferating feeder cells. UVC irradiation of K562 artificial antigen presenting cells (aAPCs) resulted in signi

Modeling effects of roadway lighting photometric criteria on nighttime pedestrian crashes on roadway segments.

INTRODUCTION: Nighttime crashes account for 74% of pedestrian fatalities in the United States, and reduced visibility is a significant cause of nighttime pedestrian crashes. Maintaining sufficient and uniform roadway lighting is an effective countermeasure to improve pedestrian visibility and prevent nighttime pedestrian crashes and injuries. Previous studies have not quantified the safety effects of roadway photometric patterns (i.e., average lighting level and uniformity) on nighttime pedestrian crashes on roadway segments. METHOD: This study investigated the association between two roadway

Hidden in the Dark: Dim Ambient Lighting Increases Game Play Duration and Total Spend.

It has been suggested that much like commercial environments (e.g., retailing), the situational characteristics of gambling environments form an important determinant of gambling behavior. However, no research has examined whether ambient lighting in gaming venues can have unintended consequences in terms of gambling behavior. The results of three experimental laboratory studies show that game play duration and total spend increase when ambient lighting is dim (vs. bright). Process evidence suggests that this phenomenon occurs as ambient lighting influences risk-taking, which in turn increases

Impact assessment of energy-efficient lighting in patients with lupus erythematosus: a pilot study.

BACKGROUND: Patients with lupus erythematosus (LE) are often abnormally photosensitive. Ultraviolet (UV) exposure can not only induce cutaneous lesions but may also contribute to systemic flares and disease progression. Various forms of energy-efficient lighting have been shown to emit UV radiation. OBJECTIVES: To determine the effects of these emissions on individuals with LE. METHODS: This assessment investigated cutaneous responses to repeated exposures from three types of lighting: compact fluorescent lamp (CFL), light-emitting diode (LED) and energy-efficient halogen (EEH). The subjects
