Increasing the amount of fat in a conjugated linoleic acid-supplemented diet reduces lipodystrophy in mice.

Conjugated linoleic acid (CLA) is a naturally occurring group of dienoic derivatives of linoleic acid found in beef and dairy products. However, when 1 g CLA/100 g diet was given to mice in a low fat diet (4 g fat/100 g diet), they showed a marked decrease in fat mass, but demonstrated symptoms of lipoatrophic diabetes, i.e., marked hepatomegaly and insulin resistance. In this study, to determine whether the decrease in adipose tissue was responsible for these adverse effects, mice were fed different doses of CLA and dietary fat. In Experiment 1, mice were fed different doses of CLA (0, 0.1 an

Conjugated linoleic acid supplementation reduces adipose tissue by apoptosis and develops lipodystrophy in mice.

Conjugated linoleic acid (CLA) is a naturally occurring group of dienoic derivatives of linoleic acid found in beef and dairy products. CLA has been reported to reduce body fat. To examine the mechanism(s) of CLA reduction of fat mass, female C57BL/6J mice were fed standard semipurified diets (10% fat of total energy) with or without CLA (1% wt/wt). Terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick endlabeling (TUNEL) and DNA fragmentation analysis revealed that fat-mass decrease by CLA was mainly due to apoptosis. Tumor necrosis factor (TNF)-alpha and uncoupling protein (UCP)-2

Response of liver microsomal mixed-function oxidases to dietary linoleic acid levels in rats.

Responses of the activities of liver microsomal mixed-function oxidases (MFOs) induced by phenobarbital (PB) to dietary linoleic acid (LA) levels were investigated in rats. Diets varied in LA content (0, 1.8, 4.1, 7.0, or 13.8 energy%) were fed to rats for 16 days. The activities of MFOs did not change significantly with increasing the dietary LA level, where they reached a plateau even at 1.8 energy %, although the fatty acid composition of liver microsomal lipids reflected well that of dietary lipids. Accordingly, 2 energy % of LA as essential fatty acid seemed to be sufficient to supply the

Caecal and colonic uptake of both linoleic acid and cholesterol in rats following intestinal resection.

Caecal and colonic uptake of both linoleic acid and cholesterol were studied in rats after distal small bowel resection (DSBR). The results showed that the surgical operation increased the caecal and colonic uptake of linoleic acid. Supplementation with linolenic acid inhibited caecal and colonic uptake of linoleic acid. Experiments carried out in the presence of rotenone and ouabain suggest that facilitated diffusion is the predominant mechanism of caecal and colonic linoleic acid absorption, at least at low concentrations. An increase in caecal and colonic uptake of cholesterol was observed

Linoleic Acid Potentiates Response to Chemotherapy in Biliary Tract Cancer Through RARγ Activation.

Biliary tract cancer (BTC) remains highly challenging owing to its poor prognosis. Although gemcitabine plus cisplatin (Gem/Cis) represents a standard chemotherapy option, treatment resistance significantly limits the outcomes. Metabolic reprogramming influences therapeutic response; however, the specific metabolic determinants of chemosensitivity in BTC are poorly defined. Using untargeted metabolomics followed by targeted validation, we found that plasma linoleic acid (LA) levels were significantly elevated in patients with chemotherapy-responsive BTC. LA supplementation led to tumor accumul

Olive oil and linoleic acid as potential acaricidal agents against Psoroptes cuniculi mites.

Psoroptes cuniculi mite is an important ectoparasite in rabbit breeding. Extra virgin olive oil (EVOO) is a vegetable oil with anti-inflammatory and regenerative properties, and linoleic acid (LA) also has beneficial properties for health. The activity against P. cuniculi mites of EVOO was evaluated at concentrations of 1, 5, 10, 30, 60 and 100 %; LA was evaluated at 50, 100, 150, 200, 300 and 400 mg/mL. The effectiveness was evaluated by the immersion technique using 11 mites and four replicates, and post-treatment mortality was analyzed after 24 and 48 h. Also, the composition of EVOO was de

Linoleic acid addition prevents Staphylococcus aureus biofilm formation on PMMA bone cement.

Acrylic bone cement is widely used in vertebroplasty to treat osteoporosis-induced vertebral compression fractures. However, infection after vertebroplasty is problematic and previous work has suggested loading the bone cement with an antibiotic for prophylaxis. Linoleic acid (LA) has been investigated as a promising additive to improve the mechanical properties of bone cement for vertebroplasty, but LA could potentially also have an antibacterial effect. In this study, we evaluated the antibacterial properties of LA-loaded bone cement by comparing its antibiofilm properties with that of origi

Synergistic treatment of linoleic acid and cefazolin on Staphylococcus aureus biofilm-related catheter infections.

Staphylococcus aureus is a human pathogen that causes severe infections through biofilm formation. S. aureus biofilm is particularly susceptible to catheters in patients undergoing peritoneal dialysis. Although antibiotics are used to treat catheter infections, high-concentration treatments adversely affect human host immune systems and change the physicochemical properties of the catheters. To improve therapeutic outcomes without side effects, we combined antibiotics and natural products. In this study, we propose a combination of linoleic acid (LA) and cefazolin (CFZ) to treat S. aureus infe

Dietary linoleic acid supplementation protects against obesity-induced microglial reactivity in mice.

We investigated whether linoleic acid (LA) supplementation could modulate emotional behavior and microglia-related neuroinflammation. For that, male mice of C57BL/6J genetic background fed either a high-fat diet (HFD) or a standard diet (STD) for 12 weeks, were treated with a vehicle or LA solution for 5 weeks before being evaluated for emotional behavior using a battery of behavioral tests. The animals were subsequently sacrificed and their brains collected and processed for immunofluorescence staining, targeting microglia-specific calcium-binding proteins (IBA-1). Neuroinflammation severity

Mechanical characterization and cytocompatibility of linoleic acid modified bone cement for percutaneous cement discoplasty.

Minimally invasive spine treatments have been sought after for elderly patients with comorbidities suffering from advanced degenerative disc disease. Percutaneous cement discoplasty (PCD) is one such technique where cement is injected into a degenerated disc with a vacuum phenomenon to relieve patients from pain. Adjacent vertebral fractures (AVFs) are however an inherent risk, particularly for osteoporotic patients, due to the high stiffness of the used cements. While low-modulus cements have been developed for vertebroplasty through the addition of linoleic acid, there are no such variations

Improving cellular uptake and bioavailability of periplocymarin-linoleic acid prodrug by combining PEGylated liposome.

Periplocymarin (PPM), a cardiac glycoside isolated from Cortex periplocae, has a strong anti-tumor effect against various cancer cells. However, cardiotoxicity and rapid metabolism hinder its clinical applications. In this study, small molecule prodrug was integrated into PEGylated liposome to improve the efficiency of periplocymarin in vivo. The periplocymarin-linoleic acid (PL) prodrug was constructed by conjugating the linoleic acid with PPM via esterification, which was further facilitated to form PEGylated liposome (PL-Lip) through film dispersion. Compared with PL self-assembling nano-pr

Conjugated Linoleic Acid Stimulates Apoptosis in RH and Tehran Strains of Toxoplasma gondii, in Vitro.

BACKGROUND: The aim of this study was to evaluate the effects of conjugated linoleic acid (CLA) on apoptosis of tachyzoites of T. gondii, RH strain (type I) and the cyst-forming Tehran strain (type II) in vitro. METHODS: Toxoplasma strains were injected into the peritoneal cavity of BALB/c mice. The Tehran strain forms cysts in the brain of mice. Bradyzoites within the cysts are reactivated to proliferative tachyzoites, by dexamethasone. Tachyzoites were aspirated from the peritoneum of infected mice, and the percentage of viable parasites was estimated with trypan blue staining. Tachyzoites

Heat generates oxidized linoleic acid metabolites that activate TRPV1 and produce pain in rodents.

The transient receptor potential vanilloid 1 (TRPV1) channel is the principal detector of noxious heat in the peripheral nervous system. TRPV1 is expressed in many nociceptors and is involved in heat-induced hyperalgesia and thermoregulation. The precise mechanism or mechanisms mediating the thermal sensitivity of TRPV1 are unknown. Here, we have shown that the oxidized linoleic acid metabolites 9- and 13-hydroxyoctadecadienoic acid (9- and 13-HODE) are formed in mouse and rat skin biopsies by exposure to noxious heat. 9- and 13-HODE and their metabolites, 9- and 13-oxoODE, activated TRPV1 and

Effects of dietary conjugated linoleic acid at high-fat levels on triacylglycerol regulation in mice.

OBJECTIVE: Our aim was to investigate the effects of dietary conjugated linoleic acid (CLA) at high-fat (HF) levels on parameters related to triacylglycerol (TG) regulation and some potential impacts on liver damage. METHODS: Growing mice were fed a control diet (7% corn oil), an HF diet containing 20% corn oil, or an HF diet containing 3% CLA (HF + CLA) for 30 d. Tissue and organ weights, plasma and tissue TG levels, and parameters related to their regulation were evaluated. Liver oxidative status was also assessed. RESULTS: Dietary CLA showed detrimental and beneficial effects. CLA added t

Feedback activation of ferrous 5-lipoxygenase during leukotriene synthesis by coexisting linoleic acid.

Ferrous lipoxygenases seem to be activated through a feedback control mechanism via FA hydroperoxides generated from PUFAs by partially existing ferric lipoxygenases. However, during leukotriene synthesis, feedback activation of ferrous 5-lipoxygenase in the presence of arachidonic acid (AA) was not observed. In the present study, we examined the feedback activation of ferrous 5-lipoxygenase in the 5-lipoxygenase/AA system in the presence of linoleic aicd (LA), which is a predominant component of membrane phospholipids. When potato 5-lipoxygenase was incubated with AA and LA in the presence of

Lower plasma linoleic acids as a risk factor for gout: an integrated analysis of population-based cohort and genetic data.

Background: Gout is a nutrition-related, highly prevalent inflammatory arthritis with undesirable effects on the quality of life. The relationships between circulating fatty acids (FAs) and gout remain poorly understood. Method: We included 268 174 participants with plasma FAs measured using nuclear magnetic resonance at the baseline (2006-2010) from the UK Biobank, of which 15 194 participants had repeated measures of FAs between 2012 and 2013. Cox proportional hazards models were used to assess the association of the baseline and longitudinal changes in relative levels of plasma FAs (% total
