Luteolin increases susceptibility to macrolides by inhibiting MsrA efflux pump in Trueperella pyogenes.

Trueperella pyogenes (T. pyogenes) is an opportunistic pathogen associated with a variety of diseases in many domestic animals. Therapeutic treatment options for T. pyogenes infections are becoming limited due to antimicrobial resistance, in which efflux pumps play an important role. This study aims to evaluate the inhibitory activity of luteolin, a natural flavonoid, on the MsrA efflux pump and investigate its mechanism. The results of antimicrobial susceptibility testing indicated that the susceptibility of msrA-positive T. pyogenes isolates to six macrolides increased after luteolin treatme

Co-administration of Luteolin mitigated toxicity in rats' lungs associated with doxorubicin treatment.

Doxorubicin (DOX), is a drug against lung malignancies with undesirable side effect including oxidative, inflammatory and apoptotic effects. Luteolin (LUT), present in fruits and vegetables is pharmacologically active against oxido-inflammatory and apoptotic responses. The present study examined the effect of LUT on DOX-induced lungs and blood dysfunction in Wistars rat (sex: male; 10 weeks old, 160 ± 5 g). Randomly grouped (n = 10) rats were treated as follows: control, LUT alone (100 mg/kg; per os), DOX (2 mg/kg; i. p), and co-treated rats with LUT (50 or 100 mg/kg) and DOX for two consecuti

Luteolin exhibits broad-spectrum antiviral activity against swine enteric coronaviruses by targeting 3CLpro.

Swine enteric coronaviruses (SECoVs), including porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), and porcine deltacoronavirus (PDCoV), pose a major threat to global swine production, with current control measures remaining suboptimal. These infections are often characterized by mixed infections, highlighting the urgent need for anti-SECoV drugs. This study elucidates the potential of luteolin as a broad-spectrum antiviral agent targeting the 3 C like protease (3CLpro). Molecular docking and dynamics simulations indicated a stable and high-affinity interaction

Designing Microparticles of Luteolin and Naringenin in Different Carriers via Supercritical Antisolvent Process.

Antioxidants are contained in fruits and vegetables and are commonly obtained through food. However, it is frequently necessary to supplement the diet with substances that are often poorly soluble in water and sensitive to light and oxygen. For this reason, in this work, luteolin (LUT) and naringenin (NAR), two compounds with antioxidant activity and potential health benefits, were precipitated through the supercritical antisolvent technique using polyvinylpyrrolidone and β-cyclodextrin as the carriers. The precipitation occurred from dimethylsulfoxide using supercritical carbon dioxide as the

Luteolin Isolated from Juncus acutus L., a Potential Remedy for Human Coronavirus 229E.

The COVID-19 pandemic, caused by SARS-CoV-2, addressed the lack of specific antiviral drugs against coronaviruses. In this study, bioguided fractionation performed on both ethyl acetate and aqueous sub-extracts of Juncus acutus stems led to identifying luteolin as a highly active antiviral molecule against human coronavirus HCoV-229E. The apolar sub-extract (CH2Cl2) containing phenanthrene derivatives did not show antiviral activity against this coronavirus. Infection tests on Huh-7 cells, expressing or not the cellular protease TMPRSS2, using luciferase reporter virus HCoV-229E-Luc showed tha

Luteolin attenuates lupus nephritis by regulating macrophage oxidative stress via HIF-1α pathway.

Lupus nephritis (LN) is a serious complication of systemic lupus erythematosus (SLE) and a leading cause of mortality. Luteolin (LUT), a compound found in many vegetables, fruits, and Chinese herbal medicine, has been shown to possess anti-inflammatory, antioxidant, and immunosuppressive properties. However, the mechanisms underlying LUT's potential therapeutic effects on LN remain unclear. In this study, we investigated LUT's antagonistic effects on inflammation and oxidative stress using MRL/lpr mice and H2O2-treated macrophages (Raw264.7). Our results indicate that LUT can ameliorate pathol

Luteolin attenuates hepatic injury in septic mice by regulating P2X7R-based HMGB1 release.

P2X7 receptor (P2X7R) and NLRP3 cooperatively participate in inflammation and hepatocyte damage during hepatic injury induced by lipopolysaccharides (LPS). High-mobility group box 1 (HMGB1) released from immune cells in response to such stimuli plays a vital role in mediating inflammation via TLR4 and the receptor for advanced glycation end products (RAGE), a receptor for HMGB1. However, the correlation among P2X7R, RAGE and TLR4 in regulating the release of HMGB1 has not been elucidated. Increasing the number of daily foods is found to be beneficial for hepatocyte damage in septic hepatic inj

Cell-Based Phenotypic Drug Screening Identifies Luteolin as Candidate Therapeutic for Nephropathic Cystinosis.

BACKGROUND: Mutations in the gene that encodes the lysosomal cystine transporter cystinosin cause the lysosomal storage disease cystinosis. Defective cystine transport leads to intralysosomal accumulation and crystallization of cystine. The most severe phenotype, nephropathic cystinosis, manifests during the first months of life, as renal Fanconi syndrome. The cystine-depleting agent cysteamine significantly delays symptoms, but it cannot prevent progression to ESKD and does not treat Fanconi syndrome. This suggests the involvement of pathways in nephropathic cystinosis that are unrelated to l

Luteolin reduces adipose tissue macrophage inflammation and insulin resistance in postmenopausal obese mice.

Previously, we showed that loss of ovarian function in mice fed high-fat diet exacerbated insulin resistance and adipose tissue inflammation. In the current study, we tested whether consumption of luteolin, an anti-inflammatory flavonoid, could mitigate adipose tissue inflammation and insulin resistance in obese ovariectomized mice. Nine-week-old ovariectomized C57BL/6 mice were fed a low-fat diet, high-fat diet (HFD) or HFD supplemented with 0.005% luteolin (HFD+L) for 16 weeks. Results showed no difference in body weight or fat mass between mice fed HFD+L and those fed HFD. However, luteolin

Luteolin-mediated PI3K/AKT/Nrf2 signaling pathway ameliorates inorganic mercury-induced cardiac injury.

Inorganic mercury is a toxic metal of worldwide concern, and causes serious cardiac injury. However, effective treatment for cardiac injury induced by mercuric chloride (HgCl2) has not been fully identified. Luteolin (Lut) is a novel natural antioxidant. This study aimed to investigate the role of Lut on HgCl2-induced cardiac injury. Male Wistar rats were randomly assigned to 4 groups, control, Lut (80 mg/kg intragastrically), HgCl2 (80 mg/L, in drinking water), and HgCl2 + Lut groups. The results indicated that Lut significantly ameliorated cardiac histopathological damage, oxidative stress,

A Luteolin-Loaded Electrospun Fibrous Implantable Device for Potential Therapy of Gout Attacks.

The recurrence of acute gout attacks remains an unsolved problem in clinical therapy. In order to tackle this problem, poly(ε-caprolactone) (PCL)/gelatin composite fibrous devices loaded with luteolin are presented via electrospinning for the therapy of gout and its recurrence. The luteolin-loaded fibrous device has the capability of inhibiting metabolic activities and reducing inflammation-associated cytokine productions (TNF-a, IL-1β, IL-6) that are secreted by lipopolysaccharide stimulated RAW 264.7 macrophages. The device can also suppress the reaction activities of xanthine oxidase for 7

Luteolin inhibits the growth and arylamine N-acetyl-transferase activity in Neisseria gonorrhoeae.

Growth inhibition and arylamine N-acetyltransferase (NAT) activity in Neisseria gonorrhoeae were inhibited by luteolin, a drug which originated from herbs. The growth inhibition was based on changes in optical density (OD) using a spectrophotometer, and arylamine NAT activity with 2-aminofluorene (2-AF) was determined using high pressure liquid chromatography. The inhibition of growth in N. gonorrhoeae demonstrated that luteolin elicited a dose-dependent growth inhibition in the N. gonorrhoeae cultures.
