Mixture effects of estrogenic compounds on proliferation and pS2 expression of MCF-7 human breast cancer cells.

Humans are exposed to a variety of food-borne phytochemicals (PC) as well as synthetic chemicals (SC). Some of these compounds have been reported to have estrogenic or anti-estrogenic properties and are therefore suspected endocrine disruptors. Until now it remains unclear if non-additive effects occur in combinations with endogenous estrogens, such as 17beta-estradiol (E(2)). To investigate such interactions, several PC and SC were tested individually, in mixtures and as combinations of mixtures with E(2) for effects on ERalpha receptor mediated cell proliferation and estrogen regulated pS2 e

Oestrogen-induced genes, pLIV-1 and pS2, respond divergently to other steroid hormones in MCF-7 cells.

The level of oestrogen-responsive gene expression in breast tumours has been proposed as a predictor of the response of the tumour to endocrine (anti-oestrogen) therapy. We demonstrate that different oestrogen-responsive genes may differ in their responses to other hormones. pLIV-1 and pS2 are two oestrogen-regulated genes that are expressed in the MCF-7 human breast cancer cell line. We show that pLIV-1 mRNA, but not pS2 mRNA, is also induced, to a lesser extent, by progesterone, 5 alpha-dihydrotestosterone and dexamethasone. For pLIV-1, combinations of these hormones with oestradiol and with

Probing the specific binding of folic acid to folate receptor using amino-functionalized mesoporous silica nanoparticles for differentiation of MCF 7 tumoral cells from MCF 10A.

Folate receptor (FR) is overexpressed in various cancer cells while its expression in normal cells is restricted. The present study provides a new folic acid/folate (FA) functionalized nanomaterials to sense and the differentiation of the cancer cells from normal ones. The reported nanoprobe is based on the mesoporous materials that are functionalized with FA to specify the FR overexpressed cancerous cells. MCF 7 cell lines were used as a model to show the ability of the developed probe for cancer cell detection. The selective binding of FA to FR-positive cells causes the endocytosis of the me

Potential-resolved single-nanoparticle collision electrochemistry for simultaneous quantification of MCF-7 and SK-BR-3 cells.

Compared to detecting individual target analytes by single immunoassay, multiplex immunoassays (MIAs) can simultaneously detect multiple analytes in a single experiment, saving more time and reducing sample consumption. Single-nanoparticle collision electrochemistry (SNCE), characterized by its high throughput, sensitivity, and rapid response, has been extensively employed in the field of biosensing. Herein, an innovative potential-resolved SNCE biosensor was proposed for the simultaneous and rapid detection of two types of breast cancer cells (MCF-7 and SK-BR-3 cells) based on platinum nanopa

Integrated data-independent acquisition and thermal proteome profiling for proteomic characterization of lamotrigine-treated MCF-7 cells.

Lamotrigine, a clinically approved anticonvulsant, has recently gained interest for its potential anticancer effects. However, the molecular mechanisms underlying its activity in breast cancer remain poorly understood. Here, an integrated mass spectrometry-based proteomic strategy combining data-independent acquisition (DIA) and thermal proteome profiling (TPP) was applied to investigate the cellular effects and potential targets of lamotrigine in the human breast cancer cell line MCF-7. The DIA-based quantitative approach identified 6622 proteins, of which 142 were significantly modulated fol

A natural coumarin, benjaminin, suppresses proliferation and migration and induces apoptosis in MCF-7 breast cancer cells.

Breast cancer remains the leading cause of cancer-related mortality in women, highlighting the need for selective and effective therapeutics. Conventional chemotherapies are often limited by off-target toxicity, prompting interest in natural products, including coumarins from Calophyllum species. Benjaminin, a novel coumarin, has demonstrated cytotoxicity in multiple cancer models, yet its effects in breast cancer remain unexplored. Here, we evaluated benjaminin in MCF-7 (tumorigenic) and MCF-10A (non-tumorigenic) breast cells. Benjaminin exhibited moderate but highly selective cytotoxicity to

An acidic pH-activatable fluorescent probe for MUC1 detection and the precise imaging of MCF-7 cells.

Mucin 1 (MUC1), a pivotal cancer biomarker, is overexpressed on MCF-7 cells, which has stimulated the construction of numerous aptamer-based fluorescent probes for breast cancer diagnosis. However, MUC1 is also present in the serum of cancer patients and expressed at low levels on the surface of normal cells, which may result in false-positive signals upon recognition by aptamers. In this work, a gold nanoparticle (AuNPs)-based fluorescent probe termed b-NPs was proposed by utilizing the acidic tumor microenvironment and pH-responsive single-stranded DNA (f-DNA) with conformational switching c

NSUN7 Suppression Reduces Metastatic Potential and Restores Sensitivity to 4-OH Tamoxifen in Resistant MCF-7 Cells.

BACKGROUND/AIM: Tamoxifen remains a first-line treatment for estrogen receptor-positive breast cancer. Emerging evidence indicates that aberrant 5-methylcytosine (m5C) modification of RNAs contributes to chemotherapeutic resistance in various types of cancer, however their role in determining tamoxifen resistance in breast cancer remains elusive. MATERIALS AND METHODS: We measured global m5C RNA methylation and expression of its regulating enzymes in tamoxifen-sensitive (MCF-7) versus tamoxifen-resistant cells (MCF-7 Tam1). Expression of the most significantly dysregulated enzyme NOP2/Sun RNA

Propofol suppresses migration and vascular mimicry of breast cancer MCF-7 cells by down regulating interleukin-6.

Propofol is widely used in anesthesia, but its role in breast cancer progression remains controversial. This study investigated the molecular mechanisms of propofol in breast cancer, focusing on IL-6 and tumor microenvironment modulation. Bioinformatics analysis identified IL-6 as a potential target of propofol. MCF-7 cells were treated with varying propofol concentrations (0-100 μg/mL), and cell viability was assessed via CCK-8 assay. Propofol at 50 μg/mL significantly reduced viability, while 25 μg/mL (a non-cytotoxic dose) was selected for further experiments. Western blot confirmed propofo

Lidocaine and Bupivacaine Downregulate MYB and DANCR lncRNA by Upregulating miR-187-5p in MCF-7 Cells.

Background: Breast cancer is the most common malignancy and a leading cause of death among women. The majority of patients require surgery, and retrospective studies have revealed an association between anaesthetic techniques during surgery and clinical outcomes. Local anaesthetics (LAs) influence carcinogenesis by interacting with non-coding RNAs (ncRNAs). However, the detailed mechanisms underlying the association between LAs and ncRNAs remain unclear. Methods: In this study, the effects of two commonly used LAs, lidocaine and bupivacaine, on the malignancy of MCF-7 breast cancer cells were

Pro-Oxidant Effect of Resveratrol on Human Breast Cancer MCF-7 Cells is Associated with CK2 Inhibition.

Casein kinase 2 (CK2) plays a critical role in the proliferation and apoptosis of cancer cells. Resveratrol is a bioactive compound with anticancer and anti-inflammatory effects. This study investigated the pro-oxidant cytotoxic effects of resveratrol in association with the inhibition of CK2 activity on human breast carcinoma cells MCF-7. We showed that resveratrol and TBB, an inhibitor of CK2, decreased cell viability in a concentration dependent manner with an IC50 value of 238 µM and 106 µM after 24 h, of treatment, respectively. Resveratrol and TBB decreased CK2 activity by 1.6 and 1.4-fo

Response of MCF-7 Breast Cancer Cells Overexpressed with P-Glycoprotein to Apoptotic Induction after Photodynamic Therapy.

Multidrug resistance (MDR) has posed a significant threat to cancer treatment and has led to the emergence of a new therapeutic regime of photodynamic therapy (PDT) to curb the menace. The PDT modality employs a photosensitiser (PS), excited at a specific wavelength of light to kill cancer cells. In the present study, we used a zinc phthalocyanine tetrasulfonic acid PS to mediate the photodynamic killing of MCF-7 cells overexpressed with P-glycoprotein (P-gp) and investigate the response to cell death induction. After photodynamic treatment, MCF-7 cells undergo cell death, and indicators like

Recombinant adenoviruses expressing apoptin suppress the growth of MCF‑7 breast cancer cells and affect cell autophagy.

Autophagy and apoptosis both promote cell death; however, the relationship between them is subtle, and they mutually promote and antagonize each other. Apoptin can induce apoptosis of various tumor cells; however, tumor cell death is not only caused by apoptosis. Whether apoptin affects tumor cell autophagy is poorly understood. Therefore, the present study aimed to explore the potential mechanisms underlying the effects of apoptin using recombinant adenoviruses expressing apoptin. Reverse transcription‑quantitative polymerase chain reaction, immunoblotting, flow cytometry, fluorescence micros

Oxidative stress mediated cytotoxicity of tin (IV) oxide (SnO2) nanoparticles in human breast cancer (MCF-7) cells.

Due to unique optical and electronic properties tin oxide nanoparticles (SnO2 NPs) have shown potential for various applications including solar cell, catalyst, and biomedicine. However, there is limited information concerning the interaction of SnO2 NPs with human cells. In this study, we explored the potential mechanisms of cytotoxicity of SnO2 NPs in human breast cancer (MCF-7) cells. Results demonstrated that SnO2 NPs induce cell viability reduction, lactate dehydrogenase leakage, rounded cell morphology, cell cycle arrest and low mitochondrial membrane potential in dose- and time-dependen

Catalase expression in MCF-7 breast cancer cells is mainly controlled by PI3K/Akt/mTor signaling pathway.

Catalase is an antioxidant enzyme that catalyzes mainly the transformation of hydrogen peroxide into water and oxygen. Although catalase is frequently down-regulated in tumors the underlying mechanism remains unclear. Few transcription factors have been reported to directly bind the human catalase promoter. Among them FoxO3a has been proposed as a positive regulator of catalase expression. Therefore, we decided to study the role of the transcription factor FoxO3a and the phosphatidylinositol-3 kinase (PI3K) signaling pathway, which regulates FoxO3a, in the expression of catalase. To this end,

Griseofulvin stabilizes microtubule dynamics, activates p53 and inhibits the proliferation of MCF-7 cells synergistically with vinblastine.

BACKGROUND: Griseofulvin, an antifungal drug, has recently been shown to inhibit proliferation of various types of cancer cells and to inhibit tumor growth in athymic mice. Due to its low toxicity, griseofulvin has drawn considerable attention for its potential use in cancer chemotherapy. This work aims to understand how griseofulvin suppresses microtubule dynamics in living cells and sought to elucidate the antimitotic and antiproliferative action of the drug. METHODS: The effects of griseofulvin on the dynamics of individual microtubules in live MCF-7 cells were measured by confocal microsc

Nebivolol prevents exhausted T cells and enhances cytotoxicity against MCF-7 breast cancer cells in a β2-adrenergic receptor-dependent manner.

INTRODUCTION: Cancers often drive T cells toward an exhausted state characterized by impaired cytotoxicity and upregulation of inhibitory receptors (PD-1, TIM-3, CD38) and transcriptional regulators (TOX, NFATc1). Repeated stimulation in vitro is used to model this process, reflecting chronic antigen exposure in the tumor microenvironment. Stress-derived catecholamines further drive dysfunction through β-adrenergic receptor (β-AR) signaling. Here, we examined the impact of nebivolol, an atypical β1-AR blocker with β2-biased agonist activity, on T-cell exhaustion and cytotoxicity against breast

An electrochemiluminescent aptasensor for amplified detection of exosomes from breast tumor cells (MCF-7 cells) based on G-quadruplex/hemin DNAzymes.

Exosomes are non-invasive biomarkers for cancer diagnosis. Herein, we describe an electrochemiluminescent (ECL) aptasensor for the detection of exosomes from breast tumor cells. Mercaptopropionic acid (MPA)-modified Eu3+-doped CdS nanocrystals (MPA-CdS:Eu NCs) and H2O2 were used as ECL emitters and coreactant, respectively. The exosomes are recognized and captured by the CD63 aptamer, and then form a G-quadruplex/hemin DNAzyme, which efficiently catalyzes the decomposition of H2O2, resulting in the decreased ECL signal of MPA-CdS:Eu NCs. The exosomes from breast tumor cells (MCF-7 cells) can b
