(Macro)Molecular Crowding Effects Beyond Volume Exclusion.

Macromolecular crowding affects many areas, including protein folding, binding of small molecules, enzymatic activity, interaction with nucleic acids, protein aggregation, and protein-protein interactions. While the common belief has been that the primary impact of crowded environments on the function, structure, thermodynamics, and aggregation of a protein can be described in terms of excluded volume effects, it has now become clear that other factors, which originate from high concentrations of "inert" macromolecules in crowded solution, must be considered in order to get a clearer understan

Natural endogenous material-based vehicles for delivery of macromolecular drugs.

Natural endogenous materials (NEMs), such as cell and cell derivatives, polysaccharide, protein and peptide, and nucleic acid-derived vectors, often exhibit biocompatibility, biodegradability and natural homing ability, which can minimize adverse reactions in vivo and have the potential to improve drug delivery efficacy. Currently, a variety of drug delivery systems (DDSs) based on NEMs have been constructed for macromolecules to address the challenges posed by their inherent large size, intricate structure, low permeability, and susceptibility to harsh environments. The aim of this article is

The Hamilton Receptor in Supramolecular Polymer Sciences.

Supramolecular polymers are polymeric materials of monomeric fragments, held jointly by reversible and directional non-covalent interactions such as multiple hydrogen-bonding, charge transfer effects, host-guest interactions, metal coordination, and aromatic stacking. This review article on the Hamilton-based supramolecular polymers aims to shed light on the molecular recognition achievements by the Hamilton-based polymeric systems, evaluate Hamilton receptor's future prospects, and capitalize its potential applications in supramolecular chemistry. To the best of our knowledge, this is the fir

Detection of Macromolecular Fractions in HCN Polymers Using Electrophoretic and Ultrafiltration Techniques.

Elucidating the origin of life involves synthetic as well as analytical challenges. Herein, for the first time, we describe the use of gel electrophoresis and ultrafiltration to fractionate HCN polymers. Since the first prebiotic synthesis of adenine by Oró, HCN polymers have gained much interest in studies on the origins of life due to the identification of biomonomers and related compounds within them. Here, we demonstrate that macromolecular fractions with electrophoretic mobility can also be detected within HCN polymers. The migration of polymers under the influence of an electric field de

Reaction of nitrosonium cation with resorc[4]arenes activated by supramolecular control: covalent bond formation.

Resorc[4]arenes 1 and 2, which previously proved to entrap NO(+) cation within their cavities under conditions of host-to-guest excess, were treated with a 10-fold excess of NOBF4 salt in chloroform. Kinetic and spectral UV-visible analyses revealed the formation of isomeric 1:2 complexes as a direct evolution of the previously observed event. Accordingly, three-body 1-(NO(+))2 and 2-(NO(+))2 adducts were built by MM and fully optimized by DFT calculations at the B3LYP/6-31G(d) level of theory. Notably, covalent nitration products 4, 5 and 6, 7 were obtained by reaction of NOBF4 salt with host

Dendrimers as a scaffold for nitric oxide release.

The preparation and characterization of nitric oxide (NO)-releasing dendrimer conjugates are reported. Generation 3 and 5 polypropylenimine dendrimers (DAB-Am-16 and DAB-Am-64) were modified at the exterior to impart different amine functionalities. The ability to store NO on a dendritic scaffold using N-diazeniumdiolate NO donors was examined via the reaction of primary amine, secondary amine, and amide functionalities with high pressures of NO (5 atm). The secondary amine dendrimer conjugates exhibited a high storage capacity for NO (up to 5.6 micromol NO/mg), greatly increasing the "payload

New calixarene-bonded stationary phases in high-performance liquid chromatography: comparative studies on the retention behavior and on influences of the eluent.

The chromatographic behavior of six calixarene-bonded stationary phases is reported. Varying analyte selectivities (i.e., for phenols, substituted aromatics, polycyclic aromatic hydrocarbons, barbituric acid derivatives, xanthines) exist as a function of the ring-size of the calix[n]arenes (n=4, 6, 8) and the substitution at the "upper rim" with para-tert.-butyl groups. Although eluents with unusually high proportions of water were used, a comparison with conventional reversed-phase (RP) columns shows a predominantly reversed-phase character with remarkable selectivities of these phases. The i

Celiac sprue and macroamylasemia: potential clinical and pathophysiological implications. Case study.

In a patient with celiac sprue, abdominal complaints were ascribed to acute pancreatitis in the setting of hyperamylasemia and choledocholithiasis. The problem turned out to be macroamylasemia associated with elevated serum immunoglobulin A. The patient remained stable for 4 years with no specific intervention except gluten restriction. An association between celiac sprue or other malabsorptive states and macroamylasemia requires more study.

Chlorpromazine induced aggregation of normal human hemoglobin. Electron microscopic evidence.

Normal human hemoglobin exceeding a certain minimum concentration (called critical aggregation concentration) undergoes aggregation in presence of the psychotherapeutic drug chlorpromazine (CPZ). The critical aggregation concentration decreases with the increase of CPZ concentration. Electron micrographs of CPZ-treated hemoglobin clearly indicate that the aggregates of hemoglobin are in filamentous form of average width 75 +/- 8 A. A possible mechanism for such aggregation has been discussed.

Macromolecular markers in joint disease.

Cartilage contains a number of matrix macromolecules not present in other connective tissues. In tissue processes in disease and in normal turnover of the matrix these molecules are fragmented and released into surrounding fluids, in the case of joints into the synovial fluid. Immunoassay techniques have been developed to quantify these fragments. It appears that increased levels in synovial fluid (SF) correlate to a process in the joint cartilage. With successful therapy the level returns to normal. Interestingly, in early rheumatoid arthritis, i.e., with no discernible cartilage damage by ra

Antihistone antibodies in rheumatoid arthritis and Felty's syndrome.

Although antihistone antibodies are frequently detected in patients with systemic lupus erythematosus, they appear to be uncommon or present only at low levels in rheumatoid arthritis (RA) patients. We used an enzyme-linked immunosorbent assay to study sera from 42 patients with RA, and we confirmed that antihistone antibodies mainly occurred at low levels in these patients. In contrast, 83% of 24 patients with Felty's syndrome had antihistone antibodies, often at high levels. Thus, antihistone antibodies may be a common autoimmune abnormality in patients with Felty's syndrome but not in other

Subunit assembly of hemoglobin: an important determinant of hematologic phenotype.

Hemoglobin's physiologic properties depend on the orderly assembly of its subunits in erythropoietic cells. The biosynthesis of alpha- and beta-globin polypeptide chains is normally balanced. Heme rapidly binds to the globin subunit, either during translation or shortly thereafter. The formation of the alpha beta-dimer is facilitated by electrostatic attraction of a positively charged alpha-subunit to a negatively charged beta-subunit. The alpha beta-dimer dissociates extremely slowly. The difference between the rate of dissociation of alpha beta- and alpha gamma-dimers with increasing pH expl

Electroimmunoassay of a subunit protein in a macromolecular complex (apolipoprotein B in human plasma very low density lipoprotein); implications for other electroimmunoassay systems.

With an electroimmunoassay ("rocket") system for the apolipoprotein B component of the plasma very low density lipoprotein complex we obtained results which were similar to those obtained by a colorimetric tetramethylurea extraction method. Results were up to twice as high as those using radioimmunoassay. Low density lipoprotein containing apolipoprotein B as the only demonstrable protein component was used as the standard for these assays. This protein produced larger and higher rockets at pH 8.6 when the negative particle charge was increased by maleylation. The very low density lipoprotein

Distribution volumes of macromolecules in human ovarian and endometrial cancers--effects of extracellular matrix structure.

Elements of the extracellular matrix (ECM), notably collagen and glucosaminoglycans, will restrict part of the space available for soluble macromolecules simply because the molecules cannot occupy the same space. This phenomenon may influence macromolecular drug uptake. To study the influence of steric and charge effects of the ECM on the distribution volumes of macromolecules in human healthy and malignant gynecologic tissues we used as probes 15 abundant plasma proteins quantified by high-resolution mass spectrometry. The available distribution volume (VA) of albumin was increased in ovarian

Stable Isotope Abundance Patterns as Potential Biosignatures.

The abundance and distribution of stable isotopes of an element in a substance can provide insights regarding the source, synthesis, and environmental history of that substance. Because isotopic discrimination during chemical reactions can be unique to specific chemical pathways or environmental conditions, isotopic patterns within a substance or between related substances may provide insights into their formation. Biosynthetic pathways can create isotopic patterns that differ from patterns that arise from abiotic processes, but this is not universally true. Isotope patterns are signatures of

Biomass-Derived Plant Extracts in Macromolecular Chitosan Matrices as a Green Coating for PLA Films.

Due to the growing problem of food and packaging waste, environmental awareness, and customer requirements for food safety, there is a great need for the development of innovative and functional packaging. Among these developments, the concept of active packaging is at the forefront. The shortcoming in this area is that there is still a lack of multifunctional concepts, as well as green approaches. Therefore, this work focuses on the development of active chemical substances of natural origin applied as a coating on polylactic acid (PLA) films. Biopolymer chitosan and plant extracts rich in ph

[Defining and characterization of supramolecular complexes in TCM formulations].

The supramolecular perspective offers a path distinct from component analysis for understanding the material basis of TCM compatibility, shifting the research focus from co-existence of components to formation of structures. It is common that precipitates or colloidal particles appear after co-decocting of medicinal materials, while such phenomena mostly reflect phase changes in the system and are insufficient to directly identify stable and definable supramolecular complexes. Against this background, this paper systematically reviews the determination path of supramolecular structures induced

Osmotic Pressure-Induced Lipid Membrane Phase Separation within Macromolecular Environments.

Lipid membrane phase separation, which is a thermodynamic physical process, has attracted attention as a dynamic function of cellular interfaces. As membrane tension influences the phase separation of cellular membranes under isothermal conditions, it is essential to clarify the physicochemical mechanism involved. Living cells contain numerous macromolecules that can generate osmotic pressure across the membrane due to the semipermeable nature of the lipid bilayer. In this research, we examined how macromolecular surroundings and the transmembrane osmotic pressure influence membrane phase sepa
