Starvation as a weapon in fungal-plant warfare.

Fungal pathogens cause devastating diseases in staple crops and pose a tremendous threat to food security. Therefore, it is critical to understand the mechanisms of fungal attack and plant defense. Recent studies provide new insights into the role of nutrient manipulation for both plant and pathogen combatants. On the plant side, a defense signaling system has been discovered that provokes starvation during disease to limit access to carbohydrate by the head smut fungus Sporisorium reilianum. For pathogenic fungi, a novel class of effector proteins in the rice blast fungus Magnaporthe oryzae a

Discovery of Potential Fungal Dihydroorotate Dehydrogenase Inhibitors Via Structure-based Virtual Screening for Plant Disease Control.

Dihydroorotate dehydrogenase (DHODH), a key enzyme in the de novo pyrimidine biosynthesis pathway of phytopathogenic fungi, has emerged as a promising target for fungicide discovery. In this study, compound VS-24 was identified from a commercial chemical library using molecular docking-based virtual screening and bioactivity assays. Antifungal tests demonstrated that at a concentration of 50 mg/L, VS-24 has broad-spectrum inhibitory effects with inhibition rates of 96.0%, 76.7%, 75.3%, and 76.5% against Valsa mali, Botrytis cinerea, Gaeumannomyces graminis, and Magnaporthe oryzae, respectively

The role of the casein kinase 1 (CK1) family in fungal infections.

The Casein kinase 1 (CK1) family, as an important member of the protein kinase family, exhibits high evolutionary conservation and is widely distributed in various cell types. CK1 plays critical roles in numerous cellular biological processes, and its abnormal expression is closely associated with the development and progression of various diseases. The CK1 protein family has been extensively studied and reported as an influential factor in various mammalian diseases. Increasing evidence indicates that the CK1 family also plays an indispensable role in fungal growth and infection processes. Fo

H2S-mediated protein S-sulfhydration modulates infectivity and autophagy in the rice blast fungus.

Hydrogen sulfide (H2S) regulates cellular activities in plants and mammals through S-sulfhydration, a post-translational modification of proteins. The role of H2S and its molecular targets in fungi, however, remains unclear. Here we show that H2S, synthesized by cystathionine γ-lyase (CSE1) in the rice blast fungus Magnaporthe oryzae, is essential for optimal fungal infection. Excessive H2S, through S-sulfhydration, impairs fungal infectivity by inhibiting autophagy. Using quantitative proteomics, we identify numerous S-sulfhydrated proteins in M. oryzae, including the autophagy-related protei

Balancing rice blast resistance and growth through suppression of the E3 ubiquitin ligase OsRING80.

In crop plants, there is often a trade-off between growth and defense; modulating the balance is crucial for effective crop improvement. In rice (Oryza sativa), the DENSE AND ERECT PANICLE 2 (DEP2) mutant shows altered phenotypes such as a more vertical leaf angle, erect panicles, small and round grains, slightly lower thousand-grain weight, and dwarfism. Here, we determined that loss of DEP2 function enhances resistance to the rice blast fungus Magnaporthe oryzae. We established that DEP2 interacts with the E3 ubiquitin ligase OsRING80, which ubiquitinates DEP2 and promotes its degradation vi

CRISPR/Cas9-mediated knockout of Bsr-d1 enhances the blast resistance of rice in Northeast China.

The blast resistance allele of OsBsr-d1 does not exist in most japonica rice varieties of Jilin Province in China. The development of Bsr-d1 knockout mutants via CRISPR/Cas9 enhances broad-spectrum resistance to rice blast in Northeast China. Rice blast is a global disease that has a significant negative impact on rice yield and quality. Due to the complexity and variability of the physiological races of rice blast, controlling rice blast is challenging in agricultural production. Bsr-d1, a negative transcription factor that confers broad-spectrum resistance to rice blast, was identified in th

Targeting the TORC1/TORC2 Signaling Hub MoLst8 Cripples Fungal Pathogenesis and Reveals TF3 as a Novel Selective Inhibitor.

Rice blast, caused by Magnaporthe oryzae, remains a primary threat to global food production, making the discovery of novel antifungal targets imperative. The Target of Rapamycin (TOR) signaling network is a prime target for therapeutic intervention, but the function of its central components in M. oryzae is not fully understood. In this study, we found that MoLst8 is indispensable for fungal growth, development, and virulence in M. oryzae. Genetic deletion of MoLST8 impairs fungal growth, conidiation, and virulence by disrupting TOR signaling, triggering excessive autophagy, and damaging plas

Constitutive expression of rice WRKY30 gene increases the endogenous jasmonic acid accumulation, PR gene expression and resistance to fungal pathogens in rice.

WRKY transcription factors are crucial regulatory components of plant responses to pathogen infection. In the present study, we report isolation and functional characterization of the pathogen-responsive rice WRKY30 gene, whose transcripts accumulate rapidly in response to salicylic acid (SA) and jasmonic acid (JA) treatment. Overexpression of WRKY30 in rice enhanced resistance to rice sheath blight fungus Rhizoctonia solani and blast fungus Magnaporthe grisea. The enhanced resistance in the transgenic lines overexpressing WRKY30 was associated with activated expression of JA synthesis-related

Unveiling the Roles of LncRNA MOIRAs in Rice Blast Disease Resistance.

Rice blast disease, caused by the fungal pathogen Magnaporthe oryzae, is a major threat to rice production worldwide. This study investigates the role of long non-coding RNAs (lncRNAs) in rice's response to this destructive disease, with a focus on their impacts on disease resistance and yield traits. Three specific lncRNAs coded by M. oryzae infection-responsive lncRNAs (MOIRAs), MOIRA1, MOIRA2, and MOIRA3, were identified as key regulators of rice's response to M. oryzae infection. Strikingly, when MOIRA1 and MOIRA2 were overexpressed, they exhibited a dual function: they increased rice's su

Cysteine-rich receptor-like kinases OsCRK19 and OsCRK22 interact with OsRbohI to modulate ROS production and disease resistance in rice.

Reactive oxygen species (ROS) burst is a hallmark response of plant innate immunity. ROS production must be under tight control since inappropriate accumulation is toxic. In rice, OsRbohI is the key NADPH oxidase responsible for ROS generation. However, the upstream kinases regulating OsRbohI activity remain unknown. Here, we identified two cysteine-rich receptor-like kinases, OsCRK19 and OsCRK22, as regulators of OsRbohI. OsCRK19 and OsCRK22 bind to N- both termini of OsRbohI. OsCRK19/22-OsRbohI interactions are constitutive and not responding to PAMP elicitation. OsCRK19 and OsCRK22 phosphor

Homology Modeling and Pharmacophore Based Inhibitor Design Targeting Isocitrate Lyase of Madurella mycetomatis-A Novel Approach Against Eumycetoma.

Eumycetoma, caused by pathogenic fungi, is one of the most neglected tropical disease worldwide. The causative agent of the disease is Madurella mycetomatis. Ethnomedicinal sources reveal that several medicinal plants are used for the phytotherapeutic management of eumycetoma symptoms. M. mycetomatis uses isocitrate lyase (ICL) to promote gluconeogenesis for growth and survival in adverse host conditions. This study aims to develop a natural inhibitor molecule of ICL to target against M. mycetomatis using computational techniques. Homology modeling was used to predict the 3D structure of ICL u

Glycoside Hydrolase MoGls2 Controls Asexual/Sexual Development, Cell Wall Integrity and Infectious Growth in the Rice Blast Fungus.

N-linked glycosylation is a way of glycosylation for newly synthesized protein, which plays a key role in the maturation and transport of proteins. Glycoside hydrolases (GHs) are essential in this process, and are involved in processing of N-linked glycoproteins or degradation of carbohydrate structures. Here, we identified and characterized MoGls2 in Magnaporthe oryzae, which is a yeast glucosidase II homolog Gls2 and is required for trimming the final glucose in N-linked glycans and normal cell wall synthesis. Target deletion of MoGLS2 in M. oryzae resulted in a reduced mycelial growth, an i

Rice ubiquitin-conjugating enzyme OsUbc13 negatively regulates immunity against pathogens by enhancing the activity of OsSnRK1a.

Ubc13 is required for Lys63-linked polyubiquitination and innate immune responses in mammals, but its functions in plant immunity still remain largely unknown. Here, we used molecular biological, pathological, biochemical, and genetic approaches to evaluate the roles of rice OsUbc13 in response to pathogens. The OsUbc13-RNA interference (RNAi) lines with lesion mimic phenotypes displayed a significant increase in the accumulation of flg22- and chitin-induced reactive oxygen species, and in defence-related genes expression or hormones as well as resistance to Magnaporthe oryzae and Xanthomonas

Evidence for biotrophic lifestyle and biocontrol potential of dark septate endophyte Harpophora oryzae to rice blast disease.

The mutualism pattern of the dark septate endophyte (DSE) Harpophora oryzae in rice roots and its biocontrol potential in rice blast disease caused by Magnaporthe oryzae were investigated. Fluorescent protein-expressing H. oryzae was used to monitor the colonization pattern. Hyphae invaded from the epidermis to the inner cortex, but not into the root stele. Fungal colonization increased with root tissue maturation, showing no colonization in the meristematic zone, slight colonization in the elongation zone, and heavy colonization in the differentiation zone. H. oryzae adopted a biotrophic life
