Dietary Magnolia kobus DC. Attenuates Age-Associated Hippocampal Alterations in Mice in Association with Modulation of Iron Metabolism- and Ferroptosis-Related Markers.

Brain aging is a major contributor to cognitive decline and neurodegenerative disorders. Ferroptosis, a regulated form of cell death driven by iron-dependent lipid peroxidation, has emerged as a key mechanism associated with age-related hippocampal dysfunction. Here, we investigated whether dietary supplementation of Magnolia kobus DC. (MO) attenuates age-related hippocampal alterations in mice in association with changes in iron metabolism- and ferroptosis-related markers. Behavioral tests combined with molecular and histological analyses of hippocampal tissues from young and aged mice demons

Magnoliae flos (Shin-i) Essential Oil and Its Principal Component Eucalyptol Inhibit JAK1 to Counteract the LPS-Induced RAW264.7 Inflammation.

Magnoliae flos (Shin-i) has been traditionally utilized in Chinese medicine for rhinitis, sinusitis, and headaches. However, the underlying anti-inflammatory mechanisms of Shin-i essential oil (SIEO), especially its effects on the Janus kinase/signal transducer and activator of transcription (JAK-STAT) signaling pathway, remain inadequately elucidated. The chemical composition of SIEO was characterized using gas chromatography-mass spectrometry (GC-MS). Network pharmacology and gene ontology (GO) enrichment analyses predicted that SIEO may target the JAK-STAT signaling pathway. Cellular experi

Behavioural pharmacological characteristics of honokiol, an anxiolytic agent present in extracts of Magnolia bark, evaluated by an elevated plus-maze test in mice.

Honokiol, a neolignane derivative of Magnolia bark, has central depressant action and, at much lower doses, anxiolytic activity. We have investigated the characteristics of the behavioural effects of honokiol by means of an elevated plus-maze test. In the plus-maze test a single oral dose of 20 mg kg(-1) honokiol significantly prolonged the time spent in the open arms of the maze, suggesting anxiolytic effect. Moreover, when honokiol was administered daily for seven days and the plus-maze test was conducted 3 or 24 h after the last administration, significant prolongation of the time in the op

Deciphering the multi-target mechanism of Magnolia officinalis in irritable bowel syndrome with diarrhea: A combined network pharmacology, machine learning, and in vivo validation study.

BACKGROUND: The bark of Magnolia officinalis Rehder & E. Wilson, used in TCM for abdominal distension, pain, and diarrhea, aligns with IBS-D symptoms. It is now supported by evidence for its multi-component, multi-target action. This study aims to elucidate the core bioactive components and their mechanisms against IBS-D. To achieve this, we employed an analytical strategy that uses machine learning to refine network pharmacology predictions, rather than relying on network pharmacology alone. MATERIALS AND METHODS: This study integrates network pharmacology and machine learning to identify co

Spontaneous formation of highly concentrated phenylacetylcarbinol via in situ phase separation in deep eutectic solvents using Candida magnoliae whole cells.

Deep eutectic solvents (DESs) are promising green media for biocatalysis, yet their application in pyruvate decarboxylase (PDC)-mediated phenylacetylcarbinol (PAC) biotransformation with product separation remains limited. This study screened nine DESs using frozen-thawed whole cells (FT-WHC) of Candida magnoliae (C. mag.), followed by evaluation of water content and DES to phosphate buffer ratios. Glycerol-based DES resulted in significantly higher (p ≤ 0.05) volumetric PDC activity and PAC productivity than other DESs. Choline chloride: glycerol (ChCl: Gly (1:2) 20% (v/v) H2O) achieved the h

Magnolia kobus Extract Suppresses Porphyromonas gingivalis LPS-Induced Proinflammatory Cytokine and MMP Expression in HGF-1 Cells and Regulates Osteoclastogenesis in RANKL-Stimulated RAW264.7 Cells.

Clinical prevention is of utmost importance for the management of periodontal diseases. Periodontal disease starts with an inflammatory response in the gingival tissue, and results in alveolar bone destruction and subsequent tooth loss. This study aimed to confirm the anti-periodontitis effects of MKE. To confirm this, we studied its mechanism of action using qPCR and WB in LPS-treated HGF-1 cells and RANKL-induced osteoclasts. We found that MKE suppressed proinflammatory cytokine protein expression by inhibiting the TLR4/NF-κB pathway in LPS-PG-induced HGF-1 cells and blocking ECM degradation
