Continuous infusion of manganese improves contrast and reduces side effects in manganese-enhanced magnetic resonance imaging studies.

The ability to administer systemically high doses of manganese as contrast agent while circumventing its toxicity is of particular interest for exploratory MRI studies of the brain. Administering low doses either repeatedly or continuously over time has been shown to enable the acquisition of satisfactory MRI images of the mouse brain without apparent side effects. Here we have systematically compared the obtained MRI contrast and recorded potential systemic side effects such as stress response and muscle strength impairment in relation to the achieved contrast. We show in mice that administer

Increased manganese superoxide dismutase expression or treatment with manganese porphyrin potentiates dexamethasone-induced apoptosis in lymphoma cells.

Glucocorticoid-induced apoptosis is exploited for the treatment of hematologic malignancies. Innate and acquired resistance limits treatment efficacy; however, resistance mechanisms are not well understood. Previously, using WEHI7.2 murine thymic lymphoma cells, we found that increasing the resistance to hydrogen peroxide (H(2)O(2)) by catalase transfection or selection for H(2)O(2) resistance caused glucocorticoid resistance. This suggests the possibility that increasing H(2)O(2) sensitivity could sensitize the cells to glucocorticoids. In other cell types, increasing manganese superoxide dis

Coagulation mechanism of polyaluminum manganese chloride in alkaline conditions: Role of manganese in changing hydrolysates and coordination structure.

The development of highly efficient coagulants with broad pH adaptability remains a critical challenge in water treatment. In this study, a new type of polyaluminum chloride-manganese chloride composite coagulant (PAMCs) was synthesized, and the effect of manganese on changing the types of hydrolysis products and enhancing coagulation performance was systematically studied. A series of PAMCs with varying Al/Mn molar ratios and basicity (B = [OH-]/[M3+]) were prepared, and their interactions with humic acid (HA) were evaluated. Results demonstrated that PAMC2 (Al/Mn = 2:1, B = 2) exhibited supe

Evidence for a role of calcium and vitamin D in the regulation of manganese homeostasis and in the prevention or management of manganese-induced disease.

SLC30A10 mediates manganese (Mn) efflux in both the intestine and liver, protecting against Mn toxicity. In previous studies, we found that Slc30a10 is one of the genes most induced by 1,25(OH)2D3 in mouse intestine and human enteroids. In addition, in Slc30a10 knockout mice, we noted a significant decrease in Trpv6, a vitamin D target gene involved in intestinal calcium transport, indicating a link between Mn efflux transport in the intestine, calcium, and the vitamin D endocrine system. In this study, we examined the effect of varying levels of Mn in 3-wk-old male and female mice fed diets e

Phosphatidylinositol 3 kinase (PI3K) modulates manganese homeostasis and manganese-induced cell signaling in a murine striatal cell line.

In a recent study, we found that blocking the protein kinase ataxia telangiectasia mutated (ATM) with the small molecule inhibitor (SMI) KU-55933 can completely abrogate Mn-induced phosphorylation of p53 at serine 15 (p-p53) in human induced pluripotent stem cell (hiPSC)-differentiated striatal neuroprogenitors. However, in the immortalized mouse striatal progenitor cell line STHdhQ7/Q7, a concentration of KU55933 far exceeding its IC50 for ATM was required to inhibit Mn-induced p-p53. This suggested an alternative signaling system redundant with ATM kinase for activating p53 in this cell line

Nasal toxicity of manganese sulfate and manganese phosphate in young male rats following subchronic (13-week) inhalation exposure.

Growing evidence suggests that nasal deposition and transport along the olfactory nerve represents a route by which inhaled manganese and certain other metals are delivered to the rodent brain. The toxicological significance of olfactory transport of manganese remains poorly defined. In rats, repeated intranasal instillation of manganese chloride results in injury to the olfactory epithelium and neurotoxicity as evidenced by increased glial fibrillary acidic protein (GFAP) concentrations in olfactory bulb astrocytes. The purpose of the present study was to further characterize the nasal toxici

Long-term follow-up of workplace and well water manganese effects on iron status indexes in manganese miners.

The authors assessed the effect of water reconstitution in the workplace by evaluating the iron status of manganese mine workers during a long-term study. Subsequent analyses and biological monitoring were performed in a group of 150 manganese miners before, and 2.8 yr after, reconstitution of drinking water in the miners' workplace. The authors found significantly high concentrations of manganese in the workplace well water, as well as in the miners' blood, urine, and hair. There was a considerable prevalence of epithelial lesions, which resulted from iron deficiency, in the miners, compared

Tissue manganese concentrations and antioxidant enzyme activities in rats given total parenteral nutrition with and without supplemental manganese.

BACKGROUND: Manganese is an essential but potentially toxic mineral. Parenteral administration of manganese via total parenteral nutrition (TPN) bypasses homeostatic mechanisms (intestinal absorption and presystemic hepatic elimination). Our objective in this study was to determine the effect of supplemental manganese in TPN solutions on manganese status in a rat model. METHODS: Male Sprague-Dawley rats underwent jugular catheterization and were given 61.0 +/- 0.4 g/d TPN solution providing 0.5 +/- 0.2 nmol manganese/g (Mn-; n = 6) or 16 +/- 3 nmol manganese/g (Mn+; n = 7) for 7 days. Manganese Provision in Parenteral Nutrition: An Update.

Manganese (Mn) is an essential micronutrient required for the activity of metalloenzymes. It is an essential component of parenteral nutrition (PN), but requirements are low. Mn status is difficult to assess, with the commonest method being measurement of its concentration in whole blood. This method has limitations, including artifactually high concentrations resulting from contamination of specimen tubes. Mn toxicity is a well-recognized complication of PN, the risk of which increases if there is cholestasis or if the patient has received high doses. It usually presents with parkinsonian-lik

Competition for Manganese at the Host-Pathogen Interface.

Transition metals such as manganese are essential nutrients for both pathogen and host. Vertebrates exploit this necessity to combat invading microbes by restricting access to these critical nutrients, a defense known as nutritional immunity. During infection, the host uses several mechanisms to impose manganese limitation. These include removal of manganese from the phagolysosome, sequestration of extracellular manganese, and utilization of other metals to prevent bacterial acquisition of manganese. In order to cause disease, pathogens employ a variety of mechanisms that enable them to adapt

Manganese accumulation in the CNS and associated pathologies.

Manganese (Mn) is an essential metal for life. It is a key constituent of clue enzymes in the central nervous system, contributing to antioxidant defenses, energetic metabolism, ammonia detoxification, among other important functions. Until now, Mn transport mechanisms are partially understood; however, it is known that it shares some mechanisms of transport with iron. CNS is susceptible to Mn toxicity because it possesses mechanisms that allow Mn entry and favor its accumulation. Cases of occupational Mn exposure have been extensively reported in the literature; however, there are other ways

Effect of manganese exposure on MPTP neurotoxicities.

We used a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated mice model to evaluate whether manganese (Mn) exposure can affect MPTP-induced neurotoxicity. We randomly assigned adult male C57BL/6 mice (n=5-7 per group) the following treatments: SO, Mn(-) x MPTP(-); MO, Mn(+) x MPTP(-); SM, Mn(-) x MPTP(+); MM, Mn(+) x MPTP(+). Mn (MnCl(2).4H(2)O) was administered intraperitoneally at a dose of 2 mg/kg daily for 3 weeks. MPTP was then administered intraperitoneally at a dose of 30 mg/kg daily for 5 days in the SM and MM groups. Seven days after the last MPTP injection, the animals were

[Neuropsychologic effects of occupational exposure to manganese].

A broad use of manganese and its compounds in industry generates adverse health effects in workers exposed. When setting environmental exposure criteria, changes in the central nervous system are considered as a critical health effect. The author describes these changes and reviews the latest reports on empirical studies employing psychological methods in detecting preclinical neurotoxic effects of manganese. Results of neuropsychological studies carried out in different countries and in different occupational groups exposed to rather low manganese concentrations evidence the range of changes

Manganese, iron and lipid interactions in rats.

The interactive effects of manganese, iron and lipid on mineral status, manganese-dependent superoxide dismutase (MnSOD) activity and lipoprotein composition were investigated by feeding weanling rats two levels of manganese (0.4 or 56 micrograms Mn/g diet), two levels of iron (29 or 109 micrograms Fe/g diet) and either 12% high-linoleic acid safflower oil or 12% high-oleic acid safflower oil for 8 wk. Rats fed the manganese-deficient diets had decreased heart MnSOD activity; depressed tibia and kidney manganese concentrations; lowered plasma and high density lipoprotein (HDL) cholesterol, HDL

Hair manganese concentrations in newborns and their mothers.

The study was designed to investigate the manganese (Mn) status of mothers and their offspring at delivery. Hair Mn concentrations in 31 full-term, 18 preterm and 12 newborn infants with congenital malformations and their mothers were determined by the flameless atomic absorption technique. Both in infants with congenital malformations and their mothers, hair Mn levels were significantly lower than the full-term and preterm infant-mother pairs. With the exception of mothers of infants with congenital malformations, hair Mn concentrations in mothers were significantly higher as compared with th
