Low temperature triggers Ca2+ signalling and reprogramming of gene expression and metabolism in plants. However, how the Ca2+ signal is transduced to the downstream metabolic pathways remains unknown. The involvement of a cold-induced calmodulin-like protein, MfCML50, from Medicago falcata in regulation of cold tolerance was examined in the present study. The interaction of MfCML50 with isopiperitenol/carveol dehydrogenase (MfCDH) was identified, which was dependent upon Ca2+ and led to activated activity of MfCDH. MfCDH catalysed the production of carvone from carveol, with Km value of 10.25

Plants respond to herbivore or pathogen attacks by activating specific defense programs that include the production of bioactive specialized metabolites to eliminate or deter the attackers. Volatiles play an important role in the interaction of a plant with its environment. Through transcript profiling of jasmonate-elicited Medicago truncatula cells, we identified Emission of Methyl Anthranilate (EMA) 1, a MYB transcription factor that is involved in the emission of the volatile compound methyl anthranilate when expressed in M. truncatula hairy roots, giving them a fruity scent. RNA sequencing

BACKGROUND: Low temperature severely restricts plant growth and development. Medicago falcata is an important germplasm resource for alfalfa breeding because of its strong adaptability and tolerance to abiotic stresses, particularly low-temperature stress. MicroRNAs (miRNAs) are key regulators of stress-responsive genes and modulate plant adaptation to stress by directing transcript cleavage or translational repression. However, the roles of miRNAs in low-temperature response mechanisms in M. falcata remain largely unclear.
RESULTS: Here, we constructed miRNA libraries from M. falcata roots g

Drought constitutes a major abiotic stress limiting alfalfa productivity. To elucidate the genetic basis of drought tolerance, a genome-wide association study (GWAS) was conducted in alfalfa, which identified a CBF/DREB1 family transcription factor designated MsCBF8. Utilizing an optimized CRISPR-Cas9 system, mutants were successfully generated in the complex autotetraploid background. Combined with overexpression and Arabidopsis complementation assays, this confirmed that MsCBF8 functions as a positive regulator of drought tolerance, with its overexpression enhancing drought tolerance without

BACKGROUND: As an antioxidant, SO2 plays a critical role in maintaining the plant's redox balance. At the same time, Hg2+ as heavy metals, will accumulate in plants through soil, often disrupting physiological processes in plants. However, few studies have been reported to examine changes in Hg2+ and SO2 in plants. Here, we developed a dual-channel fluorescent probe NYD, which can differentiate between the detection of Hg2+ and SO2.
RESULTS: The probe exhibits high sensitivity (LOD, 44 nM for Hg2+ and 26 nM for SO2). When Hg2+ and HSO3- deprotect and attack the double bond by nucleophilic add

The objective of this study was to explore the microbiological factors that cause the difference in silage fermentation characteristics between grass and legume. Specifically, the effects of epiphytic microbiota from alfalfa, oat and Italian ryegrass on ensiling characteristics and microbial community of alfalfa were assessed. By γ-ray irradiation sterilization and microbiota transplantation technology, the sterile alfalfa was inoculated as follows: (i) aseptic water (STAL); (ii) epiphytic bacteria from alfalfa (ALAL); (iii) epiphytic bacteria from oat (ALOT); (iv) epiphytic bacteria from Ital

Legumes can meet their nitrogen requirements through root nodule symbiosis, which could also trigger plant systemic resistance against pests. The pea aphid Acyrthosiphon pisum, a legume pest, can harbour different facultative symbionts (FS) influencing various traits of their hosts. It is therefore worth determining if and how the symbionts of the plant and the aphid modulate their interaction. We used different pea aphid lines without FS or with a single one (Hamiltonella defensa, Regiella insecticola, Serratia symbiotica) to infest Medicago truncatula plants inoculated with Sinorhizobium mel

Alfalfa (Medicago sativa L.) is commonly used as a traditional medicine and functional food. This study investigated the anti-inflammatory potential of alfalfa and the mechanisms involved. The chloroform extract of alfalfa aerial parts inhibited lipopolysaccharide (LPS)-stimulated immune responses more than ether, butanol, or water soluble extracts. Treatment with 1 μg/mL LPS increased nitrite concentrations to 44.3 μM in RAW267.4 macrophages, but it was reduced to 10.6 μM by adding 100 μg/mL chloroform extract. LPS treatment also increased the concentrations of tumor necrosis factor-α, interl

The mean retention time (MRT) of stained hay through the whole digestive tract and its digestibility were measured in Japanese sika deer (Cervus nippon) and were compared with those in sheep when lucerne (Medicago sativa) hay was fed at 10, 20 and 30 g/kg body-weight. The recoveries in faeces of plastic particles with five specific gravities were also measured in deer. MRT for deer was significantly shorter than that for sheep at 10 and 20 g/kg feeding levels. The digestibilities of dry matter, organic matter, and neutral-detergent fibre were significantly lower for deer than for sheep at 30,

The essential oil of Medicago marina was subjected to analysis at vegetative and reproductive stages of development, with a particular emphasis on its chemical composition, α-amylase inhibition, and allelopathic activities. The results of the gas chromatography and molecular docking studies demonstrated alterations in oil yield and composition throughout the developmental stages. Before blooming, hydrocarbons were the predominant compounds, while monoterpenes were abundant during the reproductive stage, with higher concentrations of eugenol (8.86%) and β-ionone (17.67%) compared to earlier sta

The Aras Basin, located at the intersection of three major gene centers, represents one of the most important transition zones for the evolution of forage legumes. This study evaluates the genetic diversity and population structure of 74 Medicago sativa genotypes, including wild populations and commercial cultivars, using ISSR markers. The analysis revealed a broad level of genetic variability, reflecting the adaptive potential of alfalfa in this ecologically heterogeneous region. Population structure analyses consistently separated the germplasm into three genetic clusters, demonstrating clea

Arbuscular mycorrhizal fungi (AMF) are ubiquitous root symbionts that form common mycelial networks (CMN), linking multiple plants underground. CMN are thought to contribute to information exchange between plants and to neighbour-primed defences, but the transcriptomic and metabolomic responses of receiver plants connected through CMN remain poorly understood. In addition, the confounding effects of hyphal damage caused by CMN disconnection have not been clearly resolved. To assess the contribution of CMN integrity to neighbour-primed plant defences, we used the model AMF Rhizophagus irregular

Flowering of the reference legume Medicago truncatula is promoted by winter cold (vernalization) followed by long-day photoperiods (VLD) similar to winter annual Arabidopsis. However, Medicago lacks FLC and CO, key regulators of Arabidopsis VLD flowering. Most plants have two INHIBITOR OF GROWTH (ING) genes (ING1 and ING2), encoding proteins with an ING domain with two anti-parallel alpha-helices and a plant homeodomain (PHD) finger, but their genetic role has not been previously described. In Medicago, Mting1 gene-edited mutants developed and flowered normally, but an Mting2-1 Tnt1 insertion

Cadmium (Cd) is a highly toxic heavy metal that poses a serious threat to plant growth and the quality of agricultural and animal products. Therefore, elucidating the response and regulatory mechanisms of plants to Cd stress is essential for controlling the safety of agricultural and animal products at the source. In this study, we used a Cd-tolerant alfalfa cultivar, Medicago sativa 'Zhaodong' (ZD), and a Cd-sensitive cultivar, M. sativa 'Zhongmu No.1' (ZM), to investigate their differential responses to Cd stress in terms of morphology, photosynthetic function, and oxidative damage through c

The accumulation of cell wall components as plants grow limits forage nutrition for ruminants. In particular, lignin deposition in plant tissues impedes the breakdown of the cell wall carbohydrates cellulose and hemicellulose, thereby reducing the amount of energy available from forage. The highly digestible pectin component of the cell wall, however, is readily available for degradation. Information about lignin and pectin deposition in different stem cell types may provide mechanistic insights into the factors underlying variation in stem digestibility among alfalfa lines. In vitro incubatio

The infection of the model legume Medicago truncatula with Ralstonia solanacearum GMI1000 gives rise to bacterial wilt disease via colonisation of roots. The root and leaf responses to early infection (1 and 3 days post infection) were characterised to investigate the molecular mechanisms of plant resistance or susceptibility. A proteomics approach based on pools of susceptible and resistant recombinant inbred lines was used to specifically target the mechanisms for tolerance. Differential abundances were evidenced for proteins involved in defence (e.g., PR5, PR10, or Kunitz protease inhibitor

The pea aphid (Acyrthosiphon pisum) is a destructive phloem-feeding pest of alfalfa. Its overreliance on chemical insecticides necessitates the development of sustainable, plant-based resistance strategies. Plant flavonoids are important in induced defense, but their dynamic responses to aphid feeding and subsequent insecticidal effects remain unclear. Using the highly resistant alfalfa variety 'Gannong No. 5' (GN5), this study integrated behavioral assays, performance bioassays, untargeted and targeted metabolomics, and exogenous flavonoid feeding assays to investigate how pea aphid infestati

Soil salinization severely limits forage crop productivity, yet the regulatory networks that govern salt stress adaptation in alfalfa, a moderately salt-tolerant leguminous forage, remain largely unexplored. Here, we examined the physiological and transcriptomic dynamics of alfalfa leaves under 200 mM NaCl stress across three time points. Salt stress induced a progressive elevation of the Na+/K+ ratio, biphasic activation of antioxidant enzymes and concurrent accumulation of malondialdehyde. Time-course RNA-seq analysis identified 3631 differentially expressed genes (DEGs) and 132 core salt-re

Increasing evidence indicates that long non-coding RNAs (lncRNAs) play a regulatory role in plant responses to environmental stress. However, the epigenetic mechanisms behind the regulation of salt stress by lncRNAs remain largely elusive. We previously discovered that the lncRNA MtCIR2 of Medicago trnucatula negatively regulated seed germination under salt stress by altering endogenous ABA and GA concentrations via histone ubiquitination. Here we evaluated the roles of MtCIR2 in regulating response of M. truncatula seedlings to salt stress. We found that over-expression and mutation of MtCIR2

Fusarium sambucinum species complex (FSAMSC) includes not only major mycotoxin-producing pathogens but also non-pathogenic endophytic species that can promote plant growth and stress tolerance. However, the molecular mechanisms underlying these contrasting lifestyles remain poorly understood, underscoring the need for deeper molecular studies. A non-pathogenic endophytic Fusarium strain, TZ1MST22, isolated from Medicago sativa plants in the Tozeur region of Southern Tunisia, was sequenced using the Illumina NovaSeq 6000 sequencing technology. Here, we report a 39.8 Mb draft genome of TZ1MST22,
