Apolipoprotein A-I Mimetic Peptide L-4F Removes Bruch's Membrane Lipids in Aged Nonhuman Primates.

PURPOSE: Multiple evidence lines support Bruch's membrane lipid deposition as a major precursor of soft drusen and age-related macular degeneration as including a potentially treatable atherosclerosis-like progression in the subretinal pigment epithelium (RPE)-basal lamina space. We evaluated the effect of anti-inflammatory, antiatherogenic peptide L-4F on Bruch's membrane of aged nonhuman primates in a dose-escalating study. METHODS: Macaca fascicularis ≥20 years of age evaluated by color fundus photography and optical coherence tomography received monocular intravitreal injections of L-4F (

Lipid transfer proteins rectify inter-organelle flux and accurately deliver lipids at membrane contact sites.

The endoplasmic reticulum (ER) is the main center for the synthesis of various lipid types in cells, and newly synthesized lipids are delivered from the ER to other organelles. In the past decade, various lipid transfer proteins (LTPs) have been recognized as mediators of lipid transport from the ER to other organelles; inter-organelle transport occurs at membrane contact sites (MCSs) and in a nonvesicular manner. Although the intermembrane transfer reaction catalyzed by LTPs is an equilibrium reaction, various types of newly synthesized lipids are transported unidirectionally in cells. This r

Role of Membrane Lipids in the Regulation of Erythrocytic Oxygen-Transport Function in Cardiovascular Diseases.

The composition and condition of membrane lipids, the morphology of erythrocytes, and hemoglobin distribution were explored with the help of laser interference microscopy (LIM) and Raman spectroscopy. It is shown that patients with cardiovascular diseases (CVD) have significant changes in the composition of their phospholipids and the fatty acids of membrane lipids. Furthermore, the microviscosity of the membranes and morphology of the erythrocytes are altered causing disordered oxygen transport by hemoglobin. Basic therapy carried out with the use of antiaggregants, statins, antianginals, bet

CLN3P, the Batten disease protein, localizes to membrane lipid rafts (detergent-resistant membranes).

Juvenile neuronal ceroid lipofuscinosis is an inherited pediatric neurodegenerative disorder, which occurs as a result of mutations in the CLN3 gene that is located on chromosome 16p12.1. The encoded protein, CLN3P, is a putative transmembrane protein with no known function. In this study, we demonstrate that CLN3P resides on membrane lipid raft domains (detergent-resistant membranes) and provide important new data towards possible functions of the protein.

Contribution of membrane surface charge in the interaction of lead and tin derivatives with model lipid membrane.

The objective of this study was to determine the interaction of modified bilayer lipid membranes (BLMs) with lead and tin organoderivatives. The relative depolarization, of the lipid membrane, caused by organo metals, was used to estimate their activity. Dodecyltrimethylammonium bromide (TMDA) and sodium dodecylsulfonate (AS-12) were used to modify BLMs. The trialkylderivatives of tin (IV) were found to be the most active towards the lipid membrane, whereas dialkyltin (IV) and trialkyllead (IV) derivatives were less active. Also, a correlation exists between depolarization activity and the lip

Freeze-fracture study of cardiotoxin action on axonal membrane and axonal membrane lipid vesicles.

Freeze-fracture electron microscopy was used to follow morphological changes induced by Naja mossambica mossambica venom cardiotoxins on crab axonal membranes and thier lipids. It was shown that the extent of morphological changes depended drastically on the purity of cardiotoxin preparations and on their nature. Highly purified cardiotoxin induced mainly fusion of membrane or lipid vesicles. The extent of fusion and other morphological changes depended on the nature of cardiotoxin used: VII4 cardiotoxin induced only fusion while VII1 led to further modifications of membranes and liposomes. Ethephon interactions with biomimetic sperm plasma membranes and their effects on lipid packing and membrane stability.

Ethephon (ETP) is an organophosphorus plant growth regulator extensively used in agriculture, raising concerns about environmental exposure and potential reproductive toxicity. Although animal studies report reduced sperm count, morphological abnormalities, and testicular damage following ETP exposure, the molecular mechanisms underlying these effects remain poorly understood. Because membrane organization and stability are critical determinants of sperm function, we investigated simplified biomimetic sperm plasma membrane systems in the presence of ETP to elucidate membrane-level interaction

Lipid Remodeling and Metabolic Adaptation in Cancer Cell Membranes: A Platform for Membrane-Targeted Nanotherapy.

Metabolic reprogramming in cancer cells induces profound alterations in membrane lipid organization, affecting phospholipid distribution, surface charge, and overall lipid composition. These modifications contribute to key oncogenic processes, including tumor progression, immune evasion, and resistance to therapy. Importantly, they also give rise to distinct biophysical features that may serve as selective targets for therapeutic intervention. Among these, the externalization of anionic phospholipids and the overexpression of negatively charged glycoconjugates render cancer cell membranes part

Interaction of elastin-derived peptides with model lipid membranes.

Elastin-derived peptides (EDPs), generated during elastin fragmentation in aging and pathological conditions, are widely recognized for their biological activity through interactions with cell-surface receptors. However, despite their hydrophobic character and proximity to membrane-associated receptors, their capacity to directly interact with lipid bilayers has remained largely unexplored. In this study, we investigated the membrane-binding properties of representative elastin hexa and nonapeptides using a combination of molecular dynamics simulations, potential of mean force calculations, an

Lipid nanoparticle induced model endosomal membrane disruption.

Endosomal escape remains the central bottleneck limiting lipid nanoparticle (LNP)-mediated mRNA delivery. In this study, we use giant unilamellar vesicles (GUVs) mimicking the endosomal membrane to directly visualize the interaction and membrane disruptions caused by two model LNPs constituted by two different ionizable lipids: LP01 and BiP-20 (a Branched ionizable Phospholipid) that are both benchmark ionizable lipids for in vivo gene editing in liver. Despite identical TNS-measured apparent pKa (6.6) for both lipids in our conditions, LP01 LNP showed maximal membrane engagement at pH 5.8, wh

Lipid packing contributes to the confinement of caveolae to the plasma membrane.

Lipid packing is a fundamental characteristic of bilayer membranes. Yet, we lack detailed mechanistic understanding of how lipid packing directly affects membrane-associated cellular processes. Here, we address this by focusing on caveolae, small Ω-shaped invaginations of the plasma membrane, which serve as key regulators of cellular lipid sorting and mechano-responses. In addition to caveolae coat proteins, the lipid membrane is a core component of caveolae that critically impacts their biogenesis, morphology, and stability. We show that the small compound Dyngo-4a adsorbs and inserts into th

Calcium Reshapes Aβ Aggregation at Anionic Lipid Membranes.

Dysregulated calcium homeostasis is a hallmark of neurodegenerative disorders, such as Alzheimer's, Parkinson's, and Huntington's diseases. In Alzheimer's disease (AD), the aggregation of amyloid beta (Aβ) peptides at neuronal membranes is shown to be modulated by the presence of Ca2+ ions, yet the molecular mechanism by which Ca2+ reshapes Aβ aggregation at anionic membrane surfaces remains poorly understood. To address this knowledge gap, we employed coarse-grained molecular dynamics simulations to investigate the aggregation of the model amyloidogenic K16LVFFAE22 fragment of Aβ (Aβ16-22), o

The lipid bilayer strengthens the cooperative network of membrane proteins.

Membrane proteins fold and function in a lipid bilayer constituting cell membranes. Nonetheless, their structure and function can be recapitulated in diverse amphiphilic assemblies whose compositions deviate from native membranes. It remains unclear how various hydrophobic environments stabilize membrane proteins and whether lipids play any unique role in protein stability compared to other types of amphiphiles. Here, using the evolutionarily unrelated α-helical and β-barrel membrane proteins from Escherichia coli, we find that the hydrophobic thickness and the strength of amphiphile-amphiphil

Bacterial cell membrane models: choosing the lipid composition.

The reasons for the wide diversity of lipids found in natural cell membranes are still not fully understood but could potentially be exploited in treating disease and infection. This study aims to establish whether charge alone or specific chemical structure of an anionic lipid headgroup determines the structure and properties of model bacterial cell membranes. We compare different compositions of a zwitterionic lipid di-myristoyl phosphatidylethanolamine (DMPE) and two anionic lipids, di-myristoyl phosphatidylglycerol (DMPG) and tetra-myristoyl cardiolipin (TMCL). TMCL has a distinct condensi

Membrane lipid raft homeostasis is directly linked to neurodegeneration.

Age-associated neurodegenerative diseases such as amyotrophic lateral sclerosis (ALS), Parkinson's disease (PD) and Alzheimer's disease (AD) are an unmet health need, with significant economic and societal implications, and an ever-increasing prevalence. Membrane lipid rafts (MLRs) are specialised plasma membrane microdomains that provide a platform for intracellular trafficking and signal transduction, particularly within neurons. Dysregulation of MLRs leads to disruption of neurotrophic signalling and excessive apoptosis which mirrors the final common pathway for neuronal death in ALS, PD an

Electropermeabilization does not correlate with plasma membrane lipid oxidation.

The permeabilized condition of the cell membrane after electroporation can last minutes but the underlying mechanisms remain elusive. Previous studies suggest that lipid peroxidation could be responsible for the lasting leaky state of the membrane. The present study aims to link oxidation within the plasma membrane of live cells to permeabilization by electric pulses. We have introduced a method for the detection of oxidation by ratiometric fluorescence measurements of BODIPY-C11 dye using total internal reflection fluorescence (TIRF) microscopy, limiting the signal to the cell membrane. A Blue-Light-Emitting BODIPY Probe for Lipid Membranes.

Here we describe a new BODIPY-based membrane probe (1) that provides an alternative to dialkylcarbocyanine dyes, such as DiI-C18, that can be excited in the blue spectral region. Compound 1 has unbranched octadecyl chains at the 3,5-positions and a meso-amino function. In organic solvents, the absorption and emission maxima of 1 are determined mainly by solvent acidity and dipolarity. The fluorescence quantum yield is high and reaches 0.93 in 2-propanol. The fluorescence decays are well fitted with a single-exponential in pure solvents and in small and giant unilamellar vesicles (GUV) with a l

Interaction of miltefosine with the lipid and protein components of the erythrocyte membrane.

Miltefosine (MT) is an alkylphospholipid that has been approved for the treatment of breast cancer metastasis and visceral leishmaniasis, although its mechanism of action remains poorly understood. Electron paramagnetic resonance spectroscopy of a spin-labeled lipid and a thiol-specific spin label showed that MT causes an increase in the molecular dynamics of erythrocyte ghost membranes and detergent-resistant membranes (DRMs) prepared from erythrocyte ghosts. In the vesicles of lipid raft constituents, it was shown that 20 mol % sphingomyelin could be replaced by 20 mol % MT with no change in
