The Memory Binding Test Detects Early Subtle Episodic Memory Decline in Preclinical Alzheimer's Disease: A Longitudinal Study.

BACKGROUND: The asymptomatic at-risk phase might be the optimal time-window to establish clinically meaningful endpoints in Alzheimer's disease (AD). OBJECTIVE: We investigated whether, compared with the Free and Cued Selective Reminding Test (FCSRT), the Memory Binding Test (MBT) can anticipate the diagnosis of emergent subtle episodic memory (EM) deficits to an at-risk phase. METHODS: Five-year longitudinal FCSRT and MBT scores from 45 individuals matched for age, education, and gender, were divided into 3 groups of 15 subjects: Aβ-/controls, Aβ+/stable, and Aβ+/progressors (preclinical-AD

The memory trace of an intrusive trauma-analog episode.

Intrusive memories are a core symptom of posttraumatic stress disorder. Compared with memories of everyday events, they are characterized by several seemingly contradictory features: intrusive memories contain distinct sensory and emotional details of the traumatic event and can be triggered by various perceptually similar cues, but they are poorly integrated into conceptual memory. Here, we conduct exploratory whole-brain analyses to investigate the neural representations of trauma-analog experiences and how they are reactivated during memory intrusions. We show that trauma-analog movies indu

Episodic memories in anxiety disorders: clinical implications.

The aim of this review is to summarize research on the emerging role of episodic memories in the context of anxiety disorders (AD). The available literature on explicit, autobiographical, and episodic memory function in AD including neuroimaging studies is critically discussed. We describe the methodological diversity of episodic memory research in AD and discuss the need for novel tests to measure episodic memory in a clinical setting. We argue that alterations in episodic memory functions might contribute to the etiology of AD. We further explain why future research on the interplay between

Induction and measurement of episodic memories in healthy adults.

Episodic memory is that part of our memory which enables us to memorize a personal event (what) in terms of place (where) and time (when). It is expressed on verbal and behavioral level. In this report we describe a novel method to induce de novo episodic memories in humans for unique events in the laboratory and measure them with standardized ratings and non-verbal motor responses. Visual stimuli were hidden in four out of eight quadrants on a computer screen. Each of these quadrants could be highlighted by pushing a corresponding button on a keyboard which either revealed a visual stimulus f

Maintaining and regaining episodic memory in Alzheimer disease: a circuit-based perspective.

Losing track of personal experiences is a defining feature of Alzheimer disease (AD), arising from the spread of AD pathology through the brain circuits that support episodic memory. In this Review, we explore strategies to improve the function of episodic memory circuits in AD by leveraging the optimized use of neural resources. We introduce the circuit utilization framework, which builds on evidence that synaptic dysfunction, maladaptive responses and deficient adaptive plasticity contribute to episodic memory impairment. The circuit utilization framework posits that by optimizing the utiliz

Is the Papez circuit the location of the elusive episodic memory engram?

All of the brain structures and white matter that make up Papez' circuit, as well as the circuit as a whole, are implicated in the literature in episodic memory formation and recall. This paper shows that Papez' circuit has the detailed structure and connectivity that is evidently required to support the episodic memory engram, and that identifying Papez' circuit as the location of the engram answers a number of long-standing questions regarding the role of medial temporal lobe structures in episodic memory. The paper then shows that the process by which the episodic memory engram may be forme

Effects of word presentation during treadmill walking on episodic memory and gait.

Entrainment emerges when oscillatory movements synchronize with environmental stimuli processing. The purpose of this experiment was to assess how cognitive-motor entrainment during a dual-task would influence the quality of gait and affect episodic long-term memory. Twenty-one participants (22.56 y/o; 64 % F) walked at preferred paces while listening to 40-item word lists. In separate sessions, unique word lists were presented predictably on every fourth stride, unpredictably related to stepping, or predictably while standing. Memory tests administered 24-hr after encoding revealed that predi

Sex differences in Parkinson disease-associated episodic memory and processing speed deficits.

OBJECTIVES: This study aims to address a gap in the data on cognitive sex differences in persons living with Parkinson disease (PD). There is some evidence that cognitive dysfunction is more severe in male PD, however data on episodic memory and processing speed is incomplete. METHODS: One hundred and sixty-seven individuals with a diagnosis of PD were included in this study. Fifty-six of those individuals identified as female. The California Verbal Learning Test 1st edition and the Wechsler Memory Scale 3rd edition were used to evaluate verbal and visuospatial episodic memory and the Wechsle

Mapping multidimensional content representations to neural and behavioral expressions of episodic memory.

Human neuroimaging studies have shown that the contents of episodic memories are represented in distributed patterns of neural activity. However, these studies have mostly been limited to decoding simple, unidimensional properties of stimuli. Semantic encoding models, in contrast, offer a means for characterizing the rich, multidimensional information that comprises episodic memories. Here, we extensively sampled four human fMRI subjects to build semantic encoding models and then applied these models to reconstruct content from natural scene images as they were viewed and recalled from memory.

[Episodic memory and Alzheimer's disease: evolution of theories and clinical assessment tools].

Alzheimer's disease is characterized by an impairment of episodic memory in the early stages of the disease. Neuropsychological evaluation is performed for diagnostic purposes and to personalize follow-up. However, although many tests have been developed over the last few decades, not all of them seem to be able to meet this dual challenge. Through a review of the evolution of the concept of episodic memory and of knowledge about the disease, we discuss how neuropsychological tools have adapted. We question the interests and limits of existing tools for patients with Alzheimer's disease. At th

The anterior thalamic nuclei: core components of a tripartite episodic memory system.

Standard models of episodic memory focus on hippocampal-parahippocampal interactions, with the neocortex supplying sensory information and providing a final repository of mnemonic representations. However, recent advances have shown that other regions make distinct and equally critical contributions to memory. In particular, there is growing evidence that the anterior thalamic nuclei have a number of key cognitive functions that support episodic memory. In this article, we describe these findings and argue for a core, tripartite memory system, comprising a 'temporal lobe' stream (centred on th

Gene-expression correlates of the oscillatory signatures supporting human episodic memory encoding.

In humans, brain oscillations support critical features of memory formation. However, understanding the molecular mechanisms underlying this activity remains a major challenge. Here, we measured memory-sensitive oscillations using intracranial electroencephalography recordings from the temporal cortex of patients performing an episodic memory task. When these patients subsequently underwent resection, we employed transcriptomics on the temporal cortex to link gene expression with brain oscillations and identified genes correlated with oscillatory signatures of memory formation across six frequ

Relationship between brain AD biomarkers and episodic memory performance in healthy aging.

The presence of brain biomarkers can be observed decades before the first clinical symptoms of Alzheimer's disease (AD). We aimed to determine whether associations between biomarkers and episodic memory performance already exist in a healthy late middle-aged population or only in participants over 60 years old. Performance at the Free and Cued Selective Reminding Test [FCSRT], the Logical Memory test and the Mnemonic Similarity Task [MST] was determined in sixty healthy participants (50-70 y.) with a negative status for amyloid-beta (Aβ) biomarker. We assessed Aβ cortical level and tau/neuroin

Autobiographical Memory and Episodic Specificity Across Different Affective States in Bipolar Disorder.

Autobiographical memory is essential to ground a sense of self-identity, contributing to social functioning and the development of future plans, and being an essential source for the psychiatric interview. Previous studies have suggested loss of autobiographical episodic specificity in unipolar depression, but relatively fewer investigations have been conducted in bipolar disorder (BD) patients, particularly across different mood states. Similarly, there is a scarcity of systematic investigations about mood-congruent and mood-dependent memory in relation to autobiographical memory in BD. Consi

The ventral lateral parietal cortex in episodic memory: From content to attribution.

The ventral lateral parietal cortex (VLPC) shows robust activation during episodic retrieval, and is involved in content representation, as well as in the evaluation of memory traces. This suggests that the VLPC has a crucial contribution to the quality of recollection and the subjective experience of remembering, and situates it at the intersection of the core and attribution systems.

Reduced Fidelity of Neural Representation Underlies Episodic Memory Decline in Normal Aging.

Emerging studies have emphasized the importance of the fidelity of cortical representation in forming enduring episodic memory. No study, however, has examined whether there are age-related reductions in representation fidelity that can explain memory declines in normal aging. Using functional MRI and multivariate pattern analysis, we found that older adults showed reduced representation fidelity in the visual cortex, which accounted for their decreased memory performance even after controlling for the contribution of reduced activation level. This reduced fidelity was specifically due to olde

Pauses During Autobiographical Discourse Reflect Episodic Memory Processes in Early Alzheimer's Disease.

There is a large body of research on discourse production in Alzheimer's disease (AD). Some studies have focused on pause production, revealing that patients make extensive use of pauses during speech. This has been attributed to lexical retrieval difficulties, but pausing may also reflect other forms of cognitive impairment as it increases with cognitive load. The aim of the present study was to analyze autobiographical discourse impairment in AD from a broad perspective, looking at pausing behavior (frequency, duration, and location). Our first objective was to characterize discourse changes

Differential episodic and semantic memory performance in Alzheimer's disease and vascular dementias.

Differential performance on measures of episodic and semantic memory were examined in AD, cortical vascular dementia (CVaD), subcortica] vascular dementia (SVaD), and controls. Groups were matched on age, education, and gender; dementia groups also were matched on severity. Recognition/retrieval differences were found only between SVaD and AD groups, not between CVaD and AD. Thus, recognition/retrieval differences are likely secondary to subcortical pathology rather than to vascular etiology per se. Similarly, significant numbers of memory errors were associated with cortical pathology, regard
