Gonadal morphogenesis and sexual differentiation in rhea (Rhea americana).

The rhea (Rhea americana) is a docile and highly adaptable ratite with notable zootechnical potential for the production of meat, eggs, feathers, and leather. Beyond its economic relevance, captive breeding plays an important role in conservation. However, information on its embryonic and gonadal development remains limited-knowledge that is essential for improving reproductive management. This study aimed to characterize gonadal differentiation during artificial incubation, focusing on developmental stages at days 9, 12, 15, 18, 21, and 24. We provide a detailed morphological and cytological

Anti-Müllerian Hormone Is Required for Chicken Embryonic Urogenital System Growth but Not Sexual Differentiation.

In mammals, the primary role of anti-Müllerian hormone (AMH) during development is the regression of Müllerian ducts in males. These structures otherwise develop into fallopian tubes, oviducts, and upper vagina, as in females. This highly conserved function is retained in birds and is supported by the high levels of AMH expression in developing testes. In mammals, AMH expression is controlled partly by the transcription factor, SOX9. However, in the chicken, AMH mRNA expression precedes that of SOX9 , leading to the view that AMH may lie upstream of SOX9 and play a more central role in avian t

SOX9 has both conserved and novel roles in marsupial sexual differentiation.

In addition to an essential role in chondrogenesis, SOX9 is a highly conserved and integral part of the testis determining pathway in human and mouse. To determine whether SOX9 is involved in sex determination in noneutherian mammals we cloned a marsupial orthologue and studied its expression. The tammar wallaby SOX9 gene proved to be highly conserved, and maps to a region of the tammar genome syntenic to human chromosome 17. Marsupial SOX9 transcripts were detected by RT-PCR in the developing limb buds and both the developing ovary and testis from the first sign of gonadal development through

Six4, a putative myogenin gene regulator, is not essential for mouse embryonal development.

Six4 is a member of the Six family genes, homologues of Drosophila melanogaster sine oculis. The gene is thought to be involved in neurogenesis, myogenesis, and development of other organs, based on its specific expression in certain neuronal cells of the developing embryo and in adult skeletal muscles. To elucidate the biological roles of Six4, we generated Six4-deficient mice by replacing the Six homologous region and homeobox by the beta-galactosidase gene. 5-Bromo-4-chloro-3-indolyl-beta-D-galactopyranoside staining of the heterozygous mutant embryos revealed expression of Six4 in cranial

Incidental mesonephric remnant hyperplasia of the jejunal mesentery: A diagnostic challenge.

Mesonephric remnants are embryonic vestiges of the mesonephric (Wolffian) ducts which regress during normal development. These remnants have been uncommonly reported in the female and male reproductive tract as a spectrum of morphologic lesions that can be misdiagnosed as carcinoma. One case of mesonephric remnant hyperplasia of the jejunal mesentery incidentally found in a 47-year-old man is herein reported. This is the first description of mesonephric hyperplasia arisen in the mesentery. The presence of ducts, tubules, and cysts lined by bland, epithelial, cuboidal cells with scant cytoplasm

Mesonephric remnants, hyperplasia, and neoplasia in the uterine cervix. A study of 49 cases.

Hyperplasia of mesonephric remnants in the uterine cervix is an unusual condition that is often misdiagnosed as adenocarcinoma. The rare mesonephric carcinoma can be confused with more common forms of cervical adenocarcinoma. We studied 49 specimens of cervix containing mesonephric remnants, or lesions derived from them, in women 21 to 72 (mean, 38) years of age. Four cases were classified as mesonephric remnants, 31 as lobular mesonephric hyperplasia, eight as diffuse mesonephric hyperplasia, two as mesonephric ductal hyperplasia, and four as mesonephric carcinoma. In the nonneoplastic cases,

Steroidogenesis during prenatal testicular development in Spix's cavy Galea spixii.

Spix's cavy is a potentially good experimental model for research on reproductive biology and sexual development. The aim of the present study was to evaluate the ontogeny of the steroidogenic enzymes involved in testicular androgen synthesis during prenatal development. Testes were investigated on Days 25, 30, 40 and >50 of gestation. Immunohistochemistry and immunoblotting were used to establish the site and relative amount of androgenic enzymes, including 5α-reductase, cytosolic 17β-hydroxysteroid dehydrogenase (17β-HSDI) and mitochondrial microsomal 3β-hydroxysteroid dehydrogenase (3β-HSDI

Two distinct ontogenies confer heterogeneity to mouse brain microglia.

Hoxb8 mutant mice show compulsive behavior similar to trichotillomania, a human obsessive-compulsive-spectrum disorder. The only Hoxb8 lineage-labeled cells in the brains of mice are microglia, suggesting that defective Hoxb8 microglia caused the disorder. What is the source of the Hoxb8 microglia? It has been posited that all microglia progenitors arise at embryonic day (E) 7.5 during yolk sac hematopoiesis, and colonize the brain at E9.5. In contrast, we show the presence of two microglia subpopulations: canonical, non-Hoxb8 microglia and Hoxb8 microglia. Unlike non-Hoxb8 microglia, Hoxb8 mi

Morphogenesis of the gubernaculum testis in Sprague-Dawley rat. Preliminary report.

The morphology of testicular descent in the Sprague-Dawley rat from day 14 of gestation to newborn was studied; the purpose was to analyse the anatomy and related observations made with light and scanning microscopy. Adult female rats were mated during estrus. The presence of spermatozoa in vaginal smears was taken to indicate day 0 of gestation. Animals were kept in a controlled 12-hour light-darkness cycle; food and water were provided ad libitum. At day 14 genital ducts are similar in both sexes. At day 15 and 16 the gubernaculum testis is seen as a mesenchymal cord that extends from mesone

[Research Progress of Single-cell Transcriptome Sequencing Technology in Physiological Hematopoiesis--Review].

Hematopoiesis starts from the embryo and runs through the entire life of a living body, which is a multi-stage and complex dynamic process that is regulated by multiple pathways, involving a variety of cells and hematopoietic anatomical locations. During the development of the mammalian hematopoietic system, the currently known hematopoietic anatomical locations mainly include yolk sac, aorta-gonad-mesonephros, fetal liver, bone marrow, and thymus. The first three are mainly responsible for hematopoiesis during the embryonic and fetal period, while bone marrow is the main place for postnatal h

Role of Vitamin A/Retinoic Acid in Regulation of Embryonic and Adult Hematopoiesis.

Vitamin A is an essential micronutrient throughout life. Its physiologically active metabolite retinoic acid (RA), acting through nuclear retinoic acid receptors (RARs), is a potent regulator of patterning during embryonic development, as well as being necessary for adult tissue homeostasis. Vitamin A deficiency during pregnancy increases risk of maternal night blindness and anemia and may be a cause of congenital malformations. Childhood Vitamin A deficiency can cause xerophthalmia, lower resistance to infection and increased risk of mortality. RA signaling appears to be essential for express

Spatiotemporally primed coelomic epithelium and supporting-like cells establish the dual origin of granulosa cells.

Ovarian organogenesis relies on the coordinated specification of supporting and steroidogenic lineages from multipotent progenitors of the coelomic epithelium. However, how these progenitors adopt distinct fates and differentiate into pregranulosa cells and steroidogenic progenitors of theca cells remain poorly understood. Here, we show that the dynamics of ovarian somatic lineage specification are conserved between human and mouse. Coelomic epithelial cells covering the fetal ovaries are heterogeneous and already biased toward supporting or steroidogenic fates. This priming is spatially and t

The role of apoptosis in the development of AGM hematopoietic stem cells revealed by Bcl-2 overexpression.

Apoptosis is an essential process in embryonic tissue remodeling and adult tissue homeostasis. Within the adult hematopoietic system, it allows for tight regulation of hematopoietic cell subsets. Previously, it was shown that B-cell leukemia 2 (Bcl-2) overexpression in the adult increases the viability and activity of hematopoietic cells under normal and/or stressful conditions. However, a role for apoptosis in the embryonic hematopoietic system has not yet been established. Since the first hematopoietic stem cells (HSCs) are generated within the aortagonad-mesonephros (AGM; an actively remode

Histological and immunohistochemical analysis of gonadal development in the corn snake (Pantherophis guttatus).

Snakes are a group of vertebrates with many unique features, such as significant body elongation, asymmetric positioning of viscera, and the loss of limbs. They belong to the squamate reptiles, one of the vertebrate groups with the most poorly studied gonadogenesis. This study presents the results of histological analyses on gonadal development in embryos of the corn snake (Pantherophis guttatus), covering the period from the initial formation of gonadal primordia, through the phase of undifferentiated gonads, sexual differentiation, to the establishment of the structures of testes and ovaries

The Development of the Human Female Reproductive Tract. Part 1: Uterine Tube and Uterus.

The uterine tubes and uterus develop from the paramesonephric (Müllerian) ducts. Most experimental data are obtained in rodents. Since the (micro-)anatomy of the murine urogenital tract differs from that in humans, evaluation of the translatability of mouse data to human development is relevant. We studied the Müllerian ducts in serially sectioned female human embryos and fetuses between 5 and 15 weeks of development and prepared 3D-reconstructions to establish topographic relations. At 5 weeks of development, the dorsocranial peritoneal epithelium thickens locally to form a placode-like struc

Renal development: a complex process dependent on inductive interaction.

Renal development begins in-utero and continues throughout childhood. Almost one-third of all developmental anomalies include structural or functional abnormalities of the urinary tract. There are three main phases of in-utero renal development: Pronephros, Mesonephros and Metanephros. Within three weeks of gestation, paired pronephri appear. A series of tubules called nephrotomes fuse with the pronephric duct. The pronephros elongates and induces the nearby mesoderm, forming the mesonephric (Woffian) duct. The metanephros is the precursor of the mature kidney that originates from the ureteric

Prenatal development of the bovine oviduct.

In this study the development of the bovine Fallopian tube was investigated using light microscopic methods. Formation and differentiation of the Müllerian duct were studied in mesonephroi of 16 embryos and fetuses with a crown-rump lengths (CRL) of 0.9-8.4 cm. The funnel field, the rostral beginning of the Müllerian duct was first observed at a CRL of 0.9 cm. It appears as a thickening of the mesothelium on the craniolateral side of the mesonephros. During later development the Müllerian duct emerges by caudal outgrowth from the funnel field. Formation of a common basal lamina surrounding the

Expression of the Wilms' tumour gene WT1 in the developing human and in paediatric renal tumours: an immunohistochemical study.

AIMS: The Wilms' tumour gene (WT1) product is expressed during the development of the urogenital system. This study was undertaken to evaluate four anti-WT1 antibodies and use the most specific one to examine the expression of WT1 in formalin fixed, paraffin wax embedded tissues from human embryos, fetuses, and paediatric renal neoplasms. METHODS: The antibodies were assessed on paraffin sections of fetal kidney and by western blotting. Immunohistochemical techniques were optimised and performed on a range of embryonic, fetal, and infant tissues from 35 days post-conception to three months of

Pax-2 controls multiple steps of urogenital development.

Urogenital system development in mammals requires the coordinated differentiation of two distinct tissues, the ductal epithelium and the nephrogenic mesenchyme, both derived from the intermediate mesoderm of the early embryo. The former give rise to the genital tracts, ureters and kidney collecting duct system, whereas mesenchymal components undergo epithelial transformation to form nephrons in both the mesonephric (embryonic) and metanephric (definitive) kidney. Pax-2 is a transcriptional regulator of the paired-box family and is widely expressed during the development of both ductal and mese
