Potassium mediates photosynthetic efficiency in tomato through genetic regulation rather than anatomical variation.

Potassium (K) is classically viewed as a guard-cell osmoticum, yet its influence on the photosynthetic efficiency under steady-state and fluctuating light remains unresolved. We combined gas exchange, cellular anatomy, and transcriptomic profiling to explore how K deficiency limits CO2 assimilation (AN) in tomato (Solanum lycopersicum). Contrasting our expectations, the decline of AN and mesophyll conductance (gm) under K deficiency in tomato was not attributed to alternation of leaf and cellular anatomy but was linked to downregulation of carbonic anhydrases and plasma-membrane aquaporins tha

Phenotypic plasticity in Passiflora suberosa L.(Passifloraceae): induction and reversion of two morphs by variation in light intensity.

Leaf morphology may vary considerably even within a branch of Passiflora suberosa plants. Leaves are of a typical green type in shaded areas, but in open fields turn into violet, and apparently have greater thickness and trichome density. The proximate causes and the adaptive meaning, if any, for the existence of the violet morph are still unknown. By cultivating P. suberosa clones under two light regimes (total and partial exposure to sunlight), we consecutively induced (first year) and then reversed (second year) the appearance of the violet morph. We evaluated the corresponding changes in m

Single-cell transcriptomic analysis of pea shoot development and cell-type-specific responses to boron deficiency.

Understanding how nutrient stress impacts plant growth is fundamentally important to the development of approaches to improve crop production under nutrient limitation. Here we applied single-cell RNA sequencing to shoot apices of Pisum sativum grown under boron (B) deficiency. We identified up to 15 cell clusters based on the clustering of gene expression profiles and verified cell identity with cell-type-specific marker gene expression. Different cell types responded differently to B deficiency. Specifically, the expression of photosynthetic genes in mesophyll cells (MCs) was down-regulated

Identification of promoter elements involved in the cytosolic Ca(2+)-mediated photoregulation of maize cab-m1 expression.

Changes in cytoplasmic Ca2+ levels are involved in the regulation of several plant genes. However, to our knowledge, no regions of genes or specific cis elements have been shown to be involved in the regulation of plant gene expression by cytosolic Ca2+ signaling. The maize (Zea mays) gene cab-m1, which encodes a light-harvesting chlorophyll a/b-binding apoprotein, is positively photoregulated in mesophyll cells (MC) but not in bundle-sheath cells (BSC). This gene is highly preferentially expressed in maize MC versus BSC. In situ transient expression assays have revealed that exposure of tissu

Cr-induced cellular injury and necrosis in Glycine max L.: Biochemical mechanism of oxidative damage in chloroplast.

Chromium-induced toxicity and mechanisms of cell death involved in plants are yet to be fully elucidated. To understand the events of these processes, the stress response of the soybean plant using trivalent and hexavalent chromium compounds, namely, basic chromium sulphate (BCS) and potassium dichromate (K2Cr2O7) was investigated. The leaf surface morphology for stomatal aperture, wax deposition and presence of trichomes for chromium accumulation was examined by SEM-EDAX and light microscopy. The leaf mesophyll cell integrity was identified by trypan blue staining; chlorophyll autofluorescenc

Microscopic and physicochemical evaluation of Ruta angustifolia leaves.

BACKGROUND: There are thousands of species of known medicinal plants in the world. Ruta angustifolia L. has been widely used in traditional medication for jaundice and liver disease. Previous studies have shown that R. angustifolia leaves can inhibit the hepatitis C virus in Huhit culture cells, reduce the value of NS3 protein, and possess a synergistic effect in combination with antiviral drugs. Therefore, this plant is potential to be developed as a drug candidate. Characteristics of plants including microscopic, physicochemical properties, and phytochemical profiles are necessary informatio

Anoxia-induced elevation of cytosolic Ca2+ concentration depends on different Ca2+ sources in rice and wheat protoplasts.

The anoxia-dependent elevation of cytosolic Ca(2+) concentration, [Ca(2+)](cyt), was investigated in plants differing in tolerance to hypoxia. The [Ca(2+)](cyt) was measured by fluorescence microscopy in single protoplasts loaded with the calcium-fluoroprobe Fura 2-AM. Imposition of anoxia led to a fast (within 3 min) significant elevation of [Ca(2+)](cyt) in rice leaf protoplasts. A tenfold drop in the external Ca(2+) concentration (to 0.1 mM) resulted in considerable decrease of the [Ca(2+)](cyt) shift. Rice root protoplasts reacted upon anoxia with higher amplitude. Addition of plasma membr

Derrislongiracemosa (Fabaceae), a new species from Thailand with extraordinary limestone adaptations and the longest inflorescences ever recorded.

A new species of Derris Lour. (Fabaceae), Derrislongiracemosa Boonprajan & Sirich., sp. nov., is described as the species bearing the longest inflorescences recorded in the genus to date, reaching up to 155 cm in length. Endemic to limestone areas in southwestern Thailand, it is possibly the third known limestone-adapted Derris species. Compared with its closest morphological relative and a partially sympatric species, D.solorioides, D.longiracemosa exhibits a longer and differently structured inflorescence, lower numbers of ovules per ovary (fewer than five ovules), and flowers that turn pink

The Use of Fullerenol as a Seed Treatment to Maintain the Intensity of Physiological and Biochemical Processes of Wheat in Control and Low-Temperature Conditions.

It is of utmost importance to investigate the effect of carbon-based nanostructures on plants in order to fully unlock their potential for enhancing productivity and stress tolerance. In this study, a comprehensive assessment of the effect of pre-sowing treatment of wheat seeds with fullerenol (PHF [C60(OH)24], 0.1 mg/L) solution on a number of physiological and biochemical parameters in seedlings under optimal and low temperature (LT, 4 °C, 5 days) conditions was carried out. Seedlings primed with PHF differed from control variant in accelerated growth (by 27-34%), increased biomass (by 10%),

Variation in photosynthetic capacity of Sorbus rehderiana and Quercus aquifolioides along elevational gradients on the eastern edge of the Qinghai-Tibetan Plateau, China.

Elevation provides a critical environmental context for studying the photosynthetic adaptation mechanisms of plants and is of great significance for understanding plant responses and acclimation to environmental change. Although elevational variation strongly influences photosynthetic capacity, the underlying regulatory mechanisms remain insufficiently understood. This study addresses this gap by systematically examining the photosynthetic and anatomical adaptations of two dominant tree species with contrasting functional types, Sorbus rehderiana Koehne (deciduous broad-leaved tree) and Quercu

The MRE11-ATM-SOG1 DNA damage signaling pathway confers rice immunity to Xanthomonas oryzae.

Plants are constantly exposed to microbial pathogens in the environment. One branch of innate plant immunity is mediated by cell-membrane-localized receptors, but less is known about associations between DNA damage and plant immune responses. Here, we show that rice (Oryza sativa) mesophyll cells are prone to DNA double-stranded breaks (DSBs) in response to ZJ173, a strain of Xanthomonas oryzae pv. oryzae (Xoo). The DSB signal transducer ataxia telangiectasia mutated (ATM), but not the ATM and Rad3-related branch, confers resistance against Xoo. Mechanistically, the MRE11-ATM module phosphoryl

Machine Learning-Guided Stress Atlases Reveal Co-Expression Rewiring and Divergent Cellular Deployment of Abiotic Stress Programs in Rice and Wheat.

Abiotic stress limits cereal productivity, yet whether conserved stress-responsive genes retain similar transcriptional network organization and cellular deployment across cereal species remains unknown. Here, we developed a machine learning-guided comparative framework to integrate public leaf transcriptomes of rice (Oryza sativa) and wheat (Triticum aestivum) under major abiotic perturbations. Using harmonized compendia comprising 787 rice and 337 wheat samples, we found that rice samples resolved into more discrete stress-associated transcriptional states, whereas wheat samples formed a mor

AtWRKY22 promotes susceptibility to aphids and modulates salicylic acid and jasmonic acid signalling.

Aphids induce many transcriptional perturbations in their host plants, but the signalling cascades responsible and the effects on plant resistance are largely unknown. Through a genome-wide association (GWA) mapping study in Arabidopsis thaliana, we identified WRKY22 as a candidate gene associated with feeding behaviour of the green peach aphid, Myzus persicae The transcription factor WRKY22 is known to be involved in pathogen-triggered immunity, and WRKY22 gene expression has been shown to be induced by aphids. Assessment of aphid population development and feeding behaviour on knockout mutan

Single-cell transcriptomics and DNA methylomics reveal cell type-specific heat stress response mechanisms in alfalfa.

Heat stress severely limits crop yield and threatens global food security; it also significantly influences the growth and development of alfalfa. Although there have been some studies on heat stress at the organ and tissue levels, the heat stress response mechanisms on the cell type level still remain unclear in alfalfa. Here, single-nucleus RNA sequencing (snRNA-seq) was performed on 61,835 high-quality leaf cells under control and heat-stressed conditions (1 and 48 h), revealing distinct transcriptional responses across 5 major cell types identified from 17 cell clusters. A total of 5,098 d

Whiteflies glycosylate salicylic acid and secrete the conjugate via their honeydew.

During insect feeding, a complex interaction takes place at the feeding site, with plants deciphering molecular information associated with the feeding herbivore, resulting in the upregulation of the appropriate defenses, and the herbivore avoiding or preventing these defenses from taking effect. Whiteflies can feed on plants without causing significant damage to mesophyll cells, making their detection extra challenging for the plant. However, whiteflies secrete honeydew that ends up on the plant surface at the feeding site and on distal plant parts below the feeding site. We reasoned that thi

Epidermal and subepidermal changes during the formation of hairy galls induced by Eriophyidae on Avicennia schaueriana leaves.

Leaf-galling Eriophyidae (Acarina) may promote simple or complex alterations in the organs of their host plants, such as an increase in indumentum density or the reorganization of epidermis and ground system tissue patterns. To test if hairy galls of Eriophyidae on Avicennia schaueriana (Acanthaceae) are related to complex changes, leaf galls in distinct developmental phases were compared to non-galled leaves using anatomical, histochemical, and histometric analyses. Quantitative comparisons of preferential gall induction sites and gall area according to distinct leaf portions were made to eva

An RNA virus-encoded zinc-finger protein acts as a plant transcription factor and induces a regulator of cell size and proliferation in two tobacco species.

Plant viruses cause a variety of diseases in susceptible hosts. The disease symptoms often include leaf malformations and other developmental abnormalities, suggesting that viruses can affect plant development. However, little is known about the mechanisms underlying virus interference with plant morphogenesis. Here, we show that a C-4 type zinc-finger (ZF) protein, p12, encoded by a carlavirus (chrysanthemum virus B) can induce cell proliferation, which results in hyperplasia and severe leaf malformation. We demonstrate that the p12 protein activates expression of a regulator of cell size and

A versatile phenotyping platform combining three-photon microscopy, optimized tissue clearing, and deep-learning signal separation for full-depth multi-organelle phenotyping.

Imaging the internal structures of plant organs at the subcellular level is essential for understanding key processes such as photosynthesis, growth, and stress responses. However, conventional optical microscopy struggles to visualize deep tissues owing to intense light scattering and spectral crosstalk from autofluorescent components-most prominently chlorophyll. To overcome these barriers, we present an integrated platform that combines deep-penetrating three-photon microscopy (3PM), tissue clearing, and a custom deep-learning network (SE-UNet) for intelligent signal separation. First, our

Hydraulic Signaling in Plants: From Physical Perturbation to Distributed Perception and Context-Dependent Decoding.

Hydraulic perturbations are among the earliest plant-wide consequences of drought, salinity and wounding, yet they are often treated as passive outcomes rather than as biologically interpreted inputs. This review distinguishes hydraulic state, hydraulic perturbation and hydraulic signal, and evaluates how organ-scale pressure and water-potential changes are converted into local membrane tension, wall strain, turgor and water-flux cues. We propose, as a testable model rather than an established mechanism, a distributed architecture comprising OSCA/TMEM63 and other mechanosensitive channels, cel
