Obesity-driven metabolic reprogramming and immune dysfunction in renal cancer.

Renal cell carcinoma (RCC), specifically clear cell renal cell carcinoma (ccRCC), is a metabolic tumor wherein the physiological state of the host is central to tumor development, progression, and therapeutic resistance. Obesity has emerged as one of the major risk factors associated with RCC; however, its impact on RCC is more complex than simply the accumulation of excess body fat. Obesity transforms the renal tumor microenvironment through metabolic rewiring and alterations in inflammation, vasculature, and anti-tumor immunity. The expansion of adipose tissue in obesity alters the renal mic

Combating VEGFA-siRNA-Induced Metabolic Reprogramming via Glucose Utilization Deprivation.

Vascular endothelial growth factor (VEGF) inhibitors suppress tumor energy supply, but their efficacy is often limited by the restoration of tricarboxylic acid (TCA) cycle activity and enhanced glycolysis. Here, a synergistic strategy is established using an in-house-designed ionizable lipid nanoparticle (LNP) to co-encapsulate VEGFA-targeting siRNA (siVEGFA) and glucose oxidase (GOx), thereby enhancing siRNA efficacy by depleting both aerobic and anaerobic glucose utilization. At the cellular level, the optimal formulation, iVG128, inhibits energy production, suppresses microvessel formation,

Natural products modulate renal metabolic reprogramming in diabetic kidney disease.

Diabetic kidney disease (DKD) is a major cause of end-stage renal disease worldwide. Its pathogenesis is highly complex and involves dysfunction across multiple renal cell types and metabolic pathways, thereby underscoring the urgent need for novel therapeutic strategies. In recent years, renal metabolic reprogramming has been increasingly recognized as a central driver of DKD progression. This review systematically examines the mechanisms of metabolic reprogramming in different renal cell types and highlights their contribution to renal injury. Particular emphasis is placed on the ability of

PMVK drives hepatocarcinogenesis via SP1 phosphorylation-mediated lipid metabolic reprogramming.

Metabolic reprogramming is a hallmark of cancer, yet the critical drivers and mechanisms in hepatocellular carcinoma (HCC) remain incompletely understood. Through a metabolism-focused CRISPR screen, we identified phosphomevalonate kinase (PMVK), a mevalonate pathway enzyme, as a key regulator of HCC stemness and progression. PMVK directly phosphorylates the transcription factor SP1 at Thr355, which enhances SP1's DNA binding affinity and promotes its interaction with the master lipid regulator SREBP1/2, driving a transcriptional program essential for de novo cholesterol and fatty acid synthesi

Statin-Induced Coenzyme Q Deficiency Induces Metabolic Reprogramming in Astrocytes.

Statins are commonly used cholesterol-lowering drugs, but their effects on astrocyte oxidative metabolism are poorly understood. To investigate this, rat astrocytes were exposed to 200 nM atorvastatin or simvastatin for 6 days and then assessed for changes in coenzyme Q (CoQ) homeostasis, mitochondrial function, and energy metabolism. Both statins comparably decreased cellular CoQ9 and CoQ10 levels (~35%), with greater losses of their reduced antioxidant forms (60-75%). Lower intracellular and mitochondrial levels of reactive oxygen species (ROS) were accompanied by the upregulation of nuclear

Energetic Decoding of VSMC Plasticity: Metabolic Reprogramming and Phenotypic Switching.

Phenotypic switching of vascular smooth muscle cells (VSMCs) constitutes a pivotal event in the initiation and progression of various vascular diseases, including vascular calcification, atherosclerosis, aneurysm, and pulmonary arterial hypertension. The transition from a contractile to a synthetic phenotype is commonly characterized by increased proliferation, migration, and extracellular matrix synthesis, thus generating substantial bioenergetic demands. In recent years, accumulating evidence has indicated that metabolic reprogramming has emerged as a key regulatory dimension of VSMC plastic

Metabolic reprogramming of Th17/Treg imbalance in autoimmune thyroid diseases.

Autoimmune thyroid diseases (AITDs), including Hashimoto's thyroiditis and Graves' disease, arise from thyroid-specific autoimmunity driven by a breakdown of immune tolerance and dysregulated T-cell responses. Within this immune network, imbalance between T helper 17 (Th17) cells and regulatory T (Treg) cells has emerged as a major determinant of persistent inflammation and defective immune restraint. These two subsets are supported by distinct but interconnected metabolic programs. Th17 cells preferentially engage glycolytic and anabolic pathways to sustain inflammatory activity, whereas Treg

Amino acid transporters in hepatocellular carcinoma: Gatekeepers of metabolic reprogramming.

Hepatocellular carcinoma (HCC), an aggressive cancer predisposed to notable metabolic alterations, including an imbalance in amino acid transporters, remains a global concern. Amino acid transporters such as proton-assisted amino acid transporter (PAT), sodium-coupled neutral amino acid transporter (SNAT), cysteine/glutamate transporter (xCT), L-type amino acid transporter (LAT1), and Alanine-serine-cysteine transporter 2 (ASCT2). The tumour microenvironment (TME), cellular signalling pathways, tumour survival, and proliferation all depend heavily on these transporters. Current investigation o

Melatonin Deficits Result in Pathologic Metabolic Reprogramming in Differentiated Neurons.

Differentiation from neural progenitor to mature neuron requires a metabolic switch, whereby mature neurons become almost entirely dependent upon oxidative phosphorylation (OXPHOS) for ATP production. Although more efficient with respect to ATP production, OXPHOS produces additional reactive oxygen species, as compared to glycolysis; thus, endogenous mechanisms to quench free radicals are essential for the maintenance of neuronal health. Melatonin is synthesized in neuronal mitochondria and has a dual role as a free radical scavenger and as an inhibitor of mitochondrial-triggered cell death an

Ocean warming drives tissue-wide metabolic reprogramming in a fish.

Ocean warming and marine heatwaves are predicted to have adverse impacts on marine organisms. Yet, knowledge of the molecular mechanisms that underpin successful or failed acclimation to increasing temperatures remains incomplete. We conducted an aquaria-based study of early-life stage clownfish comprising of six thermal regimes, measuring the metabolic, and multi-tissue transcriptional response of Amphiprion ocellaris using seven tissues. Sampling at 31°C increased metabolic rates in fish reared at 28°C; however, these effects were reduced with increasing developmental rearing at 31°C. Transc

Metabolic reprogramming in cholangiocarcinoma cancer stem cells: Emerging therapeutic paradigms.

Cholangiocarcinoma (CCA) is an aggressive malignancy characterized by limited therapeutic options and poor prognosis, largely attributed to the presence of cancer stem cells (CSCs). These CSCs serve as pivotal drivers of tumor heterogeneity, chemotherapy resistance, and disease recurrence. CSCs in CCA exhibit remarkable plasticity, a characteristic sustained through metabolic state alterations and intricate interactions with the tumor microenvironment (TME), which collectively enhance their self-renewal and survival potential. While advancements have been made in understanding metabolic reprog

MRPL12 K163 acetylation inhibits ccRCC via driving mitochondrial metabolic reprogramming.

Renal cell carcinoma (RCC) is a common urological tumor, with clear cell renal cell carcinoma (ccRCC) being the most prevalent subtype. Metabolic reprogramming plays a critical role in ccRCC progression, making it a promising target for therapeutic intervention, though effective treatments remain unavailable. Our previous studies have shown that mitochondrial ribosomal protein L12 (MRPL12) contributes to various metabolic diseases, including diabetic kidney disease and HCC, by regulating mitochondrial biosynthesis. In this study, we demonstrated that MRPL12 is acetylated at lysine 163 (K163) i

Epigenetic Regulation of G6PD Drives Metabolic Reprogramming in Intrahepatic Cholangiocarcinoma.

Intrahepatic cholangiocarcinoma (ICC) is a highly aggressive malignancy with poor prognosis and significant molecular heterogeneity. This study investigates the role of tumor-specific enhancers in metabolic reprogramming, focusing on glucose-6-phosphate dehydrogenase (G6PD) and the pentose phosphate pathway (PPP). Using native elongating transcript-cap analysis of gene expression and single-cell RNA sequencing, tumor-specific enhancers driving G6PD overexpression were identified in ICC tumor epithelial cells. Functional assays demonstrated that G6PD promotes tumor proliferation by enhancing PP

Vascular Smooth Muscle Cell Metabolic Reprogramming in Arteriovenous Fistula Failure.

Hemodialysis is the most commonly used renal replacement therapy worldwide, and arteriovenous fistulas (AVFs) are the preferred vascular access. The functional patency of AVFs directly determines the dialysis efficiency and quality of life of patients with end-stage renal disease (ESRD). However, in clinical practice, the failure of AVFs seriously affects the treatment prognosis of these patients. Vascular smooth muscle cells (VSMCs), as the main component of the vascular media, not only maintain the integrity and tension of the vascular wall under physiological conditions but also play a cruc

Metabolic reprogramming in Helicobacter pylori infection: from mechanisms to therapeutics.

Helicobacter pylori (H. pylori), a key gastric mucosal pathogen, causes chronic gastritis, peptic ulcers, and gastric cancer. H. pylori remodel the gastric microenvironment through metabolic reprogramming to drive pathogenesis. CagA+ strains disrupt lipid metabolism, increasing non-alcoholic fatty liver disease, cardiovascular, and Alzheimer's risks via PPAR interference, GBA1 demethylation, and altered FABP1/APOA1 expression, reversible by eradication. In glucose metabolism, H. pylori promote carcinogenesis via Lonp1-induced glycolysis, PDK1/Akt dysregulation, and HKDC1/TGF-β1/MDFI-mediated e

Androgens drive SLC1A5-dependent metabolic reprogramming in polycystic ovary syndrome.

Polycystic ovary syndrome is the primary cause of female infertility. Growing evidence suggests that dysregulation of amino acid metabolism plays a significant role in the onset and progression. However, the underlying mechanism remains unclear. In this study, we conduct targeted metabolite profiling of human follicular fluid and granulosa cells. A significant increase in glutamine uptake is observed in patients with hyperandrogenic polycystic ovary syndrome, mediated by the upregulation of SLC1A5, a specific glutamine transporter. We find that androgen excess primarily activates SLC1A5 expres

3D Modeling: Insights into the Metabolic Reprogramming of Cholangiocarcinoma Cells.

Developing accurate in vitro models that replicate the in vivo tumor environment is essential for advancing cancer research and therapeutic development. Traditional 2D cell cultures often fail to capture the complex structural and functional heterogeneity of tumors, limiting the translational relevance of findings. In contrast, 3D culture systems, such as spheroids, provide a more physiologically relevant context by replicating key aspects of the tumor microenvironment. This study aimed to compare the metabolism of three intrahepatic cholangiocarcinoma cell lines in 2D and 3D cultures to ident
