Advancing forensic accuracy: mitigating methemoglobin interference in postmortem carbon monoxide analysis using sodium dithionite reduction.

Analyzing carbon monoxide concentration within an individual is crucial. The analysis of CO content in a tissue sample is performed using gas chromatography. The concentration is calculated based on a linear equation derived from the calibration curve created with the CO-fortified sample. However, when methemoglobin (MetHb) is formed from putrefaction, it inhibits CO binding to the sample and may lead to inaccurate results. MetHb results from the iron oxidation of normal heme hemoglobin (HHb), and by treating the sample with a reducing agent, it can be converted back to HHb. To investigate the

Predictors of methemoglobin formation in high-dose inhaled nitric oxide therapy: A retrospective cohort study.

BACKGROUND: High-dose inhaled nitric oxide (iNO), defined as doses exceeding 20 parts per million (ppm) and reaching up to 300 ppm, has demonstrated promising antimicrobial properties. However, elevated concentrations of nitric oxide (NO) can increase the formation of methemoglobin (MetHb), which can impair oxygen transport. Understanding factors influencing MetHb formation is essential to guide safe clinical use of iNO therapy. METHODS: We conducted a retrospective analysis of 660 high-dose iNO treatments administered to 71 individuals between April 2020 and January 2022 under three prospect

Changes of hexose monophosphate pathway and methemoglobin reductase enzyme activity after radiation in guinea pigs.

HMP pathway activity changes occurring after exposure to ionizing radiation (LD50 dose) have been investigated. The study was carried out on 18 experimental guinea pigs subjected to 5 successive exposures of 150 rads 3 or 4 days apart. The control animals were sham radiated but were otherwise treated identically as those of the experimental groups. Blood samples were taken by cardiac puncture before radiation and 30 min after each exposure of 150 rads. The red cells were re-suspended in their own plasma and HMP pathway activity was measured in the suspension. The pathway activity showed a cons

NADH-methemoglobin reductase (cytochrome b5 reductase) levels in two groups of American blacks and whites.

BACKGROUND: Sickle cell trait, glucose-6-phosphate dehydrogenase (G6PD) deficiency and alpha-thalassemia trait are common genetic abnormalities among the American Black population. Under oxidative stress, the presence of any of these conditions would predispose the hemoglobin (Hb) to oxidation resulting in accelerated methemoglobin (metHb) formation. It was hypothesized that red cells phenotypic for these genetic variants should have more or different levels of metHb reductase (cytochrome b5 reductase) activity. METHODS: To test this hypothesis, we measured the red cell metHb reductase activi

Evaluation of the Methemoglobin-Haptoglobin Protein Complex for Detoxification of Cyanide, Azide, and Sulfide in In Vitro Binding Models.

Cyanide, azide, and sulfide are potent anion inhibitors of mitochondrial enzymes, causing rapid disruption of cellular energy metabolism and potentially fatal outcomes. Current treatments for these toxic anions are limited, highlighting the need for broadly effective antidotes. Methemoglobin (methHb), the ferric form of hemoglobin (Hb), can bind these ligands with high affinity, acting as a mechanistic scavenger that prevents their interaction with mitochondrial targets. Binding methHb to haptoglobin (Hp), an endogenous Hb-binding plasma protein, offers a strategy to enhance methHb stability a

Consequences of methemoglobinemia in pregnancy in newborns, children, and adults: issues raised by new findings on methemoglobin catabolism.

OBJECTIVE: The aim of this review is to warn about the effects of methemoglobin and its catabolic products and the toxic effects caused by environmental oxidants that cause high-risk pregnancy and may later impair the health of newborns, children and adolescents. METHODS: In our study of pregnant women (n = 36) whose methemoglobin level was >1.5 g/l, we took blood samples from their newborns to determine the frequency of sister chromatid exchange (SCE) by cultivating lymphocytes. The research took place at the Department of Biology and Medical Genetics of the School of Medicine in Rijeka (Cro

Effect of iron and phosphate on bacterial cyanide formation determined by methemoglobin in two-dimensional gradient microcultivations.

Cyanide formation by microorganisms is typically observed during early stationary growth phase and is facilitated by the presence of iron(III) and inorganic phosphate. Extracellular free cyanide in aqueous solutions might readily react with methemoglobin added and can be determined by UV/VIS spectroscopy. As alternative to existing methods, this provided the basis for an analytical method which has not been used previously for the determination of cyanide in bacterial cultivations. We successfully applied the technique to study the combined effect of both iron(III) and phosphate on the cyanide

[Cyanosis in children caused by inherited methemoglobinemia due to deficiency of NADH-dependent methemoglobin reductase in erythrocytes].

Two cases of the congenital methemoglobinemia in children due to the deficiency of NADH-dependent methemoglobin reductase in erythrocytes. These children were referred to the Cardiological Ward at the Child Health Centre with suspected cyanotic heart defect. Cardiological examinations excluded heart defect but an increased blood methemoglobin level and decreased activity of NADH-dependent methemoglobin reductase were found, that caused methemoglobinemia. Methylene blue and vitamin C diminished cyanosis. These cases advocate inclusion of methemoglobinemia into differential diagnosis of cyanotic

[Optical spectroscopy of methemoglobin from the Lampetra fluviatilis river lamprey and its complexes reduced at 77K].

The absorption spectra from met-haemoglobin from lamprey Lampetra fluviatilis and its complexes with F-, CN-, N3- and imidazole were studied in non-equilibrium states arising in low-temperature (77K) reduction of water-glycerol of oxidized forms of protein with thermolyzed electrons. The Soret band was shown to be not split in low-temperature absorption spectra of reduced met-haemoglobin and its complexes unlike that of the equilibrium deoxyhaemoglobin. It was concluded that met-haemoglobin has a higher symmetry of the active centre as compared to deoxyhaemoglobin.

Anomalously slow cyanide binding to Glycera dibranchiata monomer methemoglobin component II: implication for the equilibrium constant.

In comparison to sperm whale metmyoglobin, metleghemoglobin a, methemoglobins, and heme peroxidases, the purified Glycera dibranchiata monomer methemoglobin component II exhibits anomalously slow cyanide ligation kinetics. For the component II monomer methemoglobin this reaction has been studied under pseudo-first-order conditions at pH 6.0, 7.0, 8.0, and 9.0, employing 100-250-fold mole excesses of potassium cyanide at each pH. At 20 degrees C, with micromolar protein concentrations, kobsd varies between 9.11 x 10(-5) s-1 at pH 6.0, 100-fold KCN mole excess, and 1.12 x 10(-2) s-1 at pH 9.0, 2

Nitroxides catalytically inhibit nitrite oxidation and heme inactivation induced by H2O2, nitrite and metmyoglobin or methemoglobin.

Stable nitroxide radicals have multiple biological effects, although the mechanisms underlying them are not fully understood. Their protective effect against oxidative damage has been mainly attributed to scavenging deleterious radicals, oxidizing reduced metal ions and reducing oxyferryl centers of heme proteins. Yet, the potential of nitroxides to protect heme proteins against inactivation while suppressing or enhancing their catalytic activities has been largely overlooked. We have studied the effect of nitroxides, including TPO (2,2,6,6-tetramethylpiperidin-N-oxyl), 4-OH-TPO, 4-oxo-TPO and

The value of methemoglobin reduction test as a screening test for neonatal glucose 6-phosphate dehydrogenase deficiency.

UNLABELLED: Glucose 6-phosphate dehydrogenase (G-6-PD) deficiency is common in the Thai population and is the cause of neonatal hyperbilirubinemia and hemolytic anemia. This X-linked disorder is much more common in males than females. The objectives of this study were to compare the result of the screening methemoglobin reduction test (MRT) with the gold standard G-6-PD activity, and also to determine the prevalence of G-6-PD deficiency in the cord blood and blood of neonates with hyperbilirubinemia. Five hunderd and twenty two randomly selected cord blood (350 males, 172 females) and 229 peri

Comparison of nitrite treatment and stroma-free methemoglobin solution as antidotes for cyanide poisoning in a rat model.

The standard nitrite/thiosulfate regimen for cyanide poisoning was tested in our rat model. By modifying the treatment regimen and the nitrite solution an effective antidote against an LD90 of cyanide could be produced. However, this treatment was effective against two times the LD90 only when administered ten minutes prior to cyanide injection. These results are in marked contrast to our results with stroma-free methemoglobin solutions (SFMS) which showed SFMS to be a highly effective antidote against four times the LD90 when administered 30 seconds after an intravenous injection of cyanide.

Proton NMR study of methemoglobin and its isolated chains. Effect of the subunit association on the structure of the subunits.

In order to investigate the effect of the alpha beta subunit contacts on the subunit structure of human adult methemoglobin, the hyperfine shifted proton NMR spectra of several high spin complexes (water, cyanate, thiocyanate, formate, fluoride, and nitrite) and low spin complexes (imisazole, azide, and cyanide) of hemoglobin and its isolated subunits were characterized at 220 MHz and 22 degrees C. The spectra of ferric low spin derivatives of the isolated subunits were approximately superimposable on the corresponding hemoglobin spectra. On the other hand, the high spin spectra of the isolate
