Involvement of transmitters in the anxiolytic action of urocortin 3 in mice.

Urocortin 3 (Ucn 3) was tested for anxiolytic action in mice an elevated plus maze. For detection of the possible involvement of neurotransmitters, the mice were pretreated with receptor blockers: haloperidol, phenoxybenzamine, propranolol, atropine, methysergide, bicuculline or naloxone. The peptide was administered into the lateral brain ventricle; the receptor blockers were applied intra- peritoneally. Ucn 3 alone elicited dose-dependent bell-shaped anxiolytic action. The most effective dose was 0.5 μg. In the combined testing a 0.5 μg dose was used. Haloperidol, propranolol, atropine, meth

Headache currents commentary.

What Happens to the Old Headache Medicines? Rapoport AM, MD. Old headache medicines never die; they either fade away or come back in disguise. The disguise is often a new route of administration, which may work better, faster, more completely, with fewer adverse events, and/or have certain other advantages. The clinical aspects of 3 of the oldest headache medicines (ergotamine tartrate, dihydroergotamine, and methysergide) will be discussed here. Sumatriptan will then be discussed as the prototype of the newest category of acute care therapy (triptans) for migraine. It will be compared with th

Characterization of serotonin receptors in the iris + ciliary body of the albino rabbit.

Experiments were undertaken to determine if serotonin (5-HT) radioligand binding sites were present in a membrane fraction of iris + ciliary body from adult, albino rabbits. The total binding of 3H-5-HT, 3H-spiroperidol (SPI) and 3H-ketanserin (KET), all at 2 nM, was determined in the absence and in the presence of ketanserin, mianserin, methysergide or 5-methoxytryptamine (5-MT), all at 1 microM. Except for ketanserin, which displaced 15% of 3H-KET binding, none of the agents altered 3H-SPI or 3H-KET binding. Reductions of 12% and 14% in 3H-5-HT binding were achieved by ketanserin and mianser

Neural control of the sphincter of Oddi. A physiological role of 5-hydroxytryptamine in the regulation of basal sphincter of Oddi motor activity in the cat.

The effect of 5-hydroxytryptamine (5-HT) on the sphincter of Oddi (SO) was studied in the cat. The SO had two motor responses to 5-HT: the most common was an initial contraction followed by a more prolonged relaxation, and the other was an exclusive relaxation. Tetrodotoxin did not impair the magnitude of the net contraction induced by 5-HT, but it completely blocked the relaxation. Methysergide partially inhibited the SO contraction in response to submaximal doses of 5-HT (5-20 micrograms/kg). Atropine decreased the SO excitatory response to all doses of 5-HT. The combination of atropine and

Partial inhibition of the abstinence syndrome in morphine tolerant-dependent mice following pharmacological denervation.

Mice were chronically treated with either atropine, methysergide or pentobarbital in order to induce sensitivity changes resulting from adaptative adjustments in the central nervous system (CNS), and to examine the degree of tolerance to and physical dependence on morphine several days after the discontinuation of pretreatments. Subsequently to the chronic blockade of muscarinic or serotonergic receptors, the intensity of tolerance was unaffected, but some manifestations of the abstinence behavior induced by naloxone were reduced in part. This attenuation of the abstinence syndrome in the pret

Pharmacological analysis of certain mechanisms of morphine addiction.

The effects of L-Dopa, Methysergid, Diphenhydramine hydrochloride and LSD on the development of morphine dependence and the abstience syndrome after its withdrawal, were tested in experiments on 200 male Wistar albino rats. L-Dopa had no efect on the development of physical morphine dependence, while Methysergid prevented its development. Applied in rats during the abstinence syndrome, LSD intensified their aggressivity with no influence on the analgesic effect of morphine. Diphenhydramine reduced the aggressiveness of the rats during the abstinence syndrome. Biochemical tests show that in mor

Pharmacological characterization of 5-hydroxytryptamine-induced motor activity (in vitro) in the guinea pig gastric antrum and corpus.

In order to characterize the receptor subtypes involved in 5-hydroxytryptamine (5-HT)-induced circular muscle motor responses of the guinea pig gastric antrum and corpus, we examined the effects of several antagonists in vitro. 5-HT evoked concentration-dependent contractions of the gastric antrum and relaxations of the corpus. 5-HT induced antral contractions were abolished by pretreatment with atropine and tetrodotoxin. Characterization of the receptors involved in the 5-HT-induced excitation of canine antral longitudinal muscle.

1. We aimed to characterize the 5-HT receptors involved in the 5-HT-induced effect on electrically induced contractions of dog antrum longitudinal muscle in vitro. 2. In the presence of L-NOARG (0.1 mM), electrical field stimulation (EFS) induced atropine- and tetrodotoxin-sensitive contractions. Tetrodotoxin or atropine left any agonist tested ineffective. These EFS-induced contractions were on average enhanced by 5-HT (0.3 microM), however, pronounced variation in the response to 5-HT was observed. There were non-significant trends of the selective 5-HT3 receptor antagonist granisetron (1 mi

Analgesia induced by 2- or 100-Hz electroacupuncture in the rat tail-flick test depends on the activation of different descending pain inhibitory mechanisms.

EA was applied bilaterally to the Zusanli and Sanyinjiao acupoints in lightly anesthetized rats. EA increased tail-flick latency, where the effect of 2-Hz EA lasted longer than that produced by 100-Hz EA. The 2-Hz EAIA was inhibited by naloxone or

Antinociception elicited by aversive stimulation of the inferior colliculus.

We have shown that the inferior colliculus is involved in the integration of defensive reactions. Electrical and chemical stimulation of this structure elicits fear and escape behavior, expressed respectively by immobility (freezing) and wild running, followed by jumps. In this study, we analyzed whether the defensive behavior integrated at this level of the midbrain tectum is also followed by antinociception and its chemical mediation. In addition, we further addressed whether or not the aversive states and the stress-induced analgesia share the same neural substrates in the inferior collicul

Pharmacological characterization of 5-Hydroxytryptamine-receptor subtypes in circular muscle from the rat stomach.

5-Hydroxytryptamine (5-HT) modulates gastric motility and gastric emptying via a variety of 5-HT receptor subtypes. However, regional and functional differences among 5-HT receptor subtypes in the rat stomach are not fully investigated. Thus, we aimed to characterize 5-HT receptor subtypes involved in the 5-HT-induced contractions in the isolated antral, corporal and fundic circular muscles of the rat stomach by measuring the contractile force. 5-HT induced concentration-dependent contractions in the antrum, corpus and fundus. 5-HT-induced antral contractions were partly blocked by atropine an

Role of serotonin, histamine, and thromboxane A2 in platelet-induced contractions of coronary arteries and aortae from rabbits.

The present study was undertaken to clarify the underlying mechanisms responsible for contractions of isolated coronary arteries and aortae from rabbits in response to thrombin-stimulated autologous platelets. Thrombin-stimulated platelets evoked potent contractions of both arteries in a platelet concentration-related manner. Pretreatment of platelets with aspirin, which almost completely inhibited thromboxane A2 synthesis but not the release reaction of biologic monoamines from platelets, caused only slight suppression of platelet-induced contractions of both arteries. Ketanserin as well as m

Joint manipulation reduces hyperalgesia by activation of monoamine receptors but not opioid or GABA receptors in the spinal cord.

Joint manipulation has long been used for pain relief. However, the underlying mechanisms for manipulation-related pain relief remain largely unexplored. The purpose of the current study was to determine which spinal neurotransmitter receptors mediate manipulation-induced antihyperalgesia. Rats were injected with capsaicin (50 microl, 0.2%) into one ankle joint and mechanical withdrawal threshold measured before and after injection. The mechanical withdrawal threshold decreases 2 h after capsaicin injection. Two hours after capsaicin injection, the following drugs were administered intrathecal

The anorectic action of peripherally administered 5-HT is enhanced by vagotomy.

Peripherally administered 5-HT produced a greater suppression of food intake in rats with subdiaphragmatic vagotomy than in sham-operated controls. The enhanced anorexia to 5-HT in vagotomised rats and the anorexia in sham-operated controls were reversed by methysergide, indicating the involvement of 5-HT receptors in the observed anorexia in both groups of animals. Thus the increased suppression of food intake in vagotomised rats cannot be explained in terms of non-specific effects of 5-HT. Both vagotomised and sham-operated rats showed an equivalent degree of anorexia when treated with fenfl

Neural control of the sphincter of Oddi. Physiologic role of enkephalins on the regulation of basal sphincter of Oddi motor activity in the cat.

The effect and physiologic role of enkephalins on the sphincter of Oddi was studied in the cat. Leucine- and methionine-enkephalin caused an initial sphincter of Oddi contraction followed by a more prolonged relaxation. The excitatory effect of leucine-enkephalin was antagonized partially by atropine and by methysergide infusion. It was completely blocked by a combination of atropine and methysergide infusion, by tetrodotoxin, by 5-hydroxytryptamine depletion induced by reserpine, and by 5-hydroxytryptamine tachyphylaxis. Naloxone infusion decreased sphincter of Oddi motor activity, which was

Inhibition of hind-paw edema and cutaneous vascular plasma extravasation in mice by acetylshikonin.

Acetylshikonin, a naphthoquinone isolated from the Chinese herb medicine, tzu ts'ao, was demonstrated to inhibit the polymyxin B-induced hind-paw edema in normal as well as in adrenalectomized mice. Liver glycogen content was increased in adrenalectomized mice pretreated with dexamethasone, but not with acetylshikonin. Like diphenhydramine, methysergide and isoproterenol, acetylshikonin reduced the plasma exudation evoked in dorsal hind-paw skin by antidromic stimulation of the saphenous nerve, and in passive cutaneous anaphylactic reaction, bradykinin-, substance P-, compound 48/80-, histamin

Comparative effects of morphine and nalorphine on colonic motility in the conscious dog.

The effects of increased doses (0.05-02 mg/kg) of morphine and nalorphine on colonic motility were investigated in 15-22 h fasted dogs fitted with two strain gauge transducers on the transverse colon at 8 and 15 cm from the ileo-colonic junction. These effects were compared to those obtained after previous administration of naloxone (0.3 mg/kg), atropine (0.1 mg/kg) and methysergide (0.1 mg/kg). Both morphine and nalorphine, at a dose of 0.1 mg/kg increased the colonic motility index by 285 and 248% from 0 to 60 min respectively after their administration, these effects lasting 3.2 and 2.6 h.
