Desiccating environmental stress exacerbates autoimmune lacrimal keratoconjunctivitis in non-obese diabetic mice.

The non-obese diabetic (NOD) mouse is prone to develop autoimmune disease, including Sjögren's syndrome. The purpose of this study was to determine if desiccating environmental stress exacerbates the development of Sjögren's syndrome-like lacrimal keratoconjunctivitis in the NOD.B10.H2(b) mouse. Four-week-old male mice were used as young controls. Sixteen-week-old male mice were untreated or subjected to desiccating stress with a fan alone or with a fan plus subcutaneous injections of the anticholinergic agent scopolamine for 5 or 10 days to inhibit tear production. Mice spontaneously develope

4-week aerobic exercise training regulates systemic macrophage polarization in obese mice.

BACKGROUND: Obesity is accompanied by chronic low-grade inflammation, largely driven by imbalances in macrophage polarization. While pro-inflammatory M1 macrophages accumulate in adipose tissue and circulation, contributing to insulin resistance and metabolic disruption, alternatively activated M2 macrophages exert anti-inflammatory and tissue-protective effects. Exercise is widely recognized as a non-pharmacological strategy to improve metabolic health; however, the extent to which short-term aerobic training influences systemic macrophage polarization in obesity is not fully understood. This

Central FGF19 reduces hypothalamic inflammation and improves leptin signaling in obese mice.

Obesity is a chronic metabolic disorder characterized by systemic and hypothalamic inflammation that impairs neuronal signaling and disrupts the fine-tuning of energy balance regulation. Leptin resistance, a hallmark of obesity, arises from this inflammatory state and contributes to increased food intake and reduced energy expenditure (EE). Fibroblast growth factor 19 (FGF19), a gut-derived hormone with beneficial metabolic effects in peripheral tissues, also acts centrally to reduce body weight. However, the mechanisms underlying its hypothalamic action remain unclear. In this study, we inves

[Butyrate alleviates Clostridium difficile infection in high-fat diet-induced obese mice].

OBJECTIVES: To investigate the mechanism by which obesity exacerbates Clostridium difficile infection (CDI) and explore the value of gut microbiota metabolite butyrate as an alternative therapy. METHODS: In mouse models of high-fat diet (HFD)-induced obesity and CDI and normal diet-fed mouse models of CDI, the effect of butyrate treatment on hepatic lipid deposition and liver function was assessed using Oil Red O staining and by measuring aspartate aminotransferase and alanine aminotransferase levels. Superoxide dismutase (SOD) activity and malondialdehyde (MDA) of the mice were determined, a

Liraglutide-Driven Weight Loss Modulates Placental Remodeling in Obese Pregnancies in Mice.

BACKGROUND: The placenta stands at the maternal-fetal interface and is a key organ regulating the intrauterine environment. In pregnancies exposed to obesity, placental function, signaling, and nutrient handling are adversely altered. Pre-conception weight loss is a potential intervention to alter an obesogenic milieu of pregnancy, which we investigated in a mouse model of maternal obesity using diet or administration of the glucagon-like peptide-1 (GLP-1) receptor agonist liraglutide. METHODS: Pre-pregnancy weight loss in C57BL/6 high-fat diet (HFD)-fed dams was induced in the pre-pregnancy

LNP-mRNA vaccine prevents type 1 diabetes in non-obese diabetes mice.

Islet-antigen-specific tolerization is a key goal of experimental immunotherapies for type 1 diabetes. mRNA-based vaccines have demonstrated the feasibility of RNA delivery in inducing antigen tolerance in autoimmune diseases. In this study, mRNA vaccine, encoded tandem glutamic acid decarboxylase 65 (GAD65) epitopes and cholera toxin B subunit (CTB-GADIII), prepared by an in vitro transcription (IVT) system and encapsulated with lipid nanoparticles (LNP), was intramuscularly administered to non-obese diabetic (NOD) and cyclophosphamide (Cy)-NOD mice respectively. The results showed that the m

[Punicalagin inhibits hepatic lipid deposition in obese mice via AMPK/ACC pathway].

Hepatic lipid deposition is one of the basic manifestations of obesity, and nowadays pharmacological treatment is the most important tool. Punicalagin(PU), a polyphenol derived from pomegranate peel, is a potential anti-obesity substance. In this study, 60 C57BL/6J mice were randomly divided into a normal group and a model group. After establishing a model of simple obesity with a high-fat diet for 12 weeks, the successfully established rat models of obesity were then regrouped into a model group, an orlistat group, a PU low-dose group, a PU medium-dose group, and a PU high-dose group. Dietary Nitrate and Corresponding Gut Microbiota Prevent Cardiac Dysfunction in Obese Mice.

UNLABELLED: Impaired heart function can develop in individuals with diabetes in the absence of coronary artery disease or hypertension, suggesting mechanisms beyond hypertension/increased afterload contribute to diabetic cardiomyopathy. Identifying therapeutic approaches that improve glycemia and prevent cardiovascular disease are clearly required for clinical management of diabetes-related comorbidities. Since intestinal bacteria are important for metabolism of nitrate, we examined whether dietary nitrate and fecal microbial transplantation (FMT) from nitrate-fed mice could prevent high-fat d

Enzymatic hydrolysate of porphyra enhances the intestinal mucosal functions in obese mice.

Intestinal mucosal immunity is important to human body; however, obesity induced by high-fat diet may bring a series of problems, such as chronic inflammation which may damage intestinal mucosal immunity. In this study, the effects of two different enzymatic hydrolysates of porphyra on the function of intestinal mucosal were explored in obese mice. The results showed that 10 consecutive weeks of high-fat dietary intake resulted in weight gain and intestinal abnormalities in C57BL/6 mice. However, the administration of enzymatic hydrolysate of porphyra effectively protected the intestinal mucos

Lower melanoma pulmonary metastatic burden in obese mice: role of FGF-21.

Obesity is a risk factor for malignant melanoma. The lungs are main target organs for metastization and their immune response is a key modulator of this mechanism. The concept that the metastatic potential of some types of cancer is reduced or inhibited by obesity, known as the obesity paradox, drives major concerns on the prognosis of metastasized patients. The aim of this study was to investigate how high-fat diet (HFD)-induced obesity affects melanoma metastization. C57Bl6/J mice were fed with HFD or standard diet for 180 days and inoculated intravenously with B16F10 melanoma cells. Upon 21

Gut Microbiota Reshaped by Pectin Treatment Improves Liver Steatosis in Obese Mice.

Pectin, a soluble fiber, improves non-alcoholic fatty-liver disease (NAFLD), but its mechanisms are unclear. We aimed to investigate the role of pectin-induced changes in intestinal microbiota (IM) in NAFLD. We recovered the IM from mice fed a high-fat diet, treated or not with pectin, to perform a fecal microbiota transfer (FMT). Mice fed a high-fat diet, which induces NAFLD, were treated with pectin or received a fecal microbiota transfer (FMT) from mice treated with pectin before (preventive FMT) or after (curative FMT) being fed a high-fat diet. Pectin prevented the development of NAFLD, i

Sevoflurane-induced hyperglycemia is attenuated by salsalate in obese insulin-resistant mice.

PURPOSE: Perioperative hyperglycemia is common and is associated with significant morbidity. Although patient characteristics and surgery influence perioperative glucose metabolism, anesthetics have a significant impact. We hypothesized that mice that were obese and insulin-resistant would experience greater hyperglycemia in response to sevoflurane anesthesia compared with lean controls. We further hypothesized that sevoflurane-induced hyperglycemia would be attenuated by salsalate pre-treatment. METHODS: Lean and obese male C57BL/6J mice were anesthetized with sevoflurane for 60 min with or

Chronopharmacology of simvastatin on hyperlipidaemia in high-fat diet-fed obese mice.

The chronopharmacology refers to the utilization of physiological circadian rhythms to optimize the administration time of drugs, thus increasing their efficacy and safety, or reducing adverse effects. Simvastatin is one of the most widely prescribed drugs for the treatment of hypercholesterolaemia, hyperlipidemia and coronary artery disease. There are conflicting statements regarding the timing of simvastatin administration, and convincing experimental evidence remains unavailable. Thus, we aimed to examine whether different administration times would influence the efficacy of simvastatin. Hi

Regulation of lymphatic function and injury by nitrosative stress in obese mice.

OBJECTIVE: Obesity results in lymphatic dysfunction, but the cellular mechanisms that mediate this effect remain largely unknown. Previous studies in obese mice have shown that inducible nitric oxide synthase-expressing (iNOS+) inflammatory cells accumulate around lymphatic vessels. In the current study, we therefore tested the hypothesis that increased expression of iNOS results in nitrosative stress and injury to the lymphatic endothelial cells (LECs). In addition, we tested the hypothesis that lymphatic injury, independent of obesity, can modulate glucose and lipid metabolism. METHODS: We

Maternal N-Acetyl Cysteine Intake Improved Glucose Tolerance in Obese Mice Offspring.

Exposure to certain environmental factors during the early stages of development was found to affect health in adulthood. Among other environmental factors, oxidative stress has been suggested to be involved in fetal programming, leading to elevated risk for metabolic disorders, including type 2 diabetes; however, the possibility that antioxidant consumption during early life may affect the development of diabetes has scarcely been studied. The aim of this study was to investigate the effects of N-acetyl-l-cysteine (NAC) given during pregnancy and lactation on the susceptibility of offspring t
