Recovery of reproductive function of female zebrafish from the toxic effects of microcystin-LR exposure.

Fish has a strong resistance to microcystins (MCs), cyclic heptapeptide cyanotoxins, known as endocrine disrupting chemicals (EDCs) which are released during cyanobacterial blooms and many laboratory and field studies have found the hepatic recovery of fish from the MCs exposure. The aim of the present study was to investigate the recovery mechanisms of reproductive function of adult zebrafish (Danio rerio) from microcystin-LR (MC-LR) exposure. Therefore, adult female zebrafish were exposed to 0, 1 or 50 μg/L of MC-LR for 21days and transferred to MC free water for another 21 days to investiga

Anabaena/Dolichospermum as the source of lethal microcystin levels responsible for a large cattle toxicosis event.

Thirty-two 14-month old steers died during a period of four days (19-23 June 2017) after drinking from Junipers Reservoir (southeastern Oregon, USA) during a cyanobacterial bloom. Clinical and histopathological findings were consistent with acute liver disease, and microcystin-LR was present at 3000 μg/L in a reservoir water sample and at 7100 μg/L in the rumen contents of one of the mortalities. Serum biochemistry and histological examination indicated severe liver damage consistent with microcystin toxicosis. Microscopic observation of reservoir water samples, limited to frozen or poorly sto

Pulmonary and hepatic injury after sub-chronic exposure to sublethal doses of microcystin-LR.

We had previously shown that microcystin-LR (MCLR) could induce lung and liver inflammation after acute exposure. The biological outcomes following prolonged exposure to MCLR, although more frequent, are still poorly understood. Thus, we aimed to verify whether repeated doses of MCLR could damage lung and liver and evaluate the dose-dependence of the results. Male Swiss mice received 10 intraperitoneal injections (i.p.) of distilled water (60 μL, CTRL) or different doses of MCLR (5 μg/kg, TOX5), 10 μg/kg (TOX10), 15 μg/kg (TOX15) and 20 μg/kg (TOX20) every other day. On the tenth injection res

Metabolic response to oral microcystin-LR exposure in the rat by NMR-based metabonomic study.

Microcystin-LR (MCLR), a potent hepatotoxin, is causing increased risks to public health. Although the liver is the main target organ of MCLR, the metabolic profiling of liver in response to MCLR in vivo remains unknown. Here, we comprehensively analyzed the metabolic change of liver and ileal flushes in rat orally gavaged with MCLR by 1H nuclear magnetic resonance (NMR). Quantification of hepatic MCLR and its glutathione and cysteine conjugates by liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) was conducted. Metabonomics results revealed significant associations o

LASSBio 596 per os avoids pulmonary and hepatic inflammation induced by microcystin-LR.

Cyanobacterial blooms that generate microcystins (MCYSTs) are increasingly recognized as an important health problem in aquatic ecosystems. We have previously reported the impairment of pulmonary structure and function by microcystin-LR (MCYST-LR) exposure as well as the pulmonary improvement by intraperitoneally injected (i.p.) LASSBio 596. In the present study, we aimed to evaluate the usefulness of LASSBio 596 per os on the treatment of pulmonary and hepatic injuries induced by MCYST-LR. Swiss mice received an intraperitoneal injection of 40 μl of saline (CTRL) or a sub-lethal dose of MCYST

Microcystin-LR as an Emerging Aquatic Threat: A Literature Review of Ecotoxicological Effects in Fish and Remediation Methods.

Water pollution, intensified by anthropogenic activities, compromises aquatic ecosystems, especially through eutrophication, which favors the proliferation of microcystin-LR-producing cyanobacteria (MC-LR). We conducted a systematic and narrative literature review involving 72 studies on the effects of MC-LR on fish and 105 on remediation methods, aiming to understand the ecotoxicological impacts of this cyanotoxin and evaluate mitigation strategies. Our results show that most research is conducted under laboratory conditions, mainly with Danio rerio. MC-LR causes multiple adverse effects in f

Self-Assembled Living Microreactors for Selective Microcystin Removal via Cascade Sieving, Adsorption and Biodegradation.

Microcystin contamination caused by harmful cyanobacterial blooms is a growing global challenge for water safety. Here we reported a self-assembled living microreactor that encapsulated microcystin-degrading bacteria within a biochar-reinforced double-network hydrogel, enabling integrated molecular sieving, adsorption, and enzymatic degradation. The system exhibited size- and charge-selective removal of microcystin over coexisting organic matters, while maintaining high degradation efficiency under harsh stress conditions, including pH (5-9), inorganic ions (10-200 mg/L), and natural organic m

Schisandrin B alleviates hepatic damage and hepatic lipid metabolic dysregulation induced by microcystin-LR.

Microcystin-LR (MC-LR) is a common biological pollutant found in aquatic environments; it induces severe liver injury, inflammation and lipid metabolic disorders. The potential mechanisms underlying MC-LR pathogenesis is still unclear, and effective therapeutic drugs remain to be explored through in-depth research. This study was designed to systematically evaluate and clarify the protective effects and mechanisms of Schisandrin B (SchB) against MC-LR-induced hepatotoxicity. The animal study indicated that SchB alleviated MC-LR-triggered body weight loss, hepatomegaly, liver dysfunction, infla

Prenatal microcystin-LR exposure alters pancreatic proteome and impairs insulin secretion in offspring mice.

This study examines the toxic effects of microcystin-LR (MC-LR), a cyanobacterial toxin, on glucose metabolism in the pancreatic β cells of offspring following maternal exposure in mice. Female mice were exposed to varying concentrations of MC-LR for 12 weeks and the period of gestation. While no adverse effects were noted in the mothers, the neonates displayed significantly lower blood glucose levels that persisted into puberty, along with elevated fasting insulin levels. The results indicate a differential expression of pancreatic proteins, particularly those involved in the PPAR signaling p

A MOF-818-based nanozyme-linked aptasensor for microcystin-LR detection in edible algae powder.

The development of simple, rapid, and sensitive detection methods for microcystin-leucine-arginine (MC-LR) is crucial for ensuring food safety. A novel nanozyme-linked colorimetric aptasensor was developed using metal-organic framework (MOF) material MOF-818. The aptamer and MOF-818 with complementary DNA (cDNA) on its surface (MOF-818/cDNA) served as bioreceptors and signal reporters, respectively. Without MC-LR, MOF-818/cDNA bound to the aptamer, catalyzing the oxidation of 3,5-Di-tert-butylcatechol (3,5-DTBC) and producing yellow-colored products. While in the presence of MC-LR, it bound to

Pathogenic bacteria contribute to antibiotic resistance genes dissemination in microcystin-LR exposed intestine of zebrafish.

Cyanobacterial blooms and antibiotic resistance genes (ARGs) in aquatic ecosystems are emerging public health concerns. Microcystin-LR (MC-LR), a representative cyanotoxin released from cyanobacterial blooms, has poorly understood mechanisms regarding its influence on the dissemination of ARGs. In this study, zebrafish were exposed to MC-LR at environmentally relevant concentrations to investigate the mechanisms underlying the influence of MC-LR on the prevalence and dissemination of ARGs in the intestine, with a focus on intestinal microbiota and pathogenic bacteria. Overall, MC-LR exposure s

Co-exposure of microcystin and nitrite enhanced spermatogenic disorders: The role of mtROS-mediated pyroptosis and apoptosis.

Microcystins (MCs) and nitrites are coexisted in the environment and have reproductive toxicity. The combined toxic effect and mechanism of MCs and nitrite on spermatogenesis remain largely unclear. In the present study, co-exposure to microcystin-leucine arginine (MC-LR) and sodium nitrite (NaNO2) aggravated testicular damage of Balb/c mice and mitochondrial impairment of spermatogonia, Sertoli cells, and sperm. Furthermore, MC-LR and NaNO2 reduced sperm density with a synergistic effect. In addition, MC-LR and NaNO2 synergistically induced oxidative stress in the reproductive system by decre

Microcystin-LR induces lactate production disruption via altering the m6A modification in Sertoli cells.

We have previously reported the toxicity of microcystin-LR (MC-LR) to the male reproductive system, which results in functional changes in mouse testes. In this study, mice were orally exposed to MC-LR at 1, 7.5, 15, or 30 μg/L daily for 180 days. We found an increase in germ cell apoptosis in the seminiferous tubules and low-quality sperm in the epididymis. A decrease in lactate dehydrogenase A (Ldha) expression in testes through high-throughput sequencing was observed. We validated that MC-LR disrupted lactate production in Sertoli cells by suppressing the expression of Ldha. Further studies

Microcystin-LR Does Not Alter Cell Survival and Intracellular Signaling in Human Bronchial Epithelial Cells.

Changes in ecological and environmental factors lead to an increased occurrence of cyanobacterial water blooms, while secondary metabolites-producing cyanobacteria pose a threat to both environmental and human health. Apart from oral and dermal exposure, humans may be exposed via inhalation and/or swallowing of contaminated water and aerosols. Although many studies deal with liver toxicity, less information about the effects in the respiratory system is available. We investigated the effects of a prevalent cyanotoxin, microcystin-LR (MC-LR), using respiratory system-relevant human bronchial ep

Microcystin-leucine-arginine induces liver fibrosis by activating the Hedgehog pathway in hepatic stellate cells.

Microcystin-leucine-arginine (MC-LR), produced by cyanobacteria, accumulates in the liver through blood circulation. We investigated the impact of MC-LR on liver fibrosis. Mice received a daily injection of MC-LR at various concentrations for 14 consecutive days aa and then mouse liver was obtained for histopathological and immunoblot analysis. Next, a human hepatic stellate cell line (LX-2) was treated with MC-LR at various concentrations followed by measurement of cell viability, cell cycle and relevant protein expression levels. Our data confirmed the induction of mouse liver fibrosis after

Acute microcystin-LR exposure interfere thyroid hormones homeostasis in adult zebrafish (Danio rerio).

Microcystin-LR (MC-LR) in the aquatic environment may disturb thyroid hormone (TH) homeostasis. It is not clear how MC-LR affects downstream biological processes after TH disturbance. After exposure to 50, 100, 200 and 400 μg/L MC-LR for 24, 48, 72, or 96 h, alterations of the TH metabolism of adult zebrafish at thyroxine (T4), triiodothyronine (T3) levels, and iodothyronine deiodinase (Dio) activity, were observed. After exposure to MC-LR at 400 μg/L, T3 and T4 levels decreased significantly in females (p < 0.05) and returned to normal levels at 96 h. In males, T4 levels were not significantl

Sub-Chronic Microcystin-LR Liver Toxicity in Preexisting Diet-Induced Nonalcoholic Steatohepatitis in Rats.

Microcystin-LR (MCLR) is a hepatotoxic cyanotoxin reported to cause a phenotype similar to nonalcoholic steatohepatitis (NASH). NASH is a common progressive liver disease that advances in severity due to exogenous stressors such as poor diet and toxicant exposure. Our objective was to determine how sub-chronic MCLR toxicity affects preexisting diet-induced NASH. Sprague-Dawley rats were fed one of three diets for 10 weeks: control, methionine and choline deficient (MCD), or high fat/high cholesterol (HFHC). After six weeks of diet, animals received vehicle, 10 µg/kg, or 30 µg/kg MCLR via intra
