Capillary-assisted microfluidic biosensing platform captures single cell secretion dynamics in nanoliter compartments.

Cancer cells continuously secrete inflammatory biomolecules which play significant roles in disease progression and tumor metastasis toward secondary sites. Despite recent efforts to capture cancer cells' intercellular secretion heterogeneity using microfluidics, the challenges in operation of these systems as well as the complexity of designing a biosensing assay for long-term and real-time measurement of single cell secretions have become grand research barriers. Here, we present a new capillary-based microfluidic biosensing approach to easily and reliably capture ~500 single cells inside is

A microfluidics platform for studying ion and solute transport by isolated synaptic vesicles.

Loading of synaptic vesicles with neurotransmitters, an essential step in synaptic transmission, is mediated by vesicular neurotransmitter transporters that are fuelled by an electrochemical proton gradient across the vesicle membrane. While advances have recently been made in the structural and mechanistic understanding of these transporters, it is difficult to study transport kinetics and associated ion fluxes, largely due to technical constraints in accessing intracellular transporters. Here we describe a microfluidics platform that enables rapid solution changes, thus resolving solute flux

Simulation-Based Microfluidic Deformation Mapping for Region-Dependent Apparent Young's Modulus Estimation of Single Cells.

High-throughput microfluidic deformation assays enable label-free single-cell mechanophenotyping by quantifying how cells deform under controlled hydrodynamic loading. These approaches commonly extract deformation-related observables, such as projected area, axis ratio, and deformation index, and use them as indicators for cellular mechanical properties. However, deformation is not solely determined by stiffness; it is a coupled outcome of cell size, local hydrodynamic stress, and intrinsic mechanical response. Therefore, we present a simulation-based microfluidic framework for estimating regi

Microfluidic droplet-based liquid-liquid extraction and on-chip IR spectroscopy detection of cocaine in human saliva.

We present a portable microsystem to quantitatively detect cocaine in human saliva. In this system, we combine a microfluidic-based multiphase liquid-liquid extraction method to transfer cocaine continuously from IR-light-absorbing saliva to an IR-transparent solvent (tetrachloroethylene) with waveguide IR spectroscopy (QC-laser, waveguide, detector) to detect the cocaine on-chip. For the fabrication of the low-cost polymer microfluidic chips a simple rapid prototyping technique based on Scotch-tape masters was further developed and applied. To perform the droplet-based liquid-liquid extractio

A Dam-Inspired Liquid-Metal Fractal Microchannel Strain Sensor for Synergistic Hysteresis-Sensitivity Regulation and Multidirectional Sensing.

Flexible liquid-metal microfluidic strain sensors offer high stretchability and a wide sensing range for wearable electronics, yet their performance is often limited by viscoelastic hysteresis and low sensitivity caused by elastomeric substrates and uniformly deformed conductive channels. Here, we present a dam-inspired microfluidic strain sensor integrating a Peano fractal microchannel with embedded micropillar arrays. The fractal channel mimics a meandering riverbed to introduce multidirectional stress components that suppress hysteresis, while the micropillar arrays function as regulating "

Capture, Confine, Characterize: High-Throughput Dielectrophoresis-Based Single-Cell Microfluidics Platform to Analyze Mammalian and Yeast Cells Using Raman Spectroscopy.

Single-cell analysis technologies are pivotal in unraveling complex bihological mechanisms, yet existing platforms are often limited to sequencing-based end-point measurements, which fail to capture live cell dynamics. Here, we present a microfluidic-microelectronic device, the Microfluidic Dielectrophoretic Arresting System (MiDAS) that employs dielectrophoresis (DEP) for high-throughput single-cell and droplet trapping in a compact array. We tested multiple trap geometries, including a 20 µm-diameter DEP trap for polymer microbeads, fungal and mammalian cells, and a 40 µm-diameter trap for w

Dark-Pi Imaging System Permits Open-Source Label-Free Microfluidic Monitoring of Platelet Aggregation by Cellular Light Scattering.

Measuring platelet function is important for patient stratification to judge bleeding vs. thrombotic risk and for research into antiplatelet drugs to prevent cardiovascular disease. Variability in platelet function is not fully understood, and large studies of inter-individual variation are making gradual progress using laboratory measurements, but rapid and high-performance hematological tests are also needed. We present here a novel microfluidic technology for platelet function analysis that images light scatter by platelets, using a low-cost, open-source, high-throughput and customizable da

Microfluidic platform for understanding Parkinson's disease and α-synuclein conformation.

Microfluidic systems are an innovative engineering solution that is increasingly being used in a wide range of scientific fields. These systems use fluids in microchannels (1 to 300 microns) to analyze extremely small volumes of sample and reagent, allowing precise delivery and mixing while maintaining accurate results. Parkinson's disease (PD) poses significant diagnostic challenges, with early detection being critical to improved treatment outcomes. A key pathological feature of PD is the presence of Lewy bodies composed of α-synuclein (αSyn) fibrils. Recent research has shown that αSyn olig

Screening of angiotensin converting enzyme inhibitors from natural products via origami microfluidic paper-based analytical devices with colorimetric detection.

We report the screening of angiotensin converting enzyme (ACE) inhibitors on an origami microfluidic paper-based analytical device (μPAD) using colorimetric detection. The hydrolysis product reacts with ninhydrin, resulting in a purple color at the detection zones. Images of the μPADs are captured using a common cell phone and analyzed with Photoshop software. This platform allows six independent colorimetric reactions to take place simultaneously, and the IC50 values can be obtained in a single run within 22 min. The relative standard deviations of inhibition efficiencies are generally lower t

Microfluidics-guided localized low-temperature modulation of axonal signal propagation.

Low-temperature stimulation is recognized as a promising approach for neuromodulation, with the potential to suppress or slow neural activity. However, its impact on the spatial and electrophysiological properties of axonal conduction remains poorly understood. Conventional methods have lacked the spatial resolution necessary to isolate axon-specific responses to localized cooling. To overcome these limitations, we developed a microfluidic platform that integrates a microelectrode array (MEA) with a rapid and spatially confined cooling module. This platform enables real-time, phase-resolved mo

An Integrated Rotation-Programmed and Gravity-Driven Microfluidic Platform for Direct Bacterial Detection from Whole Blood.

For the precise molecular detection of bloodstream infection, sample processing is critical, as it involves purifying pathogenic DNA to circumvent the inhibitory effect of complex blood components. To integrate the cumbersome workflow encompassing the entire process to realize automated operation, passive methods such as gravity-driven microfluidics eliminate additional driving and controlling modules to simplify device setup and operations. Nevertheless, gravity-driven microfluidics encounters challenges in handling complex flow control and necessitates a relatively large volume of fluid to i

A plug-and-play microfluidic device for hydrogel fiber spinning.

Hydrogel fibers are promising biomaterials for a broad range of biomedical applications, including biosensing, drug delivery, and tissue engineering. Different types of microfluidic devices have been developed for hydrogel fiber spinning, however, they often require skillful fabrication procedures with special instruments such as 3D printers and clean-room facilities. On the other hand, microfluidic devices with predetermined and fixed configurations are susceptible to clotting, contamination, and damage, thereby creating a significant barrier for potential users. Herein, we describe a plug-an

Microwave-assisted extraction, separation, and chromogenic detection of laced marijuana for presumptive point-of-interdiction testing.

Presumptive drug screening enables timely procurement of search and arrest warrants and represents a crucial first step in crime scene analysis. Screening also reduces the burden on forensic laboratories which often face insurmountable backlogs. In most scenarios, on-site presumptive drug screening relies on chemical field tests for initial identification. However, even when used appropriately, these test kits remain limited to subjective colorimetric analysis, produce false positive or negative results with excessive sample quantities, and are known to cross-react with numerous innocuous subs

Highly fluorescent, monolithic semiconductor nanorod clusters for ultrasensitive biodetection.

We have developed highly fluorescent, monolithic colloidal CdSe seeded CdS nanorod clusters comprising thousands of nanorods. Their use in the sandwich assay detection of a model protein yields a thousand-fold improvement in the detection limit compared to individual nanorods, making them suitable for the detection of low abundance molecular targets.

A microfluidic system for studying the behavior of zebrafish larvae under acute hypoxia.

Oxygen is essential for metabolism of animals and is a vital component of their natural habitats. Hypoxic conditions in tissue, when oxygen levels are lower than normal, change a variety of cellular processes, while environmental hypoxia can have physiological and behavioral effects on the whole animal. Larval zebrafish respond to oxygen deprivation with a characteristic set of physiological changes and motor behaviors, making them a convenient vertebrate model to study hypoxia responses. However, to date, hypoxia studies in zebrafish are limited by the existing experimental setups, which only

Microfluidic tactile sensors for three-dimensional contact force measurements.

A microfluidic tactile sensing device has been first reported for three-dimensional contact force measurement utilizing the microfluidic interfacial capacitive sensing (MICS) principle. Consisting of common and differential microfluidic sensing elements and topologically micro-textured surfaces, the microfluidic sensing devices are intended not only to resolve normal mechanical loads but also to measure forces tangent to the surface upon contact. In response to normal or shear loads, the membrane surface deforms the underlying sensing elements uniformly or differentially. The corresponding var

Applications of planar microfluidic devices and gas chromatography for complex problem solving.

The application of planar microfluidic devices in GC for the separation of components of interest otherwise difficult to separate in a single analysis is presented. A variety of configurations were used for parallel chromatography, column effluent splitting, back flushing, selectivity tuning, valve less switching and column isolation, heart cutting, and comprehensive multidimensional chromatography. The synergies of recently commercialized planar microfluidic devices combined with the resolving power of fused-silica capillary columns are demonstrated. Difficult separations were accomplished in

Fish and Chips: a microfluidic perfusion platform for monitoring zebrafish development.

We have developed a multi-channel microfluidic perfusion platform for culturing zebrafish embryos and capturing live images of various tissues and organs inside the embryo. The Fish and Chips was micro-fabricated in silicon and glass for reproducibility and accuracy of the microfluidic structure. The microfluidic platform consists of three parts: a microfluidic gradient generator, a row of eight fish tanks, in which the fish embryos are individually placed, and eight output channels. The fluidic gradient generator supports dose-dependent drug and chemical studies. A unique perfusion system ens

Sickle cell vasoocclusion and rescue in a microfluidic device.

The pathophysiology of sickle cell disease is complicated by the multiscale processes that link the molecular genotype to the organismal phenotype: hemoglobin polymerization occurring in milliseconds, microscopic cellular sickling in a few seconds or less [Eaton WA, Hofrichter J (1990) Adv Protein Chem 40:63-279], and macroscopic vessel occlusion over a time scale of minutes, the last of which is necessary for a crisis [Bunn HF (1997) N Engl J Med 337:762-769]. Using a minimal but robust artificial microfluidic environment, we show that it is possible to evoke, control, and inhibit the collect
