Quantifying the spatio-temporal image degradation under motion blur in fluorescence microscopy.

The spatial resolution of optical imaging systems is fundamentally restricted by the diffraction limit. However, in widefield live-cell microscopy, the achievable resolution is further constrained by the specimen motion, which indicates the existence of a fundamental spatio-temporal resolution trade-off between signal accumulation during the full frame integration and the resulting motion blur. To improve the fidelity with which moving objects can be imaged, a quantitative understanding of this spatio-temporal trade-off is necessary. Here, we present a systematic analysis of motion-induced res

Two-Photon Fluorescence and Second-Harmonic Generation Microscopy: Principles and Biomedical Applications.

Two-photon microscopy (TPM) has revolutionized biological imaging by enabling high-resolution, deep-tissue visualization with reduced photobleaching and photodamage. This review discusses complementary nonlinear optical imaging modalities such as two-photon fluorescence (TPF), second-harmonic generation (SHG), and fluorescence lifetime imaging (FLIM) microscopy that together provide structural, molecular, and metabolic information. The review begins by introducing the basic principles, instrumentation, and capabilities of two-photon fluorescence microscopy and FLIM. Followed by the development

Spatial analysis of tissue immunity and vascularity by light sheet fluorescence microscopy.

The pathogenesis of cancer and cardiovascular diseases is subjected to spatiotemporal regulation by the tissue microenvironment. Multiplex visualization of the microenvironmental components, including immune cells, vasculature and tissue hypoxia, provides critical information underlying the disease progression and therapy resistance, which is often limited by imaging depth and resolution in large-volume tissues. To this end, light sheet fluorescence microscopy, following tissue clarification and immunostaining, may generate three-dimensional high-resolution images at a whole-organ level. Here

Quantitative Monitoring of GPCR-Mediated Spatiotemporal IP3 Dynamics Using Confocal Fluorescence Microscopy.

Activation of G protein-coupled receptors upon chemoattractant stimulation induces activation of multiple signaling pathways. To fully understand how these signaling pathway coordinates to achieve directional migration of neutrophils, it is essential to determine the dynamics of the spatiotemporal activation profile of signaling components at the level of single living cells. Here, we describe a detailed methodology for monitoring and quantitatively analyzing the spatiotemporal dynamics of 1,4,5-inositol trisphosphate (IP3) in neutrophil-like HL60 cells in response to various chemoattractant f

A versatile Wavelet-Enhanced CNN-Transformer for improved fluorescence microscopy image restoration.

Fluorescence microscopes are indispensable tools for the life science research community. Nevertheless, the presence of optical component limitations, coupled with the maximum photon budget that the specimen can tolerate, inevitably leads to a decline in imaging quality and a lack of useful signals. Therefore, image restoration becomes essential for ensuring high-quality and accurate analyses. This paper presents the Wavelet-Enhanced Convolutional-Transformer (WECT), a novel deep learning technique developed specifically for the purpose of reducing noise in microscopy images and attaining supe

Evaluation of Blood-CSF Barrier Transport by Quantitative Real Time Fluorescence Microscopy.

PURPOSE: Transporters at the blood-cerebrospinal fluid (CSF) barrier (BCSFB) play active roles in removing drugs and toxins from the CSF. The goal of this study is to develop a fluorescence microscopy approach to quantitatively study the transepithelial transport processes at the murine BCSFB in real time. METHODS: Choroid plexus (CP) tissues were isolated from mouse lateral ventricles and incubated with anionic (fluorescein-methotrexate, 8-fluorescein-cAMP) or cationic (IDT307) fluorescent probes. The CSF-to-blood transport was imaged and quantified using compartmental segmentation and digit

Three-dimensional wide-field fluorescence microscopy for transcranial mapping of cortical microcirculation.

Wide-field fluorescence imaging is an indispensable tool for studying large-scale biodynamics. Limited space-bandwidth product and strong light diffusion make conventional implementations incapable of high-resolution mapping of fluorescence biodistribution in three dimensions. We introduce a volumetric wide-field fluorescence microscopy based on optical astigmatism combined with fluorescence source localization, covering 5.6×5.6×0.6 mm3 imaging volume. Two alternative configurations are proposed exploiting multifocal illumination or sparse localization of point emitters, which are herein seaml

A polymer index-matched to water enables diverse applications in fluorescence microscopy.

We demonstrate diffraction-limited and super-resolution imaging through thick layers (tens-hundreds of microns) of BIO-133, a biocompatible, UV-curable, commercially available polymer with a refractive index (RI) matched to water. We show that cells can be directly grown on BIO-133 substrates without the need for surface passivation and use this capability to perform extended time-lapse volumetric imaging of cellular dynamics 1) at isotropic resolution using dual-view light-sheet microscopy, and 2) at super-resolution using instant structured illumination microscopy. BIO-133 also enables immob

Nitroxide amide-BODIPY probe behavior in fibroblasts analyzed by advanced fluorescence microscopy.

A novel synthesized nitroxide amide-BODIPY prefluorescent probe was used to study cellular redox balance that modulates nitroxide/hydroxylamine ratio in cultured human fibroblasts. FLIM quantitatively differentiated between nitroxide states of the cytoplasm-localized probe imaged by TIRF, monitoring nitroxide depletion by hydrogen peroxide; eluding incorrect interpretation if only fluorescence intensity is considered.

Green-to-red photoconvertible Dronpa mutant for multimodal super-resolution fluorescence microscopy.

Advanced imaging techniques crucially depend on the labels used. In this work, we present the structure-guided design of a fluorescent protein that displays both reversibly photochromic and green-to-red photoconversion behavior. We first designed ffDronpa, a mutant of the photochromic fluorescent protein Dronpa that matures up to three times faster while retaining its interesting photochromic features. Using a combined evolutionary and structure-driven rational design strategy, we developed a green-to-red photoconvertible ffDronpa mutant, called pcDronpa, and explored different optimization st

Two-photon fluorescence microscopy imaging of cellular oxidative stress using profluorescent nitroxides.

A range of varying chromophore nitroxide free radicals and their nonradical methoxyamine analogues were synthesized and their linear photophysical properties examined. The presence of the proximate free radical masks the chromophore's usual fluorescence emission, and these species are described as profluorescent. Two nitroxides incorporating anthracene and fluorescein chromophores (compounds 7 and 19, respectively) exhibited two-photon absorption (2PA) cross sections of approximately 400 G.M. when excited at wavelengths greater than 800 nm. Both of these profluorescent nitroxides demonstrated

Heterogeneous amylin fibril growth mechanisms imaged by total internal reflection fluorescence microscopy.

Total internal reflection fluorescence microscopy has been used to visualize the fibrillization of amylin, a hormone which in aggregated forms plays a role in type 2 diabetes pathology. Data were obtained at acidic pH where fibrillization is hindered by the charging of histidine 18 and at slightly basic pH where the loss of charge on the histidine promotes aggregation. The experiments show three types of aggregate growth processes. In the earliest steps globular seeds are formed with some expanding radially during the course of the reaction. The dimensions of the globular seeds as well as thei

Fluorescence microscopy and flow cytometry in measuring activated caspases in human spermatozoa.

Staining of spermatozoa with the fluorescein-Val-Ala-Asp-fluoromethylketone has already been performed on ejaculated sperm samples, using fluorescence microscopy (FM) or flow cytometry (FCM) in order to score activated caspases. This assay may help in assessing apoptosis and its role in male fertility. The present study compares the above two techniques in order to adopt the most accurate method for detection in human frozen-thawed testicular, epididymal and ejaculated spermatozoa. The analyses were carried out on frozen/thawed testicular (n = 14), epididymal (n = 8) and ejaculated (n = 10) sp

Fluorescence microscopy: established and emerging methods, experimental strategies, and applications in immunology.

Cutting-edge biophysical technologies including total internal reflection fluorescence microscopy, single molecule fluorescence, single channel opening events, fluorescence resonance energy transfer, high-speed exposures, two-photon imaging, fluorescence lifetime imaging, and other tools are becoming increasingly important in immunology as they link molecular events to cellular physiology, a key goal of modern immunology. The primary concern in all forms of microscopy is the generation of contrast; for fluorescence microscopy contrast can be thought of as the difference in intensity between th

Observing three-dimensional human microvascular and myogenic architecture using conventional fluorescence microscopy.

Microangiography and vascular casting have previously been used to demonstrate the three-dimensional architecture of human uterine microvasculature. However, a limitation of these perfusion-dependent techniques is the difficulty in identifying surrounding tissue components. We have previously shown that it is possible to visualise microvascular networks on the cut surfaces of fresh tissue specimens by diffusive labelling of vascular endothelium with fluorescently conjugated UEA-1 lectin. Unlike perfusion methods that are limited to accessible vascular networks, diffusive fluorescence labelling

Clinical results after doxorubicin extravasation treated with excision guided by fluorescence microscopy.

Doxorubicin (DR) and epirubicin (ER) produce progressive tissue necrosis when extravasation occurs. Early detection and excision of all affected tissue is important. The clinical experience with fluorescence microscopic guided detection and excision in 24 patients is evaluated. 9 patients with fluorescence negative specimens were kept under observation without excision. None developed necrosis. Wide excision was performed on 15 patients with fluorescence positive specimens. Sequelae, defined as impaired function of the affected limb at the last control examination in the out-patient clinic, we

Comparison of acridine orange fluorescent microscopy and gram stain light microscopy for the rapid detection of bacteria in cerebrospinal fluid.

OBJECTIVE: Bacterial meningitis is a life threatening condition that requires prompt recognition and treatment. Currently, Gram stain is widely used for the microscopic detection of bacterial pathogens in cerebrospinal fluid (CSF). In Nepal, fluorescent microscopes have been installed in laboratories as a part of the National tuberculosis control program. However, information on the utility of the acridine orange (AO) stain for the direct detection of bacteria in CSF samples in Nepal is not available. Therefore, this study aims to compare Gram stain and AO stain for the rapid detection of bact

Light-Sheet Fluorescence Microscopy Imaging of Whole-Mount Immunostained Mouse Embryos and Limbs.

Animal development is the complex process that enables the formation of a pluricellular organism starting from a fertilized zygote. During this process, different tissues need to grow and differentiate in a coordinated manner in order to generate a fully functional organism. Oftentimes, technical or experimental difficulties cause imaging studies to focus specifically on the development of a specific tissue as if it were independent of the development of closely related structures. Further, classical analyses are performed on histological sections in two dimensions, which often only cover part

Assessing cellular metabolic dynamics with two-photon NAD(P)H fluorescence polarization microscopy.

Altered metabolism enables adaptive advantages for cancer cells, driving the need for improved methods for noninvasive long-term monitoring of cellular metabolism. Here, we present a fast live-cell NADH imaging method that provides a real-time measurement of the fractional level of unbound NADH and show that it is a robust indicator of a cell's metabolic status. The method, two-photon fluorescence polarization ratiometric microscopy (FPRM), is easy and inexpensive to implement and more than an order of magnitude faster than fluorescence lifetime imaging microscopy (FLIM), a common means of ass
