Kinematic analysis of Toxoplasma gondii motility.

Toxoplasma gondii tachyzoites execute a complex and little understood combination of rapid movements to reach and penetrate human or other animals cells. In the present study, computer-assisted simulation was used to quantitatively analyze the motility of these parasites in three-dimensional space with spatial and temporal resolutions in the micrometer and subsecond ranges. A digital model based on electron-micrographs of a serially sectioned tachyzoite was animated according to a videomicrographed sequence of a characteristic repetitive movement. Keyframe animation defined over 150 frames by

Rapid Motility Arrest of the Lyme Disease Spirochete, Borrelia burgdorferi, by Vaccine-Induced OspA Antibodies.

Outer surface protein A (OspA)-based Lyme disease vaccines elicit antibodies that inhibit transmission of infectious Borrelia burgdorferi spirochetes from ticks to humans, although the mechanism by which this occurs is unclear. Here we demonstrate using high-resolution, single-cell fluorescence video microscopy that B. burgdorferi motility is arrested within minutes by human OspA-specific monoclonal antibodies and polyclonal OspA antisera from mice immunized with a candidate OspA mRNA lipid nanoparticle vaccine.

[Experimental studies for the improvement of facial nerve regeneration].

Using a combination of the following, it is possible to investigate procedures to improve the morphological and functional regeneration of the facial nerve in animal models: 1) retrograde fluorescence tracing to analyse collateral axonal sprouting and the selectivity of reinnervation of the mimic musculature, 2) immunohistochemistry to analyse both the terminal axonal sprouting in the muscles and the axon reaction within the nucleus of the facial nerve, the peripheral nerve, and its environment, and 3) digital motion analysis of the muscles. To obtain good functional facial nerve regeneration,

Objective skill assessment for cataract surgery from surgical microscope video.

OBJECTIVE: Video offers an accessible method for automated surgical skill evaluation; however, many platforms still rely on traditional six-degree-of-freedom (6-DOF) tracking systems, which can be costly, cumbersome, and challenging to apply clinically. This study aims to demonstrate that trainee skill in cataract surgery can be assessed effectively using only object detection from monocular surgical microscope video. METHODS: One ophthalmologist and four residents performed cataract surgery on a simulated eye five times each, generating 25 recordings. Recordings included both the surgical mi

Dermatoscopy: alternative uses in daily clinical practice.

Dermatoscopy, also known as dermoscopy, epiluminescence microscopy, or surface microscopy, is a noninvasive technique allowing rapid and magnified (× 10) in vivo observation of the skin with the visualization of morphologic features often imperceptible to the naked eye. Videodermatoscopy (VD) represents the evolution of dermatoscopy and is performed with a video camera equipped with lenses providing higher magnification (× 10 to × 1000). Over the past few years, both dermatoscopy and VD have been demonstrated to be useful in a wide variety of cutaneous disorders, including ectoparasitic infest

3D+t human sperm flagellum centerline dataset.

The study of human sperm motility has been a topic of interest for decades due to its crucial role in fertility and reproductive health. While most analyses rely on 2D+t imaging of head trajectories, sperm naturally swim in three dimensions (3D), driven by complex flagellar motion. However, the lack of comprehensive 3D+t datasets has limited progress in this field. To address this, we present 3D-SpermFlagella, the first large-scale 3D+t dataset of human sperm flagellum centerline annotations. This dataset contains 135 tracked and annotated sperm, derived from our previously published multifoca

3D digital image microscope system-assisted vasovasostomy and vasoepididymostomy in rats.

Optimal vision and ergonomics are essential factors contributing to the achievement of good results during microsurgery. The three-dimensional (3D) digital image microscope system with a better 3D depth of field can release strain on the surgeon's neck and back, which can improve outcomes in microsurgery. We report a randomized prospective study of vasoepididymostomy and vasovasostomy using a 3D digital image microscope system (3D-DIM) in rats. A total of 16 adult male rats were randomly divided into two groups of 8 each: the standard operating microscope (SOM) group and the 3D-DIM group. The

Videodermoscopy: a useful tool for diagnosing congenital triangular alopecia.

Congenital triangular alopecia, despite its name, usually presents in children between 3 and 6 years of age, but adult patients have been reported. It is not uncommon for triangular alopecia to be misdiagnosed as alopecia areata and treated for such. This is especially true when a lesion of triangular alopecia presents in an area of the scalp other than the typical fronto-temporal hairline or later in adulthood. Videodermoscopy may serve as a useful tool to perform the right diagnosis as it can highlight signs not seen by the unaided eye and may be able to spare the patient from a biopsy.

In vivo length oscillations of indirect flight muscles in the fruit fly Drosophila virilis.

We have used high-speed video microscopy to measure in vivo length oscillations of the indirect flight muscles of the fruit fly Drosophila virilis during tethered flight. The changes in muscle strain were measured by tracking the deformation of the thoracic exoskeleton at the origin and insertion of both the dorsal longitudinal (DLM) and the dorsal ventral (DVM) muscles. The mean peak-to-peak strain amplitudes were found to be 3.5% for the DLMs and 3.3% for the DVMs, although the strain amplitude within individual cycles ranged from 2 to 5%. These values are consistent with the small number of

Analysis of geometric parameters of thrombogenesis at its final stage in rats suggests that cessation of the thromb growth depends both on altered hydrodynamic parameters and on the interaction with the biochemical factors released from the vascular wall and present in the blood flow.

DYNC2H1 mutation causes Jeune syndrome and recurrent lung infections associated with ciliopathy.

Asphyxiating thoracic dystrophy, also known as Jeune syndrome, is included in a group of syndromic skeletal ciliopathies associated with mutations in genes encoding proteins involved in the formation or function of motile cilia. Herein, we report a 6-mo-old male admitted to hospital with recurrent lung infections, thoracic dystrophy, and respiratory distress that was diagnosed as Jeune syndrome; DYNC2H1 mutation was detected via genetic analysis and ciliary dysfunction was noted via high-speed video microscopy.

Diagnostic Methods in Primary Ciliary Dyskinesia.

Diagnosing primary ciliary dyskinesia is difficult. With no reference standard, a combination of tests is needed; most tests require expensive equipment and specialist scientists. We review the advances in diagnostic testing over the past hundred years, with emphasis on recent advances. We particularly focus on use of high-speed video analysis, transmission electron microscopy, nasal nitric oxide and genetic testing. We discuss the international efforts that are in place to advance the evidence base for diagnostic tests.

Discriminating single-molecule binding events from diffraction-limited fluorescence.

Single-molecule localization microscopy enables high-resolution imaging of molecular interactions, but discriminating molecular binding types has traditionally relied on complex strategies, such as multiple dyes, time-division techniques, or kinetic analysis, that are asynchronous, invasive, or time-consuming. Here, we uncover previously overlooked spatiotemporal information embedded within diffraction-limited fluorescence, enabling synchronous classification of individual binding event videos using only a single fluorescent dye. Building on this insight, we propose a Temporal-to-Context Convo

A patch-based method for repetitive and transient event detection in fluorescence imaging.

Automatic detection and characterization of molecular behavior in large data sets obtained by fast imaging in advanced light microscopy become key issues to decipher the dynamic architectures and their coordination in the living cell. Automatic quantification of the number of sudden and transient events observed in fluorescence microscopy is discussed in this paper. We propose a calibrated method based on the comparison of image patches expected to distinguish sudden appearing/vanishing fluorescent spots from other motion behaviors such as lateral movements. We analyze the performances of two

Confocal microscopy of corneal graft rejection.

Corneal allografts were transplanted into inflamed and vascularized graft beds in rabbit eyes. The grafts were examined every 4 days by slit-lamp biomicroscopy and scanning confocal microscopy. Confocal images were recorded with a video camera and computer enhanced in real-time. Layers of the cornea were visualized in serial optical sections parallel to the epithelium. In the third postoperative week, signs of graft rejection were observed; slit-lamp examination revealed a circumferential line of epithelial rejection, along with cloudiness and edema. Vessels were observed growing into the graf
