A computational model of cardiovascular physiology and heart sound generation.

A computational model of the cardiovascular system is described which provides a framework for implementing and testing quantitative physiological models of heart sound generation. The lumped-parameter cardiovascular model can be solved for the hemodynamic variables on which the heart sound generation process is built. Parameters of the cardiovascular model can be adjusted to represent various normal and pathological conditions, and the acoustic consequences of those adjustments can be explored. The combined model of the physiology of cardiovascular circulation and heart sound generation has p

[A transgenic animal model: new possibilities for cardiovascular research].

By means of molecular biology and genetic research the influence of genetic factors on a great variety of human diseases could be shown. In the field of cardiovascular research the genetic defects of a few monogenetic disorders, such as Marfan's syndrome and hypertrophic cardiomyopathy, have been characterized. In addition, candidate genes for polygenetic diseases, such as arterial hypertension and atherosclerosis, have been cloned. However, the identification of a candidate gene or its mutation does not prove its influence on the phenotype or the final cause of a particular disease. Only a ta

Cardiovascular origins of heatstroke pathophysiology: an anesthetized rat model.

Opinion is divided on issues concerning a cardiovascular origin of heatstroke pathophysiology as well as those concerning the mechanism of circulatory failure in heat. Such controversies seem to arise because of insufficient data covering the prodromal period of heatstroke. A reappraisal of the nature and sequence of early circulatory responses to heat stress was, consequently, undertaken by subjecting male albino rats to an environmental temperature of 45 degrees C at 15% relative humidity until overt circulatory failure occurred. Although the initial circulatory responses were normal for mil

Mechanistic Interpretation of Beat-to-Beat Cardiovascular Variability in Hypertension Using a Closed-Loop Cardiovascular Model.

Beat-to-beat heart rate variability (HRV) and blood pressure variability (BPV) are gaining attention as noninvasive markers of autonomic dysfunction, particularly, for improving hypertension management. However, interpretation is complicated by coexisting autonomic dysregulation and vascular mechanical alterations in hypertension. This study used a numerical model to disentangle these contributions, which cannot be isolated in vivo. A closed-loop mathematical model was developed to simulate cardiovascular variability during head-up tilt (HUT), incorporating four heart chambers, systemic and pu

Modelling maternal cardiovascular adaptation to pregnancy: a scoping review.

BACKGROUND: The maternal cardiovascular system undergoes a profound transformation during pregnancy. Disordered cardiovascular adaptation is associated with pre-eclampsia and fetal growth restriction; however, these complications remain challenging to predict and manage. Computational modelling presents an opportunity to study the maternal circulation with unprecedented flexibility. We conducted a scoping review to assess the potential utility of computational models for studying maternal cardiovascular adaptation to pregnancy and providing insights into disease. We aimed to identify research

A coupled hydrodynamic model of the cardiovascular and cerebrospinal fluid system.

Coupling of the cardiovascular and cerebrospinal fluid (CSF) system is considered to be important to understand the pathophysiology of cerebrovascular and craniospinal disease and intrathecal drug delivery. A coupled cardiovascular and CSF system model was designed to examine the relation of spinal cord (SC) blood flow (SCBF) and CSF pulsations along the spinal subarachnoid space (SSS). A one-dimensional (1-D) cardiovascular tree model was constructed including a simplified SC arterial network. Connection between the cardiovascular and CSF system was accomplished by a transfer function based o

Cardiovascular function in a rat model of diet-induced obesity.

The obesity-prone/obesity-resistant rat model has been used to study mechanisms responsible for obesity-related abnormalities in renal function and blood pressure, but whether this model exhibits cardiac dysfunction has not been determined. We tested the hypothesis that obesity-prone rats would display cardiovascular abnormalities seen in other diet-induced obese models (ie, hypertension, tachycardia, left ventricular hypertrophy, increased collagen deposition, reduced cardiac contractility, and increased end diastolic pressure). Male Sprague-Dawley rats were fed a control diet or a moderate f

Are multimodal AIs predicting proxies for age and sex in sleep-based cardiovascular risk models?

Several recent works have focused on predicting risk for incident cardiovascular disease (CVD) from routine sleep studies using AI. These works aim to utilize the breadth of multimodal sleep study signal data to better identify and stratify people who are more at risk for disease and specific groups that would benefit from therapy. The predominant sleep study cohorts used for CVD risk predictive modeling are the Sleep Heart Health Study (SHHS), the Multi-Ethnic Study of Atherosclerosis (MESA), and the Osteoporotic Fractures in Men Study (MrOS). In this perspective, we show that these datasets

Targeting osteoblast fatty acid metabolism attenuates skeletal and cardiovascular complications in chronic kidney disease models.

Chronic kidney disease (CKD) disrupts mineral homeostasis, leading to impaired skeletal mineralization and cardiovascular pathology, yet the mechanism linking these processes remains undefined. Here we identify glycerol-3-phosphate acyltransferase 2 (GPAT2) as a regulator that couples free fatty acid (FFA) partitioning in osteoblasts to systemic phosphate balance. In mouse and human CKD, elevated osteoblast GPAT2 routes FFA away from mitochondrial oxidation, limiting phosphate incorporation into bone, and toward lysophosphatidic acid synthesis, increasing production of the phosphaturic hormone

Artificial Intelligence in Cardiovascular Ultrasound: Clinical Applications, Foundation Models, and the Path to Precision Cardiology.

Cardiovascular ultrasound is a cornerstone of noninvasive cardiac and vascular assessment, yet conventional interpretation remains operator-dependent, variable, and limited in sensitivity for subclinical disease. Artificial intelligence (AI), particularly machine learning (ML), deep learning (DL), and, most recently, vision-language and foundation models, offers tools to automate, standardize, and extend ultrasound analysis. This narrative review examines the role of AI-enhanced cardiovascular ultrasound in the transition from descriptive imaging toward predictive and personalized medicine. We

Development and evaluation of cardiovascular disease risk prediction models for patients with type 2 diabetes.

To facilitate treatment decisions in people at risk of Cardiovascular Disease (CVD), several risk equations such as the Pooled Cohort Equations and Predicting Risk of Cardiovascular Disease Events (PREVENT) equations have been developed to estimate CVD risk for primary prevention patients. However, it is unclear whether these equations achieve high predictive accuracy and fairness in patients with type 2 diabetes (T2D), and whether a T2D-specific risk equation is needed. Accordingly, we developed a Weibull Accelerated Failure Time (AFT) survival model for predicting the 3-year CVD risk in 23,7

Cardiovascular events and mortality after breast cancer diagnosis: multistate models in a nationwide cohort study.

OBJECTIVE: To study the progression of newly diagnosed breast cancer patients towards cardioembolic disorders (CDs) and ischaemic or structural heart disease (ISH) within a multistate framework that includes death as an absorbing state, in order to evaluate long-term cardiovascular burden and to examine whether the observed patterns may reflect short-term acute events or longer-term effects potentially associated with treatment. CD and ISH occurrences following breast cancer treatment may have different underlying biological mechanisms, potentially leading to different survival patterns across

Integrating mental health and cardiovascular wellness: synergistic impacts and the promise of comprehensive care models.

This review explores the bidirectional relationship between mental health disorders and cardiovascular disease (CVD), highlighting the potential of integrated healthcare models to improve outcomes. While CVD remains the leading cause of global mortality, traditionally linked to risk factors like hypertension and diabetes, emerging evidence shows that mental health conditions, especially depression and anxiety, significantly increase CVD risk through mechanisms such as chronic stress, inflammation, and neuroendocrine dysregulation. Activation of the hypothalamic-pituitary-adrenal axis and sympa

Preclinical research models for evaluating the biocompatibility of bioresorbable metallic cardiovascular stents: A comparative review.

Cardiovascular stents are widely used to treat atherosclerosis by relieving vascular obstruction and providing structural support after coronary angioplasty. Bioresorbable metallic stents represent a promising alternative to conventional corrosion-resistant stents, which are linked to late-stage complications such as in-stent restenosis and thrombosis. Due to the diversity of stent materials and designs, rigorous evaluation of their interactions with the vascular environment in relevant preclinical models is essential before clinical translation. However, current studies employ highly variable

Development of postoperative delirium prediction models in patients undergoing cardiovascular surgery using machine learning algorithms.

Associations between delirium and postoperative adverse events in cardiovascular surgery have been reported and the preoperative identification of high-risk patients of delirium is needed to implement focused interventions. We aimed to develop and validate machine learning models to predict post-cardiovascular surgery delirium. Patients aged ≥ 40 years who underwent cardiovascular surgery at a single hospital were prospectively enrolled. Preoperative and intraoperative factors were assessed. Each patient was evaluated for postoperative delirium 7 days after surgery. We developed machine learni

A descriptive investigation of clinical practice models used by cardiovascular genetic counselors in North America.

Cardiovascular genetic counseling has expanded as an established genetic counseling specialty over the last 20 years. Despite guidelines recommending genetic counseling for heritable cardiac diseases, there have been limited descriptions of the practice model types used for different clinical indications seen in this genetic counseling subspecialty. We aimed to describe current clinical practice models used by cardiovascular genetic counselors and to document practice model strengths, challenges, and areas for improvement. Genetic counselor respondents (n = 63) who self-reported seeing cardiov

Development and validation of risk prediction models for multiple cardiovascular diseases and Type 2 diabetes.

Accurate risk assessment of an individuals' propensity to develop cardiovascular diseases (CVDs) is crucial for the prevention of these conditions. Numerous published risk prediction models used for CVD risk assessment are based on conventional risk factors and include only a limited number of biomarkers. The addition of novel biomarkers can boost the discriminative ability of risk prediction models for CVDs with different pathogenesis. The present study reports the development of risk prediction models for a range of heterogeneous CVDs, including coronary artery disease (CAD), stroke, deep ve

Comprehensive Analysis of Animal Models of Cardiovascular Disease using Multiscale X-Ray Phase Contrast Tomography.

Cardiovascular diseases (CVDs) affect the myocardium and vasculature, inducing remodelling of the heart from cellular to whole organ level. To assess their impact at micro and macroscopic level, multi-resolution imaging techniques that provide high quality images without sample alteration and in 3D are necessary: requirements not fulfilled by most of current methods. In this paper, we take advantage of the non-destructive time-efficient 3D multiscale capabilities of synchrotron Propagation-based X-Ray Phase Contrast Imaging (PB-X-PCI) to study a wide range of cardiac tissue characteristics in

Share and enjoy: anatomical models database--generating and sharing cardiovascular model data using web services.

Sharing data between scientists and with clinicians in cardiac research has been facilitated significantly by the use of web technologies. The potential of this technology has meant that information sharing has been routinely promoted through databases that have encouraged stakeholder participation in communities around these services. In this paper we discuss the Anatomical Model Database (AMDB) (Gianni et al. Functional imaging and modeling of the heart. Springer, Heidelberg, 2009; Gianni et al. Phil Trans Ser A Math Phys Eng Sci 368:3039-3056, 2010) which both facilitate a database-centric
