Low molecular weight IgM in B cell lymphoproliferative disorders.

Circulating low molecular weight (LMW) IgM was demonstrated in five of 38 patients with B cell lymphoproliferative disorders. These five patients all had malignant disease and could be subdivided into two groups. In the first group were three patients, each with an associated serum IgM paraprotein; two had Waldenström's macroglobulinemia, and one lymphocytic lymphoma. The two patients of the second group did not have IgM paraproteins; one had lymphocytic lymphoma and one chronic lymphocytic leukemia. Both these patients also had acquired C1 esterase inhibitor deficiency, a previously recognise

[Low molecular weight nuclear RNA of human gastric adenocarcinoma].

Low molecular weight nuclear RNAs (LMWN RNAs) isolated by thermal phenol fractionation from malignant tumor of the stomach and the mucous membrane of patients with gastric ulcer have been studied. There were found some differences in the relative content of U1 and U2 nucleoplasmic fractions as well as of loosely bound to a nucleolus U3 fraction of LMWN RNAs. The changes observed resemble those obtained earlier for experimental animal tumors.

High molecular weight forms of human ACTH are glycoproteins.

The RIA-ACTH in a normal human pituitary, 2 ectopic ACTH-secreting tumors and plasma from a patient with Nelson's syndrome and one with ectopic ACTH syndrome was divided into 3 molecular weight classes after gel exclusion chromatography. The largest component appeared in or near the void volume and was designated "big" RIA-ACTH. The second, designated "intermediate" RIA-ACTH, eluted between the void volume and standard human 1-39 ACTH (mol. wt. 4,541). An immunoreactive material designated "little" ACTH-coeluted with standard human ACTH. A significant fraction (29-61%) of "big" RIA-ACTH from t

Low molecular weight proteinuria in association with paroxysmal myoglobinuria.

A patient with paroxysmal myoglobinuria presented with low molecular weight (LMW) proteinuria in association with an episode of exertional myoglobinuria. Since no signs of acute renal failure were present, the cause was probably competition between myoglobin and other LMW proteins for proximal tubular reabsorption. Agarose gel electrophoresis was found to be an excellent method for the investigation of myoglobinuria since this technique not only allowed the separation of myoglobin from hemoglobin but also myoglobin from metmyoglobin.

Molecular and pharmacologic profile of tinzaparin and a comparable low-molecular-weight bacterial sulfaminoheparosan.

Low-molecular-weight heparins (LMWH) represent depolymerized porcine mucosal heparin derivatives, which are commonly used for the management of thrombotic disorders. Because of their widespread usage, the supplies of the raw material namely unfractionated heparin are nearly exhausted. Porcine mucosal tissue is almost exclusively used for the preparation of these agents. Thus, there is a timely need for the production of heparin like drugs from other sources. Fermentation techniques have been used to produce carbohydrates such as dextran and innulin for therapeutic purposes. Bacterial cell wall

Molecular cloning of a novel high molecular weight mucin (MG1) from human sublingual gland.

A human sublingual gland cDNA library was screened with a polyclonal antiserum against deglycosylated MG1 and a positive clone, pSM2-1, was isolated which codes for 196 amino acids in the carboxyl-terminal region of this mucin. This region is cysteine-rich and contains a C2-like domain upstream of the extreme carboxyl-terminal domain in which the arrangement of cysteines is nearly identical to that in human von Willebrand factor, human intestinal mucin MUC2, human tracheobronchial mucin MUC5 and porcine and bovine submaxillary gland mucins. Northern analyses with pSM2-1 showed MG1 transcripts

[The effect of low molecular weight substances on the human skin. Molecular mechanisms and their consequences].

Interactions between low molecular weight compounds with cells of the skin result in reactions with different proteins which enable the uptake, metabolism and efflux of these compounds. It is unlikely, that small molecular weight compounds can achieve pharmacological concentrations within cells by diffusion alone. The pattern of influx proteins of keratinocytes is different from that of hepatocytes. If the balance between these systems is disturbed, the skin may become unable to function as a protective organ which can result in diseases including cancer or-more frequently-allergic contact der

Cellular and Molecular Effects of High-Molecular-Weight Heparin on Matrix Metalloproteinase 9 Expression.

Blood sampling with different anticoagulants alters matrix metalloproteinase (MMP-) 9 expression, thus influencing its concentration and diagnostic validity. Here, we aimed to evaluate the effects of different anticoagulants on MMP-9 regulation. MMP-9 expression was assessed in response to ethylenediaminetetraacetic acid, citrate, and high-/low-molecular-weight heparin (HMWH, LMWH) in co-culture experiments using THP-1, Jurkat, and HT cells (representing monocytes, T, and B cells). Triple and double cell line co-culture experiments revealed that HMWH treatment of THP-1 and Jurkat led to a sign

PEG Middle-Block Molecular Weight as a Quantitative Design Variable for Stereocomplex Formation Efficiency in High-Molecular-Weight PLLA-PEG-PLLA/PDLA Blends.

Achieving complete stereocomplex (SC) formation in high-molecular-weight PLLA/PDLA blends by injection molding remains a challenge because the rapid mold-cooling rate suppresses the slow enantiomeric chain-recognition process. Here, we demonstrate that the PEG middle-block molecular weight in PLLA-PEG-PLLA triblock copolymers is the decisive kinetic control variable governing SC formation efficiency (f SC). Copolymers with PEG M n = 400, 4000, and 35,000 g/mol were synthesized by bulk ring-opening polymerization (ROP) at two PLLA-block M n targets (100,000 and 200,000 g/mol) and blended 1:1 wi

High molecular weight polyethylene glycol cellular distribution and PEG-associated cytoplasmic vacuolation is molecular weight dependent and does not require conjugation to proteins.

Conjugation of therapeutic proteins with high molecular weight polyethylene glycols (HMW PEGs) is used to extend the half-life of biologics. To evaluate the effects of HMW PEGs in animals, we used an immunohistochemical procedure to study the tissue distribution and toxicity of unconjugated HMW PEGs in rats given 100 mg/kg (10K)PEG, (20K)PEG, or (40K)PEG intravenously. Both the PEG cellular distribution and the histology were different between groups. In (10K)PEG and (20K)PEG groups, PEG immunoreactivity was most prominent in the renal tubule epithelium and in alveolar macrophages and hepatic

Therapeutic timing and molecular properties influence low-molecular-weight heparin (LMWH) efficacy in murine sepsis.

Sepsis is a life-threatening condition caused by a dysregulated host response to infection. Beyond anticoagulation, heparin has anti-inflammatory, anti-chemotactic, and cytoprotective properties which may provide therapeutic benefits in sepsis. In COVID-19-associated sepsis, therapeutic-dose heparin improved outcomes in moderately but not critically ill patients, highlighting the importance of treatment timing. The study objectives are: (1) To identify the therapeutic window for initiating therapeutic-dose low molecular weight heparin (LMWH) in a murine model of sepsis. (2) To determine whethe

Low molecular weight heparins in the treatment of unstable angina.

Unstable angina is in most cases caused by partial or complete coronary artery occlusion due to the disruption of an atherosclerotic plaque and resultant thrombus formation. Platelet aggregation and thrombin formation are key events in the development of acute coronary syndromes. An immediate antithrombotic approach is essential to prevent fatal and non-fatal myocardial infarction, and the combination of aspirin and unfractionated heparin has been the treatment of choice in the past years. Low molecular weight heparins have improved pharmacokinetic and pharmacodynamic properties over unfractio

The enhancement of PCR amplification by low molecular weight amides.

Amplification of a DNA target by the polymerase chain reaction (PCR) often requires laborious optimization efforts. In this regard, the use of certain organic chemicals such as dimethyl sulfoxide, polyethylene glycol, betaine and formamide as cosolvents has been found to be very helpful. Unfortunately, very little is known about the precise structural features that make these additives effective and, accordingly, the number of such chemicals currently known to enhance PCR is limited. In order to address these issues, we decided to focus on formamide and undertook an extensive study of low mole

Low-molecular-weight heparins in the management of unstable angina.

Low-molecular-weight heparins (LMWHs) are derivatives of unfractionated heparin (UFH) and have a more favourable pharmacokinetic profile and a more predictable anticoagulant effect than UFH. These features allow the subcutaneous injection of weight-adjusted doses of LMWHs without monitoring of blood coagulation, and this results in cost savings. Clinical trials have shown that combined therapy with aspirin and LMWHs is significantly more effective than aspirin alone in reducing the risk of cardiac events in the acute treatment of patients affected by unstable angina. LMWHs have also been shown

Influence of intravascular molecular weight of hydroxyethyl starch on platelets.

Complications concerning the blood coagulation have been observed repeatedly after administration of highly substituted, high molecular weight hydroxyethyl starch (HES), but it has not been examined as to how intravascular molecular weight and degree of substitution of HES influence platelet number and volume after repeated administration. Thirty patients with cerebrovascular diseases were treated for 10 days with hemodilution. 500 to 1500 ml of HES 200/0.62(n=10), HES 200/0.5(n=10) or HES 40/0.5(n=10) were infused daily. During the first days, the number of platelets was not lowered beyond th
