The Fermentable Oligosaccharides, Disaccharides, Monosaccharides, and Polyols Diet Is Effective in Irritable Bowel Syndrome With Constipation: A Single Center Tertiary Care Clinical Practice Evaluation.

BACKGROUND/AIMS: : The low fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAP) diet is a well-researched treatment for diarrhea-predominant irritable bowel syndrome (IBS-D), but its value for constipation-predominant IBS (IBS-C) is uncertain. Clinicians may hesitate to recommend it for IBS-C, fearing it could worsen constipation or offer little relief. This study aims to assess the diet's effectiveness across all IBS subtypes, specifically investigating its impact on stool patterns in IBS-C and the correlation with symptom relief. METHODS: : This prospective ser

A diet low in fermentable oligo-, di-, monosaccharides and polyols improves abdominal and overall symptoms in persons with all subtypes of irritable bowel syndrome.

BACKGROUND: A diet low in fermentable oligo-, di-, monosaccharides and polyols (LFD) improves symptoms in patients with irritable bowel syndrome (IBS). Previous studies have focused on patients with IBS and diarrhea (IBS-D). It is unclear whether LFD is effective for IBS with constipation (IBS-C) or IBS with mixed bowel habits (IBS-M). This open-label, real-world study evaluates the relative effectiveness of the LFD among IBS subtypes. METHODS: This study analyzes data from a service that provides low-FODMAP meals to individuals with IBS. Participants met with a registered dietitian and compl

Multifaceted approach to the impact of drying techniques on Lycium barbarum polysaccharide yield, quality, and bioactive retention: insights from HPLC monosaccharide quantification.

Lycium barbarum (goji berry, LB) is a nutrient-dense fruit that requires drying for preservation, but this process can degrade its bioactive compounds. This study compares the effects of hot-air drying (HAD), infrared drying (IRD), and pulsed vacuum drying (PVD) on drying kinetics, microstructure, and polysaccharide yield, with HPLC used for monosaccharide quantification and FT-IR for structural analysis. PVD significantly reduced drying time to 11 h, representing a 50.0% reduction compared to HAD (22 h), a 47.6% reduction compared to IRD (21 h), and a 42.1% reduction compared to HAD+ (19 h),

Metabolic Abnormalities in Normal Weight Children Are Associated with Increased Visceral Fat Accumulation, Elevated Plasma Endotoxin Levels and a Higher Monosaccharide Intake.

Being overweight has been identified as the main risk factor for the development of metabolic disorders in adults and children. However, recent studies suggest that normal weight individuals are also frequently affected by metabolic abnormalities with underlying mechanisms not yet fully understood. The aim of the present study was to determine if dietary pattern and markers of intestinal permeability, as well as inflammation, differ between normal weight healthy children and normal weight children suffering from metabolic abnormalities. In total, 45 normal weight children aged 5⁻9 years were i

N-palmitoyl-D-glucosamine, a Natural Monosaccharide-Based Glycolipid, Inhibits TLR4 and Prevents LPS-Induced Inflammation and Neuropathic Pain in Mice.

Toll-like receptors (TLRs) are key receptors through which infectious and non-infectious challenges act with consequent activation of the inflammatory cascade that plays a critical function in various acute and chronic diseases, behaving as amplification and chronicization factors of the inflammatory response. Previous studies have shown that synthetic analogues of lipid A based on glucosamine with few chains of unsaturated and saturated fatty acids, bind MD-2 and inhibit TLR4 receptors. These synthetic compounds showed antagonistic activity against TLR4 activation in vitro by LPS, but little

Low Fermentable Oligosaccharides, Disaccharides, Monosaccharides, and Polyols Diet Improves Colonic Barrier Function and Mast Cell Activation in Patients With Diarrhea-Predominant Irritable Bowel Syndrome: A Mechanistic Trial.

BACKGROUND & AIMS: A diet low in fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (LFD) is the most efficacious dietary therapy for irritable bowel syndrome (IBS). However, the mechanisms by which fermentable oligosaccharides, disaccharides, monosaccharides, and polyols drive IBS pathophysiology are unclear. METHODS: Patients with Rome IV diarrhea-predominant IBS (IBS-D) underwent barrier function evaluation pre- and post-LFD along with assessment of mast cell number and activation profile. Finally, fecal supernatants (FS) were administered intracolonically to wild-ty

An innovative method for simultaneous separation and determination of monosaccharide and sugar alcohol isomers associated with glycometabolism in beagle plasma using gas chromatography-mass spectrometry.

BACKGROUND: Tracking changes in plasma concentrations of monosaccharides and sugar alcohols related to glycometabolism, which plays a role as the main source of energy, is of great significance for the treatment of metabolic diseases. However, analysis of monosaccharides and sugar alcohols has always been a difficult task due to the high polarity of the polyhydroxy structure and the presence of a large number of isomers. So far, no suitable method has been developed for simultaneous determination of monosaccharides and sugar alcohols in beagle plasma. RESULTS: This work performed an innovativ

The Role of Intestinal Barrier Function in Overweight Patients with IBS with Diarrhea Undergoing a Long-Term Low Fermentable Oligo-, Di-, and Monosaccharide and Polyol Diet.

Overweight and obesity have been suggested as significant factors in irritable bowel syndrome (IBS) development. However, the relationship between overweight/obesity and IBS is unclear. It is known that a modified intestinal barrier, especially the permeability of the small intestine (s-IP), can play a significant role in the pathogenesis of both obesity and IBS. Moreover, dietary interventions are essential for treating both pathologies. We evaluated the gastrointestinal (GI) symptoms and the urinary and circulating markers of GI barrier function and integrity, the markers of intestinal dysbi

Chiral resolution of monosaccharides as 1-phenyl-3-methyl-5-pyrazolone derivatives by ligand-exchange CE using borate anion as a central ion of the chiral selector.

Six reducing monosaccharides (mannose, galactose, fucose, glucose, xylose, and arabinose) were derivatized with 1-phenyl-3-methyl-5-pyrazolone (PMP) and chiral resolution of these racemic PMP-monosaccharides was studied by ligand-exchange CE using borate anion as a central ion of the chiral selector and (S)-3-amino-1,2-propanediol (SAP) as a chiral selector ligand. PMP-mannose, PMP-galactose and PMP-fucose were successfully enantioseparated. Lowering the capillary temperature increased the resolution of PMP-mannose system, but decreased that of PMP-galactose and PMP-fucose systems. Whereas the

The diagnosis of the Sanfilippo C syndrome, using monosaccharide and oligosaccharide substrates to assay acetyl-CoA: 2-amino-2-deoxy-alpha-glucoside N-acetyltransferase activity.

Glucosamine, galactosamine, mannosamine, several disaccharides and a tetrasaccharide were evaluated as substrates for the N-acetyltransferase involved in the pathogenesis of the Sanfilippo C syndrome. Glucosamine and alpha-D-glucosaminide disaccharides and a tetrasaccharide derived from heparin were exo-N-acetylated by homogenates of cultured skin fibroblast from normal individuals at pH 6.0 in the presence of acetyl-CoA, whereas fibroblast homogenates prepared from a Sanfilippo C patient failed to catalyse the N-acetyltransferase from acetyl-CoA to these substrates. Purification and characterization of a potentially novel polysaccharide FMP-2 from Ficus microcarpa fruit: comparative extraction, LC-QTOF-MS monosaccharide profiling, and antioxidant and anti-inflammatory bioactivities.

Ficus fruit polysaccharides were extracted from four species (F. racemosa, F. microcarpa, F. amplissima, and F. auriculata) using hot-water, enzymatic, ultrasonication, and ultrasonic-enzyme-assisted methods, representing the first comparative extraction-purification framework applied to this nutritionally neglected indigenous resource. The maximum polysaccharide content was obtained from F. auriculata (3.55%) and F. microcarpa (3.45%), with hot-water extraction proving to be the most effective. F. microcarpa showed outstanding carbohydrate content (835.46 mg GE/g extract), low secondary metab
