Intra-and intercellular calcium waves in cultured human myometrium.

Intracellular calcium waves were observed in cultured human myometrium using confocal laser scanning microscopy in the epifluorescence mode. Intracellular calcium waves were observed following stimulation of the cells with 10 mU ml-1 oxytocin in physiological bathing solution, or following substitution of tetraethylammonium chloride for NaCl. Intracellular wave speeds were 18.9 +/- 8.2 microns s-1 in physiological bathing solution. Intracellular calcium waves were also observed in cells bathed in calcium-free solutions, with no significant differences in wave speeds. Calcium waves that crossed

Melatonin together with noradrenaline augments contractions of human myometrium.

The hormone melatonin is known to influence the circadian rhythm, and it probably also mediates some of the physiological changes that occur in the body at night. Inasmuch as uterine activity is greater during darkness, we investigated whether melatonin could modulate uterine contractility. Biopsies were performed during caesarean sections to obtain uterine tissue from women who had reached full term. The obtained samples were mounted in organ baths, and spontaneous contractions were recorded. Melatonin alone did not change myometrial contractility, whereas melatonin in combination with noradr

Title: β3 Adrenergic Receptor Signaling in the Human Myometrium.

Preterm labor leading to preterm birth is the leading cause of infant morbidity and mortality. Although β2 adrenergic agonists fail to provide adequate tocolysis, the expression of the β3 adrenergic receptor in myometrium and its unique signaling suggest a role for β3 agonist in the management of preterm labor. Western blot analysis showed that the β3 adrenergic receptor expression increased in human pregnancy myometrium compared to nonpregnant tissues (p < 0.0001). There was no difference in β3 adrenergic receptor expression throughout pregnancy (p > 0.05). The addition of the β3 agonist mira

Molecular evidence of functional progesterone withdrawal in human myometrium.

Progesterone suppresses uterine contractility acting through its receptors (PRA/B). The mechanism by which human labour is initiated in the presence of elevated circulating progesterone has remained an enigma since Csapo first theorized of a functional withdrawal of progesterone in 1965. Here we report that in vitro progesterone-liganded nuclear PRB forms a complex including JUN/JUN homodimers and P54(nrb)/Sin3A/HDAC to repress transcription of the key labour gene, Cx43. In contrast, unliganded PRA paradoxically activates Cx43 transcription by interacting with FRA2/JUND heterodimers. Furthermo

Rheological properties of myometrium: experimental quantification and mathematical modeling.

This work answers some questions related to detection of rheological properties of soft tissues exemplified in myometrium, stressed by external tensile force. In the first stage of the experiment the tissue samples were ciclically stressed and response loops were recorded. This test proved severe plastical deformation of samples, which is not usually being stated for living tissues. In addition to course, growth and stabilizing this deformation also energetical losses of individual hysteresis loops of the response were evaluated. In the second stage of the experiment the tissue samples were ex

Tissue prothrombinase activity in myometrium from timed-pregnant rats.

OBJECTIVE: Fgl2 and thrombin potentially play roles during inflammation-induced preterm delivery. These studies sought to demonstrate functional prothrombinase enzyme activity in rat uterus, consistent with the expression of Fgl2 in this tissue. METHODS: Myometrial and other tissue obtained from non-pregnant and timed-pregnant rats was homogenized in Tris-buffered saline. Prothrombinase activity was determined based on the kinetic metabolism of a chromogenic thrombin substrate. Homogenates were incubated with prothrombin followed by the addition of the thrombin substrate. Thrombin activity wa

Expression of stretch-activated potassium channels in human myometrium.

Using RT-PCR and a semiquantitative method to detect relative differences in mRNA expression, we examined uterine smooth muscle from both pregnant and non-pregnant women for the expression of members of weak inwardly rectifying 2-pore potassium channel family (TWIK), TREK and TRAAK. We show for the first time that TRAAK transcript is present in uterine smooth muscle, albeit at relatively low levels. The expression of TREK is far more robust. Trek-1 but not TREK-2 was expressed in both pregnant and non-pregnant myometrium. Comparison of TREK-1 expression in several hysterectomy samples relative

The cyclic-ADP-ribose signaling pathway in human myometrium.

Human myometrial contraction plays a fundamental role in labor. Dysfunction of uterine contraction is an important cause of failure in progression of labor. The mechanisms of control of uterine contractions are not completely understood. It appears that intracellular Ca(2+) mobilization may play an important role during uterine contraction. Several mechanisms of intracellular Ca(2+) mobilization have been described. However, in human uterus only the inositol 1,4,5-trisphosphate-induced Ca(2+) release has been extensively studied to date. In view of the identification of the presence of functio

Calcium homeostatic pathways change with gestation in human myometrium.

A rise in intracellular calcium is the primary trigger for contractile activity in pregnant human myometrium. It is hypothesized that key proteins involved in myometrial calcium homeostasis are gestationally regulated and play an important role in the preparation for labor. The aims of the study were to investigate the role of sarcoplasmic reticulum Ca ATPases (SERCAs) in regulating spontaneous contractile activity in myometrium, and to determine the expression of SERCA isoforms 2a and 2b, and the plasma membrane Ca ATPase (PMCA), at term and during labor. Western blot analysis demonstrated th

Cocaine and beta-adrenergic receptor function in pregnant myometrium.

OBJECTIVE: Our purpose was to evaluate the in vitro effect of cocaine on beta-adrenergic receptor function in pregnant human myometrium. STUDY DESIGN: Myometrium was obtained from 25 pregnant women at term who were undergoing cesarean section but who were not in labor. Membrane preparations were assayed for cyclic adenosine monophosphate production after exposure to isoproterenol, sodium fluoride, or forskolin in the presence and absence of either cocaine or benzoylecgonine, a major metabolite of cocaine. RESULTS: Cocaine inhibited isoproterenol-induced cyclic adenosine monophosphate product

Proposed signaling role of arachidonic acid in human myometrium.

The objective of this study was to test the hypothesis that, in human myometrial cells (HMC), PGF2 alpha and oxytocin promote the release of arachidonic acid (AA) which, in turn, acts to mobilize intracellular Ca2+. Primary monolayer cultures of HMC were labeled with [3H]arachidonic acid ([3H]AA) to isotopic equilibrium before exposure to PGF2 alpha or oxytocin. Radiolabeled phospholipids were separated on thin layer chromatography and quantitated by scintillation counting. Prostanoids were analyzed by high performance liquid chromatography. Calcium release was quantitated in digitonin-permeab

Endothelin receptors: binding and phosphoinositide breakdown in human myometrium.

Endothelin (ET) binding sites and phosphoinositide hydrolysis induced by ET peptides were studied in the nonpregnant human myometrium. Saturation binding experiments revealed that the proportion of [125I]ET-1 binding sites was 4-fold higher than that of [125I]ET-3 binding sites, whereas Kd values were not significantly different. In competition binding studies, unlabeled peptides displaced [125I]ET-1 binding with the following order of affinity ET-1 > sarafotoxin 6b > ET-3 >> sarafotoxin 6c, whereas very similar Ki values were obtained with the four peptides for the displacement of [125I]ET-3

Topographical distribution of prostaglandin E receptors in human myometrium.

The binding of radiolabelled prostaglandin (PG) F2 alpha and PGE2 by human myometrium was measured in vitro and the distribution and characteristics of the binding sites in non-pregnant and pregnant uteri were studied. PGF2 alpha binding sites were of low affinity (Kd 30 nM) and could be occupied by PG of the E series with higher affinity than PGF2 alpha itself. PGE binding sites were of high affinity (Kd 1.5 nM) and highly specific for PG of the E series, suggesting that they represent true PGE receptors. The concentration of PGE receptors was higher in non-pregnant than in pregnant uteri at

Cellulose Plant-Derived Scaffolds as a Tool for Myometrium Modeling.

The myometrium is the smooth muscle layer of the uterus, whose dysfunctions are involved in various pathologies leading to infertility, such as adenomyosis and uterine fibroids. Developing relevant in vitro models of the myometrium is crucial for investigating the pathogenesis of these diseases. In this study, we propose a novel approach for cultivating mouse myometrial smooth muscle cells (SMCs) using plant-derived cellulose scaffolds. The scaffolds were obtained through the decellularization of green onion leaf, celery stalk, or bluegrass leaf, subsequently coated with collagen type I. We fo

Collagen and elastic fiber remodeling in the pregnant mouse myometrium†.

The myometrium undergoes progressive tissue remodeling from early to late pregnancy to support fetal growth and transitions to the contractile phase to deliver a baby at term. Much of our effort has been focused on understanding the functional role of myometrial smooth muscle cells, but the role of extracellular matrix is not clear. This study was aimed to demonstrate the expression profile of sub-sets of genes involved in the synthesis, processing, and assembly of collagen and elastic fibers, their structural remodeling during pregnancy, and hormonal regulation. Myometrial tissues were isolat

Biochemical properties of H+-Ca2+-exchanger in the myometrium mitochondria.

Some biochemical properties of the H+-Ca2+-exchanger in uterine smooth muscle mitochondria have been described. The experiments were performed on a suspension of isolated mitochondria from the myometrium of rats. Methods of confocal microscopy, spectrofluorimetry and photon correlation spectroscopy were used. Fluo-4 probe was used to record changes in ionized Ca2+ in the matrix and cytosol; pH changes in the matrix were evaluated with BCECF. It was experimentally proved that in the myometrium instead of Na+-Ca2+-exchanger the H+-Ca2+-exchanger functions. It was activated at a physiological pH

Conflicting Nongenomic Effects of Progesterone in the Myometrium of Pregnant Rats.

Recently, it has been suggested that progesterone affects the contractile activity of pregnant myometrium via nongenomic pathways; therefore, we aimed to clarify whether progesterone causes and/or inhibits pregnant myometrial contractions via nongenomic pathways. Our in vitro experiments using myometrial strips obtained from rats at 20 days of gestation revealed that progesterone caused myometrial contractions in a concentration- and time-dependent manner at concentrations up to 5 × 10-7 M; however, this effect decreased at concentrations higher than 5 × 10-5 M. Similarly, progesterone enhance

Mass lesions of the myometrium: interpretation and management of unexpected pathology.

PURPOSE OF REVIEW: Uterine leiomyomas are the most common benign uterine smooth muscle tumors. On the basis of imaging, these masses are often presumed to be benign conventional leiomyomas and surgical excision is a common treatment choice. After myomectomy or hysterectomy for presumed leiomyomas, the surgical pathology report may reveal an unexpected diagnosis of another type of mesenchymal tumor. These can range from a variant of benign smooth muscle tumors to smooth muscle tumors of uncertain malignant potential to malignant sarcomas. This review describes these variant pathologies and revi

Anoctamin Channels in Human Myometrium: A Novel Target for Tocolysis.

BACKGROUND: Spontaneous preterm labor leading to preterm birth is a significant obstetric problem leading to neonatal morbidity and mortality. Current tocolytics are not completely effective and novel targets may afford a therapeutic benefit. OBJECTIVE: To determine whether the anoctamin (ANO) family, including the calcium-activated chloride channel ANO1, is present in pregnant human uterine smooth muscle (USM) and whether pharmacological and genetic modulation of ANO1 modulates USM contraction. METHODS: Reverse transcription-polymerase chain reaction (RT-PCR), quantitative RT-PCR, and immun
