A novel naphthoquinone compound triggers DNA damage-induced apoptosis on cholangiocarcinoma through upregulation of BAX.

Cholangiocarcinoma (CCA), a devastating malignancy originating from the bile ducts, is of significant clinical importance due to its rising incidence and poor prognosis. Quinones as being naturally occurring compounds and their frequent utility in anticancer drug development studies seem to be potential sources for the discovery of new chemotherapeutics. In this study, a synthetic naphthoquinone derivative newly synthesized and previously published by our group, named as MK13, has been tested against intrahepatic-CCA (iCCA) cell lines (CCLP1 and HUCCT1). Cell viability was measured with the MT

Scaffold-leaping optimization of naphthoquinone derivatives as ferroptosis inducer against non-small lung cancer.

Ferroptosis, an iron-dependent programmed cell death pathway, has emerged as a promising therapeutic target for cancer. Herein, a series of naphthoquinone derivatives were designed via scaffold-leaping optimization from lead compound QD394, synthesized and assessed for their anti-proliferation activity against five cancer cell lines, and the structure-activity relationship (SAR) were described. Of these compounds, I-21 was identified as most active, demonstrating significant anticancer efficacy in vitro (IC50 = 0.76 μM against A549 cell lines). This cytotoxic effect can be counteracted by ferr

Combinatorial screening for therapeutics in ATTRv amyloidosis identifies naphthoquinone analogues as TTR-selective amyloid disruptors.

Hereditary ATTR amyloidosis is caused by the point mutation in serum protein transthyretin (TTR) that destabilizes its tetrameric structure to dissociate into monomer. The monomers form amyloid fibrils, which are deposited in peripheral nerves and organs, resulting in dysfunction. Therefore, a drug that dissolves amyloid after it has formed, termed amyloid disruptor, is needed as a new therapeutic drug. Here, we first established a high throughput screening system to find TTR interactors from the LOPAC1280 compound library. Among the hit compounds, thioflavin T-based post-treatment assay deter

One-pot synthesis of pyrimidine linked naphthoquinone-fused pyrroles by iodine-mediated multicomponent reactions.

Herein we report iodine-mediated multicomponent reactions for the synthesis of naphthoquinone-fused pyrroles tethered with a pyrimidine moiety. The reaction of aryl methyl ketones (3) or terminal aryl alkynes (5) in the presence of molecular iodine in DMSO medium followed by sequential addition of barbituric acids (2) and 2-amino-1,4-naphthoquinone (1) provides the corresponding three-component hybrid molecules 4 having naphthoquinone-fused pyrroles tethered with a barbituric acid moiety. This three-component reaction proceeds via metal-free C-H oxidation followed by multi-component cyclizatio

Effects of 1,4-naphthoquinone aged carbon black particles on the cell membrane of human bronchial epithelium.

Black carbon (BC) and polycyclic aromatic hydrocarbons (PAHs), which are major atmospheric pollutants in China, are hazardous to humans following inhalation. BC can be oxidized by PAHs forming secondary particles of which the health effects are unknown. In this study, carbon black (CB) was used to simulate BC to reveal the adverse effects of 1,4-naphthoquinone aged carbon black (CB/1,4-NQ) particles on the membrane of human bronchial epithelial (16HBE) cells. It was showed that, the cell viability, cell membrane fluidity, membrane potential and mitochondrial membrane potential were significant

Novel hybrid 1,4-naphthoquinone-endoperoxide antimalarial hits: Synthesis, stereoisomeric separation, and biological evaluation.

Hybrid molecules, chemical entities with two or more pharmacophore domains, are widely investigated for their antiparasitic potential. First-in-class antimalarial hybrids were designed and tested against Plasmodium falciparum. Our multistep synthesis, initiated by a Mn(III)-mediated radical reaction, led to new hybrids containing both the pharmacophores of atovaquone (naphthoquinone) and artemisinin (endoperoxide) separated by a piperazine linker. With 2 or 3 asymmetric carbons, 4 to 8 theoretical configurational isomers were expected. For two of our 5 final products, chiral chromatography sep

Naphthoquinone derivatives exhibit apoptosis-like effect and anti-trypanosomal activity against Trypanosoma evansi.

Trypanosoma evansi is an extracellular flagellate blood protozoan parasite and an etiological agent of animal trypanosomosis. Presently, only a few drugs are registered and have been used for the treatment of animal trypanosomosis, but they show severe toxic effects and also face the problem of drug resistance. Naphthoquinones (NTQ) are considered as fortunate structures in the field of medicinal chemistry as they have been reported for their antitrypanosomal potential against other trypanosomes-T. brucei and T. cruzi. In the present study, six naphthoquinones (NTQ1-NTQ6) derivatives were eval

Computer-aided design of 1,4-naphthoquinone-based inhibitors targeting cruzain and rhodesain cysteine proteases.

Chagas disease and Human African Trypanosomiasis (HAT) are caused by Trypanosoma cruzi and T. brucei parasites, respectively. Cruzain (CRZ) and Rhodesain (RhD) are cysteine proteases that share 70% of identity and play vital functions in these parasites. These macromolecules represent promising targets for designing new inhibitors. In this context, 26 CRZ and 5 RhD 3D-structures were evaluated by molecular redocking to identify the most accurate one to be utilized as a target. Posteriorly, a virtual screening of a library containing 120 small natural and nature-based compounds was performed on

Ytterbium-decreasing Streptomyces sp. and its naphthoquinone-pigment production in the presence of rare-earth elements.

We screened for oligotrophic microorganisms capable of decreasing the concentration of ytterbium (Yb), a representative of the heavy rare-earth elements, in a culture medium. From 476 strains of oligotrophic microorganisms (grown on 1/100 diluted nutrient agar) isolated from soil and river water samples, 5 strains capable of reducing the concentration of Yb in diluted nutrient broth containing 5 ppm Yb were selected. Among them, a strain capable of reducing the concentration of all rare-earth elements to a great extent was identified as Streptomyces sp. (strain YB-1). This strain produced redi

Burkholderia pseudomallei produces 2-alkylquinolone derivatives important for host virulence and competition with bacteria that employ naphthoquinones for aerobic respiration.

Melioidosis is caused by Burkholderia pseudomallei, an opportunistic Gram-negative pathogen that inhabits soil and water in tropical and subtropical regions. B. pseudomallei infections often occur following contact with contaminated water or soil or by inhalation of contaminated dust and water droplets. There is limited knowledge about how B. pseudomallei is able to survive in harsh environmental conditions and compete with the microbes that inhabit these niches. Previous research demonstrated that 3-methyl-2-alkylquinolones (MAQs), and their corresponding N-oxides (MAQNOs), are produced by B.

Comparison of the effects on mitochondrial function of a series of 2-methyl substituted 1,4-naphthoquinones to their 6-methyl counterparts.

Seven derivatives of 2-methyl-1,4-naphthoquinone were compared to an equivalent series of 6-methyl-1,4-naphthoquinone derivatives for their abilities to interact with mitochondrial membranes and enzyme systems. The substituents on the methyl groups included various electron-withdrawing and -donating groups. The 6-methyl quinones were, in general, more potent inhibitors of NADH-oxidase and succinoxidase, depleted sulfhydryl groups more completely, produced greater rates of cyanide (CN-)-insensitive respiration (respiratory bursts), and induced more severe large-amplitude mitochondrial swelling

Synthesis, Biological Evaluation and Molecular Modeling Studies of Naphthoquinone Sulfonamides and Sulfonate Ester Derivatives as P2X7 Inhibitors.

ATP acts in the extracellular environment as an important signal, activating a family of receptors called purinergic receptors. In recent years, interest in the potential therapeutics of purinergic components, including agonists and antagonists of receptors, has increased. Currently, many observations have indicated that ATP acts as an important mediator of inflammatory responses and, when found in high concentrations in the extracellular space, is related to the activation of the P2X7 purinergic receptor. In this sense, the search for new inhibitors for this receptor has attracted a great dea

Application of "macromolecular crowding" in vitro to investigate the naphthoquinones shikonin, naphthazarin and related analogues for the treatment of dermal scars.

Pathological scarring is an intractable problem for both patients and clinicians. A major obstacle for the development of scar remediation therapies is the paucity of suitable in vivo and in vitro models. The "Scar-in-a-jar" model was previously established by our colleagues based on the principle of "Macromolecular crowding". This has been demonstrated to be an extracellular matrix-rich in vitro model offering a novel tool for studies related to the extracellular matrix. In the study reported herein, we have optimised this approach to model human dermal fibroblasts derived from hypertrophic t
