Morphostatic Speciation within the Dagger Nematode Xiphinema hispanum-Complex Species (Nematoda: Longidoridae).

Dagger nematodes of the genus Xiphinema include a remarkable group of invertebrates of the phylum Nematoda comprising ectoparasitic animals of many wild and cultivated plants. Damage is caused by direct feeding on root cells and by vectoring nepoviruses that cause diseases on several crops. Precise identification of Xiphinema species is critical for launching appropriate control measures. We deciphered the cryptic diversity of the Xiphinema hispanum-species complex applying integrative taxonomical approaches that allowed us to verify a paradigmatic example of the morphostatic speciation and th

Steinernema pwaniensis n. sp., a new entomopathogenic nematode (Nematoda: Steinernematidae) from Tanzania.

A new species of entomopathogenic nematode, Steinernema pwaniensis n. sp., belonging to the glaseri group, is described from Tanzania. The infective juveniles of S. pwaniensis n. sp. are characterized by a body length of 978 μm (808-1131), distance from anterior end to excretory pore of 86 μm (80-95) and a tail length of 87 μm (75-95). The ratios a, c and E% of S. pwaniensis n. sp. are substantially lower than those of all other African 'glaseri' group members. The first-generation males of S. pwaniensis n. sp. can be distinguished by a large spicule length of 92 μm (80-97) and by the absence

Molecular Characterization of Nippostrongylus brasiliensis (Nematoda: Heligmosomatidae) from Mus musculus in India.

Mus musculus (Rodentia: Muridae) has generally been infected with a rodent hookworm Nippostrongylus brasiliensis. In this report, we present morphological and molecular identification of N. brasiliensis by light and scanning electron microscopy and PCR amplification of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene and the protein sequences encoded by cox1 gene, respectively. Despite the use of N. brasiliensis in many biochemistry studies from India, their taxonomic identification was not fully understood, especially at the species level, and no molecular data is available in GenBank

New zoonotic cases of Onchocerca dewittei japonica (Nematoda: Onchocercidae) in Honshu, Japan.

BACKGROUND: Zoonotic infections with Onchocerca species are uncommon, and to date only 25 clinical cases have been reported worldwide. In Japan, five previous zoonotic infections were concentrated in Oita, Kyushu (the southern island), with one previous case in Hiroshima in the western part of Honshu (the main island). The causative agent in Japan was identified as Onchocerca dewittei japonica Uni, Bain & Takaoka, 2001 from Japanese wild boars (Sus scrofa leucomystax Temminck, 1842). Here we report two infections caused by a female and male O. dewittei japonica, respectively, among residents o

Interaction of microbial populations in Steinernema (Steinernematidae, Nematoda) infected Galleria mellonella larvae.

Infection of Galleria mellonella larvae with the entomopathogenic nematodes Steinernema feltiae (A21 and R strains) and Steinernema glaseri (Dongrae) resulted in several species of bacteria, including the respective bacterial symbiont, Xenorhabdus spp., growing in the infected insect cadavers. These other bacteria were Enterococcus in all three nematode infections studied and Acinetobacter in the S. feltiae infections. The respective populations of these bacteria changed with time. Following infection of G. mellonella larvae with any one of the Steinernema sp., only Enterococcus bacteria were

Identification of a potential protective microfilarial antigen from Litomosoides carinii (Nematoda, Filarioidea).

The proteins of the filarial parasite Litomosoides carinii that are presented to the host in the course of a natural infection were assessed by surface and intrinsic labelling, microfilarial agglutination assays and ELISA. Surface labelling revealed several proteins present on all parasite stages which were recognised by infected animals. Only one major surface protein of 55 KD was found on microfilariae, while in all other stages (adult males, adult females and infective larvae isolated from pool animals) a range of proteins could be labelled. Immunisation with live microfilariae obtained fro

Multi-Marker Approach for the Identification of Different Heterodera Species (Nematoda: Heteroderidae).

Cyst nematodes of the genus Heterodera are important plant-parasitic nematodes that cause significant crop losses worldwide but are often overlooked due to their non-specific symptoms and complex biology. This study assessed Heterodera diversity in Portugal using an integrative molecular approach based on four genetic markers (mtCOI, 18S rDNA, ITS, and 28S rDNA). Five valid species were identified: Heterodera cruciferae, H. mani, H. schachtii, H. trifolii, and H. zeae, with H. mani reported for the first time in the country. A distinct taxon from Coimbra (central Portugal) may represent a new

Functional Ultrastructure of the Excretory Gland Cell in Zoonotic Anisakids (Anisakidae, Nematoda).

Excretory and secretory products are crucial for parasite infectivity and host immunomodulation, but the functioning and ultrastructure of the excretory gland cell (EC) that produces these products are still scarcely understood and described. In light of growing reports on anisakiasis cases in Europe, we aimed to characterise the EC of larval Anisakispegreffii and adult Pseudoterranovaazarasi. In the latter, EC starts 0.85 mm from the head tip, measuring 1.936 × 0.564 mm. Larval EC shows a long nucleus with thorn-like extravaginations toward the cytoplasm, numerous electron-dense and -lucent s

Temporal and Spatial Distribution of Enterobius vermicularis (Nematoda: Oxyuridae) in the Prehistoric Americas.

Investigations of Enterobius sp. infection in prehistory have produced a body of data that can be used to evaluate the geographic distribution of infection through time in the Americas. Regional variations in prevalence are evident. In North America, 119 pinworm positive samples were found in 1,112 samples from 28 sites with a prevalence of 10.7%. Almost all of the positive samples came from agricultural sites. From Brazil, 0 pinworm positive samples were found in 325 samples from 7 sites. For the Andes region, 22 pinworm positive samples were found in 411 samples from 26 sites for a prevalenc

A new species of Pharyngodonidae (Nematoda) of Tropidurus torquatus (Squamata:Tropiduridae) from Brazil.

Parapharyngodon bainae n. sp. (Nematoda:Pharyngodonidae) collected from the small intestine of Tropidurus torquatus (Squamata:Tropiduridae) is described. The new species is the 47 th species in the genus and the 14 th in the Neotropical region. The morphology of male posterior end, ovary position in females, and egg size can be used to differentiate P. bainae from the other congeneric species. Parapharyngodon rotundatus , Parapharyngodon sanisfaciecaudus , and Parapharyngodon riojensis are most closely related to P. bainae. Cytochemical characterization of the third-stage larva of Wuchereria bancrofti (Nematoda: Filarioidea).

In the present paper, we report the results we obtained using several cytochemical techniques to analyze the infective larva of Wuchereria bancrofti. An imidazole osmium tetroxide solution was used to visualize unsaturated fatty acids. A highly contrasted material forming a continuous structure was observed on the larval surface and over the epicuticle. A strong reaction was observed on the esophagus and also on the inner secreted material. Carbohydrates containing vic-glycol groups were not observed on the cuticle of the third-stage larva of W. bancrofti submitted to the Thiéry technique. Usi

A new species of Trichuris (Nematoda: Trichuridae) from Phyllotini rodents in Argentina.

A new species of Trichuris is described. Trichuris pardinasi n. sp. was recovered from Phyllotis xanthopygus Waterhouse (Rodentia: Muridae: Phyllotini) in Sierra de la Ventana, Buenos Aires Province, and Pampa de Achala, Córdoba Province (Argentina). This is the first record of Trichuris parasitizing Phyllotini rodents. The new species can be differentiated from the other 10 species parasitizing rodents from South America by the absence of the spicular tube, spicular sheath with spines uniformly distributed, the length of spicule, the J-shaped proximal cloacal tube, and the nonprotrusive vulva

Preliminary data on the genetic differentiation of Onchocerca volvulus in Africa (Nematoda: Filarioidea).

Data are reported on the genetic structure of three Onchocerca volvulus populations, respectively from Mali (savanna), Ivory Coast (forest), and Zaire (forest gallery in savanna). Electrophoretic analysis, carried out on 25 gene-enzyme systems, has shown a remarkable genetic heterogeneity existing within O.volvulus. Zaire and West Africa populations appear chiefly differentiated at Mdh-1 and 6Pgdh loci, their average Nei's genetic distance being 0.11. In West Africa Nei's D found between the savanna and forest samples is 0.04. The savanna population from Zaire is more similar to the savanna on

Immunization with infective larvae of Strongyloides ratti (Nematoda) exposed to microwave radiation.

Microwaves have not been tested previously for possible application in producing immunogenic preparations of parasites. This study examines the immunizing capacity of microwave-irradiated, infective larvae of Strongyloides ratti in rats. Rats were inoculated subcutaneously with untreated, microwaved, or microwaved and homogenized larvae, or distilled water, and challenged with untreated larvae. Data were collected on egg production and worm number/rat during primary infections and on egg production, worm number/rat, worm size, and eggs in utero/worm following challenge. Our results demonstrate

Vitamin A deficiency and the development of Litomosoides carinii (Nematoda, Filarioidea) in cotton rats.

This paper demonstrates that vitamin A deficient (A-) cotton rats are more susceptible to infection with Litomosoides carinii than normal (A+) cotton rats. However, embryogenesis is frequently retarded in female worms from A- hosts. Groups of young cotton rats were depleted of their liver stores of vitamin A by feeding them a vitamin A free diet from weaning. From 6 weeks of age this diet was supplemented with either 2, 5, 10, 25 or 100 IU vitamin A per 100 g body weight per day and half the animals in each group were infected with L. carinii. At autopsy, 51 days post-infection, serum vitamin
