The Neuroscientist : a review journal bringing neurobiology, neurology and psychiatry2003eng
The persistence of neurogenesis in the adult mammalian forebrain suggests that endogenous precursors may be a potential source for neuronal replacement after injury or neurodegeneration. Limited knowledge exists, however, regarding the normal function of neurogenesis in the adult and its alteration by brain injury. Neural precursors generate neurons throughout life in the mammalian forebrain subventricular zone (SVZ)-olfactory bulb pathway and hippocampal dentate gyrus. Accumulating evidence indicates that various brain insults increase neurogenesis in these persistent germinative zones. Two b

With the aid of (H3)-thymidine autoradiography, neurogenesis was documented in the nuclear groups of the medulla oblongata, pons, and mid-brain, as well as in the brain stem reticular formation of the rabbit. Following single injections of (H3)-thymidine, counts were taken of intensely labeled neurons within the nuclei of the functional columns related to the cranial nerves, nuclei of several other functional classifications, and nuclei that did not fit into a functional category. In the brain stem as a whole, neurogenesis was found to occur between days 10.0 and 18.5 of gestation: however, th

Major depressive disorder (MDD) is associated with reduced hippocampal volume, altered connectivity and negative memory bias, suggesting disrupted hippocampal plasticity. Dysregulated adult hippocampal neurogenesis is a potential contributor, but its relevance in humans and role in MDD remain unclear. Here we investigated the molecular basis of hippocampal dysfunction in nonmedicated individuals with MDD by integrating analyses of neurogenic trajectories, cell-type- and subfield-specific gene expression, chromatin accessibility and protein expression. We identify a neurogenic lineage in the ad

Intraventricular hemorrhage (IVH) commonly occurs as an extension of intracerebral hemorrhage (ICH) into the brain ventricular system, leading to worse outcomes without effective management. Using a mouse model of IVH, we found that impaired neurogenesis is evident in the subventricular zone (SVZ), along with persistent microglia activation, leukocyte infiltration and cell death. Pharmacological depletion of microglia using PLX3397, an inhibitor of colony stimulating factor 1 receptor (CSF1R), promotes neurogenesis, and alleviated delayed functional impairments in IVH mice. Meanwhile, an eleva

The subventricular zone (SVZ) in lateral ventricles is the largest neurogenic region in adult brain containing high amounts of copper (Cu). This study aims to define the role of Cu in adult neurogenesis by chelating labile Cu ions using a well-established Cu chelator D-Penicillamine (D-Pen). A neurosphere model derived from adult mouse SVZ tissues was established and characterized for its functionality with regards to neural stem/progenitor cells (NSPCs). Applying D-Pen in cultured neurospheres significantly reduced intracellular Cu levels and reversed the Cu-induced suppression of NSPC’s diff

The majority of the enteric nervous system is formed by vagal neural crest cells which enter the foregut and migrate rostrocaudally to colonise the entire length of the gastrointestinal tract. Absence of enteric ganglia from the distal colon are the hallmark of Hirschsprung disease, a congenital disorder characterised by severe intestinal dysmotility. Mutations in the receptor tyrosine kinase RET have been identified in approximately 50% of familial cases of Hirschsprung disease but the cellular processes misregulated in this condition remain unclear. By lineage tracing neural crest cells in m

In this review, we highlight recent discoveries regarding mechanisms contributing to nerve-cancer cross-talk and the effects of nerve-cancer cross-talk on tumor progression and dissemination. High intratumoral nerve density correlates with poor prognosis and high recurrence across multiple solid tumor types. Recent research has shown that cancer cells express neurotrophic markers such as nerve growth factor, brain-derived neurotrophic factor, and glial cell-derived neurotrophic factor and release axon-guidance molecules such as ephrin B1 to promote axonogenesis. Tumor cells recruit new neural

A phenomenon called vestibular compensation occurs after peripheral vestibular loss. This process involves a mosaic of profound structural rearrangements within the vestibular nuclei. Among them, adult reactive neurogenesis is perhaps the most unexpected, as it occurs in a brain area that was never reported as neurogenic before. Both the survival and functionality of this newly generated neuronal network depend on its integration into preexisting networks in the deafferented structure. Far from being aberrant, this organization allows the brain to use inputs from other sensory modalities to fa

Stress and glucocorticoid hormones regulate hippocampal neurogenesis, but the molecular mechanisms underlying their effects are unknown. We, therefore, investigated the molecular signaling pathways mediating the effects of cortisol on proliferation, neuronal differentiation, and astrogliogenesis, in an immortalized human hippocampal progenitor cell line. In addition, we examined the molecular signaling pathways activated in the hippocampus of prenatally stressed rats, characterized by persistently elevated glucocorticoid levels in adulthood. In human hippocampal progenitor cells, we found that

Leptin is a potent AMP kinase (AMPK) inhibitor that is central to cell survival. Hence, we explored the effects of leptin on neurogenesis and angiogenesis after stroke. Neural stem cells (NSC) were grown as neurospheres in culture and treated with vehicle or leptin and neurosphere size and terminal differentiation were then determined. We then explored the effects of leptin on endogenous repair mechanisms in vivo. Sabra mice underwent photothrombotic stroke, were given vehicle or leptin and newborn cells were labeled with Bromo-deoxy-Uridine. Functional outcome was studied with the neurologica

Even in the adult brain, neural stem cells in the dentate gyrus and subventricular zone continue to produce neuronal precursors, which migrate and differentiate into functional mature neurons. This physiological neurogenesis is thought to be involved in neuronal plasticity. Moreover, recent studies indicate that adult neurogenesis can change in response to various brain insults, including psychiatric diseases, stroke, and neurodegenerative disorders. Although increased neurogenesis in these pathological conditions could contribute to the restoration and regeneration of the damaged brain, an in

OBJECTIVE: Neocortical neurons are sensitive to hypoxic-ischemic (H-I) injuries at term and their demise contributes to neurological disorders. Here we tested the hypothesis that the subventricular zone of the immature brain regenerates neocortical neurons, and that this response is sustained.
METHODS: Systemic injections of 5-bromo-2'-deoxyuridine (BrdU) and intraventricular injections of replication-deficient retroviruses were used to label newly born cells, and confocal microscopy after immunofluorescence was used to phenotype the new cells from several days to several months after perinat

Neural stem cells have recently been found in the central nervous system of adult rodents and humans. In defined conditions, these multipotent cells can generate the three major cell types of the nervous system (neurons, oligodendrocytes and astrocytes). These findings raise questions on the functional role of neural stem cells in the adult brain, and point to the possibility of novel therapeutic approaches. We have been investigating the functional consequences of neural stem cells for the adult circuits of the olfactory system. We are currently investigating this unexpected juvenile characte

Cannabinoid exposure during adolescence has adverse effects on neuroplasticity, emotional behavior, cognition, and reward sensitivity in adult rats. We investigated whether escalating doses of the cannabinoid receptor 1 (CB1 R) agonist, HU-210, in adolescence would affect adult hippocampal neurogenesis and behavioral processes putatively modulated by hippocampal neurogenesis, in adult male and female Sprague-Dawley rats. Escalating doses of HU-210 (25, 50, and 100 µg/kg), or vehicle were administered from postnatal day (PND) 35 to 46. Animals were left undisturbed until PND 70, when they were

Humans select and learn goal-directed actions through the anticipation of their outcomes. While this ideomotor principle is well established in humans, its biological instantiation and relevance for organizing naturalistic behavior remain unclear, including its neurobiological substrates such as hippocampal plasticity. Here, we leverage high-resolution, continuous behavioral tracking in mice across a month-long series of automated learning tasks to test whether ideomotor principles generalize to naturalistic behavior and how they relate to adult hippocampal neurogenesis. Mice lived in a shared

The existence of human hippocampal neurogenesis has long been disputed1-12 and its relevance in cognition remains unknown. Recent studies have established the presence of proliferating progenitors and immature neurons and a reduction in the latter in Alzheimer's disease (AD)11,13. However, their origin and the molecular networks that regulate neurogenesis and function are poorly understood. Here we studied human post-mortem hippocampi obtained from different cohorts: young adults with intact memory, aged adults with no cognitive impairments, aged adults with extraordinary memory capacity (Supe

Adult hippocampal neurogenesis is involved in hippocampal plasticity and can be enhanced by voluntary physical activity, which also elevates "brain-derived neurotrophic factor" (BDNF) levels and improves memory performance. BDNF might either be essential for the maintenance of neurogenesis, or be necessary for mediating activity-dependent increases in neurogenesis. In order to get more insight on this, we examined male heterozygous BDNF-deficient (BDNF +/-) mice with or without access to a running wheel. Only males were analyzed, since sex steroids can interact with BDNF signaling pathways. Al

Adult hippocampal neurogenesis (AHN) is the process of generating new neurons in the adult hippocampal dentate gyrus (DG). Exercise promotes AHN and improves hippocampal function through neuroplastic enhancement. The underlying regulatory factors of this process are currently being vigorously studied. However, many previous studies have used a rodent wheel-running model, in which the exercise condition (e.g., volume, intensity, duration) cannot be controlled. In contrast, treadmill running (TR) allows the precise regulation of conditions such that animals can run according to specific experime

Domestic strains of canaries (Serinus canaria) variably respond to photoperiod changes and apparently stay in breeding state for extended periods. Fife Fancy canaries are supposed to be similar to the native species living at 27-39° north where photoperiod significantly changes across the year. Our birds showed reproductive cycles when exposed to light regimes mimicking the annual cycle of photoperiod. However after 6 months in short days (SD: 8L:16D), males developed large testes, as observed by X-ray tomography, and intense singing. Switching to long days (LD: 16L:8D) did not further increas

Choline acetyltransferase (ChAT)-positive neurons in neural stem cell (NSC) niches can evoke adult neurogenesis (AN) and restore impaired brain function after injury, such as acute ischemic stroke (AIS). However, the relevant mechanism by which ChAT+ neurons develop in NSC niches is poorly understood. Our RNA-seq analysis revealed that dimethylarginine dimethylaminohydrolase 1 (DDAH1), a hydrolase for asymmetric NG,NG-dimethylarginine (ADMA), regulated genes responsible for the synthesis and transportation of acetylcholine (ACh) (Chat, Slc5a7 and Slc18a3) after stroke insult.
