TRPV1 Activation via a Three-Stage Adaptive Heat-Absorbing Hydrogel Drives Neurovascular-Immune Coupling.

Impaired neurovascular-immune coupling severely hampers chronic wound healing. Although the transient receptor potential vanilloid 1 (TRPV1) channel represents a promising therapeutic target, its therapeutic application remains limited by imprecise activation. Here, we developed a three-stage adaptive and heat-absorbing hydrogel (termed HMCG) that enables programmable and localized TRPV1 activation under near-infrared (NIR) irradiation. HMCG forms reversible boronate ester bonds between PVA and TSPBA, enabling a sol-aerosol-gel adaptive with sprayable handling, self-adaptation, and conformal c

Hypovolemia Evokes Conserved Inverse Neurovascular Coupling in the Supraoptic Nucleus Independent of Heart Failure.

Vasopressin (AVP) neurons in the hypothalamic supraoptic nucleus (SON) are activated by systemic challenges that threaten fluid balance. Beyond their classical activity-dependent release of their neuropeptide cargo into the systemic circulation, these neurons also release AVP somatodendritically, enabling local modulation of neuronal excitability and vascular tone. We previously showed that a systemic salt challenge triggers inverse neurovascular coupling (iNVC) in the SON, in which activity-dependent dendritic AVP release induces parenchymal arteriole vasoconstriction and local hypoxia. Brain neurovascular coupling in amyotrophic lateral sclerosis: Correlations with disease progression and cognitive impairment.

BACKGROUND AND PURPOSE: 'Neurovascular coupling' (NVC) alterations, assessing the interplay between local cerebral perfusion and neural activity within a given brain region or network, may reflect neurovascular unit impairment in amyotrophic lateral sclerosis (ALS). The aim was to explore NVC as a correlation between the functional connectivity and cerebral blood flow within the large-scale resting-state functional magnetic resonance imaging brain networks in a sample of ALS patients compared to healthy controls (HCs). METHODS: Forty-eight ALS patients (30 males; mean age 60.64 ± 9.62 years)

Long-term development of dynamic changes in neurovascular coupling after acute temporal lobe epilepsy.

Epilepsy is an abnormal brain state that may be induced by synchronous neuronal activation and also abnormalities in energy metabolism or the oxygen supply vascular system. Neurovascular coupling (NVC), the relationship between neuron, capillary, and penetrating artery, remains unexplored on a fine-scale with respect to the pathology process after acute temporal lobe epilepsy (TLE). Here we use two-photon microscopy (TPM) to provide high temporal-spatial resolution imaging to identify changes in NVC during spontaneous and electro-stimulated (ES) states in awake mice. Implantation of a long-ter

Changes in neurovascular coupling with cerebral perfusion pressure indicate a link to cerebral autoregulation.

Cerebral autoregulation ensures a stable average blood supply to brain tissue across steady state cerebral perfusion pressure (CPP) levels. Neurovascular coupling, in turn, relies on sufficient blood flow to meet neuronal demands during activation. These mechanisms break down in pathologies where extreme levels of CPP can cause dysregulation in cerebral blood flow. Here, we experimentally tested the influence of changes in CPP on neurovascular coupling in a hydrocephalus-type non-human primate model (n = 3). We recorded local neural and vascular evoked responses to a checkerboard visual stimul

Impaired neurovascular coupling and cognitive deficits in anti-N-methyl-D-aspartate receptor encephalitis.

Anti-N-methyl-D-aspartate receptor (anti-NMDAR) encephalitis is a recently identified autoimmune disorder with heterogeneous neurological, psychiatric, and cognitive manifestations. The NMDAR is a key signaling node for neurovascular coupling, the mechanism by which cerebral blood perfusion is enhanced to meet local metabolic requirements from increased neuronal activity. Therefore, anti-NMDAR encephalitis may disrupt neurovascular coupling and induce cognitive deficits. This study examined neurovascular coupling and cognitive function in anti-NMDAR encephalitis patients to identify prognostic

Altered neurovascular coupling and brain arginine metabolism in endothelial nitric oxide synthase deficient mice.

Nitric oxide (NO) produced by endothelial NO synthase (eNOS) is a key regulator of cerebral blood flow (CBF) dynamics. Mice with eNOS deficiency (eNOS-/-) display age-related increases in amyloid beta in the brain and memory deficits, implicating eNOS dysfunction in the neuropathogenesis and/or development of Alzheimer's disease (AD). The present study systematically investigated behavioural, CBF and brain arginine metabolic profile changes in male and female wildtype (WT) and eNOS-/- mice at 14 months of age. eNOS-/- mice displayed altered behaviour in the Y-maze and open field tests. Exhaustive exercise attenuates the neurovascular coupling by blunting the pressor response to visual stimulation.

Neurovascular coupling (NVC) is assessed as an increase response to visual stimulation, and is monitored by blood flow of the posterior cerebral artery (PCA). To investigate whether exhaustive exercise modifies NVC, and more specifically, the relative contributions of vasodilatation in the downstream of PCA and the pressor response on NVC, we measured blood flow velocity in the PCA (PCAv) in 13 males using transcranial Doppler ultrasound flowmetry during a leg-cycle exercise at 75% of maximal heart rate until exhaustion. NVC was estimated as the relative change in PCAv from the mean value obta

Neuroimmune Regulation in Posttraumatic Bone Repair: From Inflammatory Transition to Neurovascular Coupling and Functional Recovery.

Posttraumatic bone repair is not a simple osteogenic process but a spatiotemporal dynamic reconstruction involving the coordinated participation of neural, immune, vascular, and bone-lineage cells. Clinically, structural healing does not always align with pain relief, weight-bearing recovery, and improvement in motor function, suggesting that beyond mechanical stability and bone formation, neuro-immune regulation may govern overall repair quality. This article adopts a structured narrative review framework to discuss the stage-specific roles of the neuro-immune axis in post-traumatic bone repa

Preserved neurovascular coupling in early multiple sclerosis is associated with reduced white matter lesion burden.

BACKGROUND: Multiple sclerosis (MS) is a chronic autoimmune disorder characterized by inflammation, demyelination and axonal degeneration of the central nervous system. Whether these pathological changes impact neurovascular coupling (NVC), which can be compromised in various neurodegenerative disorders, remains unclear in MS. OBJECTIVES: To investigate the relationship between NVC, clinical characteristics and neuroimaging measures in patients with MS. DESIGN: We conducted a cross-sectional study of patients with MS recruited from the Comprehensive Multiple Sclerosis Clinic at Northwestern

Revealing neurovascular coupling at a high spatial and temporal resolution in the living human retina.

Neurovascular coupling (NVC) regulates local blood flow in response to neuronal activity, yet its precise characterization at the capillary level has been hindered by the lack of a noninvasive, high-resolution imaging method. Here, we introduce the adaptive optics rolling slit ophthalmoscope, a unique noninvasive, label-free, high-speed, cellular-resolution clinical imaging system for assessing retinal NVC in the living human eye. Using an off-axis phase contrast approach combined with camera-based confocal slit gating, our method provides large field-of-view imaging of arterial and venular wa

Combined optical coherence tomography and electroretinography system for imaging neurovascular coupling in the human retina.

SIGNIFICANCE: During their early stages of development, neurological and neurodegenerative diseases cause changes to the biological tissue's morphology, physiology and metabolism at the cellular level, and acute, transient changes in the local blood flow. The development of optical methods that can image and quantify such changes simultaneously and investigate the relationship among them (neurovascular coupling) in neural tissues can have a profound effect on furthering our understanding of neurodegeneration. AIM: Our aim is to develop an optical imaging platform for imaging and characterizat

Assessment of Neurovascular Coupling by Spectral Analysis of Cerebral Blood Flow Velocity With Transcranial Doppler.

OBJECTIVE: Neurovascular coupling (NVC) represents the increase in regional blood flow associated with neural activity. The aim here was to describe a new approach to non-invasive measurement of NVC by spectral analysis of the cerebral blood flow velocity (CBFV) with transcranial Doppler. METHODS: In a sample of 20 healthy participants, we monitored systolic CBFV in the left posterior cerebral artery (PCA) during off (eyes closed) and on (flickering checkerboard) periods. The contralateral middle cerebral artery was simultaneously monitored as a control. Each participant was submitted to thre

Test-retest Reliability of Neurovascular Coupling and Impairment in Epilepsy Measured by fNIRS and EEG.

Simultaneous fNIRS-EEG is a cost efficient, noninvasive, and adaptable multimodal imaging method that measures both the hemodynamics and electrical activity of the brain. Neurovascular coupling (NVC) is an important mechanism in the brain in which increased neuronal activity propels an increase in oxygenated hemoglobin as well as decrease in deoxygenated hemoglobin in the local cerebral blood flow to that specific area. Previous literature has suggested that NVC may be impaired in individuals with epilepsy. The unique approach to measure and quantify NVC via multimodal fNIRS-EEG imaging is an

The bio-functionalized membrane loaded with Ta/WH nanoparticles promote bone regeneration through neurovascular coupling.

Electrospinning technology, as a novel approach, has been extensively applied in the field of tissue engineering. Nanofiber membranes prepared by electrospinning can effectively mimic the structure and function of natural bone matrix, providing an ideal scaffold for attachment, proliferation, and differentiation of bone cells while inducing osteogenic differentiation and new bone formation. However, it lacks bioactivities such as osteoinduction, angiogenesis and the ability to promote nerve regeneration. In the presence of complex critical bone defects, a single component electrospun membrane
