Oleanolic acid alleviates ulcerative colitis by remodeling Th17 cells via targeting HSP90β-ODC1 pathway-mediated polyamine synthesis.

BACKGROUND: Ulcerative colitis (UC) is a subtype of inflammatory bowel disease (IBD) characterized by chronic inflammation of the colon and rectum. Its pathogenesis is closely associated with dysregulated mucosal immune responses and intestinal barrier dysfunction. Although oleanolic acid (OA), a natural pentacyclic triterpenoid, has shown therapeutic potential for UC, its precise molecular targets and underlying mechanisms remain poorly understood. PURPOSE: This study aimed to investigate the therapeutic effects and molecular mechanisms of OA in dextran sulfate sodium (DSS)-induced colitis i

Synthesis and anticancer studies of Ru/Ir arene complexes bearing oleanolic acid in acute promyelocytic leukemia cells.

Acute leukemia, a cancer originating in the bone marrow and blood-forming tissues, poses a significant threat to human health. Chemotherapy may cause a range of side effects and further cause greater suffering to the patients. Thus, reducing the toxicity of the drugs for treating leukemia has become a significant challenge. In this study, we developed two non‑platinum anticancer agents, ole-Ru and ole-Ir, by fusing the natural product oleanolic acid as the ligand into two metal (ruthenium and iridium) precursors. Ole-Ru and ole-Ir not only exhibited remarkable selectivity and cytotoxicity agai

Oleanolic Acid Promotes the Formation of Probiotic Escherichia coli Nissle 1917 (EcN) Biofilm by Inhibiting Bacterial Motility.

Probiotic biofilms have been beneficial in the fight against infections, restoring the equilibrium of the host's gut microbiota, and enhancing host health. They are considered a novel strategy for probiotic gut colonization. In this case, we evaluated the effects of various active substances from traditional Chinese medicine on Escherichia coli Nissle 1917 (EcN) to determine if they promote biofilm formation. It was shown that 8-64 μg/mL of oleanolic acid increased the development of EcN biofilm. Additionally, we observed that oleanolic acid can effectively suppress biofilm formation in pathog

Effects of oleanolic acid on hair growth in mouse dorsal skin mediated via regulation of inflammatory cytokines.

Oleanolic acid (OA) is a pentacyclic triterpenoid with favourable physiological activity. It is widely distributed in more than 200 species of plants. OA has garnered significant interest because of its potential biological activities, such as antioxidant, bacteriostatic, and hair growth-promoting effects. To study the effect of OA on hair growth and related mechanisms, we investigated hair growth in mice with testosterone-induced androgenetic alopecia (AGA) that were treated with three different concentrations of OA. The antioxidant, bacteriostatic, and cytotoxic effects of OA were evaluated.

Valorization of olive tree leaves: Extraction of oleanolic acid using aqueous solutions of surface-active ionic liquids.

The global olive oil industry annually generates approximately 750,000-1,500,000 tons of Olea europaea leaves as waste that are typically burned for energy production. Yet, this agricultural by-product is a rich source of oleanolic acid, a high value triterpenic acid with outstanding pharmaceutical and nutraceutical activities. The present study focuses on the extraction of oleanolic acid from dried O. europaea leaves using aqueous solutions of surface-active ionic liquids as alternative solvents. A number of imidazolium-based ionic liquids with variable chain length, different anions and opti

The novel nanocomplexes containing deoxycholic acid-grafted chitosan and oleanolic acid displays the hepatoprotective effect against CCl4-induced liver injury in vivo.

Chemical liver injury threatens seriously human health, along with the shortage of efficiency and low-toxicity drugs. Herein, the novel oral nanocomplexes composed of deoxycholic acid-grafted chitosan and oleanolic acid were constructed to reverse the CCl4-induced acute liver damage in mice. Results indicated core-shell nanocomplexes, maintained by the hydrophobic interaction between deoxycholic acid and oleanolic acid, could be dissociated in the intestine. Notably, the nanocomplexes possessed superior hepatoprotective effect in vivo, possibly due to the synergistic effect between grafted chi

Oleanolic acid alleviates intestinal injury after hepatic ischemia-reperfusion under steatosis via PPARG-dependent M2 macrophage polarization.

Liver transplantation with steatotic donor livers is often complicated by ischemia-reperfusion injury (IRI), leading to intestinal dysfunction, and reduced long-term survival. This study investigates the protective effects of oleanolic acid (OA), a gut microbiota metabolite, against intestinal injury following IRI in rats with hepatic steatosis and explores its mechanisms. 8-week-old male Sprague-Dawley rats were fed a high-fat diet to induce severe hepatic steatosis and IRI with 80-min ischemia. Gut microbiota was analyzed by 16S rDNA sequencing. Rats were treated with OA or the peroxisome pr

Mechanism of HMGB1 release inhibition from RAW264.7 cells by oleanolic acid in Prunus mume Sieb. et Zucc.

High mobility group box-1 protein (HMGB1), primarily from the nucleus, is released into the extracellular milieu either passively from necrotic cells or actively through secretion by monocytes/macrophages. Extracellular HMGB1 acts as a potent inflammatory agent by promoting the release of cytokines such as tumor necrosis factor (TNF)-alpha, has procoagulant activity, and is involved in death due to sepsis. Accordingly, HMGB1 is an appropriate therapeutic target. In this study, we found that an extract of Prunus mume Sieb. et Zucc. (Ume) fruit (Ume extract), an abundant source of triterpenoids,

Oleanolic acid, a herb extract, acts as a strong and safe enhancer in the collagen-mediated hemostatic cascade in flow.

Considerable attention has been paid steadily for efficacy and safety of herb extracts with medicinal benefits, recently. Tranexamic acid (TXA) is a well-known hemostatic agent with strong efficacy but potential thrombus risk. Here, we identified oleanolic acid (OA) as a strong enhancer in promoting adhesion, spreading or aggregation of platelets on immobilized collagen, which exceeds TXA among six herb extracts. In addition, the study demonstrated that TXA could facilitate morphological changes of platelets on collagen, accelerate and enhance intracellular calcium bursting with and without me

Effect of Nano-Oleanolic Acid Combined With Lipid-Lowering Ketones on Insulin Resistance in Rats with Gestational Diabetes.

Diabetes is a widespread metabolic syndrome, an important complication during pregnancy. Most cases are type 2 diabetes, which has attracted the attention of the World Health Organization. The typical feature of T2DM is insulin resistance (IR). Its mechanism remains unclear, but it mainly manifests through parameters like insulin sensitivity, blood glucose level, and liver stability. Oxidative stress and insulin transduction play an important role in IR. This study simulates the disease situation, establishes a high-fat and high-fructose-induced model induced by tert-butyl hydroperoxide (tBHP)
