Anosmia in a patient with Silver-Russell syndrome.

The human sense of smell is important for the awareness of danger like gas, fire or spoiled food, perception of the flavours of food and drink, and social interaction. The impact of limited olfactory capacity or total loss of the sense of smell may be associated with decline in the quality of life and depressive symptoms. Various conditions can be the cause of olfactory disorders, including congenital absence of the olfactory nerve. We present a boy with Silver-Russell syndrome suffering from anosmia.

Functional subdivisions of the olfactory system correlate with lectin-binding properties in Xenopus.

Soybean agglutinin (SBA) is known to selectively label a portion of neurons in amphibian and mammalian primary olfactory systems. Hitherto, no other distinctive features have been found to correlate with the two neuronal populations. Investigating SBA-HRP binding in olfactory mucosa and CNS of Xenopus, we noted that labelled and unlabelled structures can readily be assigned to different olfactory subsystems. The SBA negative one is utilized to detect air-borne odors, whereas major SBA-positive structures serve a role in the perception of water dissolved molecules. Some labelled fibers by-pass

Intelligent artificial olfactory nervous system with sensory adaptation capabilities.

Accurate gas identification across diverse environments is crucial for the advancement of intelligent artificial olfaction, but remains challenging for conventional data-driven learning approaches. Here, we report an artificial olfactory nervous system capable of reliably identifying novel gas stimuli regardless of environmental variations by leveraging sensory adaptation mechanisms inspired by biological olfactory systems. Inspired by biological olfactory systems, our system effectively integrates artificial olfactory receptor neurons, cortical neurons, and intermediate olfactory neurons that

[Olfaction and neurological diseases: a review of the literature].

Olfactory disorders are often misjudged and rarely rated in the clinical setting. They are nevertheless described in a wide range of neurological disorders, and their evaluation can be useful for diagnosis. Usually irreversible olfactory dysfunction is a well-known complication after head trauma. Severe changes in olfactory tests are observed in Parkinson's disease. Dysfunction is present at disease onset and evidenced with all behavioral tests. Regarding other parkinsonian syndromes, olfactory performances are severely impaired in Lewy body disease, less pronounced in multiple system atrophy

Glutamatergic Neurons in the Piriform Cortex Influence the Activity of D1- and D2-Type Receptor-Expressing Olfactory Tubercle Neurons.

Sensory cortices process stimuli in manners essential for perception. Very little is known regarding interactions between olfactory cortices. The piriform "primary" olfactory cortex, especially its anterior division (aPCX), extends dense association fibers into the ventral striatum's olfactory tubercle (OT), yet whether this corticostriatal pathway is capable of shaping OT activity, including odor-evoked activity, is unknown. Further unresolved is the synaptic circuitry and the spatial localization of OT-innervating PCX neurons. Here we build upon standing literature to provide some answers to

Beyond aroma: A scoping review on the impact of chronic rhinosinusitis on retronasal olfaction.

BACKGROUND: Retronasal olfaction (RNO) refers to the perception of odorants inhaled through the mouth and carried through the nasopharynx to olfactory receptors within the olfactory cleft, enabling the perception of flavor. Although orthonasal olfactory dysfunction in chronic rhinosinusitis (CRS) has been widely described, the impact of CRS on RNO is less clear. In this study, we systematically review available literature to provide an update on RNO in the setting of CRS. METHODS: We systematically searched PubMed, Ovid Embase, Web of Science, and the Cochrane Library for studies examining RN

An olfactory perceptual fingerprint in people with olfactory dysfunction due to COVID-19.

The sense of smell is based on sensory detection of the molecule(s), which is then further perceptually interpreted. A possible measure of olfactory perception is an odor-independent olfactory perceptual fingerprint (OPF) defined by Snitz et al. We aimed to investigate whether OPF can distinguish patients with olfactory dysfunction (OD) due to coronavirus disease (COVID-19) from controls and which perceptual descriptors are important for that separation. Our study included 99 healthy controls and 41 patients. They rated 10 odors using 8 descriptors such as "pleasant," "intense," "familiar," "w

Olfactory receptor function.

Olfaction plays a critical role in several aspects of life. Olfactory disorders are very common in the general population, and can lead to malnutrition, weight loss, food poisoning, depression, and other disturbances. Odorants are first detected in the upper region of the nose by the main olfactory epithelium (OE). In this region, millions of olfactory sensory neurons (OSNs) interact with odor molecules through the odorant receptors (ORs), which belong to the superfamily of G protein-coupled receptors. The binding of odors to the ORs initiates an electrical signal that travels along the axons

Monorhinal and birhinal odor processing in humans: an fMRI investigation.

The olfactory nerve, also known as cranial nerve I, is known to have exclusive ipsilateral projections to primary olfactory cortical structures. However, the lateralization of olfactory processes is known to depend on the task and nature of stimuli. It still remains unclear whether olfactory system projections in humans also correspond to functional pathways during olfactory tasks without any trigeminal, perceptual, or cognitive-motor components. Twenty young healthy subjects with a normal sense of smell took part in an olfactory functional magnetic resonance imaging (fMRI) study. We used 2 ty

[Origin of olfactory and rhinosensory evoked cortical potentials in diseases of the central nervous system].

This is the first report to be published on olfactory evoked potentials in patients with well-defined lesions of the central nervous system and the trigeminal nerve. Absence of olfactory evoked potentials is seen in post-central and parietotemporal lesions. The first peak of the so-called olfactory evoked twin potential is absent in lesions of the basal nuclei and sectioning of the trigeminal or ophthalmic nerve, whereas there is no second peak in subcortico-frontal and cortico-temporal lesions. Tumours of the corpus callosum and sectioning of the maxillary and mandibular nerves do not disturb

The participation of cortical amygdala in innate, odour-driven behaviour.

Innate behaviours are observed in naive animals without prior learning or experience, suggesting that the neural circuits that mediate these behaviours are genetically determined and stereotyped. The neural circuits that convey olfactory information from the sense organ to the cortical and subcortical olfactory centres have been anatomically defined, but the specific pathways responsible for innate responses to volatile odours have not been identified. Here we devise genetic strategies that demonstrate that a stereotyped neural circuit that transmits information from the olfactory bulb to cort

Sense of smell: recognition and transduction of olfactory signals.

The primary processes in odour perception, i.e. recognition and transduction of olfactory stimuli, are mediated by the chemosensory olfactory neurons. Interaction of odorous compounds with suitable receptor proteins in the membrane of a subset of cells elicits chemo-electrical transduction pathways, including second messenger cascades and ion channels, that modulate the excitability of the sensory neurons, i.e. converting the chemical stimulus into electrical impulses. The encoded information is conveyed via the axons onto distinct glomeruli in the olfactory bulb. Olfactory sensory cells expre

Computer-controlled stimulation for functional magnetic resonance imaging studies of the neonatal olfactory system.

AIM: Olfactory sensation is highly functional early in human neonatal life, with studies suggesting that odours can influence behaviour and infant-mother bonding. Due to its good spatial properties, blood oxygen level-dependent (BOLD) contrast functional magnetic resonance imaging (fMRI) has the potential to rapidly advance our understanding of the neural activity which underlies the development of olfactory perception in this key period. We aimed to design an 'olfactometer' specifically for use with neonatal subjects for fMRI studies of odour perception. METHODS: We describe a fully automate

Assessing olfactory functions in patients with Barth syndrome.

Barth syndrome is a rare X-linked disease affecting less than 200 individuals worldwide. Several comorbidities have been associated with the pathology and, among those, cardiac myopathy and neutropenia are the most life threatening. The appropriate nutritive support is important to sustain the everyday life of Barth syndrome patients given the chronic fatigue they experience. Since they often prefer salty and fried food, and avoid vegetables and fruits, their eating habit and food preferences do not always provide the proper amount of vitamins and amino acids. It has been indeed reported that

A Nasal Aerodynamics Perspective of Retronasal Olfaction: Rodents vs. Humans.

INTRODUCTION: Odor perception can be achieved through ortho or retronasal routes, with the latter being an important component of flavor perception. There are significant olfactory differences that exist between rats and humans and by understanding the role of structural differences, further insight can be gained into the mechanism of odorant perception via ortho or retronasal routes. METHODS: 3D human and rat (Sprague Dawley) computational models were used to investigate nasal anatomy impact on ortho vs. retronasal odorant transport to the olfactory epithelium. The nasal pharynx region was m

Taste and smell function in Wolfram syndrome.

BACKGROUND: Wolfram syndrome is a rare genetic disease characterized by insulin-dependent diabetes, optic nerve atrophy, sensorineural hearing loss and neurodegeneration. Although olfactory dysfunction, a classical clinical marker of neurodegenerative processes, has been reported in Wolfram syndrome, its use as a clinical marker in Wolfram is limited due to data scarcity. In addition, it is unknown whether Wolfram syndrome affects the sense of taste. METHODS: Smell and taste perception were assessed in participants with Wolfram syndrome (n = 40) who were 15.1 ± 6.0 years of age (range: 5.1-28

Patients' Perspectives on Qualitative Olfactory Dysfunction: Thematic Analysis of Social Media Posts.

BACKGROUND: The impact of qualitative olfactory disorders is underestimated. Parosmia, the distorted perception of familiar odors, and phantosmia, the experience of odors in the absence of a stimulus, can arise following postinfectious anosmia, and the incidences of both have increased substantially since the outbreak of COVID-19. OBJECTIVE: The aims of this study are to explore the symptoms and sequalae of postinfectious olfactory dysfunction syndrome using unstructured and unsolicited threads from social media, and to articulate the perspectives and concerns of patients affected by these de
