Diffuse intrinsic pontine gliomas (DIPGs) are aggressive pediatric brain tumors, and patient survival has not changed despite many therapeutic efforts, emphasizing the urgent need for effective treatments. Here, we evaluated the anti-DIPG effect of the oncolytic adenovirus Delta-24-ACT, which was engineered to express the costimulatory ligand 4-1BBL to potentiate the antitumor immune response of the virus. Delta-24-ACT induced the expression of functional 4-1BBL on the membranes of infected DIPG cells, which enhanced the costimulation of CD8+ T lymphocytes. In vivo, Delta-24-ACT treatment of m

A unique robotic medical platform is designed by utilizing cell robots as the active "Trojan horse" of oncolytic adenovirus (OA), capable of tumor-selective binding and killing. The OA-loaded cell robots are fabricated by entirely modifying OA-infected 293T cells with cyclic arginine-glycine-aspartic acid tripeptide (cRGD) to specifically bind with bladder cancer cells, followed by asymmetric immobilization of Fe3 O4 nanoparticles (NPs) on the cell surface. OA can replicate in host cells and induce cytolysis to release the virus progeny to the surrounding tumor sites for sustainable infection

PURPOSE: The oncolytic effects of a systemically delivered, replicating, double-deleted vaccinia virus has been previously shown for the treatment of many cancers, including colon, ovarian, and others. The purpose of this study was to investigate the oncolytic potential of double-deleted vaccinia virus alone or in combination with rapamycin or cyclophosphamide to treat malignant gliomas in vitro and in vivo.
EXPERIMENTAL DESIGN: Rat (RG2, F98, C6) and human (A172, U87MG, U118) glioma cell lines were cultured in vitro and treated with live or UV-inactivated vaccinia virus. Viral gene [enhanced

Oncolytic virotherapy constitutes a promising treatment option for many solid cancers, including peritoneal carcinomatosis (PC), which still represents a terminal stage of many types of tumors. To date, the in vitro efficacy of oncolytic viruses is mostly tested in 2D-cultured tumor cell lines due to the lack of realistic 3D in vitro tumor models. We have investigated the feasibility of virotherapy as a treatment option for PC in a human ex vivo peritoneum co-culture model. Human HT-29 cancer cells stably expressing marker genes GFP and firefly luciferase (GFP/luc) were cultured on human perit

Getah virus (GetV) is an arthropod-borne alphavirus historically recognized as an emerging zoonotic pathogen of veterinary significance, particularly in livestock and equine populations across Asia and parts of the Western Pacific. Over the past several decades, its expanding ecological range, broad mosquito vector competence, and increasing frequency of animal outbreaks have positioned GetV as a growing concern for animal health surveillance, diagnostics, and vaccine development. However, beyond its established role in veterinary virology, a critical and underexplored dimension of GetV biolog

Oncolytic adenovirus (OA) has attracted increasing attention due to their specific proliferation in tumour cells and resulting in lysis of tumour cells. To further improve the antitumour effect of OA, in this study, we combined CD55-TRAIL-IETD-MnSOD (CD55-TMn), a CEA-controlled OA constructed previously, and chemotherapy to investigate their synergistic effect and possible mechanisms. MTT assay was performed to detect antitumour effects. Hoechst 33 342 and flow cytometric analysis were used to examine cell apoptosis. Western blotting was performed to examine cell pyroptosis and apoptosis mecha

KISS1 tumor suppressor protein regulates cancer cell invasion via MMP9 metalloproteinase. Downregulation of KISS1 gene expression promotes progression of breast cancer and melanoma, resulting in the development of distant metastases. In the current study, we investigated whether restoration of KISS1 expression in KISS1-deficient human metastatic breast cancer cells holds potential as an advanced anticancer strategy. To this end we engineered an infectivity-enhanced conditionally-replicative human adenovirus type 5 encoding KISS1 as an "arming" transgene in the Ad5 E3 region for an ectopic KISS

Signals mediated by the chemokine CXCL12 and its receptor CXCR4 are involved in the progression of ovarian cancer through enhancement of tumor angiogenesis and immunosuppressive networks that regulate dissemination of peritoneal metastasis and development of cancer-initiating cells (CICs). In this study, we investigated the antitumor efficacy of a CXCR4 antagonist expressed by oncolytic vaccinia virus (OVV) against an invasive variant of the murine epithelial ovarian cancer cell line ID8-T. This variant harbors a high frequency of CICs that form multilayered spheroid cells and express the hyal

The CD24-Siglec-10 interaction is recognized as an important component of the immunosuppressive network in tumors. CD24, which is highly expressed on tumor cells, promotes immune evasion through binding to Siglec-10 on immune cells. Therefore, targeting the CD24-Siglec-10 axis represents a promising therapeutic strategy. In this study, we genetically engineered an oncolytic adenovirus to express a novel recombinant protein consisting of the Ig-like V-type domain of mouse Siglec-10 and the Fc fragment of mouse IgG2a (termed as rSiglec-10(V set)), aiming to disrupt CD24-Siglec-10 interaction and

BACKGROUND: Oncolytic viruses (OVs) represent a class of naturally occurring or genetically engineered viruses that selectively infect and lyse tumor cells while sparing normal tissues. Sarcomas of bone and soft tissue are malignant neoplasms originating from mesenchymal tissues, encompassing a wide spectrum of pathological subtypes. Current standard treatments primarily involve surgical resection, radiotherapy, and neoadjuvant chemotherapy, with oncolytic virotherapy emerging as a promising therapeutic approach in this field.

Our previous clinical data showed that autologous mesenchymal stromal cells (MSCs) can be used as carriers for the systemic delivery of oncolytic adenoviruses in children with advanced cancers, a therapy known as Celyvir. Despite achieving clinical remissions, treatment responses have been heterogeneous. Here, we sought to determine whether intrinsic biological characteristics of carrier MSCs influence therapeutic outcomes. Transcriptomic profiling was performed on MSCs from responder and non-responder patients to identify pathways differentially regulated according to clinical response. Expre

Non-Hodgkin lymphoma (NHL) and leukemia are among the most common cancers worldwide. While the treatment of NHL/leukemia of B-cell origin has much progressed with the introduction of targeted therapies, few treatment standards have been established for T-NHL/leukemia. As presentation in both B- and T-NHL/leukemia patients is often aggressive and as prognosis for relapsed disease is especially dismal, this cancer entity poses major challenges and requires innovative therapeutic approaches. In clinical trials, oncolytic viruses (OVs) have been used against refractory multiple myeloma (MM). In pr

Oncolytic adenovirus (Ad) infection promotes intracellular autophagy in tumors. This could kill cancer cells and contribute to Ads-mediated anticancer immunity. However, the low intratumoral content of intravenously delivered Ads could be insufficient to efficiently activate tumor over-autophagy. Herein, we report bacterial outer membrane vesicles (OMVs)-encapsulating Ads as microbial nanocomposites that are engineered for autophagy-cascade-augmented immunotherapy. Biomineral shells cover the surface antigens of OMVs to slow their clearance during in vivo circulation, enhancing intratumoral ac

OBJECTIVE: The mainstay of treatment for patients with malignant pleural disease is fluid drainage and systemic therapy. A tumor-specific oncolytic virus or T-cell-activating interleukin-2 immunotherapy may provide an opportunity for local control. We previously developed a vaccinia virus-expressing interleukin-2, an oncolytic virus that mediated tumor regression in preclinical peritoneal tumor models with expansion of tumor-infiltrating lymphocytes. We evaluated the antitumor efficacy and immune modulatory effects of vaccinia virus-expressing interleukin-2 in malignant pleural disease.

Genetic mutations disrupting interferon (IFN) signaling have been described as an evolutionary route for tumor cells to evade immune checkpoint inhibitor (ICI) therapy. We hypothesized that loss of IFN signaling not only enables cancer cells to evade immune surveillance, but also renders them susceptible to oncolytic viruses. To experimentally address this hypothesis, we generated IFN-unresponsive mouse melanoma cell variants via CRISPR/Cas9 and investigated the impact of sequential immuno-virotherapy on their evolutionary dynamics in immunocompetent syngeneic hosts. In wild-type mice, Natural

Phytotherapy has been used to treat a different type of diseases including cancer for a long time, and it was a source for different active anti-tumor agents. Oncolytic Newcastle disease virus (AMHA1) are very promising anti-tumor therapy. Nevertheless, NDV-based monotherapeutics have not been very useful to some resistant tumors. Thus, the efficiency of oncolytic NDV must enhance by combining NDV with other novel therapies. The current study aimed to determine the possibility of improving the oncolytic effect induced by NDV through Rheum ribes rhizomes extract administration in vitro and in v

BACKGROUND: Herpes simplex virus type 1 (HSV-1) infects approximately four billion people worldwide, and the emergence of drug-resistant strains has reduced the effectiveness of existing antivirals. Targeting host pathways exploited by HSV-1 represents an attractive strategy for developing resistance-refractory antivirals.
METHODS: We evaluated the antiviral potential of doxorubicin, an FDA-approved anticancer drug, against HSV-1 using in vitro cell culture systems, an ex vivo porcine corneal model, and an in vivo murine ocular infection model. Viral replication, host signaling pathways, and

Immune checkpoint inhibitors can trigger antitumor immunity, yet most patients, including those with head and neck cancer, show limited benefit because of insufficient immune activation at the tumor site. Oncolytic virotherapy (OV) may overcome this by priming tumors at the injection site for a stronger response to immune checkpoint inhibitors. However, there are limited data on how OV influences systemic immune responses in uninjected tumors and secondary lymphoid organs (spleen, lymph nodes), largely because of the difficulty of sampling these tissues. We investigated whether immuno-PET can

Despite significant advances in our knowledge regarding the genetics and molecular biology of gliomas over the past two decades and hundreds of clinical trials, no effective therapeutic approach has been identified for adult patients with newly diagnosed glioblastoma, and overall survival remains dismal. Great hopes are now placed on combination immunotherapy. In clinical trials, immunotherapeutics are generally tested after standard therapy (radiation, temozolomide, and steroid dexamethasone) or concurrently with temozolomide and/or steroids. Only a minor subset of patients with progressive/r

PURPOSE OF REVIEW: The design of novel herpes simplex type I (HSV-1)-derived oncolytic virotherapies is a balancing act between safety, immunogenicity and replicative potential. We have undertaken this review to better understand how these considerations can be incorporated into rational approaches to the design of novel herpesvirus oncolytic virotherapies.
RECENT FINDINGS: Several recent papers have demonstrated that enhancing the potential of HSV-1 oncolytic viruses to combat anti-viral mechanisms present in the tumor microenvironment leads to greater efficacy than their parental viruses.
