Rheological properties of cryptococcal polysaccharide change with fiber size, antibody binding and temperature.

Aim:Cryptococcus neoformans is the major agent of cryptococcosis. The main virulence factor is the polysaccharide (PS) capsule. Changes in cryptococcal PS properties have been poorly elucidated. Materials & methods: We analyzed the mechanical properties of secreted PS and intact capsules, using dynamic light scattering and optical tweezers. Results: Storage and loss moduli showed that secreted PS behaves as a viscoelastic liquid, while capsular PS behaves as a viscoelastic solid. The secreted PS remains as a viscoelastic fluid at different temperatures with thermal hysteresis after 85°C. Antib

Lellouch-Lüscher Relation for Ultracold Few-Atom Systems under Confinement.

We derive an analog of the Lellouch-Lüscher (LL) relation for few-body bosonic systems, linking few-body scattering loss rates to the energies and widths of the corresponding harmonically trapped few-body states. Three-body numerical simulations show that the LL relation applies across a broad range of interaction strengths and energies and allows the determination of scattering rates within a single partial wave. Our Letter establishes a robust theoretical framework for understanding the role of the finite-volume effect in few-body observables in optical lattice and tweezer experiments, enabl

Coherent Control of the Fine-Structure Qubit in a Single Alkaline-Earth Atom.

We report on the first realization of a novel neutral atom qubit encoded in the spin-orbit coupled metastable states ^{3}P_{0} and ^{3}P_{2} of a single ^{88}Sr atom trapped in an optical tweezer. Raman coupling of the qubit states promises rapid single-qubit rotations on par with the fast Rydberg-mediated two-body gates. We demonstrate preparation, readout, and coherent control of the qubit. In addition to driving Rabi oscillations bridging an energy gap of more than 17 THz using a pair of phase-locked clock lasers, we also carry out Ramsey spectroscopy to extract the transverse qubit coheren

Dynamics of dual-orbit rotations of nanoparticles induced by spin-orbit coupling.

Spin-orbit coupling (SOC) in tightly focused optical fields offers a powerful mechanism for manipulating the complex motion of particles. However, to date, such a mechanism has only been applied to the single-orbit motion for particles, while multi-orbital dynamics have not yet been experimentally demonstrated. Here, the theoretical and experimental realization of dual-orbit rotational dynamics of nanoparticles in a tightly focused circularly polarized Laguerre-Gaussian beam is reported. Analyses reveal that the dual-orbit rotation of nanoparticles originates from SOC in a tightly focused vort

Investigation of precise particle manipulation using zeroth- and first-order acoustic Bessel fields.

Acoustic tweezers enable non-contact manipulation across a wide range of biomedical applications. In this work, an in-plane circular array acoustic tweezer device operating at 2.3 MHz is used to manipulate 90 μm polystyrene particles, which have higher density and stiffness than the surrounding water, under both focal-point (zeroth-order Bessel function shape) and vortex-trap (first-order Bessel function shape) fields. It is shown that when such fields are used for manipulation, significant positioning errors can occur. The vortex-trap field creates a convergent trap at its core, yet position

Measuring the forces that control protein interactions.

Although the force fields and interaction energies that control protein behavior can be inferred indirectly from equilibrium and kinetic measurements, recent developments have made it possible to quantify directly (a) the ranges, magnitudes, and time dependence of the interaction energies and forces between biological materials; (b) the mechanical properties of isolated proteins; and (c) the strength of single receptor-ligand bonds. This review describes recent results obtained by using the atomic force microscope, optical tweezers, the surface force apparatus, and micropipette aspiration to q

Quantifying Photochemical Propulsion in Light-Powered Janus Micromotors.

Active colloids that convert light into motion provide insight into nonequilibrium chemical systems and routes toward microscale engines. Here we quantify the propulsion dynamics and force generation of light-activated Au-TiO2 Janus particles and their optically trappable polystyrene (PS) core-shell Au-PS@TiO2 analogues. By varying particle size, metal thickness, fuel concentration, and illumination wavelength, we show how photochemical energy conversion at the Au-TiO2 interface governs propulsion, yielding average velocities 33.2 ± 2.99 μm s-1 with instantaneous velocities up to ∼100 μm s-1.

Hydrodynamic spin-orbit coupling in asynchronous optically driven micro-rotors.

Vortical flows of rotating particles describe interactions ranging from molecular machines to atmospheric dynamics. Yet to date, direct observation of the hydrodynamic coupling between artificial micro-rotors has been restricted by the details of the chosen drive, either through synchronization (using external magnetic fields) or confinement (using optical tweezers). Here we present a new active system that illuminates the interplay of rotation and translation in free rotors. We develop a non-tweezing circularly polarized beam that simultaneously rotates hundreds of silica-coated birefringent

Efficacy and Safety of Intravitreal Brolucizumab in Chronic Central Serous Chorioretinopathy: A Retrospective Cohort Study [Response to Letter].

As an important application of optical manipulation, orbital rotation of nanoparticles in optical beams can find uses in constructing micro-motors/micro-machines and the measurement of physical quantity. Here, we report an intriguing orbital rotation of nanoparticles in focused circularly-polarized Gaussian beams at the wavelength scale. Through the transverse scattering forces, off-axis trapping of nanoparticles becomes possible in the off-focal plane, allowing us to create a rotation orbit with a small radius. During the focusing of the incident circularly-polarized Gaussian beam, the spin a

Stochastic heat engine using a single Brownian ellipsoid.

Optical tweezers can confine position as well as orientation of a Brownian particle by simultaneously exerting restoring force and torque on it. Here we propose the theoretical model of a microscopic Stirling engine, using a Brownian ellipsoid as its working substance. The position and orientation degrees of freedom of the ellipsoid (in two dimensions), both being confined harmonically by the tweezers, are coupled to a hot and a cold thermal bath time-periodically. The stiffness of the force confinement is also time-periodic such that it resembles a pistonlike protocol, driving the Brownian el

Titin domains progressively unfolded by force are homogenously distributed along the molecule.

Titin is a giant filamentous protein of the muscle sarcomere in which stretch induces the unfolding of its globular domains. However, the mechanisms of how domains are progressively selected for unfolding and which domains eventually unfold have for long been elusive. Based on force-clamp optical tweezers experiments we report here that, in a paradoxical violation of mechanically driven activation kinetics, neither the global domain unfolding rate, nor the folded-state lifetime distributions of full-length titin are sensitive to force. This paradox is reconciled by a gradient of mechanical sta

In vivo detection of severity of optic nerve crush using manganese-enhanced magnetic resonance imaging in rats.

BACKGROUND: Traumatic optic neuropathy (TON) is one of the reasons for permanent vision loss. Currently, the clinical practices may not be sufficient for direct assessments and comprehensively determining the location and extent of the patients with optic nerve injury in traumatic optic neuropathy. Magnetic resonance imaging (MRI) provides a non-invasive option. However, rare reports have found whether the differentdegree of injury of the optic nerve can be detected by manganese-enhanced MRI (MEMRI). This study aimed to explore the efficacy of MEMRI in the visual pathway for different severity

Micro-Ring Resonator-Based Tunable Vortex Beam Emitter.

Light beams bearing orbital angular momentum (OAM) are used in various scientific and engineering applications, such as microscopy, laser material processing, and optical tweezers. Precise topological charge control is crucial for efficiently using vortex beams in different fields, such as information encoding in optical communications and sensor systems. This work presents a novel method for optimizing an emitting micro-ring resonator (MRR) for emitting vortex beams with variable orders of OAM. The MRR consists of a ring waveguide with periodic structures side-coupled to a bus waveguide. The

Visualization and force measurement of branching by Arp2/3 complex and N-WASP in actin filament.

To determine whether the Arp2/3 complex activated by N-WASP (VCA) branches actin filaments at the side (side branching), or at the barbed (B-)end (end branching) of the mother filaments, we have directly observed the branching process of actin filaments and examined single-molecule unbinding under optical microscope. We found that side branching was predominant, though not exclusive. At the initial stage of polymerization, the branching at the B-end occurred and subsequently the side branching started to occur. In either type of branching, the mother and daughter filaments elongated at nearly

Reconfiguration of orbital angular momentum via circularly polarized multi-focal spin-to-orbit conversion.

Circularly polarized light (CPL), when tightly focused, can generate longitudinal spin-induced orbital angular momentum (OAM) through spin-to-orbit conversion (STOC). While STOC in single-focal systems is well studied, multi-focal STOC remains underexplored. In this Letter, we proposed a multi-focal STOC strategy that leverages tightly focused CPL to generate longitudinal OAM at each focal point. Our numerical simulations demonstrate that reducing the distance between foci induces nonlinear coupling, transforming independent OAMs into a unified OAM state rather than a linear simple superpositi

In situ analysis of the higher-order genome structure in a single Escherichia coli cell.

We developed a novel method for the in situ analysis of the higher-order structure of an individual genome from a single Escherichia coli cell using laser tweezers. Initially, condensed DNA was stably grasped by a laser without any chemical modification and without physical attachment to an artificial object such as micro-plastic beads. Under optical transport, the trapped genome gradually unfolded in solution due to viscous friction. Interestingly, the nucleoid DNA from a log-phase cell is almost fully elongated, whereas in the stationary phase, unfolding of the nucleoid is characterized by s

Multivalent weak contacts shape chaperone-nascent protein interactions.

Molecular chaperones interact with non-native proteins, playing crucial roles in preventing misfolding and in enabling efficient folding in the cellular environment. Trigger factor is a bacterial chaperone that binds to ribosomes, interacting with nascent polypeptides emerging from the ribosome and guiding their early folding steps. In contrast to the central role of the chaperone in promoting folding of newly synthesized proteins, its dynamic interactions with nascent chains emerging from the ribosome remain poorly understood. Here, we use single-molecule fluorescence and optical tweezers app

Characterization of Drug Resistance in Chronic Myeloid Leukemia Cells Based on Laser Tweezers Raman Spectroscopy.

Multidrug resistance is highly associated with poor prognosis of chronic myeloid leukemia. This work aims to explore whether the laser tweezers Raman spectroscopy (LTRS) could be practical in separating adriamycin-resistant chronic myeloid leukemia cells K562/adriamycin from its parental cells K562, and to explore the potential mechanisms. Detection of LTRS initially reflected the spectral differences caused by chemoresistance including bands assigned to carbohydrates, amino acid, protein, lipids, and nucleic acid. In addition, principal components analysis as well as the classification and re

Permanent fixing or reversible trapping and release of DNA micropatterns on a gold nanostructure using continuous-wave or femtosecond-pulsed near-infrared laser light.

The use of localized surface plasmons (LSPs) for highly sensitive biosensors has already been investigated, and they are currently being applied for the optical manipulation of small nanoparticles. The objective of this work was the optical trapping of λ-DNA on a metallic nanostructure with femtosecond-pulsed (fs) laser irradiation. Continuous-wave laser irradiation, which is generally used for plasmon excitation, not only increased the electromagnetic field intensity but also generated heat around the nanostructure, causing the DNA to become permanently fixed on the plasmonic substrate.
