Investigating the molecular mechanisms governing developmental axon growth has been a useful approach for identifying new strategies for boosting axon regeneration after injury, with the goal of treating debilitating conditions such as spinal cord injury and vision loss. The picture emerging is that various axonal organelles are important centers for organizing the molecular mechanisms and machinery required for growth cone development and axon extension, and these have recently been targeted to stimulate robust regeneration in the injured adult central nervous system (CNS). This review summar

Simkania negevensis is an obligate intracellular Chlamydia-like pathogen of the respiratory tract. It infects and multiplies in a wide range of hosts, from unicellular amoeba to a variety of human cells, such as epithelial HeLa and macrophage-like THP1 cells. The Simkania-containing vacuole (SnCV) forms close contacts with the endoplasmic reticulum (ER), and recruits and affects mitochondria of the host cells. Simkania prevent ER stress and require the components of the retrograde transport, as well as several mitochondrial and peroxisomal proteins, for proper development. This review recapitu

In most eukaryotic cells, microtubules and filamentous actin (F-actin) provide tracks on which intracellular organelles move using molecular motors. Here we report that cytoplasmic movement of both mitochondria and lysosomes is slowed by F-actin meshwork formation in pancreatic duct epithelial cells (PDEC). Mitochondria and lysosomes were labeled with fluorescent Mitotracker Red CMXRos and Lysotracker Red DND-99, respectively, and their movements were monitored using epi-fluorescence and confocal microscopy. Mitochondria and lysosomes moving actively at rest stopped rapidly within several seco

Chediak-Higashi Syndrome (CHS) is a rare autosomal recessive disorder characterized by severe immunologic defects including recurrent bacterial infections, impaired chemotaxis and abnormal natural killer (NK) cell function. Patients with this syndrome exhibit other symptoms such as an associated lymphoproliferative syndrome, bleeding tendencies, partial albinism and peripheral neuropathies. The classic diagnostic feature of CHS is the presence of huge lysosomes and cytoplasmic granules within cells. Similar defects are found in other mammals, the most well studied being the beige mouse and Ale

A programme for diagnosis and prevention of lysosomal, peroxisomal, and mitochondrial [respiratory chain diseases (RCD)] diseases was developed on clinical, biochemical, and molecular approaches. The authors made postnatal diagnosis was made in 674 patients from 516 families and prenatal diagnosis in 124 fetuses in 94 families at risk. DNA analysis of mutant alleles in the mucopolysaccharidoses (MPS) I, II, and VI revealed 14, 13, and 4 new mutant alleles in IDS, ASB, IDUA genes, respectively. The pressure of a mutation process played a major role in the distribution of mutant alleles leading

Lysosome-related organelles (LROs) are cell-type-specific organelles derived from the endosomal system. Their lack of ubiquity requires that they be created during cell differentiation. To investigate how LROs are first formed, we characterized the biogenesis of Caenorhabditis elegans gut granules and found that these intestinally restricted LROs are generated de novo during a short period of early embryogenesis. Intriguingly, we found that gut granule number and size remain unchanged throughout the rest of embryonic development. The transition from formation to maintenance coincides with the

Stem cell identity has traditionally been viewed through the lens of transcriptional and metabolic programs. Recent work has revealed an additional layer of complexity: the plasma membrane and intracellular organelles maintain compartment-specific membrane potentials, pH gradients, and ion dynamics that can influence stemness and fate. In this review, we synthesize recent advances identified across diverse stem cell populations to highlight convergent and divergent bioelectric mechanisms driving cell fate decisions. We explore how plasma membrane mechanosensors convert physical cues into inter

Photoreceptor cells in the retina are highly specialized sensory cells that function as light receptors. During the conversion of light into neural signals, photoreceptors are constantly exposed to oxidative stress. Although environmental stressors, such as excessive light exposure, have been implicated in the progression of various retinal diseases, including dry age-related macular degeneration (AMD), the molecular mechanisms underlying the light-induced stress response remain incompletely elucidated. Excessive light exposure triggers the endoplasmic reticulum (ER) stress response in cells a

Neuromelanin-(NM) containing organelles are sub-cellular auto-lysosomal structures composed of three main compartments: NM pigment, protein matrix, and lipid bodies. These organelles accumulate during aging and are found predominantly in the catecholaminergic neurons of Substantia Nigra (SN) and Locus Coeruleus (LC), the main regions affected in Parkinson's disease (PD). NM serves a protective function by sequestering potentially toxic metals like Cu, Fe, and Al. However, NM released from degenerating neurons may lead to a cascade of events resulting in neuroinflammation and neurodegeneration.

Weibel-Palade bodies (WPB) are lysosome-related, secretory organelles unique to vascular endothelial cells. They serve as storage organelles for the pro-thrombotic and hemostatic glycoprotein von-Willebrand factor (VWF) as well as numerous other proteins involved in regulating local inflammatory responses and coagulation processes. WPB undergo a complex formation and maturation process mainly dictated by the post-translational maturation of VWF itself. They are born at the trans-Golgi network and then move on microtubules to the cell periphery where they are anchored at the actin cortex to awa

Artificial organelles have been developed to restore dysfunction or to introduce new-to-nature functions to the host cells, although limitations, such as instability, still exist. Here, we develop a new strategy to fabricate structurally stable polymeric "artificial organelles" by in situ crosslinking of multibranched monomers intracellularly using bioorthogonal click chemistry. The fabricated artificial organelles bearing bioorthogonal reactive centers allow for functional molecule conjugation and cargo release, enabling effective cell modification and manipulation. Triptolide (TPL), isolated from the traditional Chinese herb Tripterygium wilfordii Hook. f., is a natural compound with complex pharmacological activities. TPL has significant therapeutic effects in rheumatoid arthritis, Alzheimer's disease, diabetic nephropathy, and other diseases through anti-inflammatory and antioxidant effects. However, its pro-inflammatory and antioxidant effects produce toxicity to multiple organs such as kidney, liver, and the reproductive organs. Thus, the effects of TPL on inflammation and oxidative stress broaden the therapeutic scope of TPL, but also limit its clin

Corpora amylacea are cell-derived structures that appear physiologically in the aged human brain. While their histological identification is straightforward, their ultrastructural composition and microenvironment at the nanoscale have remained unclear so far, as has their relevance to aging and certain disease states that involve the sequestration of toxic cellular metabolites. Here, we apply correlative serial block-face scanning electron microscopy and transmission electron tomography to gain three-dimensional insight into the ultrastructure and surrounding microenvironment of cerebral Corpo

Proper protein localization is essential for critical cellular processes, including vesicle-mediated transport and protein translocation. Tail-anchored (TA) proteins are integrated into organellar membranes via the C-terminus, orienting the N-terminus towards the cytosol. Localization of TA proteins occurs posttranslationally and is governed by the C-terminus, which contains the integral transmembrane domain (TMD) and targeting sequence. Targeting of TA proteins is dependent on the hydrophobicity of the TMD as well as the length and composition of flanking amino acid sequences. We previously i

UNLABELLED: Most viruses that replicate in the cytoplasm of host cells form neo-organelles that serve as sites of viral genome replication and particle assembly. These highly specialized structures concentrate viral replication proteins and nucleic acids, prevent the activation of cell-intrinsic defenses, and coordinate the release of progeny particles. Despite the importance of inclusion complexes in viral replication, there are key gaps in the knowledge of how these organelles form and mediate their functions. Reoviruses are nonenveloped, double-stranded RNA (dsRNA) viruses that serve as tra

Advances in automated imaging microscopy allow fast acquisitions of multidimensional biological samples. Those microscopes open new possibilities for analyzing subcellular structures and spatial cellular arrangements. In this article, the authors describe a 3D image analysis framework adapted to medium-throughput screening. Upon adaptive and regularized segmentation, followed by precise 3D reconstruction, they achieve automatic quantification of numerous relevant 3D descriptors related to the shape, texture, and fluorescence intensity of multiple stained subcellular structures. Many RNA viruses remodel intracellular membranes to generate specialized sites for RNA replication. How membranes are remodeled and what properties make them conducive for replication are unknown. Here we show how RNA viruses can manipulate multiple components of the cellular secretory pathway to generate organelles specialized for replication that are distinct in protein and lipid composition from the host cell. Specific viral proteins modulate effector recruitment by Arf1 GTPase and its guanine nucleotide exchange factor GBF1, promoting preferential recruitment of phosphatidylinositol-4-kina

The disclosure of the mechanisms of nanoparticle interaction with specific intracellular targets represents one of the key tasks in nanobiology. Unmodified luminescent semiconductor nanoparticles, or quantum dots (QDs), are capable of a strikingly rapid accumulation in the nuclei and nucleoli of living human cells, driven by processes of yet unknown nature. Here, it is hypothesized that such a strong tropism of QDs could be mediated by charge-related properties of the macromolecules presented in the nuclear compartments. As the complex microenvironment encountered by the QDs in the nuclei and
