Molecular cartography of root nodule organogenesis: insights into the role of transcription factors.

Nodule organogenesis is a complex developmental process which is induced post-embryonically following rhizobia infection and relies on coordinated action of a suite of transcription factors at multiple stages starting from its initiation, de novo meristem establishment and differentiation. The rewiring of nitrate-responsive NIN-LIKE PROTEIN (NLP) homolog, NODULE INCEPTION (NIN) transcription factor through genetic adaptations in the common ancestor of the nitrogen-fixing clade is crucial to the origin of nodulation trait. Moreover, nodule formation emerged through the co-option of evolutionary

A Pleiotropic and Functionally Divergent RAC3 Variant Disrupts Neurodevelopment and Impacts Organogenesis.

RAC3 encodes a small Rho-family GTPase essential for cytoskeletal regulation and neurodevelopment, and de novo RAC3 variants typically act as gain-of-function alleles that cause severe neurodevelopmental disorders. In this study, we analyzed a fetus with multisystem congenital anomalies and identified a de novo RAC3 p.(T17R) variant by genome sequencing. To elucidate the pathogenicity of this variant, we combined in silico variant prioritization, structural and energetic modeling, and pathogenicity prediction with in vitro biochemical assays, including GDP/GTP exchange, GTP hydrolysis, effecto

Chd7 Is Critical for Early T-Cell Development and Thymus Organogenesis in Zebrafish.

Coloboma, heart defect, atresia choanae, retarded growth and development, genital hypoplasia, ear anomalies/deafness (CHARGE) syndrome is a congenital disorder affecting multiple organs and mainly caused by mutations in CHD7, a gene encoding a chromatin-remodeling protein. Immunodeficiency and reduced T cells have been noted in CHARGE syndrome. However, the mechanisms underlying T lymphopenia are largely unexplored. Herein, we observed dramatic decrease of T cells in both chd7knockdown and knockout zebrafish embryos. Unexpectedly, hematopoietic stem and progenitor cells and, particularly, lymp

The differential organogenesis and functionality of two liver-draining lymph nodes in mice.

The liver is an immunological organ. However, fundamental knowledge concerning liver-draining lymph nodes (LNs), which have been newly identified in mice as the portal and celiac LNs, is still lacking. Here, we revealed that the portal LN and celiac LN drain liver lymph through different lymphatic vessels. Although both the portal LN and celiac LN possess typical structures, they have different cell compositions. Interestingly, these two LNs form at different times during fetal development. Moreover, the organogenesis of the celiac LN, but not the portal LN, is controlled by the transcription

β-catenin is required in the neural crest and mesencephalon for pituitary gland organogenesis.

BACKGROUND: The pituitary gland is a highly vascularized tissue that requires coordinated interactions between the neural ectoderm, oral ectoderm, and head mesenchyme during development for proper physiological function. The interactions between the neural ectoderm and oral ectoderm, especially the role of the pituitary organizer in shaping the pituitary precursor, Rathke's pouch, are well described. However, less is known about the role of head mesenchyme in pituitary organogenesis. The head mesenchyme is derived from definitive mesoderm and neural crest, but the relative contributions of the

Logic programming to predict cell fate patterns and retrodict genotypes in organogenesis.

Caenorhabditis elegans vulval development is a paradigm system for understanding cell differentiation in the process of organogenesis. Through temporal and spatial controls, the fate pattern of six cells is determined by the competition of the LET-23 and the Notch signalling pathways. Modelling cell fate determination in vulval development using state-based models, coupled with formal analysis techniques, has been established as a powerful approach in predicting the outcome of combinations of mutations. However, computing the outcomes of complex and highly concurrent models can become prohibit

Kidney organogenesis and regeneration: a new era in the treatment of chronic renal failure?

The recent development of a strategy to establish human inducible pluripotent stem (iPS) cells has created a second surge in the field of regenerative research, which had been slowed by restrictions on the use of pluripotent embryonic stem cells. Research on regenerative nephrology offers hope for patients on dialysis. However, due to its anatomic complexity, the kidney is the most difficult organ for the application of regenerative medicine. Very recently, the establishment of a functional whole kidney has been attempted using various stem cells, which may lead to clinical applications. We re

Effects of feed restriction during organogenesis on embryo-fetal development in rabbit.

BACKGROUND: Appropriate maternal nutrition and body weight gain during pregnancy is well established as a major factor in healthy prenatal development in humans. Given the role of nutrition and body weight gain in normal development, pharmaceuticals intended to reduce appetite and promote weight loss will generate developmental toxicity data that may be challenging to interpret. To aid with this, the effects of feed restriction, and subsequent reduction in maternal body weight gain, on embryo-fetal development was investigated in the rabbit. METHODS: Groups of 15 pregnant New Zealand White ra

Methylmercury distribution in the pregnant rat and embryo during early midbrain organogenesis.

BACKGROUND: The period of neurogenesis represents a window of susceptibility for in utero methylmercury (MeHg) exposure. This study examined the toxicokinetics of potentially neurotoxic doses of MeHg during neurogenesis in the developing rat to provide additional information in the areas of mercury speciation and inter-study variability. METHODS: Pregnant Sprague-Dawley rats were dosed s.c. with 5-22 mg/kg MeHg on Day 11 of gestation to target rapidly dividing cells of the developing midbrain. Maternal liver, kidney, skin, blood, placenta, and the embryonic body and brain were evaluated for t

OPGL is a key regulator of osteoclastogenesis, lymphocyte development and lymph-node organogenesis.

The tumour-necrosis-factor-family molecule osteoprotegerin ligand (OPGL; also known as TRANCE, RANKL and ODF) has been identified as a potential osteoclast differentiation factor and regulator of interactions between T cells and dendritic cells in vitro. Mice with a disrupted opgl gene show severe osteopetrosis and a defect in tooth eruption, and completely lack osteoclasts as a result of an inability of osteoblasts to support osteoclastogenesis. Although dendritic cells appear normal, opgl-deficient mice exhibit defects in early differentiation of T and B lymphocytes. Surprisingly, opgl-defic

Embryotoxicity of orally administered chromium in mice: exposure during the period of organogenesis.

Administration of chromium (VI)(250, 500 and 750 ppm as potassium dichromate) via drinking water during organogenesis (days 6-14 of gestation) in mice revealed embryo- and fetotoxic effects. Reduced fetal weight, retarded fetal development, number of fetuses (live and dead) per mother and high incidences of dead fetuses and resorptions in treated mothers in the highest dosed group were evident. No significant gross structural abnormalities were observed in any of the fetuses of chromium (VI)-treated mothers. Significant incidences of reduced ossification were found in the highest dosed group.

[Role of HY antigen in human and mammalian gonadal organogenesis (author's transl)].

H-Y (histocompatibility Y) antigen plays a role in gonadal organogenesis, which is poorly understood. Indeed, it is not immunologically well-defined. The use of cytotoxicity tests cannot lead to a right quantification of its expression. In vertebrates, it is a marker of the heterogametic sex. As it is also detected in invertebrates, H-Y antigen is therefore ubiquitous and has a high phylogenic conservation. Its role, as an organizer of mammal testes, was carried out by controversial experiments of dissociation-reorganization, in gonadal cells. In gonads. H-Y antigen can be studied under 3 dive

Next-Generation Breast Organoids Capture Human Organogenesis with High-Resolution Live Imaging.

Organoids have emerged as a powerful tool for modeling tissue growth and diseases. In this study, we introduce a groundbreaking organotypic culture technique that replicates the morphology, scale, and heterogeneity of human breast tissue, and includes a mesenchymal-like stromal component. A standout feature of this approach is the use of long-term live imaging at high temporal resolution to directly observe stem cell dynamics during organogenesis, from single cells to mature organ tissue. The system is adaptable for high throughput applications and allows for genetic manipulation of the cells.

Steroidogenic Factor 1, a Goldilocks Transcription Factor from Adrenocortical Organogenesis to Malignancy.

Steroidogenic factor-1 (SF-1, also termed Ad4BP; NR5A1 in the official nomenclature) is a nuclear receptor transcription factor that plays a crucial role in the regulation of adrenal and gonadal development, function and maintenance. In addition to its classical role in regulating the expression of P450 steroid hydroxylases and other steroidogenic genes, involvement in other key processes such as cell survival/proliferation and cytoskeleton dynamics have also been highlighted for SF-1. SF-1 has a restricted pattern of expression, being expressed along the hypothalamic-pituitary axis and in ste

Ectopic TBX1 suppresses thymic epithelial cell differentiation and proliferation during thymus organogenesis.

The thymus and parathyroid glands arise from a shared endodermal primordium in the third pharyngeal pouch (3rd pp). Thymus fate is specified in the ventral 3rd pp between E9.5 and E11, whereas parathyroid fate is specified in the dorsal domain. The molecular mechanisms that specify fate and regulate thymus and parathyroid development are not fully delineated. Previous reports suggested that Tbx1 is required for thymus organogenesis because loss of Tbx1 in individuals with DiGeorge syndrome and in experimental Tbx1 deletion mutants is associated with thymus aplasia or hypoplasia. However, the t

Loss of primary cilia in late pituitary organogenesis does not cause endocrine dysfunction.

Cilia are finger-like organelles protruding from the cell membrane, and dysfunction of cilia can cause human diseases called ciliopathies with a range of phenotypes. Some ciliopathies include pituitary phenotypes, and previous studies have shown a role for non-motile primary cilia in pituitary organogenesis. In this study, we generated a pituitary gland-conditional knockout of the ciliary basal body protein CEP164 to investigate the role of primary cilia in pituitary development and postnatal function. Our mouse model shows a loss of pituitary primary cilia between e14.5 and e18.5, although pi

Marker-assisted in vivo imaging reveals prepatterning events prior to lateral root organogenesis.

Roots proliferate mainly through lateral root (LR) organogenesis events that succeed each other in a regular sequence. In the model plant Arabidopsis (Arabidopsis thaliana), live imaging of reporter lines containing the synthetic auxin-response DIRECT REPEAT5 (DR5) promoter have been instrumental for insight into prepatterning mechanisms that shape root architecture. However, depending on the reporter line in use, the course of prepatterning events leading to organogenesis may be described differently, causing confusion and misunderstandings in literature. In this study, we have setup a unique

The Lineage Differentiation and Dynamic Heterogeneity of Thymic Epithelial Cells During Thymus Organogenesis.

Although much progress has been made recently in revealing the heterogeneity of the thymic stromal components, the molecular programs of cell lineage divergency and temporal dynamics of thymic epithelial cell (TEC) development are largely elusive. Here, we constructed a single-cell transcriptional landscape of non-hematopoietic cells from mouse thymus spanning embryonic to adult stages, producing transcriptomes of 30,959 TECs. We resolved the transcriptional heterogeneity of developing TECs and highlighted the molecular nature of early TEC lineage determination and cortico-medullary thymic epi

Exposure to valproic acid inhibits chondrogenesis and osteogenesis in mid-organogenesis mouse limbs.

In utero exposure to valproic acid (VPA), a histone deacetylase (HDAC) inhibitor, causes neural tube, heart, and limb defects. Valpromide (VPD), the amide derivative of VPA, does not inhibit HDAC activity and is a weak teratogen in vivo. The detailed mechanism of action of VPA as a teratogen is not known. The goal of this study was to test the hypothesis that VPA disrupts regulation of the expression of genes that are critical in chondrogenesis and osteogenesis during limb development. Murine gestation day-12 embryonic forelimbs were excised and exposed to VPA or VPD in a limb bud culture syst
