[Assessment of bone quality. Osteocyte function and bone quality].

Osteocytes have control over bone mineralization as well as resorption by regulating osteoblast function and by modulating phosphate handling and vitamin D activation in the kidney. Since osteocytes seem to be responsible for the detection as well as repairing of microdamages that accumulate with aging, interventions that promote the survival of osteocytes may be useful for the prevention of fragility fracture.

Effect of raloxifene treatment on osteocyte apoptosis in postmenopausal women.

Increased osteocyte apoptosis, as the result of estrogen deficiency, could play a role in the decrease of bone mass and bone strength seen in postmenopausal osteoporosis. We investigated whether treatment with raloxifene of postmenopausal women with osteoporosis affects osteocyte apoptosis. Transiliac bone biopsies were obtained from 26 osteoporotic women at baseline and after 2 years of treatment with placebo or raloxifene. Immunohistochemical detection of cleaved caspase-3 was performed on sections from nondecalcified bone biopsies to visualize apoptosis. In the trabecular bone total osteocy

The pathogenesis of osteoarthritis of the hip. Evidence for primary osteocyte death.

Osteocyte viability was investigated in femoral head bone removed from 38 patients with chronic hip disease, with the use of a histochemical stain to demonstrate lactate dehydrogenase (LDH) activity in osteocytes. Where the osteocyte cytoplasm did not show LDH activity, the cell was considered dead; when several adjacent osteocytes were dead, the bone in that area was regarded as nonviable. The preoperative diagnoses were idiopathic osteoarthritis in 25, chondrocalcinosis in six, rheumatoid arthritis in two, Paget's disease in two, avascular necrosis in two, and congenital dislocation of the h

Effects of hormones on osteocyte function and perilacunar wall structure.

Microradiographical and ultrastructural aspects of periosteocytic lacunae are determined by osteocytic activity, lacunar modeling and perilacunar miniremodeling in particular. The miniremodeling activity is constituted by two alternating processes: osteolysis and osteoplasis. Modifications of lacunar shape and size and of perilacunar wall ultrastructure, caused by a direct or indirect effect of certain hormones on osteocytic activity, were observed. The lacunar modeling alterations, particularly in cases of VDH or PTH deficiency, result in an irreversible impaired calcification of the lacunar

Structure of the osteocyte capsule in rats with hypervitaminosis-D and rickets.

A histochemical and topo-optical study was made of the changes of the osteocyte capsule (OC) of the cortical and spongious bone layers of the tibia and vertebra of rats with hypervitaminosis-D and rickets. In hypervitaminosis-D the number of stained OCs was found to decrease. After treatment with high doses of vitamin D, a broadening of the OCs was noted in the area of demineralizing bone. Under the polarization microscope a decrease occurred in the proportion of birefringent OCs, more marked in birefringence than in staining. Since bone demineralization due to vitamin D was not followed by a

Transcriptome-wide splicing analysis reveals HuR-regulated post-transcriptional programs in osteocytes.

Post-transcriptional gene regulation is central to maintaining cellular homeostasis. Among its mechanisms, alternative splicing (AS) fine-tunes cellular adaptation to stress. This study employed an approach combining RNA splicing analysis with RNA-binding protein (RBP) motif enrichment in primary osteocytes cultured in high-glucose conditions. Our analysis identified the RBP human antigen R (HuR) as a top candidate associated with AS regulation. Loss of HuR reshaped the transcriptome through gene expression and splicing changes, converging on two major pathways: stress response and translation

Osteocytes Exposed to Titanium Particles Inhibit Osteoblastic Cell Differentiation via Connexin 43.

Periprosthetic osteolysis (PPO) induced by wear particles is the most severe complication of total joint replacement; however, the mechanism behind PPO remains elusive. Previous studies have shown that osteocytes play important roles in wear-particle-induced osteolysis. In this study, we investigated the effects of connexin 43 (Cx43) on the regulation of osteocyte-to-osteoblast differentiation. We established an in vivo murine model of calvarial osteolysis induced by titanium (Ti) particles. The osteolysis characteristic and osteogenesis markers in the osteocyte-selective Cx43 (CKO)-deficient

Effect of TNF-α-Induced Sclerostin on Osteocytes during Orthodontic Tooth Movement.

Osteocytes are abundant cells in bone, which contribute to bone maintenance. Osteocytes express receptor activator of nuclear factor kappa-B ligand (RANKL) and regulate osteoclast formation. Orthodontic tooth movement (OTM) occurs by osteoclast resorption of alveolar bone. Osteocyte-derived RANKL is critical in bone resorption during OTM. Additionally, tumor necrosis factor-α (TNF-α) is important in osteoclastogenesis during OTM. Sclerostin has been reported to enhance RANKL expression in the MLO-Y4 osteocyte-like cell line. This study investigated the effect of TNF-α on sclerostin expression

[TCP wear particles causes injury of periprosthetic osteocytes in the mouse calvaria].

OBJECTIVE: To study whether tricalcium phosphate(TCP) wear particles cause injuries of periprosthetic osteocytes in the mouse calvaria, and to explain its molecular mechanism. METHODS: Thirty six-week(ICR)male mice were randomly divided into sham group, model (TCP) group and 3-methyladenine (3-MA) group. A murine calvarial model of osteolysis was established by 30 mg of TCP wear particles implantation over the periosteum around the middle suture of calvaria in mice. On the second postoperative day, the autophagy specific inhibitor 3-MA (1.0 mg/kg) was subcutaneously injected to the calvaria i

Novel Insights into Staphylococcus aureus Deep Bone Infections: the Involvement of Osteocytes.

Periprosthetic joint infection (PJI) is a potentially devastating complication of orthopedic joint replacement surgery. PJI with associated osteomyelitis is particularly problematic and difficult to cure. Whether viable osteocytes, the predominant cell type in mineralized bone tissue, have a role in these infections is not clear, although their involvement might contribute to the difficulty in detecting and clearing PJI. Here, using Staphylococcus aureus, the most common pathogen in PJI, we demonstrate intracellular infection of human-osteocyte-like cells in vitro and S. aureus adaptation by f

Matrix extracellular phosphoglycoprotein (MEPE) is highly expressed in osteocytes in human bone.

The matrix extracellular phosphoglycoprotein (MEPE) gene is highly expressed in tumors that cause oncogenic hypophosphatemic osteomalacia (OHO). MEPE is also known as one of the bone-tooth matrix proteins and is associated with bone mineralization. We developed a rabbit polyclonal antibody directed against recombinant human MEPE (rhMEPE) after cloning its cDNA from the cDNA library of a nasal tumor tissue causing OHO. Using this antibody, we analyzed the distribution of MEPE in human bones by immunohistochemistry. In bone specimens from normal subjects, MEPE was predominantly expressed by oste

Monoclonal antibodies with selective reactivity against osteoblasts and osteocytes in human bone.

Monoclonal antibodies (MAb) may provide valuable tools for studying osteoblast differentiation. We therefore raised a panel of MAb reactive with cells of this phenotype using 1,25(OH)2D3-treated human trabecular osteoblast-like cells (HOBS) as the immunogen. Immunohistochemical studies on various tissues, including undecalcified cryostat sections of fetal and adult human bone, identified 11 bone cell-reactive MAb. Of these, 2 demonstrated particularly selective reactivities against osteocytes (OB/M) and osteoblasts (OB/L). These reactivities were also seen in developing bone from rat, rabbit,

Osteocyte dysregulation in periodontitis: Pathological mechanisms and therapeutic potential.

Periodontitis, a chronic inflammatory disease driving alveolar bone destruction, is critically mediated by osteocyte dysregulation. This review synthesizes current evidence revealing that osteocyte-derived RANKL and sclerostin actively promote osteoclastogenesis while suppressing osteoblast activity, directly accelerating bone resorption. Osteocyte apoptosis, ferroptosis, and senescence further exacerbate inflammation through cytokine cascades (e.g., IL-6, TNF-α) and impair regenerative capacity. Therapeutically, targeting osteocytes with anti-RANKL (e.g., denosumab) or anti-sclerostin antibod

Osteocytic Sclerostin Expression as an Indicator of Altered Bone Turnover.

Renal osteodystrophy (ROD) is a complex and serious complication of chronic kidney disease (CKD), a major global health problem caused by loss of renal function. Currently, the gold standard to accurately diagnose ROD is based on quantitative histomorphometric analysis of trabecular bone. Although this analysis encompasses the evaluation of osteoblast and osteoclast number/activity, tfigurehe interest in osteocytes remains almost nihil. Nevertheless, this cell type is evidenced to perform a key role in bone turnover, particularly through its production of various bone proteins, such as scleros

Osteocyte staining with rhodamine in osteonecrosis and osteoarthritis of the femoral head.

Death of osteocytes is synonymous of bone death. Aseptic osteonecrosis of the femoral head is a lesion characterized by the death of osteocytes occurring after major vascular changes. The evolution may lead to hip osteoarthritis, which requires total hip arthroplasty in most cases. Evolution of aseptic osteonecrosis in four radiological stages is well known. We analyzed 24 femoral heads from patients with osteonecrosis or osteoarthritis, retrieved at the time of surgery for a hip arthroplasty. The aim of the study was to clearly identify the necrotic bone from the living bone in the histologic

Osteocyte morphology and orientation in relation to strain in the jaw bone.

Bone mass is important for dental implant success and is regulated by mechanoresponsive osteocytes. We aimed to investigate the relationship between the levels and orientation of tensile strain and morphology and orientation of osteocytes at different dental implant positions in the maxillary bone. Bone biopsies were retrieved from eight patients who underwent maxillary sinus-floor elevation with β-tricalcium phosphate prior to implant placement. Gap versus free-ending locations were compared using 1) a three-dimensional finite-element model of the maxilla to predict the tensile strain magnitu

Report: Osteocyte enhancement function of bisphosphonates in prosthetic replacement.

Aseptic loosening after prosthetic replacement is the primary cause of shortened service life and lowered stability of prosthesis, and increased revision rate after joint replacement. Factors of causing the loosening of joint prosthesis include mechanical factors and biological factors. The mechanical effect of bisphosphonates (BP) is quite obvious, which can enhance osteocyte function, accelerate the generation of new bone and lower bone resorption activity of osteoclast and macrophage. In animal experiment and adjuvant therapy of patients after joint replacement, BP also shows up the functio

Osteocyte regulation of bone mineral: a little give and take.

Osteocytes actively participate in almost every phase of mineral handling by bone. They regulate the mineralisation of osteoid during bone formation, and they are also a major RANKL-producing cell. Osteocytes are thus able to liberate bone mineral by regulating osteoclast differentiation and activity in response to a range of stimuli, including bone matrix damage, bone disuse and mechanical unloading, oestrogen deficiency, high-dose glucocorticoid and chemotherapeutic agents. At least some of these activities may be regulated by the osteocyte-secreted product, sclerostin. There is also mountin

CFTR mediates Cl- transport in osteocytes to sustain cell viability and skeletal homeostasis.

Osteocytes are long-lived with underlying mechanisms largely unknown. Here, we report that osteocyte-specific knockout of cystic fibrosis transmembrane conductance regulator (CFTR) results in excessive osteocyte death, proinflammatory cytokine surge, osteoclast overactivation and bone formation impairment leading to bone loss in adult mice. Consistently in MLO‑Y4 osteocyte‑line, CFTR-knockout causes progressive cell death, which is reversed by CFTR overexpression or medium replenishment. A massive proinflammatory osteocyte secretome is evoked by CFTR-knockout, which deteriorates wild-type oste

Osteocytes regulate osteoprotegerin expression via the p38-MAPK-CREB pathway in rheumatoid arthritis.

Rheumatoid arthritis (RA) is accompanied by systemic and local bone loss, yet the regulation of osteoclast-modulating factors within human osteocytes under inflammatory conditions remains incompletely understood. As an exploratory study, we analyzed surgically resected femoral heads from patients with RA and osteoarthritis (OA). Micro-CT of standardized cancellous regions showed no significant differences in trabecular microarchitectural parameters between groups. In contrast, immunoblot analysis of cortical bone from the femoral neck demonstrated higher osteoprotegerin (OPG) protein levels in
